Abstract

Disclosed herein are cosmetic compositions. [0001] COSMETIC COMPOSITION

[0002] Field of the Invention

[0003] Disclosed herein are cosmetic compositions. More particularly, disclosed herein are cosmetic compositions comprising at least one compound of General Formula (1), including but not limited to N,N,N',N'-Tetrakis(2-hydroxyethyl)ethylenediamine (THEED), tetrahydroxypropyl ethylenediamine (THPE), and N,N,N',N'-Tetrakis(2-hydroxypentyl)ethylenediamine, and at least one skin care benefit agent selected from acetyl glutamic acid, acetyl glutamine, acetyl methionine, acetyl tyrosine, and combinations thereof for combating the harmful effects of atmospheric pollution relating to hyperpigmentation in human skin.

[0004] Background of the Invention

[0005] Chemical substances such as aromatic hydrocarbons such as dioxin and aromatic hydrocarbons (PAHs) are present in the air and atmosphere. Exposure such as by inhaling such chemical substances in the air can harm the human body in various aspects. For example, pollutants in the atmosphere may cause inflammation in the cells of the body, resulting in irritation, overproduction of certain substances like melanin (i.e., the natural skin pigment present in human skin) produced by cells, and even cancer.

[0006] The present inventors have surprisingly found cosmetic compositions comprising at least one compound of General Formula (1), including but not limited to N,N,N',N'-tetrakis(2- hydroxyethyl)ethylenediamine (THEED), tetrahydroxypropyl ethylenediamine (THPE), and N,N,N',N'-tetrakis(2-hydroxypentyl)ethylenediamine, and at least one skin care benefit agent are effective for combating the harmful effects of atmospheric pollution, particularly that relating to hyperpigmentation in human skin. “Hyperpigmentation,” as used herein, refers to an area of skin wherein greater pigmentation than that of an adjacent area of skin is present, such as in the case of a sun / age spot or post-inflammatory hyperpigmentation (PIH). Thus, cosmetic compositions comprising at least one compound of General Formula (1) and at least one skin care benefit agent as described herein, fill this pollution-combatting need in consumers struggling with pollution-induced hyperpigmentation. [0010] U.S. Patent Application Publication No. 2016 / 0324754 provides for a shaving cream composition comprising water, carbomer, ethylhexylpalmitate, glycerin, glyceryl acrylate / acrylic acid, isododecane, stearic acid, butylene glycol, ethyl alcohol, cetearyl alcohol, ceteareth-20, propanediol, Brassica Campestris, phenoxyethanol, ethylhexylglycerin, tetrahydroxypropyl ethylenediamine, Aloe Barbadensis, carboxymethylhydroxyethylcellulose, bisabolol, PEG-45M, and tocopheryl acetate.

[0012] Summary of the Invention

[0013] Accordingly, in a first aspect, the inventive composition relates to a cosmetic composition comprising at least one skin care benefit agent, and at least one compound of General Formula

[0014] where Ri, R2, R3, R4 are each, independently, a linear or branched carbon chain having 2 to 6 carbon atoms; and Rs is a carbon chain having 1 to 6 carbon atoms; in a cosmetically acceptable vehicle.

[0015] In a second aspect, there is provided a cosmetic composition comprising at least one skin care benefit agent and at least one skin care active comprising caffeine, ubiquinone, hydrolyzed rice protein, bisabolol, carnosine, Nannochloropsis Oculata extract, Gleditsia Triacanthos Seed extract, Zingiber Officinale (Ginger) Root extract, menthoxypropanediol, or a combination thereof.

[0016] In a third aspect, there is provided a method of providing a skin benefit to skin comprising: applying a cosmetic composition to the skin, wherein the cosmetic composition comprises at least one skin care benefit agent and at least one compound of General Formula (1), where R1, R2, R3, R4are each, independently, a linear or branched carbon chain having 2 to 6 carbon atoms; and R5is a carbon chain having 1 to 6 carbon atoms; in a cosmetically acceptable base, wherein a weight ratio of the skin care benefit agent to the compound of General Formula (1) is present from 75:1 to 1 :75, preferably, from 65:1 to 1 :50, and, more preferably, from 55:1 to 1 :25.

