Description

Composition for alleviating skin irritation containing a lipid complex

[0001] Cross-reference to related applications

[0002] This application claims priority to Republic of Korea Patent Application No. 10-2024-0098385, filed July 25, 2024, and Republic of Korea Patent Application No. 10-2025-0087958, filed July 1, 2025, the entire contents of which are incorporated herein by reference.

[0003] Disclosed herein is a composition comprising a lipid complex comprising ceramide or a similar ceramide and a fatty acid, which has a stable formulation and has a skin irritation alleviating effect.

[0004] Human skin is a part directly exposed to the external environment, and recently, environmental pollution has become serious, and the skin is easily damaged due to increased ultraviolet ray exposure due to ozone layer destruction, dry living spaces, individual allergies, stress, and increased exposure to chemical toxic substances. As a result, various irritants are generated, and the skin easily turns red or becomes sensitive.

[0005] Many cosmetics manufacturers are showing increasing interest in substances that soothe irritation in cosmetics, and various raw materials are being developed.

[0006] In one aspect of the present disclosure, it is intended to provide a composition that can alleviate skin sensitivity while maintaining a stable formulation by including a lipid complex as an active ingredient.

[0007] In one aspect, the present disclosure provides a composition for alleviating skin irritation, comprising a lipid complex as an active ingredient, wherein the lipid complex comprises: a nucleating agent; a pseudokeratin lipid lamellar layer comprising a ceramide or a ceramide analogue positioned on the nucleating agent; and a polymer layer on the pseudokeratin lipid lamellar layer.

[0008] In one aspect, the composition for relieving skin irritation according to the present disclosure includes a lipid complex having a structure similar to the stratum corneum of the skin, which has excellent phase stability and can contain a large amount of moisture and useful active substances, and thus can exhibit an excellent skin irritation relieving effect when applied to sensitive skin.

[0009] Figure 1 shows the skin irritation alleviation effect of a composition according to one embodiment of the present disclosure.

[0010] Hereinafter, the present disclosure will be described in more detail through the following examples. However, the following examples are provided solely for illustrative purposes to aid understanding of the present disclosure, and the scope and range of the present disclosure are not limited thereto.

[0011] In an exemplary embodiment of the present disclosure, a composition for alleviating skin irritation is provided, comprising a lipid complex as an active ingredient, wherein the lipid complex comprises: a nucleating agent; a pseudokeratin lipid lamellar layer comprising a ceramide or a ceramide analogue positioned on the nucleating agent; and a polymer layer on the pseudokeratin lipid lamellar layer.

[0012] In another exemplary embodiment of the present disclosure, a method of soothing skin irritation is provided, comprising applying a lipid complex to the skin, the lipid complex comprising: a nucleating agent; a pseudokeratin lipid lamella layer comprising a ceramide or a ceramide analog as an active ingredient; and a polymer layer on the pseudokeratin lipid lamella layer.

[0013] In another exemplary embodiment of the present disclosure, a use of a lipid complex for preparing a composition for soothing skin irritation is provided, the lipid complex comprising: a nucleating agent; a pseudokeratin lipid lamella layer comprising a ceramide or a ceramide analog as an active ingredient; and a polymer layer on the pseudokeratin lipid lamella layer.

[0014] In another exemplary embodiment of the present disclosure, a non-therapeutic use of a lipid complex for alleviating skin irritation is provided, the lipid complex comprising: a nucleating agent; a pseudokeratin lipid lamellae layer comprising a ceramide or a ceramide analog as an active ingredient; and a polymer layer on the pseudokeratin lipid lamellae layer.

[0015] The above-mentioned nucleating agent may be a colloid that is inactive and can be easily disintegrated by external stimuli.

[0016] In one embodiment, the nucleating agent may be selected from the group consisting of sugar, salt, hyaluronic acid, alginic acid, chondroitin sulfate, chitosan, chitin, polylysine, collagen, gelatin, stearic acid, cetyl alcohol, stearyl alcohol, and mixtures thereof.

