[0005] In the realm of research and development, epitope-tagged proteins enable precise visualization and tracking of recombinant proteins, facilitating a deeper understanding of their behavior and functionality.

[0006] The number of available tags in the human system is limited, and even more narrowed when only linear epitopes are counted. The number of linear epitopes suggested to be non-immunogenic is even smaller.

[0007] Hence there is a great need for further epitope tags in particular for linear epitope tags to be included into proteins expressed in the human system.

[0009] In the context of clinical adoptive immune therapy, epitope-tagged rTCRs provide significant benefits, allowing for the visualization and isolation of specific T-cell populations, thereby greatly enhancing the precision and efficacy of therapeutic interventions. This technology enables the identification and enrichment of highly functional T cells that possess the ability to recognize and eliminate cancer cells or pathogens. Epitope-tagged rTCRs are also useful for drug product characterization and release. Additionally, epitope-tagged rTCRs offer the potential for real-time monitoring of T-cell localization and persistence in vivo, optimizing treatment protocols and improving patient outcomes. In addition to their primary applications, epitope-tagged rTCRs hold the potential to serve as a vital safety switch in adoptive immune therapy. They offer a promising avenue for the depletion of malfunctional or overly potent T cells that have been transferred to patients. Particularly, epitope-tagged rTCRs may eventually function as a safeguard, allowing clinicians to selectively remove undesirable T-cell populations, thereby minimizing the risk of adverse effects and optimizing the overall safety and efficacy of adoptive T-cell therapies. Furthermore, epitope-tagged rTCRs of different sequence can be compared in a standardized manner to assess potential differences in surface expression, isolation of enriched populations of immune cells expressing rTCRs that can be compared with each other.

[0010] Overall, the application of epitope-tagged proteins, such as epitope-tagged rTCRs, such as in adoptive immune therapy, represents a powerful approach that combines both research insights and clinical benefits. By leveraging the capabilities of such tagged receptors, researchers and clinicians can advance understanding behavior of immune cells expressing rTCRs, including T cells and natural killer cells, and unlock new opportunities for targeted and personalized therapeutic interventions. In this context, a particular challenge is to identify an epitope that is not immunogenic itself.

[0014] Against this background, it is one objective of the present invention to provide new tags as well as antibodies or fragments thereof which bind and / or detect specific populations of epitope-tagged proteins.

[0015] It is another objective of the present invention to provide antibodies or fragments thereof for use in therapy and / or analytical scenarios.

[0016] As surprisingly discovered by the present inventors, said objectives are realized by antibodies as herein disclosed and defined, which allow for the binding and / or detecting of specific populations of epitope-tagged proteins. The inventors herewith present a new epitope tag that can be used to tag any expressed protein. The provided tag can be used for visualization, enrichment, characterization and purification of the protein to be tagged. In particular for membrane proteins, the tag can be used for visualization, enrichment, characterization of the cell type in which the tagged protein is integrated and further could be used as a safety switch. The provided tag provides excellent non-immungenic characteristics and the sequence is unique and not present in the human body. The UNI-Tag can flexible be inserted internal, C-terminal or N- terminal of the desired sequence.