[0023] Detailed Description of the Invention

[0024] Cosmetic compositions disclosed herein comprise at least one skin care benefit agent. The skin care benefit agent, as described herein, is selected from acetyl glutamic acid (e.g., N-acetyl glutamic acid), acetyl glutamine (e.g., N-acetyl glutamine), acetyl methionine (e.g., N-acetyl methionine), acetyl tyrosine (e.g., N-acetyl tyrosine), and combinations thereof, including as a free acid, amide, ester, lactone, isomer, and any derivatives or salts thereof. The term “derivative(s)” as used with reference to ingredients herein refers to an amide, ester, ether, carboxyl, amino, acetyl, and / or alcohol forms of the particular ingredient. “Salts,” as used herein, includes but is not limited to ammonium, calcium, magnesium, manganese, potassium, sodium, tetraalkylammonium, trialkylammonium, and a combination thereof. In a further aspect, the skin care benefit agent may be present in any of its isomeric forms and in combinations thereof. “Isomers,” as used herein, is meant to include D-isomers, L-isomers, and / or DL- isomers for a particular compound. Thus, the skin care benefit agent comprises its D-isomer form, L-isomer form, DL-isomer form, or a combination thereof. For example, N-acetyl methionine may be present in any of its isomeric forms and in combinations thereof. For example, N-acetyl-DL-methionine (i.e., is in its DL-isomer form), N-acetyl-D-methionine (i.e., is in its D-isomer form) and N-acetyl-L-methionine (i.e., is in its L-isomer form).

THPE is commercially available, for example, as Quadrol® or Neutrol® TE from BASF. In another aspect, the acetyl methionine to THPE is present in a weight ratio of from 60:1 to 1 :0.5, or from 60:1 to 1 :1, or from 55:1 to 5:1 , or from 55:1 to 7:1 , or from 50:1 to 10:1.

More particularly, examples of such atmospheric pollutants include polycyclic aromatic hydrocarbons (PAHs), nitro-polycyclic aromatic hydrocarbons, polychlorinated biphenyls, and the like, preferably PAHs.

[0038] PAHs are chemicals that are formed during the incomplete burning of organic substances, such as fossil fuels, and is a pollutant commonly found in the air. For purposes of the cosmetic compositions disclosed herein, the pollutants are in particular polycyclic aromatic hydrocarbons (PAHs), preferably, acenaphthene, acenaphthylene, anthracene, benzanthracene, benzochrysene, benzofluoranthene, benzoperylene, benzopyrene, bromomethylnaphthalene, chrysene, corannulene, coronene, dibenzanthracene, dibenzofluroanthene, fluoranthrene, fluorene, indenopyrene, methylphenanthrene, 1 -naphthalene, naphthalene, ovalene, pentacene, phenanthrene, phenylene, phthalic acid, picene, pyrene, tetracene, triphenylene, or a combination thereof. In one aspect, the PAH is chrysene. Chrysene is commonly found in coal tar.

[0039] The “cosmetic composition”, “cosmetically acceptable composition” or “dermatologically acceptable composition”, means that the compositions or components thereof so-described are suitable for use in contact with skin, particularly human skin, without undue toxicity, incompatibility, instability, irritation, or allergic response. Such compositions may be a liquid, cream, pump or aerosol spray, serum, lotion, balm, deodorant, foam, scrub, wash, concentrates and / or powder which can be easily hydrated or applied with an implement or via a face mask or a pad composition. In one aspect, the cosmetically acceptable carrier may comprise an aqueous phase, an oil phase, an alcohol, a silicone phase, or a mixture thereof, and may be in the form of an emulsion. Emulsions can have a range of consistencies including gels, thin lotions (which may also be suitable for spray or aerosol delivery), creamy lotions, light creams, and heavy creams. Exemplary emulsions include water-in-oil emulsions, oil-in-water emulsions, silicone-in-water emulsions, water-in-silicone emulsions, polyol-in-silicone emulsions, silicone-in-polyol emulsions, polyol-in-oil emulsions, oil-in-polyol emulsions, wax-in-water emulsions and water- oil-water triple emulsions. Preferred emulsions include oil-in-water (“O / W”) emulsions and water-in-oil (“W / O”) emulsions.

