Abstract

The present disclosure relates to the field of ionic liquids, and specifically to a pyrimidine-based ionic liquid, a preparation method and use thereof. [0004] Solvents are the basis of chemistry, biology, chemical engineering and biotechnology, whilethedischarge amount of solvents is also large, specificallyaccounting for about two thirds of thetotal industrial discharge amount. Conventional organic solvents are volatile and easy to volatilize into atmosphere, thus causing many negative effects including climate change, urban air-quality degradation and human diseases. Ionic liquids are recognized as designable solvents, whereineutectic point of solvent and solute is lowered downthrough designed ion pairs, the solvent and the solute bindmainly throughionic interactions and hydrogen-bond interactionsinstead of covalent bonds, so as to changephysicochemical properties of active pharmaceuticalingredients without changing itspharmacological properties.

[0005] The most important characteristics of ionic liquids are non-volatility, non-flammability, high thermal stability, good solubility and surface activity, and they are currently recognized asgreen designer solventsthat can replace organic solvents. Due to the adjustability characteristic of properties, that is, viscosity, hydrophilicity, polarity, density, acidity and alkalinity and the like of the ionic liquids beingadjustable by designing the ion pairs, the ionic liquids have wider applicability. Therefore, replacementof conventional solventsby the ionic liquids has always been a hot spot and a research directionin the academic and industrial communities. EUREACH (registration, evaluation, authorization and restriction of chemicals) regulations and restriction list of the European Union aim at improving safety, protecting environment, and phasing out hazardous chemicals.

[0006] Functionalized ionic liquids refer to task-specific ionic liquids, and in addition to the basic characteristics of the ionic liquids such as excellent stability, low volatility, excellent substrate compatibility and structural designability, functional groups contained in the structure further make the ionic liquids become special reagents for realizing specific tasks, so as to reduce negative effectson the environment and health.

[0007] Low solubility is a characteristic of most active pharmaceutical ingredients (APIs) . In the modern medical field, on one hand, the ionic liquids as solvents provide a solutionfor avoiding problems such as transformation of multiple crystal formsand poor solubility of solid medicines; and on the other hand, ionic liquid formsa noveldrug deliverysystem withAPI (API-ILs) , and can effectively control stability and bioavailability of APIs.

[0008] It is well known that main obstacles hindering industrial application ofionic liquids are high viscosity and low stability to water and air. Most ionic liquids synthesized heretofore have high viscosity, low ionic conductivity and low diffusivity, andthe diffusivity of constituent ions isseveral orders of magnitude lower than that of watereven at high temperatures. At a certain temperature, the viscosity of the ionic liquids decreases as water content increases. However, wateras a polar solventcan alsoform hydrogen bonds as donor and acceptor and thus weaken or even destroy intermolecular and intramolecular hydrogen bonds of asystem. Particularly when a mole fraction of water exceeds 0.5-0.8, strong hydrogen bonds are formed between water molecules and ions, which could destroy the hydrogen bond interaction between the ion pairs.

[0009] According to the report of the journal Science in 2003, hundreds of millions of species of ionic liquids can be theoretically combined. Therefore, the primary challenge is finding exactly the right ionic liquid for the specific task.

[0010] Chinese patent CN108912054A discloses a mercaptopyrimidine anti-corrosive ionic liquid, a preparation method therefor and use thereof. This is an ionic liquid for a specific task in the lubricant industry, wherein the ionic liquidis prepared by a 4-step synthesis method including esterification, anion exchange and so on, or an industrial lubricant is synthesized by 5 steps. The synthesis process is complex: precipitate and solvent need to be removed, and nitrogen protection, vacuum drying and other processesare needed. Synthetic substrates and products are notsuitable for biomedical applicationand do notconformto a reaction process of atom economy.

