[0001] Inhibitors of Pseudomonas aeruginosa Elastase (LasB) for the treatment of Pseudomonas Keratitis
[0002] The present invention relates to the use of inhibitors of the Pseudomonas aeruginosa virulence factor LasB for the treatment of Pseudomonas keratitis.
[0003] P. aeruginosa is a Gram-negative bacterium, which is ranked by the WHO as one of the most critical pathogens today (World Health Organization. Global Priority List of Antibiotic-Resistant Bacteria to Guide Research, Discovery, and Development of New Antibiotics. WHO 2017). This opportunistic bacterium causes around 10% of hospital- acquired infections and has a high occurrence among immunocompromised and cystic-fibrosis patients (Magill, S. S.; Edwards, J. R.; Bamberg, W.; Beldavs, Z. G.; Dumyati, G.; Kainer, M. A.; Lynfield, R.; Maloney, M.; McAllister-Hollod, L.; Nadle, J.; et al. N. Engl. J. Med. 2014, 370, 1198-1208; Richards, M. J.; Edwards, J. This task is complicated by the high intrinsic resistance of the pathogen (Hancock, R. E. W.; Speert, D. P. Drug Resist. Updat. 2000, 3, 247-255; Strateva, T.; Yordanov, D. J. Med. Microbiol. 2009, 58, 1133-1148).
[0004] Keratitis is an inflammation of the cornea, characterized by rapidly advancing destruction of the stroma that can lead to complications like thinning or perforation of the cornea, but also to blindness (doi: 10.4103 / meajo.MEAJO_264_16; 10.1016 / j.micpath.2016.12.013). Consequently, the main unmet medical need is the liquefactive necrosis of the cornea with major stromal melting and perforation in Pseudomonas keratitis, and in particular, in contact lens-associated keratitis (CLAK). Epidemiology indicates more than two million cases of microbial keratitis yearly (doi: 10.1016 / j .survophthal.2018.12.003), with P. aeruginosa being one of the major causal agents, especially among contact lens wearers (DOI 10.1186 / s12886-017-0612-2). Besides being a health threatening condition, it is also an economic burden (doi: 10.1038 / s41433-020-01360-6). Current solutions to treat P. aeruginosa keratitis rely on the treatment with classical antibiotics, such as chloramphenicol, gentamicin, kanamycin, levofloxacin and ofloxacin.
[0005] A new approach in the field of the development of anti-infectives is the anti-virulence strategy, which also applies to LasB. The aim of this strategy is not to kill the bacteria, but to reduce their pathogenicity, thereby reducing selection pressure. This approach e.g. has the advantage that it is effective even if pseudomonads that are resistant to standard-of-care (SOC) antibiotics need to be treated; or by inhibiting the extracellular LasB even after the bacteria have died. Several LasB inhibitors are disclosed in WO 2023 / 166039 A1 and WO 2022 / 043322 A1 and documents cited therein.
[0006] The role of P. aeruginosa elastase (LasB) - which is able to hydrolyse a vast array of host proteins, causing damage to tissues, disrupting host immune responses, and promoting inflammation (doi: 10.1016 / j.drudis.2021.02.026) - in keratitis has been discussed in the literature. Thereby, Ohman et al (doi: 10.1093 / infdis / 142.4.547), Preston et al (doi: 10.1128 / iai.65.8.3086-3090.1997) and Hobden et al (doi: 10.1089 / 10445490260099674) concluded, that LasB does not contribute to pathogenicity in corneal infections in mice. This also contradicts older publications from 1978 to 1990 in which eyes were treated (in vivo) with extracellular proteases of P. aeruginosa (doi: 10.1128 / iai.19.2.630-648.1978; Kreger and Gray, 1978), LasB enzyme (PMID: 6409834; Kessler et al, 1983) or a P. aeruginosa strain called PA-28 (doi: 10.1128 / AAC.34.11 .2065; Bums et al., 1990) - where a relevant role of LasB in the course of the disease is suspected. However, these trials suffer from inconsistencies such as the use of a protease mixture (Kreger and Gray), questionable conclusions based on the treatment with peptidic inhibitors like phosphoram idon (Kessler), which is a "notoriously promiscuous zinc endopeptidase inhibitor, inhibiting human endothelin-converting enzyme, neutral endopeptidases, and MMPs, among others" (comment in doi: 10.1016 / j .drudis.2021 .02.026, Everett and Davies, 2021 ). As a result, unspecific effects may have occurred due to these off-target activities. To conclude, particularly the above-mentioned publications by Ohman and the more recent publications by Preston as well as Hobden strongly suggest that LasB is not a suitable target for the treatment of Pseudomonas keratitis.