[0017] In one embodiment, the nucleating agent may be included in an amount of 15 to 45 wt% based on the total weight of the lipid complex. When the amount of the nucleating agent is used within the above range, not only can aggregation that may occur during the process of coating a pseudokeratin lipid lamellar layer, polymer layer, etc. on the surface of the nucleating agent be reduced, but the coating rate can also be increased, thereby enhancing the structural stability of the lipid complex, which is the final product.

[0018] For example, the nucleating agent may be at least 15 wt%, at least 20 wt%, at least 25 wt%, at least 30 wt%, at least 35 wt%, or at least 40 wt%, and may be at most 45 wt%, at most 40 wt%, at most 30 wt%, at most 25 wt%, or at most 20 wt%, based on the total weight of the lipid complex.

[0019] As mentioned above, the stratum corneum is located on the nucleus. The stratum corneum of human skin is composed of keratinocytes and intercellular lipids, and functions as a unique barrier to the skin. The intercellular lipids of the keratinocytes play the most crucial role in this barrier function, and have a multilayered lamellar structure. The pseudokeratinocyte lipid lamellar layer is a layer that substantially mimics the structure of intercellular lipids of the keratinocytes with a lamellar structure.

[0020] The above-mentioned stratum corneum lipid lamellae layer contains ceramides, which function to protect the body from stress by removing damaged cells when cells are damaged due to internal or external stress. As the skin ages, the content of ceramides within the stratum corneum decreases, which can lead to loss of skin moisture, exposure to external stimuli such as ultraviolet rays or chemicals, and exfoliation of keratinocytes, which can make the skin surface sensitive. Therefore, by supplying ceramides or ceramide analogs to the skin from the outside, the degree of irritation of sensitive skin can be alleviated.

[0021] In one embodiment, the ceramide or ceramide analogue may include at least one selected from the group consisting of ceramide 1 (ceramide EOP), ceramide 2 (ceramide NS), ceramide 3 (ceramide NP), ceramide 4, ceramide 5 (ceramide AS), ceramide 6 (ceramide AP), Hydroxypropyl Bispalmitamide MEA, Dihydroxyisopropyl Palmoylpalmamide, and Cetyl-PG Hydroxyethyl Palmitamide.

[0022] For example, the ceramide or ceramide analogue may include one or more of Hydroxypropyl Bispalmitamide MEA and Cetyl-PG Hydroxyethyl Palmitamide.

[0023] In one embodiment, the pseudokeratin lipid lamellar layer may be included in an amount of 40 to 80 wt% based on the total weight of the lipid complex.

[0024] For example, the keratin-like lipid lamella layer may be at least 40 wt%, at least 50 wt%, at least 60 wt%, or at least 70 wt%, and may be at most 80 wt%, at most 70 wt%, at most 60 wt%, or at most 50 wt%, based on the total weight of the lipid complex.

[0025] In one embodiment, the concentration of the ceramide or ceramide analog may be 15 to 45 wt% based on the total weight of the lipid complex. When this range is satisfied, the formulation may exhibit a skin irritation-alleviating effect while being stable and free of skin toxicity.

[0026] For example, the concentration of the ceramide or ceramide analog may be at least 15 wt%, at least 20 wt%, at least 25 wt%, at least 30 wt%, at least 35 wt%, or at least 40 wt%, and may be at most 45 wt%, at most 40 wt%, at most 30 wt%, at most 25 wt%, or at most 20 wt%, based on the total weight of the lipid complex.

[0027] In one embodiment, the pseudokeratin lipid lamellae may further comprise a fatty acid; cholesterol or a derivative thereof; or a mixture thereof.

[0028] In one embodiment, the fatty acid may be an unsaturated or saturated fatty acid having 8 to 30 carbon atoms. For example, the fatty acid may include one or more selected from the group consisting of stearic acid, myristic acid, arachidonic acid, palmitic acid, oleic acid, and behenic acid.