[0011] Chinese patent CN115298190A discloses a cholineionic liquid solvent, in which a synthesis process requires a volatile organic solvent chloroform, synthesisneeds to be performed under a nitrogen atmosphere, hydrochloric acidis needed for performing phase separation, the hydrochloric acid needs to be washed with pure water, and chloroform and water need to be further removed. Due to hydrophobicity, it is cloudy when meeting water, and needs to be  solubilizedand co-emulsifiedwith an organic solvent such as isopropanol and sorbitan monolaurate, so that it cannot completely replace the organic solvents.

[0012] Therefore, the present disclosure aims atdeveloping a functionalized ionic liquid solvent designed for a specific biomedical task, so as to replace the conventional volatile organic solvents that threatenthe environment and health.SUMMARY

[0013] The present disclosure provides a pyrimidine-based ionic liquid, a preparation method therefor and use thereof, aiming at solving the problems of high viscosity, poor stability to water and air and poor compatibilityto solutes of the ionic liquid.

[0015] In the first aspect, examples of the present disclosure provide a pyrimidine-based ionic liquid, which is named as [A] X, where [A] represents a cationic moiety and X represents an anionic moiety; and astructural formula of the pyrimidine-based ionic liquid is shown as follows:

[0017] In the second aspect, examplesof the present disclosureprovide a preparation method for the above pyrimidine-based ionic liquid, including: mixing and dissolving a raw material for forming cations [A] and a raw material for forming anions X.

[0018] In the third aspect, examplesof the present disclosureprovide use of the abovepyrimidine-based ionic liquid as a solvent for dissolving a low-water-solubility API.

[0025] In order to make the objectives, technical solutions and advantages of the examples in the present disclosure clearer, the technical solutions in the examples of the present disclosure will be described clearly and completely below. Examples, for which no specific conditions are specified, are performed according to conventional conditions or conditions recommended by manufactures. Where manufacturers of reagents or apparatuses used are not specified, they are conventional products commercially available.

[0026] A pyrimidine-based ionic liquid, a preparation method therefor and use thereof provided in the examples of the present disclosureare specifically described below.

[0027] According tothe precedingdescription, it can be seen that the high viscosity of the ionic liquids is a technical difficultythat makes it hardto widely apply the ionic liquids. In order to solve the technical difficultyof high viscosity of the ionic liquids, the examples of the present disclosureadopt a de novo design strategy of aqueous ionic liquid. Stability ofaqueous ionic liquid depends on interaction between cations and anions and between ions and water. The smaller the ion size is, the more charges thesurface carries, the stronger the interaction between ions, and the more stable the aqueousionic liquid. In the above, the action of anions is crucial because water has strong interaction with anions, anda hydrogen bond structure of “anion…HOH…anion” is formed in a ratio of 1: 2, which dominatesstructural stability adjustment of the aqueous ionic liquid. On the other hand, hydrogen bond structures formed between water and anions are crucial to activity and stability of proteins. Therefore, for the development of a chiraltask-specificionic liquid solvent designed for achievingabiomedical specific task, it is a feasible scheme to screen negatively chargedsmall biological molecules.

[0028] The examples of the present disclosurescreen ion pairs on a larger scale by designing a series of ion pairs and combinedmoleratios thereof, including by theoretical and computational chemical methods. It is observed that a liquid-state temperature window period (ΔT) of the ionic liquid is turned from deviation to expansion, a water content is increased from 0.1 to 0.8 mole fraction or more, and finallythe task-specificpyrimidine-basedaqueousionic liquid solvent having low viscosity, stability to water and air and good compatibilitywith solutes is obtained.

[0029] It is found frommolecular simulation experiments that strong hydrogen bonding is formed between chosenion pairs, and bonding energy of hydrogen bonds thereof is much lower than that of covalent bonds. Therefore, the pyrimidine-basedionic liquid [A] X provided by the examples of the present disclosure is of a self-assembledstructure driven by non-covalent bonds at normal temperature and pressure. That is, due to strong coordination ability generated by structural particularity of the anions, strong hydrogen bonds formed between molecules are a main force driving molecular recognition and self-assembly, and such strong electrostatic interactionis sufficientto counteract weakening or destruction effects of polarity of water on intermolecular hydrogen bonds; and athermodynamically stablepyrimidine-based ionic liquid [A] X isfinally formed. Moreover, it isalso surprisingly found that the pyrimidine-based ionic liquid [A] X provided by the examples of the present disclosure naturally has strong dissolving ability , and can completely replace the conventional organic solvents for dissolving typical low-water-solubility APIs.