[0007] The inventors of the present invention have however surprisingly found that LasB also plays an important role in this disease process, especially in the early and acute phase of infection, and that inhibition of this protease has a favorable effect on the progression of the disease (see Figures 1-9). The contradiction to the findings by other groups can possibly be explained by the fact that for the first time experiments were conducted using a non-toxic, highly selective small molecule with IC50 in the single-digit nanomolar range to treat an eye infection with a relevant clinical isolate of P. aeruginosa, which has been proven to express LasB (quantified using qPCR). Accordingly, the present invention provides inhibitors of the P. aeruginosa virulence factor LasB for use in the treatment of Pseudomonas keratitis.

(1-(((S)-1 -((4-Methoxyphenyl)amino)-3-methyl-1 -oxobutan- 2-yl)amino)-4-methyl-1-oxopentan-2-yl)phosphonic acid (139 mg, 0.30 mmol) was deprotected by bromotrimethylsilane (0.28 mL, 2.1 mmol) in DCM (3.0 mL). The exoU- and exoT-positivity of the PA54 strain, foregrounds its keratitogenic potential.

Then, three parallel, vertical, 1-2 mm long scratches were added to the cornea with a PA54-loaded 27-Gauge needle (B. Braun). Afterwards, 5 pL (~5X107CFU / mL) of the inoculum were pipetted onto the scratched eye and left untouched for 20 min (Preston et al., 1995; Wu et al., 2017). Finally, 1.2 mg / kg body weight naloxone hydrochloride (Naloxon Inresa, 0.4 mg / mL, Inresa Arzneimittel GmbH, Freiburg, Germany), 0.5 mg / kg flumazenil (Flumazenil Inresa 0.5 mg, 0.1 mg / mL, Inresa Arzneimittel GmbH) and 2.5 mg / kg atipamezole hydrochloride (Antisedan, 5 mg / ml, Vetoquinol GmbH, Ismaning, Germany) were dissolved in 0.9% NaCI and injected subcutaneously, awakening the mouse.
[0109] Documentation and Treatment. Documentation was performed every 24 h. Keratitis severity was visualized using a stereomicroscope (Leitz ELVAR, Wetzlar, Germany) at 16x magnification combined with a high-resolution microscope digital camera (Seben GmbH, Berlin, Germany) and evaluated using a previously described clinical scoring scheme (Lyu et al., 2020). Briefly, a score of 0 indicates no infiltrates, 1 indicates that less than 50% of the cornea is semitransparent, 2 indicates that more than 50% of the cornea is semitransparent, 3 indicates that less than 50% of the cornea is opaque, 4 indicates that more than 50% of the cornea is opaque, and 5 indicates that either spontaneous corneal perforation or phthisis bulbi has occurred. The body weight of the mice was measured daily and used as welfare indicator. General condition and behavior were also recorded daily. Treatment was performed every eight hours starting six hours after infection along 72h. Anesthesia was reached using 3% isoflurane inhalation (Isoflurane-Piramal, Piramal Critical Care Deutschland GmbH, Halbergmoss, Germany) under stable oxygen supply (1.0-1.2 L / min). Then a volume of 5 pL of the appropriate substance was applied to the infected eye. A two-minute incubation period followed. The animal was finally reawakened by isoflurane-removal. Four treatment groups were created. The sham treatment group was applied phosphate buffered saline containing 1 % DMSO. The meropenem treatment group was applied a 0.9% NaCI solution containing 500 pg meropenem / mL and 1 % DMSO. The LasB-inhibitor treatment group was applied a 0.9% NaCI solution containing 1 mg LasB-inhibitor / mL and 1 % DMSO. The combination treatment group was applied a 0.9% NaCI solution containing 1 mg LasB-inhibitor / mL, 500 pg meropenem / mL, and 1 % DMSO.
