Abstract
The present invention relates to the field of sustained drug release technology, and particularly, to a method for preparing a BMP-2 controllable sustained-release floc based on self-regulation of lysozyme, and use thereof. The method of the present invention comprises: preparing a chitosan solution; sequentially adding a protein stabilizer and glycerol to the chitosan solution acquired in step A1 to prepare a chitosan composite solution; acquiring a non-specific binding solution; preparing a BMP-2 / chitosan solid particle by means of lyophilization; and preparing a PLGA / PEG composite solution to give a suspension, and performing post-treatment on the suspension to form the BMP-2 controllable sustained-release floc. The carrier in the BMP-2 controllable sustained-release floc prepared by the preparation method can be specifically degraded by the lysozyme in the human body, such that the release amount and release speed of BMP-2 can be dynamically adjusted according to the progress of bone healing, thereby achieving long-term sustained release in the BMP-2 controllable sustained-release floc. The present invention solves the problems of insufficient sustained-release effects of BMP-2 in the prior art, the sudden release of the factor, and the uncontrollable sustained-release duration and dose due to the use of hydrophilic materials.