[0037] Specifically, a mole ratio of [A] to X is (1: 1) to (4: 1) , for example, any value between (1: 1) and (4: 1) , such as 1: 1, 1.5: 1, 2: 1, 2.5: 1, 3: 1, 3.5: 1 and 4: 1, preferably (2: 1) to (3: 1) .

[0038] The pyrimidine-basedionic liquid isaqueous ionic liquid; and preferably, the pyrimidine-based ionic liquid is an anionic chiral, task-specific ionic liquid.

[0039] Further, the examples of the present disclosure provide a preparation method for the above pyrimidine-based ionic liquid, including: mixing and dissolving a raw material for formingcations [A] and a raw material for forminganions X.

[0040] In the above, awhole preparation process is carried out under a non-vacuum (normal temperature and pressure) condition. The raw material for forming the cations [A] and the raw material for forming the anions X are dissolved usingwater. Atemperatureof the mixing and dissolving is 30-50 ℃, and time is 10-30 min. Alternatively, no solvent is added in the process of preparing the pyrimidine-based ionic liquid, and the two raw materials are directly mixed to form the required ionic liquid.

[0041] The pyrimidine-based ionic liquid [A] X provided in the examples of the present disclosure is designed according to The 12 Principles of Green Chemistry, synthesized by using non-toxic and harmless natural bio-renewable raw materials, does not generate cytotoxicity, and is suitable for biomedical application. It is synthesized by one stepat normal temperature and pressure without solvent or only using water as a solvent. No waste is generated in the synthesis process, and zero discharge of industrial production can be realized. The present disclosure conforms to the definition of green chemistryof the US Environmental Protection Agency in the early 1990s, “design of chemical products using (preferably renewable) raw materials and processes to reduce or eliminate the use and generation of toxic and / or hazardous reagents and solvents in the manufacture and application of chemical products” .

[0042] The pyrimidine-based ionic liquid [A] X provided by the examples of the present disclosure can be applied as a solvent for dissolving low-water-solubility APIs and biomass. It hasexcellent solubilityfordissolving low-water-solubility APIs. In the above, the low-water-solubility APIsareAPIs with solubility less than 3 g at 20 ℃, preferably less than 1 g, more preferably less than 0.001 g; for example, the low-water-solubility APIs include, but arenot limited to, any one  of ibuprofen, salicylic acid, azelaic acid, antimicrobial lipids, retinal, retinoic acid, as well as lignin, vanillin, gallic acid and cytochrome c.

[0043] It should be appreciated that the solubilityof solvents is generally solute-specific and that solubility is exponentially related to concentration of solvents, that is, the solubility of any solute is largely dependent on the mole fraction (xi) of the ionic liquid solvent [A] X. The pyrimidine-based ionic liquid [A] X provided by the examples of the present disclosure, as a solvent system consisting of a series of ionic liquids with various polarity, can be applied toa quitedifferent and wide range of solutes, but a testing range of the examples of the present disclosure is limited. Therefore, ibuprofen, salicylic acid, azelaic acid, antimicrobial lipids, retinal, retinoic acid, as well as lignin, vanillin, gallic acid and cytochrome c listed in application examples of the present disclosure are used only as model compounds fortestingsolvent propertiesof the pyrimidine-based ionic liquid [A] X provided in the examples of the present disclosure, and thus should not be construed as limiting the scope.

[0044] To sum up, the pyrimidine-based ionic liquid provided by the examples of the present disclosure has the following characteristics.