[0110] Animal Sacrifice. Laboratory animals were sacrificed by intraperitoneal injection of ketamine hydrochloride / xylazine hydrochloride (Ursotamine, 100 mg / mL, Serumwerk Bernburg AG, Germany; Rompun 2%, xylazine, 25 ml, Bayer AG) at a dose of 20 mg / kg body weight three days after infection. The vena cava served was punctured to collect blood. Anticoagulation was achieved by Ca2+-deprivation using a drop of 0.5 M ethylenediaminetetraacetic acid (EDTA). [0111] Eye Homogenization. The removed eyes were homogenized using a POLYTRON PT 1200 E dispenser (Kinematika AG, Lucerne, Switzerland). To avoid bacterial carryover, the eyes were cleaned with distilled water, 70% ethanol and 0.9% NaCI solution, respectively. Microbiological Evaluation. Serial dilutions were plated on blood agar for microbiological evaluation of the homogenates. A blood stripe also incubated on blood agar served as a control for potential systemic spread of the pathogen. In both cases, counting was performed after 18h of incubation at 37°C.
[0112] Flow Cytometry. The eye homogenates, upon receipt, were sieved using a 70 pm cell filter (Falcon, Corning) to prepare a single cell suspension. Total cell count and cell viability were determined using a NucleoCounter® NC-200™ (Chemomatec, Kaiserslautern, Germany). Single cell suspensions were fixed using 1 % paraformaldehyde, centrifuged, and resuspended in FACS buffer (0.1 mM EDTA plus 1 % fetal bovine serum in PBS). CD16 / CD32 antibodies (Thermo Fisher Scientific) were then added to the samples and incubated at 4 °C for 40 min to block Fc receptors. The surface markers were stained using the following fluorescent dye-conjugated antibodies: F4 / 80-PE (clone BM8), CD45- APC (clone 30-F11 ), CD11 b-FITC (clone M1 / 70), CD11 c APC-Cy7 (clone N418), Ly6G PE-Cy7 (1A8-Ly6g), and NK1.1-PerCP- Cy5.5 (clone PK136) (Biolegend) and incubated at 4 °C for 45 min. The blood samples were first mixed with an ACK-Lysis Buffer (Thermo Fisher Scientific) for 20 min. The lysis was terminated in time by adding DPBS. Centrifugation was used to selectively collect the leukocytes. Total cell count estimation and surface marker staining were performed as described above for the eye samples. The BD FACSverse machine was used for flow cytometry analysis, whereas the resulting data were analyzed using Flowjo v10.6.2 (BD).
[0113] Enzyme-linked Immunoadsorption Assay. After centrifugation of the homogenates at 2500 g and 4 °C for 10 min, the cell-free suspension was frozen at -70 °C. Quantification of mKC, mTNF-a, mMPO and mlL-10 in the centrifuged homogenate was performed using R&D ELISA kits according to the manufacturer's protocol. Briefly, 96-well microplates were coated with the respective capture antibody and incubated at rt overnight. The following day, the wells were blocked with Delent reagent for 2h, incubated with the samples for 2h, incubated with the capture antibody for 2h and finally incubated with streptavidin-HRP for 20 min. The reaction was started by adding the substrate solution and stopped after 20 min with 2NH2SO4. The absorbance was measured at 450 nm. A serially diluted standard was used in parallel to calculate the concentration of each protein.
