Background Art
[0002] Tumor Treating Fields (TTF) is a physical therapy based on bioelectricity. [0025] In the present disclosure, unless otherwise specified, the use of terms such as "first" and "second" to describe various elements does not intend to limit the positional relationship, timing relationship, or importance relationship of these elements. Such terms are only used to distinguish one element from another. In some examples, the first element and the second element may refer to the same instance of the element, and in certain cases, based on the context description, they may also refer to different instances.
[0026] In the description of the various examples in this disclosure, the terms used are for the purpose of describing specific examples only and are not intended to be limiting. Unless the context clearly indicates otherwise, if the number of elements is not specifically defined, the element can be one or more. As used herein, the term "plurality" means two or more, and the term "based on" should be interpreted as "at least partially based on". In addition, the terms "and / or" and "at least one of..." cover any one of the listed items and all possible combinations thereof.
[0027] For tumor electrotherapy, the greater the electric field intensity that can be provided for the target tumor, the better the effect of the electric field interfering with the mitosis of tumor cells. In specific implementation, the magnitude of the provided electric field intensity depends to a large extent on the setting position of the pair of electrode patches used to apply the electric field. For example, whether the electrode patches can accurately cover the tissue area where the lesion is located, or the relative position setting method between the two electrode patches, etc.
[0028] The corresponding electric field intensity values can be compared by setting the electrode patches at different body parts of the animal experimental subject to determine the position of the electrode patch with the highest electric field intensity value. However, due to the movement of the animal experimental subject itself and the influence of surrounding environmental factors, the accuracy of the measured electric field intensity value is not high. Moreover, this method requires moving the electrode patches multiple times and performing measurements, which will consume a large amount of manpower and material resources.
[0029] Based on this, the present disclosure proposes a computer simulation method for determining the position of electrode patches. By performing three-dimensional reconstruction on a mouse with a brain tumor, a corresponding mouse simulation model is obtained, and simulation electrode patch pairs applied with simulated alternating current signals are sequentially set at multiple preset positions in the head region of the mouse simulation model, so as to be able to obtain multiple electric field intensity values corresponding to different preset positions as research data, and to determine the target electrode patch setting position that can provide the most effective treatment according to the magnitude of the electric field intensity value and determine the electrode sheet corresponding to the target electrode patch position according to the target electrode patch position. Thus, real objective interference factors are excluded, and accurate electric field intensity measurement data is provided. In addition, through the computer simulation method, a large number of repeated experiments can be carried out in a short time, which can effectively improve efficiency and save resources.
[0030] The computer simulation method for determining the position of electrode patches according to the present disclosure will be further described below with reference to the accompanying drawings.
[0031] Figure 1 is a flowchart of a computer simulation method for determining the position of electrode patches according to an exemplary embodiment of the present disclosure.
[0032] As Figure 1 shown, the computer simulation method for determining the position of electrode patches includes:
[0033] Step S101: Obtain a mouse simulation model, which is obtained by three-dimensional reconstruction of a mouse with a brain tumor.
[0034] Step S102: Sequentially set simulation electrode patch pairs at multiple pairs of candidate positions within the head region of the mouse simulation model, and apply a simulation alternating current signal to the simulation electrode patch pairs.
[0035] Step S103: For each pair of candidate positions among the multiple pairs of candidate positions, record the electric field intensity value of the simulation electric field established between the simulation electrode patch pairs when the simulation electrode patch pairs are set at this pair of candidate positions.
[0036] Step S104: Select a pair of candidate positions with the maximum electric field intensity value from the multiple pairs of candidate positions as the target electrode patch position.
[0037] Step S105: Determine the electrode patch corresponding to the target electrode patch position according to the target electrode patch position.
[0038] In the above method, since the mouse simulation model constructed by three-dimensional reconstruction can be reused, the accurate target electrode patch position can be determined according to factors such as the size, shape, and brain tumor location of the mouse simulation model. In addition, since no actual animal experiments are required, the process can be simplified and the efficiency can be improved.
[0039] According to some embodiments, each pair of candidate positions includes a first position and a second position, and the first position and the second position are spaced apart from each other. Specifically, by setting the first position and the second position at intervals, it is possible to avoid a large amount of alternating current concentrating on a certain part of the mouse simulation model due to overlapping electrode patch settings, changing the corresponding electric field distribution, and thus affecting the magnitude of the finally obtained electric field intensity value. Moreover, overlapping of electrode patches may also cause the temperature at the overlapping part to be too high, resulting in the situation of burning the mouse in reality and reducing the effectiveness of the simulation test results.
