[0001] The present application claims priority to PCT Application No. PCT / CN2024 / 095262 filed on May 24, 2024, the content of which is incorporated herein by reference in its entirety.FIELD OF THE INVENTION

[0002] The invention relates to antibodies and antigen-binding fragments thereof that bind to SSTR2, and uses thereof.BACKGROUND OF THE INVENTION

[0003] Somatostatin (SST) is a cyclic peptide hormone that regulates neurotransmission and hormone secretion. SST binds to G-protein-coupled somatostatin receptors (SSTRs) , inhibiting adenylyl cyclase via inhibitory G-proteins. SSTRs are class A G-protein-coupled receptors (GPCRs) and there are five isoforms (SSTR1-SSTR5) . Each SSTR isoform is expressed in different tissues and organs and has a distinct function.

[0004] Among these receptor subtypes, Somatostatin receptor 2 (SSTR2) is the most investigated because of its predominance. SSTR2 is a human somatostatin receptor and is highly implicated in hormone disorders, cancers, and neurological diseases (Heo, Yoon, et al. eLife 2022; 0: e76823) . In particular, SSTR2 represents a diagnostic biomarker and potential therapeutic target in many cancers, such as lung cancer, neurologic tumors, and neuroendocrine tumors (NETs) (Jia-Hua He et al., Am J Transl Res 2021; 13 (10) : 11223-11234) . SSTR2 is also a very good target for therapeutics such as antibody and ADC due to its distribution profile.SUMMARY OF THE INVENTION

[0005] In a first aspect, the invention provides an antibody that binds to SSTR2, or an antigen binding fragment thereof, wherein the antibody comprises a heavy chain variable region (VH) and a light chain variable region (VL) , and wherein

[0366] Example 1: SSTR2 is highly expressed in many tumor samples with IHC staining

[0367] Anti-SSTR2 antibody ZA-0587 (ZSGB-BIO cat. ZA-0587) was used in this Example. For IHC protocol, immunohistochemical staining was performed on 4 μm sections using the ZA-0587 (1: 200 dilution) rabbit monoclonal primary antibody (ZSGB-BIO) on the Ventana Benchmark Ultra Autostainer according to the manufacturer’s protocol. Briefly, antigen retrieval was performed by placing unstained slides in Ventana Ultra CC1 buffer (Tris-EDTA / EGTA, pH 9) at 64 ℃ for 95 min followed by incubation of tissues with the primary antibody (ZA-0587) at 36 ℃ for 16 min. Antigen-antibody reaction was visualized using  Universal DAB Detection Kit. Hematoxylin staining for 8 min followed by a bluing reagent (Ventana Medical Systems, Tucson, AZ) for 4 min was used as a counter stain. To ensure antibody specificity, isotype controls were performed as above except primary antibodies were replaced with rabbit IgG. Immunohistochemical staining was evaluated semi-quantitatively for both percent positivity and intensity and an H-score ranging from 0-300 was derived based on the percentage of tumor cells membrane stained multiplied by the intensity (0-3) of staining.

[0372] For generation of a SSTR2-specific immune response, we developed three strategies to immunize mice. 1) 10 mice (5 balb / c, 5 C57bl / 6) were immunized five times with a mixture of the 1st extracellular domain (cQTEPYYDLTSNA, aa 33-44, SEQ ID NO: 83) and the 2nd extracellular domain (cALVHWPFGKAICRVV, aa 104-118, SEQ ID NO: 85) peptides. To elicit a strong immune response, peptides were fused to Keyhole Limpet Hemocyanin (KLH) . 2) 10 mice (5 balb / c, 5 C57bl / 6) were immunized four times with 50ug or 25ug / mouse of HEK293 derived Viral-like particles (VLPs) expressing human SSTR2 (Kactus Biosystems) . The third immunization strategy was a DNA prime-cell boost approach. 3) 10 mice (5 balb / c, 5 C57bl / 6) were immunized three times (1st, 2nd and 4th injection) with 3ug of pcDNA3.4 / mouse containing the full-length of human SSTR2 gene (Gene ID: 6752) , and boosted twice (3rd and 5th injection) with 5x106 SSTR2-expressing CHO-K1 cells (GenScript ProBio)  / mouse.

