Abstract

The present disclosure relates to the field of oral pouched products for delivering active substances and / or for providing a pleasant oral experience. More specifically, the present disclosure relates to a saliva-permeable pouch comprising a moist non-tobacco composition for oral use, said moist non-tobacco composition comprising: i) a carrier material comprising a plant-based fiber, such as microcrystalline cellulose (MCC); and ii) a composition comprising an extract of Dracocephalum moldavica L. and uses thereof.

[0003] The present disclosure relates to the field of oral pouched products for delivering active substances and / or for providing a pleasant oral experience. It relates to oral products that, similar to snuff products, are provided in small pouches for easy adaption in the oral cavity, such as under the lip of a user. The products are tobacco free and are intended to provide positive effects to the user. More specifically the present disclosure relates to oral pouched products comprising active substances that are intended to provide the user with positive effects on the skin.

[0004] BACKGROUND

[0005] As of today, there are many products on the market for providing a pleasant oral experience, such as a taste experience, and for delivering active substances, such as nicotine to a user.

[0006] Smokeless tobacco products for oral use, such as snuff, are very common in some countries, particularly Sweden. These products are available in loose form or as portion-packed in a soft material forming a small pouch. The pouch is typically used by the user by placing the pouch containing the smokeless tobacco product in the mouth, such as between the upper or lower gum and the lip, or in the cheek and then maintaining it there for a while until the contents of the product have been released into the mouth. Accordingly, the pouch material is permeable and releases parts of the smokeless tobacco products continuously when placed in the mouth of the user. The pouch material holds the snuff product in place and should be formed from a material convenient and / or comfortable for the user to have positioned against the sensitive mucous membrane in the mouth.

[0007] Smokeless tobacco products are associated with a general odour that is not pleasant for the user or to the surroundings of the user. More importantly, snuff products provide damage to the gum of the long-term user.

[0008] Moldavian dragonhead (Dracocephalum moldavica L.) is an annual herbaceous plant. Its flowers and leaves, fresh and dried, have been used to prepare fresh herbal tea. In addition, it has traditionally also been used as an herbal drug to treat headaches, stomach and liver disorders and / or congestions and it has also been shown to have cardioprotective effects. Antioxidant activity of the herb has also been studied. An extract of Dracocephalum moldavica L. may be prepared from organically cultivated aerial parts of the herb. Extracts of Dracocephalum moldavica L. have found use in various applications. Recently, an extract of Moldavian dragonhead was shown to have a collagen-boosting effect preserving a youthful collagen expression and mass during aging in a reference model of Caenorhabditis elegans. Moreover, a daily food supplementation of Moldavian dragonhead dry extract provided to humans in an open pilot trial of eight weeks was also shown to significantly increase skin moisturization and elasticity in the study group (Evaluation of the collagen-boosting effects of a Moldavian dragonhead extract, Journal of Community Medicine and Public Health Reports, ISSN: 2692-9899, Franziska Wandrey et al.).

[0009] There is constant need for new products for improving skin health, elasticity and general appearance. There is also a constant need for products that can deliver active substances known to have effects on skin health, elasticity and general appearance such that that the active substances achieve their best effects in the body.

[0010] Users of pouched non-tobacco products are also constantly seeking alternatives to nicotine- containing products providing pleasant and stimulating effects to the users.

[0011] SUMMARY OF THE DISCLOSURE

[0012] The above-mentioned needs have now been addressed by the present disclosure providing an oral pouched non-tobacco product that presents the user with an active composition that have proven effects on skin texture and tone and as further set out herein.

[0013] The active composition is administered through the oral mucosa when the oral pouched non- tobacco product is positioned in the oral cavity, particularly under the lip.

[0014] The present disclosure relates to a saliva-permeable pouch comprising a moist non-tobacco composition for oral use, said moist non-tobacco composition comprising: i) a carrier material comprising a plant-based fiber, such as microcrystalline cellulose (MCC); and ii) a composition comprising an extract of Dracocephalum moldavica L.

[0015] There is also provided the use of a composition comprising an extract of Dracocephalum moldavica L in an in an oral moist non-tobacco composition, preferably wherein said moist non-tobacco composition is comprised in a saliva-permeable pouch for positioning in the mouth, such as under the lip and / or for chewing by the user.

Further moisture may be provided to the composition by for example moisturizers such as propylene glycol.

[0024] A “pouch” according to the present disclosure is a form of a small, soft, sealed but permeable bag for keeping a moist non-tobacco composition of the present disclosure. The pouch is saliva-permeable. The permeability allows saliva to enter the moist non-tobacco composition and substances from the moist non-tobacco composition to travel through the material for release into the oral cavity, or the mouth, of the user. The pouch is made from a material that is comfortable for the user to position under the lip or in the mouth in general. Typically, the pouch is made from or comprises a cellulosic material. The pouch may also be freely positioned elsewhere in the mouth or oral cavity, i.e., other than under the lip of a user. Sometimes the terms oral cavity and mouth are used interchangeably. The material may also in addition to, or alternatively, be made from a material that is particularly suitable for chewing wherein the material has some resistance to chewing but is still permeable to saliva. A “cellulose pouch” as referred to herein means that the material for the pouch includes cellulose, but it may also include other constituents suitable for the purpose of the pouch. Suitable sizes for the pouch are further described herein.

[0025] “Saliva-permeable”, in the context of a pouch as described herein, relates to that said pouch is permeable in the sense that once the pouch is positioned in the mouth of a user, saliva is allowed to travel through the pouch material into the composition of the present disclosure positioned inside the pouch so as to provide further moisture to the composition but also to ensure that substances present in the composition can travel through the pouch material and into the mouth of the user. This is particularly important so that the active substance can reach the oral cavity of the user and provide the effect.

[0026] An “active substance” according to the present disclosure is a substance that exerts an effect on the body when taken up by the body. The effect may for example be an invigorating and / or a stimulating effect. Non-limiting examples of an active substances as referred to herein comprises a composition comprising an extract of Dracocephalum moldavica L.

[0027] A “carrier material” according to the present disclosure, is a material that carries the active substances and / or other ingredients and allows or facilitates release thereof when positioned in the mouth of a user. An example of such a carrier material is a plant-based fiber.

[0030] DETAILED DESCRIPTION OF THE DISCLOSURE

[0031] The present disclosure provides oral moist non-tobacco products which may be used to experience positive effects on the body and that also have an appealable taste and mouth feel. The oral moist non-tobacco products of the present disclosure are smokeless.

[0032] According to the present disclosure, active substances are administered to the oral mucus membrane in the form of saliva-permeable pouches, comprising a moist non-tobacco composition, that are placed in the oral cavity, preferably under the lip, and in contact with the oral mucous membrane. This surprisingly results in a rapid uptake of the active substances presented herein and therefore a more rapid on-set of the positive effects of the active substances. The active substances may therefore be directly taken up by the oral mucosa when delivered via the saliva-permeable pouches comprising a moist non-tobacco composition of the present disclosure. The user of a product of the present disclosure therefore experiences the positive effects of the active substances quicker than if the active substances, e.g., would be delivered by eating or drinking a product containing them whereby the active substances would have to be absorbed in the gastrointestinal tract.

[0033] More particularly, the present disclosure relates to a saliva-permeable pouch comprising a moist non-tobacco composition for oral use, said moist non-tobacco composition comprising: i) a carrier material comprising a plant-based fiber, such as microcrystalline cellulose (MCC); and ii) a composition comprising an extract of Dracocephalum moldavica L.

[0034] The saliva-permeable pouch comprises an extract of Dracocephalum moldavica L. that may provide the user with improvements in skin moisture, elasticity and / or density. In addition, it may also provide the user with improvements in skin density, skin elasticity, skin firmness and / or reduction of wrinkles. Hence a saliva-permeable pouch may be particularly useful for cosmetic purposes on the skin in this regard.

[0035] Accordingly, a moist non-tobacco composition of the present disclosure comprises an extract of Dracocephalum moldavica L. (Moldavian dragonhead). Dracocephalum moldavica L. is known for its high content in flavonoid glucuronides and its antioxidative effects. Tilianin, luteolin-7-glucuronide, and apigenin-7-glucuronide have been discussed as the major actives of the herb. Moldavian dragonhead is a perennial plant native to Central Asia and has been introduced and naturalized to Eurasia and Northern America.

[0036] Surprisingly, the inventor found that it was possible to deliver an extract of Dracocephalum moldavica L through the oral mucosa of the oral cavity via a non-tobacco composition presented in an oral saliva-permeable pouch. The texture of the composition was suitable for the purpose, and the extract was shown to work positively and to exert its properties in this context. To the inventor’s best knowledge, this is the first time a composition comprising ingredients with beneficial properties on the skin and as further set out herein has been delivered to a user through a saliva-permeable pouch comprising a moist non-tobacco composition. Previous compositions comprising this extract are e.g., available as food supplements. It is envisaged that regular use of the saliva-permeable pouch according to the present disclosure may present an alternative to food supplements containing said extract and that it may also constitute an improved route of administration, and thereby effect, of the extract to the user when this is exerted through the oral cavity. Regular users of oral pouched non-tobacco products comprising active substances (such as caffeine) may also use the product of the present disclosure and thereby, in addition to the stimulating effects of caffeine or other active substances, obtain benefits to the skin. An extract useful in the context of the present disclosure may be prepared as disclosed in Wandrey F et al. Journal of Community medicine and Public Health Reports, Volume 2 Issue 7, ISSN: 2962-9899, 06 August 2021 , Materials and methods section. The water-soluble extract of Dracocephalum moldavica L prepared by this method may be spray-granulated onto isomalt and used further. It is also envisaged that alternative preparations may be used that would result in the same properties as the herein described extract.

[0037] Additional processes and process optimizations for producing an extract of Dracocephalum moldavica L can be found in Edris Rahmati et al. 2020 (Process optimization of spray-dried Moldavian balm (Dracocephalum moldavica L.) extract powder, Food Sci Nutr. 2020 Dec; 8(12): 6580-6591.), incorporated by reference herein.

[0038] An extract of Dracocephalum moldavica L. may be prepared by extracting aerial parts of Dracocephalum moldavica L with about 20% to 40% (w / w), such as about 30 % ethanol (w / w) by percolation. The final extracted dry mass may be about 1% to 3%, such as about 1.5 %. More specifically, it may be prepared as follows (and as also described in Wandrey F et al., Journal of Community medicine and Public Health Reports, Volume 2 Issue 7, ISSN: 2962-9899, 06 August 2021 , Materials and methods section, incorporated by reference herein). For example, about 3% Dracocephalum moldavica L aerial parts can be extracted with about 30 % ethanol (w / w) by percolation. Following the first extraction of about 3 hours at 50°C, the supernatant is stored and replaced by fresh solvent. Following further extraction for 3 hours at about 50°C, the supernatants are pooled, leading to a final extracted dry mass of 1.5%.

[0039] The described liquid extract or a powder version prepared by spray-granulation of the liquid extract on an isomalt carrier may be used for the moist non-tobacco composition, more particularly the composition of ii) of the moist non-tobacco composition, of the present disclosure (DracoBelle™ Nu, provided by Mibelle Group Biochemistry, Buchs, Switzerland).

[0040] The amount of said composition of ii) of said moist non-tobacco composition as disclosed herein may be from about 25 mg to about 55 mg per g of moist non-tobacco composition, such as from about 35 mg to about 45 mg, such as about 40, 41, 42, 43, 44, 45, 46, 47, 48, 49 or 50 mg per g of moist non-tobacco composition. The composition of ii) may be provided as a dry carrier composition, such as a dry isomalt composition. In other words, said composition of ii) may comprise a carrier for the extract, such as an isomalt carrier, and wherein the extract is provided as a dry extract on said carrier. Such a carrier composition, such as an isomalt composition, may contain from about 5 wt% to about 15 wt% of dried extract of Dracocephalum moldavica L. If the composition of ii) is provided as a pure dried extract of Dracocephalum moldavica L the composition of ii) may be provided in an amount of from about 1 mg to about 9 mg per g of moist non-tobacco composition, such as from about 2 mg to about 6 mg, such as about 2 mg, 3 mg, 4 mg, 5 mg or 6 mg per g of moist non-tobacco composition. If the composition of ii) is provided as a pure dried carrier composition, additional carrier material of i) may be added to the moist non-tobacco composition and optionally some water or as deemed suitable to provide an appropriate texture to the moist non-tobacco composition. The total flavonoid glucuronide content of a composition of ii) may be > 150 pg / g based on the total weight of the composition of ii).

[0041] A moist non-tobacco composition of the present disclosure comprises a carrier material comprising a plant-based fiber. One example of a plant-based fiber as referred to herein is a cellulosic fiber material, or a derivative thereof. Cellulose fibers are fibers made with ethers or esters of cellulose. These can e.g., be obtained from the bark, wood or leaves of plants, or from other plant-based material.

[0042] The cellulosic fiber material may also contain hemicellulose and lignin, with different percentages of these components altering the mechanical properties of the fibers. Examples of suitable cellulosic materials, or derivatives thereof, are carboxymethyl cellulose (CMC) and microcrystalline cellulose (MCC). The plant-based fiber is however not intended to be limited thereto. Sometimes, alternatives to plant-based fibers may also be used in the context of the present disclosure, wherein such alternatives may provide the same functionality.

[0060] A saliva-permeable pouch comprising a moist non-tobacco composition of the present disclosure may be prepared in the following exemplary manner. The ingredients of the moist non-tobacco composition are mixed to a homogenous blend followed by addition of liquid (such as water). By infusing the carrier material, the plant-based fiber, with all the ingredients in a specialized blender, all components of the composition are merged into one aggregate moist non-tobacco composition.

[0088] Other ingredients of the non-tobacco composition of Table 1 can easily be obtained from suitable manufacturers and are provided as dry ingredients except for the aromas which are provided as aqueous solutions. Propylene glycol is added as a moisturizer.

Claims

CLAIMS1. A saliva-permeable pouch comprising a moist non-tobacco composition for oral use, said moist non-tobacco composition comprising: i) a carrier material comprising a plant-based fiber, such as microcrystalline cellulose (MCC); and ii) a composition comprising an extract of Dracocephalum moldavica L.

2. The saliva-permeable pouch of claim 1, wherein the amount of said composition of ii) is from about 25 mg to about 55 mg per g of moist non-tobacco composition, such as from about 35 mg to about 45 mg, such as about 40, 41 , 42, 43, 44, 45, 46, 47, 48, 49 or 50 mg per g of moist non-tobacco composition.

3. The saliva-permeable pouch of claim 1 or 2, wherein said composition of ii) comprises a carrier for the extract, such as an isomalt carrier, and wherein the extract is provided as a dry extract on said carrier.

4. The saliva-permeable pouch of any one of claims 1 to 3, wherein the amount of carrier material of i) is from about 400 mg to about 600 mg per g of moist non-tobacco composition, such as from about 400 mg to about 500 mg per g of moist non-tobacco composition, such as from about 450 mg to about 550 mg per g of moist non-tobacco composition, such as about 400 mg, 425 mg, 450 mg, 475 mg, 500 mg, 525 mg or 550 mg per g of moist non-tobacco composition.

5. The saliva-permeable pouch of any one of claims 1 to 4, wherein said moist non- tobacco composition further comprises one or more of the following ingredients: iii) caffeine, iv) vitamin B5, and v) vitamin B12.

The saliva-permeable pouch of claim 5, wherein the moist non-tobacco composition comprises caffeine in an amount of from about 20 mg to about 200 mg per g of moist non-tobacco composition, such as from about 20 mg to about 150 mg, from about 20 to about 100 mg, such as about 20, 30, 40, 50, 60, 70, 80, 90, 100, 150, 175, or 200 mg per g of moist non-tobacco composition.

7. The saliva-permeable pouch of any one of claims 1 to 6, wherein said moist non- tobacco composition further comprises one or more of water, sweetener(s), salt, moisturiser(s) and aroma(s).

8. The saliva-permeable pouch of any one of claims 1 to 7, wherein the total weight of said moist non-tobacco composition in said saliva-permeable pouch is from about 0.3 g to about 1.5 g, such as about 0.6 g.

9. Use of a composition comprising an extract of Dracocephalum moldavica L in a moist non-tobacco composition.

10. The use of claim 9, wherein said moist non-tobacco composition is comprised in a saliva-permeable pouch for positioning in the mouth, such as under the lip and / or for chewing by the user.

11. Non-medical use of a moist non-tobacco composition comprising an extract of Dracocephalum moldavica L for improving skin moisture, tone and / or elasticity, wherein said moist non-tobacco composition is comprised in a saliva-permeable pouch for positioning in the mouth, such as under the lip and / or for chewing by the user.

12. Non-medical use of a moist non-tobacco composition comprising an extract of Dracocephalum moldavica L for the prevention or improvement of premature skin aging, wherein said moist non-tobacco composition is comprised in a saliva- permeable pouch for positioning in the mouth, such as under the lip and / or for chewing by the user.

13. Non-medical use of a moist non-tobacco composition comprising an extract of Dracocephalum moldavica L for promoting collagen production in a subject, wherein said moist non-tobacco composition is comprised in a saliva-permeable pouch for positioning in the mouth, such as under the lip and / or for chewing by the user.

14. Non-medical use of any one of claims 11 to 13, wherein said saliva-permeable pouch is as defined in any one of claims 1 to 8.