[0040] Figure 2 It is a schematic diagram showing the setting positions of the simulation electrode patches in the head region of the mouse simulation model according to an exemplary embodiment. In Figure 2 In the illustrated example, the first position is located between the left ear and the left eye of the mouse simulation model, and the second position is located between the right ear and the right eye of the mouse simulation model. The electrode patches 111 and 112 are respectively attached to the first position and the second position.
[0041] This example targets a mouse simulation model with a brain tumor (e.g., glioma). Therefore, by setting the first position and the second position between the ears and eyes on the left and right sides of the mouse's head, respectively, the electrode patches can be located on both sides of the brain tumor. At this time, the electric field generated is likely to act on the brain tumor location of the mouse simulation model, effectively reducing the number of candidate locations that need to be tested and improving the efficiency of the simulation test.
[0042] Moreover, in actual implementation, this setting method can also improve the comfort and feeding problems of the mouse when the electrode patch is applied to the mouse's head, further improving the effectiveness of the final target electrode patch position.
[0043] According to some embodiments, the simulated alternating electrical signal includes a first alternating electrical signal and a second alternating electrical signal with opposite polarities, and the first alternating electrical signal and the second alternating electrical signal are respectively applied to two simulated electrode patches in the simulated electrode patch pair.
[0044] Specifically, by setting two simulated alternating electric signals with opposite polarities to be applied to the simulated electrode patches, an electric field can be formed between a pair of candidate positions.
[0045] In one example, the first alternating electrical signal and the second alternating electrical signal satisfy at least one of the following conditions: a frequency between 150 kHz and 300 kHz, an intensity greater than or equal to 20 Vpp, and a duration greater than or equal to 72 hours.
[0046] The above conditions are the setting ranges suitable for mouse animal experiments summarized through a large number of experiments. Based on this, the amount of calculation for simulation testing can be further reduced, thereby saving resources and improving efficiency.
[0047] Figure 3 is a partial flow chart illustrating a computer simulation method for determining electrode patch positions according to an exemplary embodiment. Figure 3 As shown, according to some embodiments, before step S101, the above-mentioned computer simulation method also includes: step S1001, using computer tomography technology to image mice with brain tumors to obtain three-dimensional imaging information; step S1002, constructing a mouse simulation model based on the three-dimensional imaging information.
[0048] In one example, a simulation electrode patch can be constructed by COMSOL Multiphysics (a multi-physics simulation software), and a simulated alternating electric signal applied thereto can be set. At the same time, in order to adapt to COMSOL Multiphysics, a mouse can be three-dimensionally imaged based on computed tomography techniques (such as CT, OCT, etc.), and the obtained three-dimensional imaging information can be imported into COMSOL Multiphysics to construct a corresponding mouse simulation model, so as to implement the computer simulation method for determining the position of the electrode patch of the present disclosure.
[0049] It should be noted that the above COMSOL Multiphysics simulation software is only for illustrative purposes and is not limited thereto. In the specific implementation process, other adaptable means and simulation software can also be used to perform three-dimensional imaging and corresponding electric field simulation tests on mice with brain tumors.

[0063] Referring to Figures 6 to 10 , the specific structure of a tumor electric field therapy system including at least a pair of electrode patches manufactured according to the above method of the present invention will now be described. It should be understood that the target electrode positions determined by the computer simulation method of the present disclosure can be applied to the electrode patches of the tumor electric field therapy system. The electrode patches are manufactured after being specially designed according to the target electrode patch application positions determined by the method of the present invention.
[0064] Figure 6 A schematic structural diagram of a tumor electric field therapy system 200 is shown. As Figure 6 shown, the tumor electric field therapy system 200 includes an electric field generator 201, an adapter 202 electrically connected to the electric field generator 201, an animal experiment adapter board 203 electrically connected to the adapter 202, and at least one group of animal head electrodes 204 electrically connected to the animal experiment adapter board 203.
[0065] The electric field generator 201 generates at least one set of alternating electric signals with opposite polarities. In one example, the electric field generator 201 generates a set of alternating electric signals X1 and X2 with opposite polarities. The frequency range of the above-mentioned alternating electric signals X1 and X2 is between 150 kHz and 300 kHz, the intensity of the alternating electric signal is not less than 20 Vpp, and the application time of the alternating electric signal is not less than 72 h.