[0373] 2-2 Screening strategies

[0374] 27000 hybridoma clones were generated and screened using FACS on human SSTR2-overexpressed cells HEK293-huSSTR2 (HEK293 cells expressing human SSTR2, GenScript ProBio) and CHO-K1-huSSTR2 (CHO-K1 cells expressing human SSTR2, GenScript ProBio) or cynomolgus macaques SSTR2-overexpressed cells CHO-K1-cynoSSTR2 (GenScript ProBio) and HEK293-cynoSSTR2 (GenScript ProBio) , respectively. Binding on CHO-K1 cells expressing human SSTR1 (CHO-K1-huSSTR1 GenScript ProBio) , SSTR3 (CHO-K1-huSSTR3 GenScript ProBio) , SSTR4 (CHO-K1-huSSTR4 GenScript ProBio) or SSTR5 (CHO-K1-huSSTR5 GenScript ProBio) of these hybridoma clones were also assessed with FACS for selectivity checking. Three candidate hybridoma clones were selected from the above screen, as listed in Table 3.

[0382] The binding profiles of the candidate clones screened as described in Example 2-2 were shown here. All tested SSTR2-expressing cells were cultured according to ATCC guidance. 1x105 cells per well were seeded into 96-well V-Bottom plates. Cells were washed twice with stain buffer (BD #554656) and pelleted at 300×g and 2-8℃ for 3 minutes. Cells were resuspended and incubated with 100 μL / well supernatant harvested from each hybridoma subclone at 2-8℃ away from light for 40 minutes. After two washes, cells were next resuspended with 100 μL / well of 1 μg / mL cold diluted Alexa 647 AffiniPure Goat Anti-Mouse IgG (H+L) (Jackson #115-605-062) incubated at 2-8℃ away from light for 30 minutes. After two washes with stain buffer, the cells were resuspended with 100 μL / well of stain buffer and analyzed by Beckman CytoFlex flow cytometer. For each sample, 10000 events were collected in the APC channel. A nonlinear regression analysis with four adjustable parameters was conducted using GraphPad Prism 9.2 software. The related results were shown in Table 4.

[0394] A range of humanized 183G11A12 and 221E9C10 clones were generated with subtle differences in framework and complementary determining region (CDR) sequences using CDR grafting in which the CDRs of 183G11A12 and 221E9C10 clones were grafted into human frameworks. The amino acid sequences of the murine antibody VHs and VLs were subjected to a BLAST database search and the closest human immunoglobulin heavy or light chain homologues were identified and considered candidates for CDR grafting. The chimeric clones from 18311A12 were constructed into IGHV3-48 and IGKV4-1; the chimeric clones from 221E9C10 were constructed into IGHV3-23 and IGKV2-30. The CDRs determined according to Kabat were identified and considered as the relevant CDRs. The Kabat CDRs of the parent antibodies were maintained in the humanized antibodies.

[0407] The epitope binning of humanized antibodies 183. LZ4, 183. HZ4 and their parental chimeric antibody 183G11A12, humanized antibodies 221.3.1, 221.3.2 and their parental chimeric antibody 221E9C10, as well as isotype control and the positive control BMK2, chimeric antibody 152A12F2 were tested by ELISA assay. Briefly, 96 well plate was coated with huSSTR2, the oversaturated parental chimeric antibody 183G11A12, 221E9C10 or isotype control were added and incubated. Then added oversaturated tested humanized antibodies (183. LZ4, 183. HZ4; or 221.3.1, 221.3.2) and the positive control (BMK2, 152A12F2) into designated wells respectively. The absorbance at 450 nm and 650 nm were measured with Microplate Reader (BioTek Synergy 2) . We use the following formula to calculate the signal rise rate: signal rise rate (%) = (OD450 sample-OD450 isotype)  / OD450 isotype*100%.