In one aspect, mixtures and / or combinations of any of the above-described materials may also be used. In one aspect, suitable fatty materials may be included in the cosmetic composition, as described herein, in an amount may range from 1 to 40%, or from 3 to 30%, or from 4 to 20%, or from 5 to 18%, or from 6 to 17%, or from 7 to 15%, or from 1 to 18%, or from 2 to 15%, or from 3 to 15%, or from 5 to 12% by weight of the composition. It is also within the scope of the cosmetic compositions to include sunscreens and photostabilizers as desirable cosmetically acceptable fatty materials. The sunscreens and photostabilizers that may be used in the cosmetic compositions include such materials as octylmethoxycinnamate (also known as ethylhexyl methoxycinnamate or OMK; commercially available as PARSOL® MCX), and ethylhexyl salicylate (also known as octisalate). Other sunscreens desirable for use include p-aminobenzoic acid (PABA), octyl dimethyl-PABA (also known as ethylhexyl dimethyl PABA), homomenthyl salicylate (also known as homosalate), menthyl anthranilate, benzophenone-4, triethanolamine salicylate, iscotrizinol (also known as diethylhexyl butamido triazone), octyl triazone (also known as ethylhexyl triazone), polysilicone-15, isopentenyl-4- methoxycinnamate (also known as isoamyl 4-methoxycinnamate), mixtures thereof or the like. Also desirable for use is octocrylene. In a preferred embodiment, the oil in the cosmetically acceptable vehicle comprises ethylhexyl triazone, homosalate, octisalate, octylmethoxycinnamate, or a mixture thereof.

[0043] In a preferred embodiment, illustrative examples of fatty materials that may be used in the compositions disclosed herein include, caprylic / capric triglycerides, a cyclic silicone (e.g., cyclopentasiloxane, cyclohexasiloxane), dimethicone, ethylhexyl triazone, homosalate, isodecyl neopentanoate, isopropyl myristate, isopropyl palmitate, octisalate, octocrylene, octylmethoxycinnamate, silicones, triolein, and a mixture thereof. More preferably, the oil comprises caprylic / capric triglycerides, dimethicone, isopropyl myristate, or a mixture thereof. Most preferably, the oil comprises caprylic / capric triglycerides, isopropyl myristate, or a mixture thereof.

[0044] Cosmetic compositions in the form of an emulsion, and suitable for use in the invention, typically have an aqueous phase, which may also include one or more organic liquids that are miscible with water at room temperature (25°C). Seventy-two (72) hours later, culture supernatants were collected to determine cellular cytotoxicity by a Lactate Dehydrogenase (LDH) assay kit made commercially available by Invitrogen, while cultured cells were harvested and processed for estimation of melanin content using conventional techniques known to one skilled in the art.

[0074] 30 pL of culture supernatant was collected from each treatment well and transferred to a 96-well plate. To determine maximum LDH, the lysis buffer provided in the LDH assay kit (10X) was added to the well containing cells in a 1 :10 dilution to enable release of maximum LDH from the cells. To the 30 pL of culture supernatant collected, 30 pL of a mixture of the LDH assay buffer and substrate solution was made based on the number of samples and added. The plate was incubated for thirty (30) minutes. Then, 30 pL of stop solution was added to stop the reaction. Once the reaction was stopped, the reading was recorded at a wavelength of 490 nm in a TECAN plate reader. After ascertaining non-cytotoxic doses, treatments were progressed for melanin estimation.

[0076] After culture supernatants were collected and processed for cellular cytotoxicity by the LDH assay, adherent cells were lysed with a solution containing 1 N NaOH and 10% DMSO prepared in water and placed for one (1) hour at 60°C in a shaker incubator. The lysed cell suspension was cooled to room temperature (i.e. , about 20 to 25°C), mixed thoroughly, and transferred to a 384- well plate. Melanin content was measured at 405 nm in a TECAN plate reader. Melanin content was reported as % control ± SE.