[0066] Reference Figure 7A and Figure 7B , the adapter 202 is used to transmit the alternating electric signals generated by the electric field generator 201 to the animal experiment adapter board 203. The animal experiment adapter board 203 is used to output the alternating electric signals transmitted by the adapter 202 and generated by the electric field generator 201 in multiple parallel groups, so that control experiments can be carried out synchronously to improve the accuracy and effectiveness of the basic research data and conclusions of animal experiments. The animal experiment adapter board 203 has at least one adapter board body 210, at least a pair of first connectors 220 provided on opposite sides of the adapter board body 210, multiple groups of second connectors 230, and a bracket 240 for suspending the adapter board body 210. Both the first connector 220 and the second connector 230 are electrically connected to the adapter board body 210. Each first connector 220 of the animal experiment adapter board 203 is electrically connected to a corresponding alternating electric signal transmitted by the adapter 202 through a corresponding cable (not shown). Different first connectors 220 transmit different alternating electric signals. Each second connector 230 is connected to all the first connectors 220 through the adapter board body 210. Each second connector 230 of the animal experiment adapter board 203 is electrically connected to a corresponding animal head electrode 204 respectively, so as to transmit the alternating electric signals in parallel to the corresponding animal head electrode 204 to act on the head of a mouse for animal experiments on tumor electric field therapy.
[0067] In one example, the animal experiment adapter board 203 has a pair of first connectors 220, where one first connector 220 transmits the alternating electric signal X1 and the other transmits the alternating electric signal X2. The animal experiment adapter board 203 has 10 second connectors 230 connected in parallel to the alternating electric signals X1 and X2, so as to realize the parallel output of the alternating electric signals X1 and X2 in 5 paths, enabling multiple animals to conduct experiments simultaneously under the electric field with the same parameters and improving the accuracy of the experiment.
[0068] In one example, the first connector 220 is pluggably connected to a corresponding cable (not shown), and the cable (not shown) connected to the first connector 220 is pluggably connected to the adapter 202. The second connector 230 is pluggably connected to a corresponding part of the animal head electrode 204. The first connector 220 has a plurality of first sockets 221, one of the first sockets 221 is connected to an alternating electric signal, and the remaining first sockets 221 are reserved sockets, which may not conduct signals or electricity, or may be used to configure a temperature monitoring circuit. The second connector 230 has a plurality of second sockets 231, two of the second sockets 231 are used to connect a set of alternating electric signals X1 and X2, and the remaining second sockets 231 are reserved sockets, which may not conduct signals or electricity, or may be used to configure a temperature monitoring circuit.
[0069] In summary, the electric field generator 201 generates a set of alternating electric signals X1 and X2 with opposite polarities, and the alternating electric signals X1 and X2 are routed through the adapter 202, the animal experiment adapter board 203, and the corresponding animal head electrode 204. The animal head electrode 204 is affixed to the corresponding body surface of the mouse head, and a set of alternating electric signals with opposite polarities generated by the electric field generator 201 are applied to the mouse head, and an alternating electric field is formed on the mouse head for tumor electric field therapy. In other examples, the tumor electric field therapy system 200 may not be provided with the adapter 202, and the electric field generator 201 is directly electrically connected to the animal experiment adapter board 203, and a set of alternating electric signals with opposite polarities generated by the electric field generator 201 are directly transmitted to the animal experiment adapter board 203.
[0070] As Figure 8 shown, the animal head electrode 204 is used for in-vivo experiments on animal head tumors, including: paired electrode functional components 10, a cable assembly 20 electrically connected to each electrode functional component 10, and a backing 30 and an adhesive 40 respectively disposed on opposite sides of the corresponding electrode functional component 10. The adhesive 40 of the animal head electrode 204 is affixed to the corresponding body surface of the mouse head, and the corresponding electrode functional component 10 is firmly configured on the corresponding body surface of the mouse head through the backing 30. The cable assembly 20 is electrically connected to the animal experiment adapter board 203 to transmit the alternating electric signal generated by the electric field generator 201 to the electrode functional component 10 affixed to the corresponding body surface of the mouse head, so as to form an alternating electric field on the mouse head for tumor electric field therapy.
[0071] As Figure 9As shown, each electrode functional component 10 includes a flexible circuit board 11, a dielectric element 12 and a first insulating support plate 13 provided on opposite sides of the flexible circuit board 11. The flexible circuit board 11 includes a main body portion 113 and a wiring portion 114 horizontally and laterally extending from the main body portion 113. The dielectric element 12 and the first insulating support plate 13 are respectively located on opposite sides of the main body portion 113 of the flexible circuit board 11. The dielectric element 12 is electrically connected to the main body portion 113 of the flexible circuit board 11, so that the animal head electrode 204 transmits the alternating electric signal generated by the electric field generator 201 to the body surface corresponding to the mouse head through its dielectric element 12, and then an alternating electric field for tumor electric field therapy is formed on the mouse head. A circular conductive core 111 is provided on one surface of the main body portion 113 facing the dielectric element 12. The dielectric element 12 is electrically connected to the conductive core 111 of the main body portion 113 through a conductive bonding material. The conductive bonding material is a solder material or a conductive adhesive. The dielectric element 12 is made of a high dielectric material, which can block the conduction of direct current and allow the conduction of alternating current. Preferably, in this example, the dielectric element 12 is a dielectric ceramic sheet.