[0045] (1) Green process: the pyrimidine-based ionic liquid [A] X provided by the examples of the present disclosure is synthesized by a green process at normal temperature and pressure with chiral natural products as substrates and water as solvent. It has the characteristics of common ionic liquids, such as almostnegligiblelow vapor pressure, namely, non-volatile characteristic, and can replace conventional volatile organic solvents. In the above, the anions Xarepyrimidine carboxylic acids, the cations [A] areaminoalcohols, and is a low-volatility organic compound additive in biomedical processes and a chiral ligand for synthesis of chiral medicalintermediates

[0046] (2) Atom economy: acid-base neutralizationreaction is a typical 100%atom economy reaction. In the pyrimidine-based ionic liquid [A] X provided by the examples of the present disclosure, a pKa value (about 1.3-3.14) of theanionic moietyis far smallerthan that (about 8.0-9.8) of thecationic moiety, so that the [A] X molecules as reaction substrates can successfully generate the ionic liquid [A] X at normal temperature and pressurethrough acid-base neutralization reaction, with yield being100%. That is, the reaction process for preparing the pyrimidine-based ionic liquid [A] X provided by the examples of the present disclosure conforms to the classical acid-base theory, and has 100%atom economy.

[0047] (3) Rapidreactivity: mostionic liquids reported so far need to be synthesized bymultiple steps, have a long reaction time, and meanwhilerequire an organic solventas a reaction mediumor need washing and purification, which wastes resources and pollutes environment. The pyrimidine-based ionic liquid [A] X provided by the examples of the present disclosure can be  synthesized by one step at normal temperature and pressure without dehydration or purification. The specific step is as follows: dissolving the raw material for forming the cations [A] and the raw material for forming the anions Xusingwater, and conventionally stirring under a conditionof 30-50 ℃for 10-30 minuntil the raw materials are completely dissolved; or one-step synthesis by mixing the raw material for forming the cations [A] and the raw material for forming the anions Xwithout solvent and heating the same to 80-120 ℃, which is, however, time-consuming. Therefore, rapid synthesis by a one-stepdirect method withwater as a solvent is preferred.

[0048] (4) Low viscosity at room temperature: the viscosity of most reported ionic liquids is 1-3 orders of magnitude higher than that of conventional organic solvents, whichbecomes one of the important factors hindering the industrial application of the ionic liquids. As the present disclosure adopts de novo synthetic route of aqueous ionic liquid, and the anions X pyrimidine carboxylic acid anionshavea flexible structure with interchangeof R and S configurations, and delocalized chargeson pyrimidine rings increase ionic mobility and ionic conductivity, the pyrimidine-based ionic liquid [A] X provided by the examples of the present disclosure is endowed withnatural low viscosity, thuscreating a condition for application in the biomedical industry. Similarly, due to the zwitterionic properties of some ions that make up the cationic moiety, which have separate charge centers, the intra-molecular charge-transfer effect increases the degree of freedom of each ion. For example, the pyrimidine-basedionic liquid [A] X provided in Examples 5-8 has lower viscosity and higher ionic conductivity.

[0049] (5) Stability and compatibility withsolutes: in thepyrimidine-basedionic liquid [A] X, the anions Xarederived from the extremelyhalophilic bacteriaand act as natural osmoprotectantsagainst extremes for microorganisms. For example, 2-methyl-1, 4, 5, 6-tetrahydropyrimidine carboxylic acid has two isomers, namely 2-methyl-1, 4, 5, 6-tetrahydropyrimidine carboxylic acid and 2-methyl-3, 4, 5, 6-tetrahydropyrimidine carboxylic acid, andexhibits three zwitterionic states in an aqueous solution. Suchzwitterionic compound contains one carboxyl group and two amino groups which bind with water through strong hydrogen bonds, so that more water molecules are in a bound-water state and solvation effects of water is restricted. Meanwhile, strong hydrogen bonds and multi-hydrogen bondsis formed between carboxylanions and cations, whichstrong electrostatic interactionfurther counteracts the weakening or destruction effects of polarity of water on the intermolecular hydrogen bonds. This is molecular basis for the pyrimidine-based ionic liquid [A] X provided by the examples of the present disclosure to have a high water content, and the water content thereof can be up to mole fraction of 0.90or more. Therefore, the pyrimidine-based ionic liquid [A] X provided by the  examples of the present disclosure is stable to water and air and has good compatibility with solutes, and thus hasapplication value.