[0029] In one embodiment, the cholesterol or a derivative thereof may include at least one selected from the group consisting of cholesterol, cholesteryl chloride, cholesteryl octanoate, cholesteryl nonanoate, cholesteryl oleyl carbonate, cholesteryl isostearyl carbonate, and mixtures thereof.

[0030] In one embodiment, the pseudokeratin lipid lamellae may comprise ceramide or a ceramide analog, a fatty acid, and cholesterol or a derivative thereof.

[0031] In one embodiment, the ceramide or ceramide analog; fatty acid; and cholesterol or a derivative thereof; may be included in a weight ratio of 1:0.1 to 10:0.01 to 1.

[0032] For example, the weight ratio of the ceramide or ceramide analog; fatty acid; and cholesterol or a derivative thereof may be 1:0.1 to 10:0.01 to 1, 1:0.2 to 5:0.01 to 0.9, 1:0.5 to 3:0.01 to 0.5, or 1:0.8 to 2:0.01 to 0.1.

[0033] A polymer layer is located on the pseudokeratin lipid lamellar layer. The polymer layer surrounds the outside of the pseudokeratin lipid lamellar layer and protects the pseudokeratin lipid lamellar layer, thereby significantly improving the structural stability of the lipid complex.

[0034] The polymer layer is not particularly limited as long as it does not easily dissolve in a solvent, such as water or a polar organic solvent, and can be evenly coated on the surface of the pseudokeratin lipid lamellar layer, and may be a natural polymer or a synthetic polymer.

[0071] Exam example

[0072] 1. Test method for skin irritation relief effect of ceramide-containing lipid complex

[0073] To confirm the skin irritation relief effect of a lipid complex containing ceramide, a single skin irritation test was conducted on 25 healthy adults (2 males, 23 females, average age: 42.1 years) (February 20, 2024 - February 23, 2024, Safety Lab, Amorepacific Technology Research Center).

[0074] On the first day of the study, a patch was attached to the upper back of the subject, and after 48 hours, the patch was removed. The first reading was performed 30 minutes later, and the second reading was performed 24 hours later. In order to determine the intensity of skin irritation of the sample, the average skin reactivity was calculated by assigning weights according to the degree of positive skin reaction and determining the skin irritation of the sample. The test was performed based on the terminology presented in [Table 1], which is a modified test standard from the ICDRG, CTFA guideline (1981) and the test standard of Frosch & Kligman (1979). The test results were calculated as the average value of the reaction for each subject.

[0081] Referring to Tables 3 and 4 below, the vehicle prescription containing 60% DI Water, 39.6% BG, and 0.4% CARBOMER showed a 1-point reaction in 5 subjects in the first test and a 1-point reaction in 4 subjects in the second test, showing an average stimulation score of 0.36.

[0082] In the second prescription containing 6.55% Hydroxypropyl Bispalmitamide MEA (the content of Hydroxypropyl Bispalmitamide MEA in the lipid complex is 32.8%, and the content of Hydroxypropyl Bispalmitamide MEA in the entire composition is 6.55% by calculating the ratio of 32.8% of the 20% content of lipid complex in the composition, and the same content hereinafter), 2 people scored 1 point in the first test and 2 people scored 1 point in the second test, and the average stimulation was evaluated as 0.16. In addition, in the 3rd prescription containing CERAMIDE NP 6.55%, 0 people scored 1 point in the first test, 2 people scored 1 point in the second test, and the average stimulation was evaluated as 0.08. In the 4th prescription containing Cetyl-PG Hydroxyethyl Palmitamide 6.55%, 2 people scored 1 point in the first test, and 3 people scored 1 point in the second test, and the average stimulation was evaluated as 0.16.

[0083] Therefore, all prescriptions 2-4 showed a decrease in irritation level compared to the vehicle, and when the lipid complex containing Hydroxypropyl Bispalmitamide MEA, Ceramide NP, and Cetyl-PG Hydroxyethyl Palmitamide was included, irritation was reduced by 55.56%, 77.78%, and 55.56%, respectively.