[0114] Bioanalytical sample preparation of keratitis samples for determination of concentration of HIPS-7178 in eye and serum. All PD serum and eye homogenate samples were analyzed via HPLC-MS / MS using an Agilent 1290 Infinity II HPLC system and coupled to an AB Sciex QTrap6500+ mass spectrometer. First, a calibration curve was prepared by spiking different concentrations of HIPS-7178 into the respective matrix (mouse serum (pooled, from CD-1 mice) for plasma samples, isotonic sodium chloride solution for eye homogenate samples). Caffeine was used as an internal standard. In addition, quality control samples (QCs) were prepared for HIPS-7178 with the respective matrix. The following extraction procedure was used: 7.5 pL of a serum sample (calibration samples, QCs or PD samples) was extracted with 37.5 pL of methanol containing 12.5 ng / mL of caffeine as internal standard for 5 min at 2,000 rpm on an Eppendorf MixMate® vortex mixer. Then samples (serum) were spun down at 13,000 rpm for 5 min. Supernatants were transferred to standard HPLC-glass vials. 50 pL of an eye homogenate sample (calibration samples, QCs or PD samples) were extracted with 50 pL of methanol and 1 pL caffeine (concentration 1 pg / mL in methanol) for 5 min at 800 rpm on an Eppendorf MixMate® vortex mixer. Then samples (eye homogenate) were spun down at 4,000 rpm for 40 min at 4 °C. Supernatants were transferred to 96well V-bottom plates (Greiner). HPLC conditions were as follows: column: Agilent Zorbax Eclipse Plus C18, 50x2.1 mm, 1.8 pm; temperature: 30 °C; injection volume: 5 pL; flow rate: 700 pL / min; solvent A: water + 0.1 % formic acid; solvent B: acetonitrile + 0.1 % formic acid; gradient: 99% A at 0 min and until 1 min, 99% - 0% A from 1.0 min to 2.2 min, 0% A until 4 min. Mass spectrometric conditions were as follows: Scan type: MRM, negative and positive mode; Q1 and Q3 masses for caffeine and HIPS-7178 can be found in table given below. Peak areas of each sample and of the corresponding internal standard were analyzed using MultiQuant 3.0 software (AB Sciex). Peak areas of the respective sample were normalized to the internal standard peak area. The MS / MS pairs used for quantification are marked with a ‘Q’ in the table, the other MS / MS pairs for the respective compound were used for qualification. Peaks of PD samples were quantified using the calibration curve. The accuracy of the calibration curve was determined using QCs independently prepared on different days.

All samples were analyzed via HP LC-MS / MS using an Agilent 1290 Infinity II HPLC system and coupled to an AB Sciex QTrap7500 mass spectrometer. Eye homogenate samples from the PD study with HIPS-7178 were extracted as follows: 15 pL of an eye homogenate sample was treated with the Sciex Protein Preparation Kit (Darmstadt, Germany, AB Sciex). In brief, 30 pl of digestion buffer (0.1 M Tris, pH 8, 4 mM CaCL), then 2.5 pl Denaturant (10 % N-octyl-glucoside) and 5 pl reducing agent (50 mM of tris (2-caroxyethyl)-phosphine were added to each sample. Then the mixture was incubated for 1 hour at 60°C at 150 rpm on a rotary shaker. Then samples were shortly spun down. Next, 2.5 pl cysteine blocking reagent (200 mM methyl methane- thiosulfanate) were added and the mixture was incubated for 10 min at room temperature. Then, 50 pl of digestion buffer was added and finally 10 pl of trypsin ( / dissolved in 0.1 % formic acid). The mixture was incubated for 3 hours at 37°C. Then samples were quickly spun down and reaction was terminated with 5 pl of a quenching solution containing 10% formic acid in water. Then samples were spun down at 4,000 rpm at room temperature for 10 min. Supernatants were transferred to new 96well- Greiner-V-bottom plates. [0128] Effects of LasB on the human cornea
[0129] The following experiments illustrate the effects of LasB on the human cornea. The latter were provided as ex vivo samples by the Eye Clinic of the Saarland University Hospital. Collagen is the main structural component of the human cornea, making up around 70-80% of its dry weight. The extent of collagen degradation in the cornea is measured by quantifying hydroxyproline (HP). HP is a non-essential amino acid that is released from collagen in case of its destruction. While HP levels remain low in the absence of LasB, they increase significantly with rising protease concentrations (Figure 5). This effect is also reflected in the increase in corneal opacity - an observation that aligns with clinical findings in P. aeruginosa infections.
[0130] Figure 5 shows effects of different concentrations of LasB on human corneal samples ex vivo. The extent of collagen degradation in the cornea is measured by quantifying hydroxyproline, which is a decomposition product of collagen.