[0072] As Figure 10 shown, a circular metal layer 121 is attached to the middle of one surface of the dielectric element 12 facing the conductive core 111 of the main body portion 113 of the flexible circuit board 11, so as to facilitate welding the dielectric element 12 to the conductive core 111 of the main body portion 113 of the flexible circuit board 11 through a solder material. The edge of the metal layer 121 is spaced from the edge of the dielectric element 12, that is, the diameter of the metal layer 121 is smaller than the diameter of the dielectric element 12.
[0073] The first insulating support plate 13 is adhesively disposed on the surface of the main body portion 113 away from the dielectric element 12 to increase the mechanical strength of the main body portion 113 of the flexible circuit board 11. The thickness of the first insulating support plate 13 is 0.1 - 0.5 mm, so as to make the electrode functional component 10 thinner and lighter to adapt to animal experiments and facilitate the attachment of the animal head electrode 204. Preferably, the first insulating support plate 13 is made of polyimide material. The first insulating support plate 13 made of polyimide material has excellent mechanical properties and flexibility.
[0074] The dielectric element 12 and the first insulating support plate 13 are arranged in one-to-one correspondence along the thickness direction of the main body portion 113 of the flexible circuit board 11. Both the dielectric element 12 and the first insulating support plate 13 are circular sheet-like structures. The diameter of the first insulating support plate 13 is substantially the same as the diameter of the main body portion 113 of the flexible circuit board 11. The diameter of the dielectric element 12 is slightly smaller than the diameter of the main body portion 113 of the flexible circuit board 11. The range of the diameters of the first insulating support plate 13 and the main body portion 113 of the flexible circuit board 11 is 6 - 12 mm, so as to make the electrode functional component 10 thinner and lighter to adapt to animal experiments and facilitate the application of the animal head electrode 204. In this example, the diameters of the first insulating support plate 13 and the main body portion 113 of the flexible circuit board 11 are 6.7 mm. The range of the diameter of the dielectric element 12 is 4 - 10 mm. In this example, the diameter of the dielectric element 12 is 6 mm.
[0075] As Figure 11 shown, the wiring portion 114 of the flexible circuit board 11 is in a strip-like sheet structure, and the wiring portion 114 of the flexible circuit board 11 extends horizontally and laterally from the main body portion 113 of the flexible circuit board 11. The wiring portion 114 of the flexible circuit board 11 is electrically connected to the cable assembly 20. A conductive pad 112 is provided at one end of the wiring portion 114 of the flexible circuit board 11 away from the main body portion 113. The conductive pad 112 and the conductive core 111 are located on the same side surface of the flexible circuit board 11. A conductive trace 115 is connected between the conductive pad 112 and the conductive core 111. The conductive trace 115 is embedded inside the flexible circuit board 11 to electrically connect the conductive pad 112 and the conductive core 111. The conductive core 111 and the conductive pad 112 are serially connected through the conductive trace 115.
[0076] A second insulating support plate 14 is provided on the surface of the wiring portion 114 of the flexible circuit board 11 away from the conductive pad 112. The second insulating support plate 14 and the conductive pad 112 are arranged in one-to-one correspondence along the thickness direction of the wiring portion 114 of the flexible circuit board 11. The size of the second insulating support plate 14 is larger than the size of the conductive pad 112. The width of the second insulating support plate 14 is the same as the width of the wiring portion 114 of the flexible circuit board 11, and the width of the conductive pad 112 is smaller than the width of the wiring portion 114 of the flexible circuit board 11. The length of the second insulating support plate 14 is greater than the length of the conductive pad 112.
[0077] Similar to the first insulating support plate 13, the second insulating support plate 14 and the first insulating support plate 13 are located on the same side surface of the flexible circuit board 11, and the second insulating support plate 14 also increases the mechanical strength of the wiring portion 114 of the flexible circuit board 11. The material of the second insulating support plate 14 is the same as the material of the first insulating support plate 13. The thickness of the second insulating support plate 14 is the same as the thickness of the first insulating support plate 13.