[0050] The pyrimidine carboxylic acids serving as X anionshaveunique tautomer and zwitterionic states, that is, isomers (I, II) of 2-methyl-1, 4, 5, 6-tetrahydropyrimidine carboxylic acid and 2-methyl-3, 4, 5, 6-tetrahydropyrimidine carboxylic acid, and exist in the aqueous solution in three zwitterionic states (III, IV, V) :

[0051] (6) Excellentsolubility: hydrogen bonds of ionic liquidshavequite important non-covalent interaction, and remarkablyaffect physicochemical propertiesand dissolving propertyof the ionic liquids. Due to structural asymmetry of the pyrimidine-based ionic liquid [A] X system provided by the examples of the present disclosure, where theanion X component is presentin three zwitterionic states in theaqueous solution, carboxyl anions and amino cations in pyrimidine carboxylic acid molecules can bind with surrounding water molecules to form strong hydrogen bonds and multi-hydrogen bonds, and the hydrogen bond interactions exist between cations and anions, so that the pyrimidine-based ionic liquid [A] X provided by the examples of the present disclosure is endowed withstrongdissolving ability.

[0052] The pyrimidine-based ionic liquid [A] X provided by the examples of the present exhibits neutral to basic and various polarity in physicochemical properties. A dielectric constant is remarkablylower than that of water and close to that of organic solvents such as benzene, toluene, carbon tetrachloride, chloroform, ether, and ethanol, that is, it exhibits characteristics of a hydrophilic ionic liquid solvent with strong solubility, and can be mixed with water in any mixing ration; the surface tension is between that of water and that of conventional surfactantssuch as anionic surfactant N-acylamino acid, indicating thatthe pyrimidine-based ionic liquid [A] X provided by the examples of the present disclosure has the characteristics of common surfactants. Thischaracteristic can greatly changeenvironmental pollution and health damage caused by organic solvents and surfactants in the biomedical, chemical engineeringand fine chemicalindustrials.

[0053] (7) Biocompatibility: in the pyrimidine-based ionic liquid [A] X provided by the examples of the present disclosure, pyrimidine carboxylic acids that constitutethe anionic moiety X, and it can be classified as an amino acid derivative in chemistry, so that it has the characteristics of better environmental friendliness, biocompatibility, biodegradability and the like. The pyrimidine carboxylic acidshaveR and S chiral configurations, thus can provide a stable chiral center, and facilitate stabilizing biomacromolecules such as proteins, nucleic acids and DNA bases. It belongs to anionic chiral, task-specific, biocompatible ionic liquid solvent. Compared with the conventional organic solvents, it is more suitable for biomedical application.

[0054] Pyrimidine carboxylic acidsas the anionicmoiety Xare chiral molecules which havea negatively charged carboxyl group attached to a pyrimidine ring containing a delocalized positive charge, have R and S enantiomers, and providea stable chiral center:

[0055] Further, in the pyrimidine-based ionic liquid [A] X provided by the examples of the present disclosure, as the carboxyl group of the anions X can form a hydrogen bond with the amino group of proteins, it has a unique behavior in regulating stability of proteins. This is because in ahigher-order structure of biologically active proteins, abinding force between subunits mainly comes fromhydrogen bonds and ionic bonds. Therefore, anion species existing in the ionic liquid play a key role in activity and stability of the dissolved proteins in the ionic liquid, and sucharomatic X anionic moiety greatly contributes to the characteristics of the [A] X system dissolving and stabilizing protein and forms astructural basis of the pyrimidine-based ionic liquid [A]X provided by the examples of the present disclosure.