[0131] When adding 5 or 25 pM of LasB inhibitors such as HIPS-7178 to the LasB treated samples, the HP levels are significantly lower compared to the positive control (where LasB but no inhibitor was added) (Figure 6). These experiments demonstrate two aspects: 1. LasB contributes drastically to the damage of the collagen-containing human cornea and 2. this degradation can be prevented by inhibiting the metalloprotease.
[0132] Figure 6 shows the protective effect of HIPS-7178 on the integrity of human corneal samples ex vivo, as demonstrated by the quantification of hydroxyproline levels. PC, positive control only containing LasB and no inhibitor; NC, negative control without LasB and compound. The collagen degradation of LasB was reduced in presence of HIPS-7178, almost reaching the levels of the NC.
[0133] Hydroxyproline quantification. The assay was conducted using the Hydroxyproline Assay Kit (Colorimetric) from abeam (ab222941 ). Firstly, the donated cornea was removed from their incubator storage flask and placed into a petri dish filled with MEM- medium (Gibco, 11095080; with 10% FBS, 1 % Pen / Strp, 1 % NEAA and 1 % Sodium Pyruvate). The whole cornea was split into equally sized quarters using a sterile scalpel and pincers. Afterwards, a 24-well plate is prepared by adding a base volume of 530 pL MEM-medium to each used well. The cornea quarters were then moved to one well each and the respectively planned amounts of LasB, inhibitor as well as MEM-medium are added, summing up to a total volume of 600 pL. After mixing every sample well by pipetting up and down multiple times, the 24-well plate were incubated at 37 °C and 5% CO2 for 24 h.
[0134] After the incubation time ended, the entire volume as well as the cornea piece of each well were transferred to separate 5 mL reaction tubes and treated in the ultrasonic bath for 10 min. The rest of the procedure as well as the detection were done according to the kit’s manual. In detail, for duplicate measurements 2x 100 pL of each sample were added to separate 2 mL SafeSeal polypropylene reaction tubes, mixed with 100 pL 10 M NaOH respectively and hydrolyzed at 120 °C over 1 h. During this time frame the assay kit was equilibrated at room temperature for 30 minutes before preparing a 0.1 pg / pL hydroxyproline stock solution using materials the kit provides. This stock is used to create a dilution series of final hydroxyproline amounts from 0 to 1 pg in 0.2 pg steps. A duplicate of this dilution series was added to a 96-well plate with 2x 10 pL per step. After the hydrolysis was done, each sample is cooled on ice for 5 min before adding 100 pL of 10 M HCI, shaking the tubes to mix everything and centrifuging them for 5 min at 10.000 g. Like the calibration standards, 10 pL of each sample are pipetted onto the 96-well plate. Following this, the plate was placed onto a Thermomixer at 75°C until all samples evaporated. Meanwhile, a stock of 94 pL Oxidation buffer and 6 pL Chloramine T is prepared per standard and sample. 100 pL of the reaction solution are transferred to the respective wells after the samples completely evaporated. Afterwards, the plate was incubated for 20 min at room temperature before adding 50 pL of the Developer solution to every sample and incubating the plate at 37°C for 5 min. Next, 50 pL of DMAB Concentrate are pipetted into each sample. The plate is sealed with a foil, placed on a thermomixer, shortly shook at 500 rpm and incubated for a final time at 65°C for 45 min. Lastly, the absorbance at 560 nm is measured using the Pherastar platereader by BMG Labtech.