[0078] The length of the wiring portion 114 of the flexible printed circuit board 11 is set according to the size of the experimental mouse, so as to better fix the animal head electrode 204 on the mouse head. The length of the wiring portion 114 of the flexible printed circuit board 11 ranges from 20 to 100 mm. The width of the wiring portion 114 of the flexible printed circuit board 11 ranges from 1 to 5 mm. In this example, the wiring portion 114 of the flexible printed circuit board 11 is 38 mm long and 2 mm wide. It should be noted that the length direction of the wiring portion 114 of the flexible printed circuit board 11 and the length direction of the second insulating support plate 14 are both consistent with the extending direction of the wiring portion 114.
[0079] As Figure 12 shown, the cable assembly 20 includes a pair of first cables 21 electrically connected to the corresponding electrode functional components 10, a conductive slip ring 23 electrically connected to the first cables 21, a pair of second cables 24 electrically connected to the conductive slip ring 23, and a pair of connectors 25 respectively electrically connected to the corresponding second cables 24. The numbers of the second cables 24 and the connectors 25 are both the same as the number of the first cables 21.
[0080] One end of the first cable 21 is welded to the conductive pad 112 provided on the flexible printed circuit board 11 of the electrode functional component 10, and the other end is electrically connected to the conductive slip ring 23. A section of heat shrinkable tube 22 is fixedly covered around the welding portion of the first cable 21 and the conductive pad 112. The heat shrinkable tube 22 plays an insulating and protective role for the welding portion of the first cable 21 and the conductive pad 112, preventing the welding portion of the first cable 21 and the conductive pad 112 from being exposed to contact with other conductive substances and causing a short circuit. The heat shrinkable tube 22 provides strength support for the welding portion of the first cable 21 and the conductive pad 112, preventing the welding portion of the first cable 21 and the conductive pad 112 from breaking. The heat shrinkable tube 22 is covered around the welding portion of the first cable 21 and the conductive pad 112, so as to prevent dust and water from entering the welding portion of the first cable 21 and the conductive pad 112.
[0081] A plurality of first cables 21 arranged in pairs are rotatably electrically connected to the corresponding second cables 24 through the conductive slip ring 23. The first cables 21 and the second cables 24 can both rotate around the central axis of the conductive slip ring 23 without causing the first cables 21 to be intertwined, which can prevent the first cables 21 from being intertwined when the animal head electrode 204 is attached to the skin surface of the mouse head and the mouse moves inside the mouse cage, thus affecting normal activities. The conductive slip ring 23 is fixedly arranged on the mouse cage (not shown).
[0082] The connector 25 is electrically connected to the animal experiment adapter board 203. The connector 25 is connected to the animal experiment adapter board 203 by plugging. Each connector 25 has a conductive terminal. Each conductive terminal is connected one-to-one with a different one of a set of signal output terminals corresponding to the animal experiment adapter board 203. The number of connectors 25 is consistent with the number of electrode functional components 10. In this example, the number of connectors 25 is 2, which are respectively used to electrically connect the electric field generator 201 to generate a different one of a set of alternating electrical signals X1 and X2 with opposite polarities.
[0083] The backing 30 is a mesh fabric. The preferred backing 30 is a non-woven fabric. One side surface of the backing 30 has a biocompatible adhesive. The electrode functional component 10 is centrally adhered to the backing 30 through a layer of biocompatible adhesive to enhance the adhesion of the corresponding part of the electrode functional component 10 to the mouse skin. The backing 30 has a main body 31 and a plurality of strips 32. The plurality of strips 32 are all extended in the direction outward from the center of the main body 31 to better wrap the electrode functional component 10 of the animal head electrode 204 to the surface of the mouse head skin. The main body 31 is roughly circular. The edge of the assembly of the first insulating support plate 13, the main body 113 of the flexible circuit board 11 and the dielectric element 12 is arranged in an interval shape with the edge of the main body 31 of the backing 30. The assembly of the first insulating support plate 13, the main body 113 of the flexible circuit board 11 and the dielectric element 12 is roughly located in the middle of the main body 31 of the backing 30. The plurality of strips 32 of the backing 30 are arranged in a staggered manner with the connection portion 114 of the flexible circuit board 11. In this example, there are four strips 32 of the backing 30. The four strips 32 of the backing 30 are arranged in a substantially "cross" shape.
[0084] The adhesive member 40 has double-sided adhesiveness. The adhesive member 40 is arranged in a circular sheet shape, and its size is slightly larger than the size of the dielectric element 12. The adhesive member 40 covers the surface of the dielectric element 12 on one side away from the flexible support plate 11. The adhesive member 40 is applied to the surface skin of the mouse's head as a pasting layer to keep the skin surface moist and relieve local pressure. Preferably, the adhesive member 40 is a conductive hydrogel, which can serve as a conductive medium.
