Description

COMPLEMENT FACTOR B INHIBITORSFIELDThe present disclosure in some aspects relates generally to compounds capable of inhibiting the complement alternative pathway and particularly to inhibition of Factor B, and methods of using such compounds for treating or preventing diseases or disorders associated with complement alternative pathway activation.BACKGROUNDThe complement system is a crucial component of the innate immunity system and comprises a group of proteins that are normally present in an inactive state. These proteins are organized in three activation pathways: the classical, the lectin, and the alternative pathways (V.M. Holers, In Clinical Immunology: Principles and Practice, ed. R.R. Rich, Mosby Press; 1996, 363-391) . Factor B may be a suitable target for the inhibition of this amplification of the complement pathways because its plasma concentration in humans is typically about 200 μg / mL (or about 2 μM) , and it has been shown to be a critical enzyme for activation of the alternative complement pathway (P.H. Lesavre and H.J. MOiier-Eberhard. J. Exp. Med., 1978; 148: 1498-1510; J.E. Volanakis et al., New Eng. J. Med., 1985; 312: 395-401) . However, there remains needs for efficacious and safe treatment for modulating the complement system activation. The present application addresses such needs.SUMMARYIn one aspect, provided herein is a compound of Formula (I) :or a stereoisomer, diastereomer, or pharmaceutically acceptable salt thereof, whereinX is S or Se;R1 is H, halo, cyano, -NO2, C1-C6 alkyl, halo C1-C6 alkyl, C1-C6 alkoxy, C2-C6 alkenyl, C2-C6 alkenyl-O-, C2-C6 alkynyl, C2-C6 alkynyl-O-, -N (Ra) 2, -C (O) N (Ra) 2, -CH2C (O) R7, -C (O) R7, -S (O) -C1-C6 alkyl, -S (O) 2-C1-C6 alkyl, -CH2NHC (O) -C1-C6 alkyl, -OCH2C (O) R7, C6-C10 aryl or 5-to 12-membered heteroaryl, wherein each of C1-C6 alkyl, C1-C6 alkoxy, C2-C6 alkenyl, C2-C6 alkynyl, C6-C10 aryl or 5-to 12-membered heteroaryl is independently and optionally substituted with one or more independently selected -OH, halo, -CN, C1-C6 alkyl or C3-C6 cycloalkyl, wherein each of C1-C6 alkyl, halo C1-C6 alkyl and C3-C6 cycloalkyl is independently and optionally substituted with one or more hydroxyl;R2 is H; halo; -NO2; -ORa; -SRa; -N (Ra) 2; C3-C6 cycloalkyl; 3 to 12-membered heterocyclyl; C1-C6 alkyl; halo C1-C6 alkyl; hydroxy C1-C6 alkyl; amino C1-C6 alkyl; C1-C6 alkoxy; halo C1-C6 alkoxy; C2-C6 alkenyl; C2-C6 alkenyl-O-; C2-C6 alkynyl; C2-C6 alkynyl-O-; -C (O) -C1-C6 alkyl; -S (O) -C1-C6 alkyl; -S (O) 2-C1-C6 alkyl; -CH2NHC (O) -C1-C6 alkyl; -CH2C (O) R7; -OCH2C (O) R7 , wherein each of C1-C6 alkyl, halo C1-C6 alkyl, hydroxy C1-C6 alkyl, amino C1-C6 alkyl, C1-C6 alkoxy, halo C1-C6 alkoxy, C2-C6 alkenyl or C2-C6 alkynyl is independently and optionally substituted with one or more independently selected -OH, halo, amino, -CN, C1-C6 alkoxy or C3-C6 cycloalkyl optionally substituted with one or more independently selected -OH, halo, -CN; and each of C3-C6 cycloalkyl or 3 to 12-membered heterocyclyl is independently and optionally substituted with one or more independently selected -OH, halo, -CN;R3 is hydrogen, halo, cyano, -NO2, C1-C6 alkoxy, hydroxy C1-C6 alkoxy, halo C1-C6 alkoxy, C1-C6 alkyl, hydroxy C1-C6 alkyl or halo C1-C6 alkyl, each of which is independently and optionally substituted with one or more independently selected -OH, halo, -CN, or C3-C6 cycloalkyl; or C3-C6 cycloalkyl optionally substituted with one or more independently selected -OH, halo, or -CN;n is 0, 1, or 2;R4 is C6-C10 aryl or 5-to 12-membered heteroaryl, each of which is optionally substituted by R6 and optionally further substituted by one or more substituents selected from halo, -OH, CN, cyanomethyl, C1-C6 alkoxy, hydroxy C1-C6 alkyl or C1-C6 alkyl, wherein each of C1-C6 alkoxy, hydroxy C1-C6 alkyl or C1-C6 alkyl is independently and optionally substituted with one or more independently selected -OH, halo, -CN, or C3-C6 cycloalkyl;R5 is hydrogen, C1-C6 alkyl, hydroxy C1-C6 alkyl, or C3-C6 cycloalkyl, wherein the C1-C6 alkyl, hydroxy C1-C6 alkyl or C3-C6 cycloalkyl is independently and optionally substituted with one or more independently selected -OH, halo, -CN;R6 is -N (Ra) 2, -C (O) N (Ra) 2, -SON (Ra) 2, -SO2N (Ra) 2, -C (O) R7, -CH2C (O) R7, -C(O) NHSO2 (C1-C4 alkyl) , -C (O) NHSO (C1-C4 alkyl) , -SO2NHC (O) (C1-C4 alkyl) , -SONHC (O) (C1-C4 alkyl) , -SO2N (H) m (C1-C4 alkyl) 2-m, -SON (H) m (C1-C4 alkyl) 2-m, -SO2 (C1-C4 alkyl) , -SO (C1-C4 alkyl) , -B (R8) 2, cyano, halogen, 5-to 12-membered heteroaryl, 3-to 12-membered heterocyclyl, C1-C6 alkyl or C1-C6 alkoxy, wherein each of 5-to 12-membered heteroaryl, 3-to 12-membered heterocyclyl, C1-C6 alkyl or C1-C6 alkoxy is independently and optionally substituted with one or more independently selected -OH, halo, -CN, or C3-C6 cycloalkyl;m is 0, 1, or 2;R7 is -OH, -O-C1-C6 alkyl, -N (Ra) 2, 3 to 6-membered heterocyclyl, C1-C4 alkoxy, amino, mono-or di-C1-C4 alkylamino;Ra, at each occurrence, is independently H, C1-C6 alkyl, C3-C6 cycloalkyl, wherein each of C1-C6 alkyl or C3-C6 cycloalkyl is independently and optionally substituted with halo, -OH or C3-C6 cycloalkyl;wherein the hydrogens in the compound of Formula (I) are each independently and optionally substituted with D.In some embodiments, n is 0 and the compound is of Formula (I-1-A) or Formula (I-1-B) :or a stereoisomer, diastereomer, or pharmaceutically acceptable salt thereof.In some embodiments, n is 1 and the compound is of Formula (I-2-A) or Formula (I-2-B) :or a stereoisomer, diastereomer, or pharmaceutically acceptable salt thereof.In some embodiments, n is 2 and the compound is of Formula (I-3-A) or Formula (I-3-B) :or a stereoisomer, diastereomer, or pharmaceutically acceptable salt thereof.In some embodiments, R4 is a phenyl substituted by R6. In some embodiments, R4 is a phenyl substituted by one R6. In some embodiments, R4 is a phenyl subs