A method for treatment of Pseudomonas keratitis, which comprises administering to a subject in need of such treatment a therapeutically effective amount of a compound of formula (I):whereinX is a group of formula -PO(OH)2;R1is an optionally substituted cycloalkyl group, an optionally substituted heterocycloalkyl group, an optionally substituted aryl group or an optionally substituted heteroaryl group or an optionally substituted aralkyl group or an optionally substituted heteroaralkyl group; or a group of formula -CH(R6)-C(=O)- NH-R7, or a group of formula -C(Me)2-CH2-C(=O)-NH-R7, or a group of formula -CH(R6)-CH2-C(=O)-NH-R7, or a group of formula -CH(R6)-R8;R2is an alkyl, alkenyl, alkynyl, heteroalkyl, cycloalkyl, heterocycloalkyl, alkylcycloalkyl, heteroalkylcycloalkyl, aryl, heteroaryl, aralkyl or heteroaralkyl group, all of which may optionally be substituted;R6is hydrogen or an alkyl, alkenyl, alkynyl, heteroalkyl, cycloalkyl, heterocycloalkyl, alkylcycloalkyl, heteroalkylcycloalkyl, aryl, heteroaryl, aralkyl or heteroaralkyl group, all of which may optionally be substituted;R7is an optionally substituted cycloalkyl group, an optionally substituted heterocycloalkyl group, an optionally substituted aryl group, an optionally substituted heteroaryl group, an optionally substituted aralkyl group or an optionally substituted heteroaralkyl group;R8is an optionally substituted cycloalkyl group, an optionally substituted heterocycloalkyl group, an optionally substituted aryl group, an optionallysubstituted heteroaryl group, an optionally substituted aralkyl group or an optionally substituted heteroaralkyl group; andR1ais hydrogen, or, if R1is a group of formula -CH(R6)-C(=O)-NH-R7, R1aand R6together may be a group of formula -(CH2)3- or -(CH2)4-; or a salt thereof.
4. The compound for use according to claim 1 or the use according to claim 2 or the method according to claim 3, wherein the compound is a compound of formula (II):wherein R1and R2are as defined in claim 1 , 2 or 3, or a salt thereof.

The compound for use according to claim 1 or 4 or the use according to claim 2 or 4 or the method according to claim 3 or 4, wherein R2is a C1-6 alkyl group; a heteroalkyl group containing from 1 to 6 carbon atoms and 1 , 2, 3 or 4 heteroatoms selected from 0, S and N; a C4-10 alkylcycloalkyl group; or a C7-12 aralkyl group; all of which may optionally be substituted.
6. The compound for use according to claim 1 or 4 or the use according to claim 2 or 4 or the method according to claim 3 or 4, wherein R2is an optionally substituted benzyl group, or a group of formula -CH2CH(CH3)2.
7. The compound for use according to claim 1 or 4 or the use according to claim 2 or 4 or the method according to claim 3 or 4, wherein R2is a group of formula - CH2CH(CH3)2.
8. The compound for use according to any one of claims 1 or 4 to 7 or the use according to any one of claims 2 or 4 to 7 or the method according to any one of claims 3 to 7 wherein R1is an optionally substituted aryl group or an optionally substituted heteroaryl group.
9. The compound for use according to any one of claims 1 or 4 to 7 or the use according to any one of claims 2 or 4 to 7 or the method according to any one of claims 3 to 7 wherein R1is an optionally substituted phenyl group, an optionally substituted naphthyl group or an optionally substituted heteroaryl group containing one or two rings and from 5 to 10 ring atoms selected from C, 0, N and S.
10. The compound for use according to any one of claims 1 or 4 to 7 or the use according to any one of claims 2 or 4 to 7 or the method according to any one of claims 3 to 7 wherein R1is an optionally substituted phenyl group.
11. The compound for use according to any one of claims 1 or 4 to 7 or the use according to any one of claims 2 or 4 to 7 or the method according to any one of claims 3 to 7 wherein R1is a group of formula -Cy1-L-Cy2, wherein Cy1is an optionally substituted cycloalkylene group containing 1 or 2 rings and from 3 to 7 carbon ring atoms, an optionally substituted heterocycloalkylene group containing 1 or 2 rings and from 3 to 7 ring atoms selected from C, N, 0 and S, an optionally substituted phenylene group, or an optionally substituted heteroarylene group containing 5 or 6 ring atoms selected from C, N, 0 and S; Cy2is a cycloalkyl, heterocycloalkyl, alkylcycloalkyl, heteroalkylcycloalkyl, aryl, heteroaryl, aralkyl or heteroaralkyl group, all of which may optionally be substituted; and L is a bond or -O-, -S-, -NH-, -CH2-, -CO-, -NHCO-, -CO-NH-, - CH2-CO-NH-, -NH-CO-CH2-, -CH2-O-CO-NH-, -NH-CO-O-CH2-, -O-CO-NH-, -NH-CO-O-, -NHS02-, -SO2NH-, -CH2-SO2-NH-, -NH-SO2-CH2-, -S-CH2-, -CH2- S-, -NH-CH2-, -CH2-NH-, -O-CH2- or -CH2-O-.

