Description

[0001] MULTILAYER PACKAGING FOR STORING SEMEN EXTENDERS

[0002] FIELD OF THE INVENTION

[0003] The present invention relates to the use of a multilayer packaging, preferably a film or a pouch, for storing semen extenders.

[0004] BACKGROUND AND PRIOR ART

[0005] Semen extender are often used in assisted reproduction technology (ART) to increase the volume of the semen until the required dose while preserving the functional characteristics of the sperm cells and in particular their fertilizing ability. At the present time, semen extender are usually stored in sealed pouches and can only be stored in very specific and limited ranges of temperature, humidity and pressure and this for a very limited period of time.

[0006] Beyond these very specific and limited storage conditions and time, the quality of semen extender decreases which results in a decrease of the semen motility and increase of semen agglutination.

[0007] The present invention aims to overcome the above-mentioned drawbacks of the sealed pouches of the prior art used for storing semen extenders.

[0008] Surprisingly and unexpectedly, the present inventors have discovered that the multilayer packaging, preferably the film or the pouch, according to the present invention makes it possible to dispense with current very specific and limited storage conditions and times while preserving the quality of semen extender and thus while maintaining the semen motility and avoiding the semen agglutination after long-term storage and extreme storage conditions.

[0167] The resulting desiccant films according to the present invention have then been exposed to 3 different treatments (see table 2 below).

[0168] Spermatozoa motility has been assessed using CASA (Computer Assisted Sperm Assessment) method performed on LEJA slides after 7 days, 10 days, and 13 days after the different treatments.

[0169] Table 2 Results:

[0170] No yellowing of the semen extender was observed for any of the conditions of table 2, whether treated or not, except for PRIMXcell Ultra TG after four cycles of 1 hour at 75°C.

[0171] Spermatozoa motility and progressivity results are shown in figures 5 and 6.

[0172] As shown in figures 5 and 6:

[0173] - No significant difference in spermatozoa motility was observed between the different conditions on day 7 (D7), day 10 (D10), and day 13 (D13). The yellowed area had no impact on motility results for PRIMXcell Ultra TG after four cycles of 1 hour at 75°C (see figure 5); and

[0174] - No significant difference in spermatozoa progressivity was observed between the different conditions on day 7 (D7), day 10 (D10), and day 13 (D13). The yellowed area had no impact on motility results for PRIMXcell Ultra TG after four cycles of 1 hour at 75°C (see figure 6).

[0175] 2 / Antibacterial activity

[0176] The antibacterial activity of semen extenders (PRIMXcell Ultra (commercialized by IMV Technologies, Saint-Ouen-Sur-lton, France) or NutriXcell ultra commercialized by IMV Technologies (Saint-Ouen-Sur-lton, France)) against different bacterial strain (E.coli, Klebsiella pneumoniae, Enterococcus faecalis, Staphylococcus aureus and Streptococcus zooepidemicus) stored in the multilayer packaging of the present invention at 65°C for 30 min and at 75°C for 1h has been further analyzed.

[0177] Method:

[0178] - Reconstitution of lyophilized samples with 6 mL of sterile ultrapure water.

[0179] - Antibiograms performed based on the AFNOR NF U47-107 standard (agar diffusion method):

[0180] • Inoculation of Mueller-Hinton agar plates with the five bacterial strains.

[0181] • Placement of cellulose disks on the surface of the agar plates.

[0182] • Application of antibiotic solutions onto the disks until "saturation" (7.5 pL per disk).

[0183] • Incubation of the plates for 24 hours at 37°C ± 2°C.

[0184] • Measurement of inhibition diameters (manual reading).

[0185] Results :

[0186] The measured inhibition diameters are expressed in mm. Note that the diameter of the cellulose disk is 6 mm.

[0187] The results are presented in tables 3 and 4.

[0188] Table 3

[0189] Table 4