A cosmetic composition comprising: a) at least one skin care benefit agent, and b) at least one compound of General Formula (1):where Ri, R2, R3, R4 are each, independently, a linear or branched carbon chain having2 to 6 carbon atoms; and Rs is a carbon chain having 1 to 6 carbon atoms; in a cosmetically acceptable vehicle; and wherein the skin care benefit agent is acetyl glutamic acid, acetyl glutamine, acetyl methionine, acetyl tyrosine, or a derivative, salt, or combination thereof.

2. The cosmetic composition according to claim 1 , wherein the compound of General Formula (1) is selected from N,N,N',N'-tetrakis(2-hydroxyethyl)ethylenediamine (THEED), tetrahydroxypropyl ethylenediamine (THPE), and N,N,N',N'-tetrakis(2- hydroxypentyl)ethylenediamine.

3. The cosmetic composition according to claim 1 or 2, wherein the compound of General Formula (1) is tetrahydroxypropyl ethylenediamine.

4. The cosmetic composition according to any of the preceding claims, wherein the skin care benefit agent is, acetyl methionine, acetyl tyrosine, or a derivative, salt, or combination thereof, more preferably, acetyl methionine.

The cosmetic composition according to any of the preceding claims, wherein the skin care benefit agent is present in the cosmetic composition disclosed herein in an amount of from 0.0001 to 10%, preferably, from 0.0005 to 8%, and, more preferably, from 0.001 to 6% by weight of the cosmetic composition.P0000896WG GPL256. The cosmetic composition according to any of the preceding claims, wherein the cosmetic composition comprises from 0.00001 to 10%, preferably, from 0.0001 to 7.5%, and, more preferably, from 0.0001 to 5% by weight of a compound having General Formula (1).

7. The cosmetic composition according to any of the preceding claims, wherein the skin care benefit agent to the compound of General Formula (1) is present in a weight ratio of from 75:1 to 1 :75, preferably, from 65:1 to 1 :50, and, more preferably, from 55:1 to 1 :25.

8. The cosmetic composition according to any of the preceding claims, wherein the skin care benefit agent to the compound of General Formula (1) is present in a weight ratio of from 60:1 to 1 :0.5, preferably, from 60:1 to 1 :1 , and, more preferably, from 55:1 to 5:1.

9. The cosmetic composition according to any of the preceding claims, wherein the cosmetically acceptable vehicle comprises at least 40% by weight aqueous phase.

10. The cosmetic composition according to any of the preceding claims further comprising at least one skin care active and / or functional ingredient.

11. A cosmetic composition comprising: a) at least one skin care benefit agent, and b) at least one skin care active comprising caffeine, ubiquinone, hydrolyzed rice protein, bisabolol, carnosine, Nannochloropsis Oculata extract, Gleditsia Triacanthos Seed extract, Zingiber Officinale (Ginger) Root extract, menthoxypropanediol, or a combination thereof; wherein the skin care benefit agent is acetyl glutamic acid, acetyl glutamine, acetyl methionine, acetyl tyrosine, or a derivative, salt, or combination thereof.

12. The cosmetic composition according to claim 11 , wherein the cosmetic composition further comprises at least one of a vitamin B3 compound, a resorcinol compound, a sunscreen, or a combination thereof.

The cosmetic composition according to claim 11 or 12, wherein the skin care benefit agent is acetyl methionine, acetyl tyrosine, or a derivative, salt, or combination thereof, and, more preferably, acetyl methionine.P0000896WG GPL2614. Use of the cosmetic composition according to any of the preceding claims for at least one skin care benefit, preferably, skin tone and / or skin texture, more preferably, for reducing pigmentation of skin.

A method of providing a skin benefit to skin comprising: a) applying a cosmetic composition to the skin, wherein the cosmetic composition comprises at least one skin care benefit agent and at least one compound of General Formula (1):where Ri, R2, R3, R4 are each, independently, a linear or branched carbon chain having 2 to 6 carbon atoms; and Rs is a carbon chain having 1 to 6 carbon atoms; in a cosmetically acceptable base; wherein a weight ratio of the skin care benefit agent to the compound of General Formula (1) is present from 75:1 to 1 :75, preferably, from 65:1 to 1 :50, and, more preferably, from 55:1 to 1 :25; and further wherein the skin care benefit agent is acetyl glutamic acid, acetyl glutamine, acetyl methionine, acetyl tyrosine, or a derivative, salt, or combination thereof.