The compound for use or the use or the method according to claim 11 , wherein Cy2is an optionally substituted phenyl group, an optionally substituted biphenyl group, an optionally substituted naphthyl group, an optionally substituted heteroaryl group containing one or two rings and 5, 6, 9 or 10 ring atoms selected from C, 0, N and S, an optionally substituted cycloalkyl group containing from 3 to 7 ring atoms, an optionally substituted heterocycloalkyl group containing from 3 to 7 ring atoms selected from C, N, 0 and S, an optionally substituted heterocycloalkylaryl group containing 9 or 10 ring atoms selected from C, N, S and 0, or a group of formula -CH(CH2Ph)Ph; and / or wherein L is a bond or -NHCO-, -CO-NH-, -CH2-CO-NH-, -NH-CO-CH2-, - NHSO2- or -SO2NH-; and / or wherein Cy1is a 1 ,4-phenylene group.
13. The compound for use according to any one of claims 1 or 4 to 7 or the use according to any one of claims 2 or 4 to 7 or the method according to any one of claims 3 to 7 wherein R1is a group of formula -CH(R6)-C(=O)-NH-R7or a group of formula -CH(R6)-R8.

The compound for use or the use or the method according to claim 13, wherein R6is hydrogen or a C1-6 alkyl group, a C3-7 cycloalkyl group, a heterocycloalkyl group containing from 3 to 7 ring atoms selected from C, N, 0 and S, a phenyl group or a heteroaryl group containing 5 or 6 ring atoms selected from C, N, S and 0, or a group of formula -CH2-R6a, wherein R6ais a C3-7 cycloalkyl group, a heterocycloalkyl group containing from 3 to 7 ring atoms selected from C, N, 0 and S, a phenyl group or a heteroaryl group containing 5 or 6 ring atoms selected from C, N, S and 0.
15. The compound for use or the use or the method according to claim 13, wherein R6is a group of formula -CH(CH3)2 or a cyclopropyl group.
16. The compound for use or the use or the method according to claim 13, 14 or 15, wherein R7is an optionally substituted phenyl group or an optionally substituted C3-7 cycloalkyl group.
17. The compound for use or the use or the method according to claim 13, 14 or 15, wherein R7is an optionally substituted phenyl group.

The compound for use or the use or the method according to claim 13, 14 or 15, wherein R8is an optionally substituted heteroaryl group containing 5 to 10 ring atoms selected from C, N, 0 and S (such as an optionally substituted benzimidazole group or an optionally substituted triazole group or an optionally substituted imidazole group).

The compound for use according to claim 1 or the use according to claim 2 or the method according to claim 3, wherein the compound is a compound of formula (II):whereinR1is a group of formula -CH(R6)-C(=O)-NH-R7or a group of formula -CH(R6)- R8;R2is a group of formula -CH2CH(CH3)2;R6is a group of formula -CH(CH3)2 or a cyclopropyl group;R7is an optionally substituted phenyl group; andR8is an optionally substituted heteroaryl group containing 5 to 10 ring atoms selected from C, N, 0 and S; or a salt thereof.
20. The compound for use or the use or the method according to claim 13, 14, 15 or 19, wherein R8is a group of the following formula:wherein D is an optionally substituted aryl group or an optionally substituted heteroaryl group.21 . The compound for use or the use or the method according to claim 20, wherein D is an optionally substituted phenyl group.
22. The compound for use or the use or the method according to any one of the preceding claims, for use in the definitive adjunctive therapy of keratitis, especially contact lens-associated keratitis (CLAK), due to confirmed P. aeruginosa.
23. The compound for use or the use or the method according to any one of the preceding claims, wherein the compound of formula (la) is administered in combination with an antibiotic like chloramphenicol, gentamicin, kanamycin, neomycin, levofloxacin, moxifloxacin, tobramycin, ofloxacin, oxytetracyclin and meropenem.
