This invention discloses a reagent kit and method for the joint detection of glucose, cellobiose, and xylose in cellulose hydrolysate, belonging to the field of biomass conversion and analytical detection technology. The method includes the following steps: setting up three types of detection channels: a glucose oxidase detection channel that specifically responds to the glucose component; a β-glucosidase-glucose oxidase combined detection channel that responds to the total signal of glucose and cellobiose; and a pyranose oxidase detection channel that simultaneously responds to the combined signals of glucose, cellobiose, and xylose. Enzymatic colorimetric reactions are performed on the cellulose hydrolysate sample and the multi-component gradient standard solution in the three types of detection channels, respectively. A ternary linear calibration model is constructed based on the absorbance data of the three types of channels of the multi-component gradient standard solution. This method provides a rapid, low-cost, and highly specific detection method for glucose, cellobiose, and xylose in cellulose biomass hydrolysate, and has practical application value.

A method for the targeted synthesis of rare ginsenosides Rg3 and Rh2

This invention discloses a method for the targeted conversion and synthesis of rare ginsenosides Rg3 and Rh2. Specifically, fresh Panax notoginseng leaves are microwave-dried for 1-3 minutes to inactivate their endogenous β-glucosidase. The dried Panax notoginseng leaves are extracted with anhydrous methanol, and the extract is resuspended in an acetate-sodium acetate buffer solution after drying. α-arabinofuranase, β-xylosidase, and β-glucosidase are added to the resuspended solution, and an enzymatic reaction is carried out at 35-40℃ to directionally convert the original ginsenosides into rare ginsenosides Rg3 and Rh2. This method can achieve the inactivation of endogenous enzymes and the stable retention of original saponins in Panax notoginseng leaves in a very short time. Compared with the natural drying combined with exogenous enzymes, the conversion ratios of Rg3 and Rh2 are increased by 3.14 and 2.4 times, respectively. Compared with fresh Panax notoginseng leaves, the contents of Rg3 and Rh2 are increased by 20.6 and 8.7 times, respectively. This method is suitable for the high-value utilization of Panax notoginseng leaf waste resources and the large-scale preparation of rare saponins.

This invention provides a microbial preparation for enhancing the aroma and quality of low-grade tobacco leaves and its preparation method, belonging to the interdisciplinary field of microbial engineering and tobacco processing technology. Bacillus subtilis is inoculated into a fermentation medium containing low-grade tobacco leaf powder, using the powder as the core inducer for directional fermentation. Combined with staged fermentation regulation, the metabolic flow of Bacillus subtilis is directionally trained to produce a complex enzyme system highly matched to the degradation needs of tobacco leaves. Compared with conventional fermentation, this invention significantly enhances the activities of protease, amylase, and β-glucosidase. This complex enzyme system can simultaneously degrade proteins and starches in low-grade tobacco leaves and hydrolyze glycosidic bonds to release latent aroma substances. The present invention aims to provide a saccharide composition suitable for a cyclic-tetrasaccharide-containing starch syrup which has low viscosity, low water activity, low coloration property, and low calorie content, and which is unlikely to cause precipitation of crystals of saccharides during storage, and to provide a use of the composition and a production method for the composition. The object is achieved by providing a cyclic-tetrasaccharide-containing saccharide composition having the following characteristics (1) to (3): (1) the saccharide composition includes a branched cyclic tetrasaccharide in addition to the cyclic tetrasaccharide, wherein the content of the cyclic tetrasaccharide with respect to the total solid content of the saccharide composition obtained by allowing glucoamylase and α-glucosidase to act on the above saccharide composition is 38% by mass or higher, on a dry solid basis; (2) the ratio of α-1,4-linked glucose in the total glucose residues constituting the saccharide composition in methylation analysis is over 9% and 15% or lower, and (3) the ratio of α-1,4,6-linked glucose in the total glucose residues constituting the saccharide composition in methylation analysis is less than 6%; and providing a use of the composition and a production method for the composition.
