Abstract

The invention discloses new application of genistein in periapical disease control. It is found that genistein can remarkable suppress local inflammatory cell infiltration and bone destruction of periapical disease caused by pulp exposure from in-vivo experiment. By reducing RANKL expression level and promoting OPG expression, local RANKL / OPG ratio of periapical focus is dynamically adjusted, and the treatment function of the periapical diseases is achieved. The result indicates that the genistein is an effective medicine treating the periapical disease and can be used for manufacturing periapical disease treatment medicine.

Description

technical field

[0001] The invention belongs to the technical field of prevention and treatment of periapical diseases, and in particular relates to a new application of genistein for preventing and treating periapical diseases. Background technique

[0002] Periapical disease is a common and frequently-occurring disease of the oral cavity. The main pathological feature is the destruction and absorption of the periapical alveolar bone. The main pathogenic factor of periapical disease comes from the infection of anaerobic bacteria and facultative anaerobic bacteria in the root canal, so root canal treatment is often used clinically to remove infected substances in the root canal. CT examinations in recent years have shown that although about 63% of the affected teeth have undergone perfect root canal treatment, the periapical bone destruction is still difficult to heal, and finally apical resection or extraction of the affected teeth may be required. Therefore, finding poten...

Embodiment 1

[0021] 1. Experimental animals and materials:

[0022] 1.1 Experimental animals

[0023] All experimental procedures complied with the guidelines of the Ethics Committee of the School of Stomatology, Wuhan University. 20 male C57BL mice of SPF grade 6-8 weeks (purchased from the Animal Experiment Center of Wuhan University, license number: SCXK (E) 2008-0004), weighing about 20 grams, were kept in separate cages, 5 mice per cage.

[0024] 1.2 Experimental materials

[0025] Genistein was purchased from Cayman Corporation (molecular formula C 15 h 10 o 5 , molecular weight 270.2, purity≧98%). Referring to relevant literature and drug instructions, the powder genistein was dissolved in DMSO (Biosharp company) one day before administration, the concentration of the mother solution was 30 mg / mL, and the working concentration of the drug was prepared with PBS to 2.5 mg / mL.

[0026] 2. Animal grouping and establishment of periapical mouse model:
