[0009] Chelating mixtures
[0010] The present invention mainly relates to mixtures with chelating properties and their application in pharmaceutical, cosmetic, personal care, animal care, homecare or industrial or institutional cleaning formulations. More specifically, the present invention relates to a mixture comprising at least two orthree chosen from the following compounds: (A) a pyrone 5 derivative, (B) a tropolone derivative, (C) a phenylpropanoid, (D) a monoterpene or monoterpenoid, (E) an aromatic acid or salt, (F) a dipeptide, and (G) an alkanediol, and its use as chelating mixture.
[0011] Chelating agents play a crucial role in various formulations, such as pharmaceuticals, cosmetics (e.g. hair care and skin care), personal care, animal care, homecare and0 industrial or institutional cleaning products, for several important reasons.
[0012] Chelating agents, like EDTA (ethylenediaminetetraacetic acid), bind to metal ions (such as calcium, magnesium, iron, and copper) that might otherwise react with ingredients in the formulation. These metal ions can catalyze oxidation reactions, leading to degradation or discoloration of the product. By sequestering these metals, chelating agents help stabilize5 the formulation and extend the shelf life of cosmetics. Metal ions in cosmetic products can promote the growth of certain microorganisms, as they can be essential nutrients for microbial activity. Chelating agents deprive microbes of these essential metals, thereby enhancing the effectiveness of preservatives and helping to maintain the product's hygiene. By binding to metal ions that could otherwise deactivate active ingredients, chelating agents help maintain the potency of key ingredients, such as antioxidants or preservatives, ensuring that the product performs as intended throughout its lifespan. Chelating agents prevent metal-induced reactions that could cause undesirable physical changes in the product, such as changes in color, texture, or viscosity. This helps maintain the cosmetic's appealing appearance and smooth application properties. In products like shampoos or cleansers, chelating agents can neutralize the impact of hard water minerals (e.g. , calcium or magnesium) that might reduce the effectiveness of surfactants, improving foaming action and rinse-off performance. They also prevent minerals from building up on the cuticle, enhance the hair’s natural shine, improve compability and reduce a rough or dull feel. When having a microfilm on the hair, penetration of color or straightening agents are hindered. Thus, the use of chelating agents is especially important when working with blonde hair, where even a slight presence of metals can lead to greenish, yellowish, or blotchy tones after lightening. In addition, chelating agents in haircare formulations help to prevent and protect the hair from damage, as especially iron and copper ions can catalyze free-radical reactions that fade color and damage proteins.
[0013] In summary, chelating agents in cosmetic formulations enhance stability, preserve product quality, and improve the overall effectiveness and appearance of the product by controlling unwanted interactions with metal ions.
[0014] However, there is no universal chelating agent that fits all needs I formulations, hence the industry continuously thrives to explore further compounds which exhibit effective chelating properties. Moreover, with the number of consumer reports on skin intolerances to cosmetics increasing, the ambition grows to reduce synthetic compounds, and in generally reduce the number of ingredients in a cosmetic product. It would thus be desirable to identify multifunctional agents providing both chelating and cosmetic effects. Desirably, the agents are at least partly natural and / or plant-based.
[0015] It is thus a main object of the present invention to provide new chelating agents as a replacement or partial replacement for existing chelating agents. An optional aim is to provide the chelating agents in the form of multifunctional compounds that combine the chelating capability with further functions that are of interest for pharmaceutical, cosmetic, personal care, animal care, homecare, and industrial or institutional cleaning formulations.
[0016] These objects are achieved by the present invention as specified in the appended claims. The invention itself, and its preferred variants, other objects and advantages, are however also apparent from the following detailed description in conjunction with the accompanying examples and figures. According to a first aspect of the present invention, a mixture comprises, or consists of, at least two or three of the following compounds:
[0017] (A) a pyrone derivative,
[0018] (B) a tropolone derivative,
[0019] (C) a phenylpropanoid,
[0020] (D) a monoterpene or monoterpenoid,
[0021] (E) an aromatic acid or salt,
[0022] (F) a dipeptide,
[0023] (G) an alkanediol.
[0024] The present invention is based on the inventors’ observation that a combination of two or three out of the aforementioned seven compounds A, B, C, D, E, F and G exhibit chelating properties. Advantageously, the effect is synergistic (as determined by the assays described herein). This means, the mixture of the invention is more effective than the sum of each respective compound alone. Therefore, the mixture is capable to function as a strong (synergistic) chelating mixture (system) alone or in conjunction with one or more further chelating agent(s) chemically distinct from the components A to G. Another advantage is that the mixtures of the invention are formed from compounds established on the cosmetic sector. They can thus be rapidly integrated in pharmaceutical, cosmetic, personal care, animal care, homecare, and industrial or institutional cleaning formulations. They not only impart cosmetically relevant properties, but have also proven to be well- tolerated for use in skin products. They are renewable or naturally occurring, and may thus be provided as mixtures free of synthetic compounds.
[0025] Any of compounds A to G is understood to respectively denote a single compound. However, this does not exclude the general possibility that mixtures of the invention may comprise one or more further compound(s) falling within the definition of one or more of compounds A to G. Moreover, any of components A to G may be provided by way of a mixture, such as a natural extract. For example, hinokitiol can be found in extracts of Hinoki cypress. Mixtures containing such extract would therefore comprise hinokitiol as compound B.
[0026] Compound A, preferably maltol, may be provided as pure substance, in diluted or dispersed form, or in the form of an extract such as a sugarcane ferment extract.
[0027] Compound B, preferably hinokitiol, may be provided as pure substance, in diluted or dispersed form, or in the form of an extract such as an extract of Hinoki cypress.
[0028] Compound C, preferably cinnamal, may be provided as pure substance, in diluted or dispersed form, or in the form of an extract such as cinnamomum cassia bark oil. Compound D may be provided as pure substance, in diluted or dispersed form, or in the form of an extract such as eucalyptus dives oil in the case of piperitone, or Litsea Cubeba oil or Lemongras oil cochin in the case of citral.
[0029] Compound E, an aromatic acid or salt, preferably anisic acid or a salt thereof, may be provided as pure substance, in diluted or dispersed form, or in the form of extract such as anis oil.
[0030] Compound F, a dipeptide, preferably carnosine, may be provided as pure substance, in diluted or dispersed form, or in the form of an extract. Although carnosine is a natural substance, it is primarily produced by synthesis.
[0031] Compound G, an alkanediol, preferably a 1 ,2-alkanediol or a 1 ,3-alkanediol, may be provided as pure substance, in diluted or dispersed form.
[0032] Moreover, the compounds chosen from A to G may be provided in the form of a (homogeneous) blend, a (heterogeneous) mixture, or as a combination in separate containers. The compounds may also be provided as a solution, for instance the compounds may be dissolved in a common solvent. A combination in separate containers is also referred to as a kit-type combination provided in separate containers. This means, compound A may be contained in a first container, compound B may be contained in a second container and compound C may be contained in a third container, etc.
[0033] Although the invention is not limited in this regard, it is preferred that all compounds in the mixtures of the invention occur in nature or are derived from renewable sources. This does not mean that the compounds are actually isolated from the nature, but they may also be synthetically derived as long as they occur naturally as such. Moreover, some compounds, for example, compounds A, B, C, D, E and G, may be of natural origin, whereas other compounds, for example compound F, the dipeptide, may be obtained by chemical synthesis.
[0034] In some embodiments, the mixture of the invention is a bio-mixture. The term “bio-mixture” means that some or all of components A to G are from bio-based sources. For example, the components A to G may be obtained from bioderived feedstock by fermentation, or from bio-based natural and renewable feedstock - such as biomass by catalytic synthesis as described in US 2019 / 0241491 A1 and US 2020 / 0189995 A1 for the component G. However, the present invention is not limited to a bio-mixture. It is rather envisaged that one, two, three or more, or even all of components A to G are derived from petrochemical or other fossil fuel sources by chemical synthesis. A mixture of bio-based and petrochemical components is also possible. It is also possible that all compounds included in the mixture occur naturally, whether the compounds of the mixture are actually of natural or synthetic origin.
[0035] The term “comprising” means that the named recitations are essential, and that additional recitations may be included. The term “consisting of’ signifies that the subject matter is closed-ended and only includes the recitations that are expressly recited. Whenever reference is made to “comprising” it is intended to cover both meanings as alternative embodiments. That is, the meaning can be either “comprising” or “consisting of’, unless the context dictates otherwise.
[0036] Compound A, a pyrone derivative, is an organic compound that is chemically based on, preferably includes, a pyrone structure, a six-membered ring containing both oxygen and carbon atoms, along with a carbonyl group within the ring. Preferred pyrone derivatives include a-Pyrone (2-pyrone) and y-Pyrone (4-pyrone). a-Pyrones are also known as 2H- pyran-2-one, and have the carbonyl group at the 2-position relative to the oxygen atom in the ring. y-Pyrones have the carbonyl group at the 4-position, making this structure 4H- pyran-4-one. Preferred examples of pyrone derivatives include coumarin, chromones, kojic acid (5-hydroxy-2-(hydroxymethyl)-4H-pyran-4-one), flavonoids, maltol, xanthones, 6- pentyl-2H-pyran-2-one, kojic acid derivatives (e.g., kojic dipalmitate), 2-ethyl-3 -hydroxy- 4H-pyran-4-one, 3-hydroxy-2-propylpyran-4-one, 3-hydroxy-2-propan-2-ylpyran-4-one, 2- acetyl-3-hydroxypyran-4-one, 2-ethyl-5-hydroxypyran-4-one, 3-hydroxy-2-(2-propen-l-y1 )- 4H-Pyran-4-one, 3-hydroxypyran-4-one, 2-hex-2-eny1-3-hydroxypyran-4-one, 3,5- dihydroxy-2-methy1 -4H-pyran-4-one (5-hydroxymaltol), 2-methy1 -4-oxo-4H-pyran-3-y1 isobutyrate (maltol isobutyrate), 2-methy1-3-(1-oxopropoxy)-4H-pyran-4-one (maltol propionate), 2-methy1 -4-oxo-4H-pyran-3-y1 butyrate (maltol butyrate), (2-methy1-4- oxopyran-3-y1) acetate, (2-ethy1-4-oxopyran-3 -y1) 2-methylpropanoate, 2-methy1-4-oxo- 4H-pyran-3-y1 2-hydroxybenzoate (salicylic acid maltol ester), 2-methy1-4-oxo-4H-pyran- 3-y1 lactate (maltol lactate), 2-methy1-4-oxo-4H-pyran-3-y1 , (Z)-(2-methy1-4-oxopyran-3- y1)-2-methylpent-2-enoate, (E)-(2-methy1-4-oxopyran-3-y1)-2-methylpent-2-enoate, and combinations thereof.
[0037] Preferably, the pyrone derivative is a 3-hydroxy-4-pyrone. It consists of a pyrone ring with a hydroxyl group attached at the 3rd position and a ketone group at the 4th position. Examples of 3-hydroxy-4-pyrones are listed above and include maltol and kojic acid, among others. Compound B, a tropolone derivative, is a compound derived from tropolone, a sevenmembered, aromatic, non-benzenoid ring structure with alternating double bonds and a hydroxyl group. Tropolone derivatives are compounds in which the basic tropolone structure is modified by substituting or attaching different functional groups to the sevenmembered ring. Examples of tropolone derivatives include hinokitiol (p-Thujaplicin), thujaplicin, eucarvone and colchicine. Preferred tropolone derivatives are tropolone and hinokitiol.
[0038] Compound C, a phenylpropanoid, is an organic compound derived from phenylalanine, characterized by a phenyl ring connected to a three-carbon chain. Examples of phenylpropanoids include cinnamic aldehyde (cinnamal), cinnamyl alcohol, hydroxycinnamaldehyde, anethol, dilapiol, allyl anisole, isoeugenol, eugenol, anethole and flavonoids.
[0039] Compound D, a monoterpene or monoterpenoid, is predominantly found in essential oils. Monoterpenes are compounds consisting of two isoprene units. Examples of (natural) monoterpenes include p-menthan, safranal, iridodial, fenchan, limonene, carvone, pinene, piperitone, citral, or one of the two isomers geranial and neral, and ocimene. Preferably, the term monoterpenes encompasses aromatic, alicycic or aliphatic structures, comprising two isoprene units. More preferably, monoterpenes are understood as aromatic or alicyclic structures, comprising two isoprene units. Monoterpenoids are a class of organic compounds derived from monoterpenes. This includes the products of (bio)chemical reactions between a monoterpene and another scaffold such as a carbohydrate as well as monoterpenes that contain additional functional groups after being (bio)chemically modified (e.g., by addition of new functional groups such as hydroxyl groups, ketones, aldehydes et cetera). Preferably the term monoterpenoid is used to describe the group of heteroatomcontaining molecules, which carbon structure comprises two fused isoprene units. More preferably, monoterpenoid is understood in this context as the group of alcohols, ketones, aldehydes or carboxylic acids of monoterpenes. Most preferably the term monoterpenoid describes the alcohols, ketones, aldehydes or carboxylic acids of aromatic or alicyclic monoterpenes. Examples of monoterpenoids include thymol, carvacrol, geraniol, menthol, linalool and eucalyptol (1 ,8-cineole), citronellol, (+) / (-)menthone, campher, camphene, grapefruit mercaptane, myrcene, perillyl alcohol.
[0040] Should there be an overlap between the class of tropolone derivatives and the class of monoterpenes or monoterpenoids, the overlapping part is to be allocated to the group of tropolone derivatives. In other words, compound D, a monoterpene or monoterpenoid, as understood herein is chemically a monoterpene or monoterpenoid other than a tropolone derivative.
[0041] Compound E, an aromatic acid or salt thereof, is characterized by an aromatic system when hydrogen atoms are substituted by carboxyl groups. Dependent on the number of carboxylgroups mono-, di-, and tri-substituted aromatic acids are distinguished. Examples include benzoic acid, anisic acid, phenylacetic acid, terephthalic acid, salicylic acid, p- aminobenzoic acid and their salts. Mono substituted aromatic acid (monobasic aromatic acid) and salts are preferred. Most preferred is anisic acid and its salts.
[0042] Compound F, a dipeptide, is a molecule consisting of two amino acids linked together by a peptide bond. Several dipeptides exist in nature such as Carnosine (p-alanyl-L-histidine), Anserine (p-alanyl-1-methyl-L-histidine), Kyotorphin (tyrosine-arginine), Aspartame (aspartyl-phenylalanine methyl ester), Balenine (p-alanyl-L-methylhistidine) and L-alanyl- L-glutamine.
[0043] Compound G, an alkanediol, is a group of substances consisting of linear or branched hydrocarbon chains containing exactly two hydroxy groups at different positions. The term “alkanediol” within the context of the present invention also includes its constitutional isomers or position isomers. Constitutional isomers are compounds that have the same molecular formula and different connectivity. Position isomers, a particular form of constitutional isomerism, are structural isomers that can be viewed as differing only on the position of a functional group on a parent structure, which in this case is the position of the two alcohol functions.

[0084] The synergistic chelating activities of the compounds contained in the mixtures of the invention have been confirmed by determining binding (complex formation) capability of cupric and / or ferrous ions of the mixtures as compared to the respective binding capability of the respective pure compounds. The binding capability was determined indirectly by measuring the amount of cupric and / or ferrous ions bound to a reference compound (Pyrocatechol Violet and / or Ferrozine) using absorbance measurement, whereby binding to the reference compound is assumed to be only possible if the binding capability of the compounds / mixture of the invention is exhausted. This gave a quantitative indication of the synergistic effect. The synergism is particularly remarkable for combinations of two orthree compounds chosen from maltol, cinnamal, hinokitiol, carnosine and a 1 ,2-alkanediol or a 1 ,3-alkanediol as specified herein. Therefore, particularly preferred mixtures comprise, or consist of, the following combinations of compounds:

[0379] Another aspect of the present invention relates to a method of conveying chelating properties to a formulation, or increasing chelating properties of a formulation comprising:
[0380] (i) Providing a combination of compounds as defined herein, or providing the individual compounds contained therein,

[0382] (iii) mixing the combination of compounds of (i), or the individual compounds contained therein, with the compounds of (ii) to provide the formulation with chelating properties or the formulation with increased chelating properties.
[0383] Another aspect of the present invention relates to the use of a combination of compounds as defined herein, or of the individual compounds contained therein, as chelators in a pharmaceutical, cosmetic, personal care, animal care, homecare or industrial or institutional cleaning formulation.
[0384] On the basis of the observation that the combination of compounds included in the mixture of the invention provides a synergistic efficacy with respect to the chelating effect, it shall be understood that, generally, features and embodiments described in conjunction with a specific aspect of the invention shall form corresponding features and embodiments of all other aspects, unless the context dictates otherwise.
[0385] In the following, the invention is explained in more detail with reference to representative examples and the drawings, which show:

[0390] A. Chelating properties of different ingredients and combinations thereof
[0391] The below listed ingredients were evaluated fortheir chelating properties in relation to the most common ions in formulation (Fe and Cu). Normally EDTA or other classic chelators are used in formulations.
[0392] 1 . Ingredients tested
[0393] 2. Test design
[0394] Sample preparation was as follows. Stock solutions were prepared. The pH value of the solutions was adjusted to 6. Final concentrations were adjusted via dilution factor. Assays for cupric ions and ferrous ions were performed by using an Assay Kit ex Zen-Bio. In both cases EDTA served as a positive control capable of chelation the respective ions in a dose dependent manner. Both were endpoint assays measuring absorbance of the formed complex. The activity was determined as the percent of total Pyrocatechol Violet (Pycrocatecholsulfonphtalein, PV) (Cu) or 3-(2-Pyridyl)-5,6-diphenyl-1 ,2,4-triazine-p,p'- disulfonic acid (Ferrozine) (Fe) I ion binding.
[0395] In the cupric ion assay, PV is able to form a complex with free cupric ions resulting in a chromophore with strong absorbance at 632 nm. Compounds that are able to chelate Cu(ll) lowerthe amount of free Cu(ll) in solution, decreasing the PV-Cu(ll) complex concentration, which results in a loss of absorbance at 632 nm. The ferrous ion assay is based on the ability of Ferrozine to form a complex with free ferrous ions resulting in a chromophore with strong absorbance at 562 nm. Compounds that are able to chelate Fe(ll) lower the amount of free Fe(ll) in solution, decreasing the Ferrozine- Fe(ll) complex concentration, which results in a loss of absorbance at 562 nm.
[0396] The results thereof are shown in the following:
[0397] 3. Results
[0398] 3.1 Ferrous ion assay a) Synergistic effect for Pentylene Glycol + Maltol + Carnosine
[0399] For the combination pentylene glycol, maltol and carnosine a synergistic effect on the chelating properties has been found, which can be easily seen from Fig. 1. Note that in order to better see a synergism, the amount of pentylene glycol, maltol and carnosine in the mixture was chosen to be half of the concentration of the respective pure substance solution. Despite lowering the concentrations, the chelating activity of the mixture was higher than the sum of each compound (in double concentration) alone. Therefore, the synergistic effect would be more remarkable if equal concentrations had been compared. b) Effect for Maltol + Cinnamal + Carnosine
[0400] The mixture of maltol, carnosine and cinnamal was found to exhibit 94.6% chelating efficiency. Summing the respective chelating efficiency determined for each respective (pure) compound up gives 85,5%. Thus, the chealting effect of the mixture is synergistic. This can also be derived from Fig. 2. Maltol, cinnamal and carnosine act together in a synergistic manner in relation to the chelating properties. c) Effect for Maltol + Cinnamal + Pentylene Glycol
[0401] The results are illustrated in Fig. 3 and the table below.
[0402] Once again, a strong synergism could be observed for the combination pentylene glycol, maltol and cinnamal. Note that the mixture contains half of the pentylene glycol included in the respective single substance solution. d) Effect for Maltol + Hinokitiol + Pentylene Glycol e) Chelating effect of anisic acid
[0403] Tested at two different concentrations (A = 50 ppm, B = 87.5 ppm), anisic acid alone was found to have strong and concentration-dependent chelating properties (cf. Fig. 4). f) Effect for Maltol + Sodium Gluconate / + / - 1,2-Hexanediol
[0404] Surprisingly, a combination of maltol and sodium gluconate leads to higher chelating properties which can even be enhanced by the addition of 1 ,2-hexanediol.
[0405] 3.2 Cupric ion assay a) Effect for Cinnamal + Hinokitiol (50 : 50% DPG : Water) b) Effect for Hinokitiol + Piperitone
[0406] Surprisingly, a combination of piperitone and hinokitiol leads to higher chelating properties compared to the single ingredients. c) Effect for Hinokitiol + Anisic Acid
[0407] Surprisingly, a combination of anisic acid and hinokitiol leads to higher chelating properties compared to the single ingredients. d) Effect for Maltol + Piperitone
[0408] Surprisingly, a combination of piperitone and maltol leads to higher chelating properties compared to the single ingredients. e) Effect for Maltol + Anisic Acid
[0409] Surprisingly, a combination of anisic acid and maltol leads to higher chelating properties compared to the single ingredients. f) Effect for Maltol + Anisic Acid + Trisodium Ethylenediamine Disuccinate
[0410] Surprisingly, a combination of maltol, anisic acid and trisodium ethylenediamine disuccinate leads to higher chelating properties compared to the single ingredients.
[0411] 4. Summary
[0412] In summary, synergistic effects have been observed for the following combinations:
[0413] - Maltol, Carnosine, Pentylene Glycol;
