Description

Technical Field

[0001] The invention relates to the field of pharmaceutical chemistry, in particular to a method for synthesizing 3-bromo-2-hydroxy-6-methylpyridine. Background Art

[0002] 3-Bromo-2-hydroxy-6-methylpyridine has a wide range of applications as a fine chemical raw material and pharmaceutical intermediate. For example, patent WO2015089192A1 reports that it can be used to synthesize lysine-specific demethylase-1 inhibitors for the treatment of cancers such as prostate cancer, breast cancer, bladder cancer, lung cancer, and melanoma. Patent WO2008101682A2 also reports that it can be used to synthesize new fungicides.

[0003] The currently reported synthesis process of 3-bromo-2-hydroxy-6-methylpyridine is to use 2-hydroxy-6-methylpyridine as raw material, first react with a bromination reagent to obtain 3,5-dibromo-6-methylpyridine-2-ol, with a yield of 88%, and then use n-butyl lithium to selectively remove a bromine atom under deep cold conditions (-60 to -90°C) to obtain a crude 3-bromo-2-hydroxy-6-methylpyridine. Due to the poor selective dehalogenation effect, the final product must be purified by column, with a yield of only 17%. (References: WO2015089192A1 and WO2016037005A1), and its synthesis route is shown in the following formula. This process is cumbersome and uses highly flammable n-butyl lithium, which is unsafe to operate. In addition, a large amount of by-products are produced during regioselective debromination, which makes post-processing of the final product difficult and it is difficult to obtain pure 3-bromo-2-hydroxy-6-methylpyridine. The overall yield is also low, less than 15%, resulting in high industrial costs. Therefore, this process is not suitable for large-scale production.

[0004]

[0005] In summary, the existing synthesis method of 3-bromo-2-hydroxy-6-methylpyridine has the problems of complex process operation, poor operation safety, low total yield, etc., which leads to high industrial cost and is not suitable for large-scale production. Summary of the invention

The next day, after the reaction is complete as detected by TLC, the reaction solution is cooled to room temperature, then poured into ice water for destruction, stirred for half an hour, and the pH value is adjusted to 7-8 with solid sodium bicarbonate, extracted three times with an organic solvent, combined the organic phases, washed the organic phases twice with saturated brine, dried over anhydrous sodium sulfate, and desolventized under reduced pressure to obtain a light yellow liquid 2,3-dibromo-6-methylpyridine; (3) reacting 2,3-dibromo-6-methylpyridine with sodium methoxide to obtain 3-bromo-2-methoxy-6-methylpyridine, wherein the specific process is as follows: adding sodium methoxide to an organic solvent, then adding 2,3-dibromo-6-methylpyridine, heating the reaction mixture to T4, 20°C≤T4≤150°C, reacting overnight, and the next day, after TLC detection of the reaction being complete, cooling the reaction mixture to room temperature, then evaporating the solvent under reduced pressure, pouring the residue into ice water, then extracting with dichloromethane three times, combining the organic phases, washing the organic phases twice with saturated brine, drying with anhydrous sodium sulfate, and desolventizing under reduced pressure to obtain a colorless liquid 3-bromo-2-methoxy-6-methylpyridine; (4) 3-bromo-2-methoxy-6-methylpyridine is reacted with an acid to remove the methyl group to obtain 3-bromo-2-hydroxy-6-methylpyridine. The specific process is as follows: 3-bromo-2-methoxy-6-methylpyridine is slowly added to the acid solution under stirring at room temperature. After the addition is completed, the reaction mixture is heated to T5 and reacted at 40°C≤T5≤120°C. After TLC detection shows that the reaction is complete, the reaction mixture is cooled to room temperature, and then the solvent is evaporated under reduced pressure. The residue is poured into ice water, and the pH is adjusted to 8 with solid potassium carbonate. The mixture is extracted with ethyl acetate for 3 times. The organic phases are combined, washed twice with saturated brine, dried over anhydrous sodium sulfate, and desolventized under reduced pressure to obtain 3-bromo-2-hydroxy-6-methylpyridine as an off-white solid.