[0410] Example 9: Assessment of the tested antibodies on internalization

[0411] To evaluate the internalization into tumor cell lines, humanized antibodies 183. LZ4, 183. HZ4, 221.3.1, 221.3.2 and their parental chimeric antibody 183G11A12 and 221E9C10, were tested with HEK293-huSSTR2, NCI-H524 and NCI-H446 cells. All these cells were cultured according to ATCC’s guidance. The indirect method of internalization (FACS surface binding) was applied in this study. The MFIs which determined SSTR2 molecules on tumor cells treated with 25 μg / mL test antibodies were assessed at 0 hour and after 24 hour-incubation, respectively. The MFIs at 0 hour were detected using the FACS procedure mentioned in Example 3. The MFIs at 24 hour were detected as bellow, after incubating with test antibody solution, cells were washed twice with 200 μL / well culture medium, resuspended with 500 μL / well culture medium and transferred to 24 wells plate to culture for 24 hours at 37℃; The second antibody Alexa 647 AffiniPure Goat Anti-Human IgG (H+L) (Jackson 109-605-088) staining was conducted with collected cells after washing twice with stain buffer; then the MFIs were measured according to the method described in Example 3. The internalization ratio of 24 hours was calculated with the formula as 100%- (MFI of 24 hours  / MFI of 0 hour) *100%. Results shown in Figure 6 demonstrated that all the tested antibodies can trigger internalization, which reached around 60%within 24 hours. The good internalization will help enhance the efficacy of antibodies antibody-drug conjugates (ADCs) , as most ADCs rely on internalization to release their toxic payloads. Additionally, it benefits the antibody drug itself by reducing the SSTR2 on the cell membrane.

[0412] Example 10: Assessment of cross reactivity and selectivity of the tested antibodies with FACS binding

[0413] To assess the cross reactivity of test antibodies to human, cynomolgus and mouse SSTR2, CHO-K1-huSSTR2, CHO-K1-cynoSSTR2 and CHOK1 cells overexpressing mouse SSTR2 (CHO-K1-mouse SSTR2, GenScript ProBio) were cultured and tested respectively, following the same method as Example 3. Due to the lack of commercially validated antibodies against mouse SSTR2 for flow cytometry applications, CS5005-22D11IgG1, a murine monoclonal antibody isolated via hybridoma screening with high binding affinity to mouse SSTR2, was employed as the positive control antibody for mouse SSTR2 binding in this study, the heavy chain and light chain sequences of the antibody were respectively shown in SEQ ID NOs: 116 and 117.

[0435] SSTR2- (negative) cells (HEK293) , SSTR2-overexpressing cells (HEK293-huSSTR2) , and SSTR2+ small cell lung cancer (SCLC) cells (NCI-H524 and NCI-H82) were resuspended to 4 x 106 cells / mL with assay buffer (1%FBS, 1%PS-RPMI 1640 medium) , and followed by adding 25 μL of the cells into each well on a 96-well white Opaque flat-bottom plate. Subsequently, the antibody samples were gradient diluted with assay buffer and added into each well with 50 μL, 105 Jurkat CD3-BsAb Reporter cells (Genomeditech GM-C17940) in 25 μL were then added to the each well as the effector cells. Further, the 96-well plates with the two cells were incubated at 37℃ and 5%CO2 for 7 h. Then incubate the plate at 20-25℃ for 10 min and add 100 μL / well LUC substrate solution into each well of the plates, incubate at 20-25℃ on a shaker at 300 rpm for another 5 min. Luminescence values in relative luminescence units (RLU) were measured using a modular multi-technology microplate reader (BioTek, Synergy 2) , and plotted versus the antibody concentrations to determine the EC50 using GraphPad Prism software (version 10.0) .

[0440] 183. LZ4-deruxtecan (DAR8) and 221.3.1-deruxtecan (DAR8) were synthesized with the protocol described below. The drug-linker mc-GGFG-DXd (deruxtecan) was alkylated to the cysteines of the reduced 183. LZ4 and 221.3.1 according to procedures described in the literature (Nakada T et al. (2016) Bioorg Med Chem Lett. 26 (6) : 1542-1545. Kang MS et al. (2021) . Chem Sci. 12 (41) : 13613-13647. ) . Briefly, after buffer-exchange into 50 mM PB containing 5 mM EDTA (pH 7.2) using Amicon ultracel filters (MWCO 25 kDa) , the antibody samples were reduced with 8-12 equivalence of TCEP (10 mM) at room temperature for 4h. Charge 12-15 equivalence of drug-linker (5 mM in DMSO) slowly (in 3-5min) at 20℃ for 30min. Quench the reaction with 15x NAC (20 mM) at 20℃ for 30min. Any residual unconjugated drug was removed by purification and the final antibody-drug conjugates were formulated in 20mM Histidine pH6.0. The samples were subsequently analyzed to detect concentration by absorbance at 280 nm. HIC / RP-HPLC to identify drug antibody ratio (DAR) and SEC to identify purity were applied to check the chemical quality of synthesized ADCs. The diagram of 183. LZ4-deruxtecan and 221.3.1-deruxtecan was shown in Figure 12, and the quality data was summarized in Table 17. All ADCs had shown synthesized with good quality and were evaluated in the following studies.

[0448] Example 17: Cytotoxicity effect of anti-SSTR2 humanized ADCs on tumor cells

[0449] The cytotoxicity of ADCs, negative control (isotype-deruxtecan) and the corresponding naked antibodies, as well as the free cytotoxin DXd (Taoshu Bio T11249L) were evaluated in vitro with NCI-H524 and NCI-H69 SCLC cell models.

[0450] Briefly, 4000 cells in 50 μL culture medium per well were seeded into 96-well flat bottom plates and incubated at 37℃ for 24 hours. The test samples were diluted with corresponding culture medium. 50 μL 2x test sample solutions were added to the 96-well plates with cells and incubated at 37℃, 5%CO2 for 6 days. 100 μL / well of CellTier-Glo (Promega #G7571) was added. The plates were shaken on an orbital shaker to induce cell lysis for 2 minutes and left to stand for 10 minutes at room temperature to stabilize signal. The luminescence intensity was detected by Microplate Reader. The viability rate was calculated as (LUM of samples  / LUM of Blank) x100%. The absolute IC50 was interpolated with A nonlinear regression analysis with four adjustable parameters was conducted using GraphPad Prism 9.2 software. The related results demonstrated that all tested ADCs effectively killed NCI-H524 and NCI-H69 SCLC tumor cells (Table 19 and Figure 15) . On SSTR2 high expressing NCI-H524 cells, 183. LZ4-deruxtecan killed tumor cells with an IC50 of 0.98nM, whilst 221.3.1-deruxtecan killed tumor cells with an IC50 of 0.45nM, both of which were showed much higher activity than Isotype-deruxtecann. On SSTR2-low expressing NCI-H69 cells, the IC50 values of 183. LZ4-deruxtecan and 221.3.1-deruxtecan were 32.00 nM and 40.36 nM, respectively, comparable to the IC50 of Isotype-deruxtecan. It therefore demonstrated the tumor cell-killing activities of SSTR2-ADCs were antigen-dependent.

[0456] Example 19: Cell Line Derived Xenograft (CDX) tumor growth inhibition of anti-SSTR2 ADCs

[0457] For in vivo efficacy in cell-line derived xenografts, 5x106 of NCI-H524, NCI-H69 and NCI-H446 cells in 0.1 ml PBS with Matrigel (1: 1) were injected subcutaneously into female nude (BALB / c-nude) mice (GemPharmatech) respectively. Mice were randomly divided into study groups (6 mice / group) and dosed with test ADCs via intravenous injection once the tumors reached 100-300 mm3. Tumor volume was calculated with the formula (volume = 1 / 2 x length x width x width) . No weight loss or treatment-related toxicities were observed during the study. All animal procedures were performed under a protocol approved by the Institutional Animal Care and Use Committee of the animal facility. Studies grouping information and efficacy were listed in Table 21. As shown in Figure 17, a significant tumor growth inhibition was observed following treatment of mice with 10 mg / kg of either 183. LZ4-ADCs or 221.3.1-ADCs. On SSTR2-high expressing NCI-H524 CDX model, 183. LZ4-deruxtecan conducted 105%TGI on day 20 with 6 CRs out of 6 mice. 221.3.1-deruxtecan conducted 104%TGI on day 20 with 2 PRs and 4 CRs out of 6 mice. On another SSTR2-high expressing NCI-H446 CDX model, 183. LZ4-deruxtecan conducted 106%TGI on day 20 with 5 PRs and 1 CRs out of 6 mice. 221.3.1-deruxtecan conducted 104%TGI on day 20 with 4 PRs and 0 CRs out of 6 mice. On SSTR2-low expressing NCI-H69 CDX model, 183. LZ4-deruxtecan conducted 104%TGI on day 22 with 2 PRs and 0 CRs out of 6 mice. 221.3.1-deruxtecan conducted 86%TGI on day 22 with no PRs and CRs out of 6 mice.
