Abstract
Description
[0001] CROSS-REFERENCE TO RELATED APPLICATIONS
[0002] This application claims the benefit of priority to U.S. Provisional Application No. 63 / 342,394, filed on May 16, 2022, entitled “Sunscreen Compositions and Methods of Use,” the contents of which are incorporated herein in their entirety for all purposes. Technical Field
[0003] The present disclosure relates to broad spectrum sunscreen compositions, and to methods of using broad spectrum sunscreen compositions to protect skin from UV radiation. Background Art
[0004] The negative effects of exposure to ultraviolet ("UV") light are well known. Long-term exposure to sunlight can cause damage to the skin and hair, such as sunburn. Exposure to UV light of certain wavelengths (e.g., about 290 nm to about 400 nm), for example, can cause long-term damage and serious conditions, such as skin cancer.
[0005] For example, ultraviolet radiation in the shorter wavelength UVB region of 290nm to 320nm is considered to be significant in causing burning. Therefore, protection against UVB radiation is traditionally the main target of sunscreen preparations. Relatively recently, further protection against UVA radiation is also desired to reduce long-term skin deterioration. Usually, UV light promotes aging by causing free radicals in the skin, such as the formation of O-, OH, superoxide anions, nitric oxide and hydrogen radicals. Free radicals attack DNA, membrane lipids and proteins to produce carbon free radicals. These then react with oxygen to produce hydroperoxy radicals, which can attack adjacent fatty acids to produce new carbon free radicals. The final result is the loss of skin elasticity and the appearance of wrinkles.
[0006] More importantly, both UVA and UVB wavelengths have been identified as potential factors that increase skin cancers such as melanoma. Skin cancer is one of the most common types of cancer and has a problematic incidence in the United States. For these and other reasons, continued use of surface sunscreens is now highly recommended and common.
[0007] In order to be considered "broad spectrum", sunscreens are subject to strict regulatory approval by various federal agencies. For example, in order to be considered a "broad spectrum" sunscreen, the current federal standard (21 C.FR201.327) requires that the average critical wavelength must be 370nm or greater. The critical wavelength is determined to be the wavelength at which the integral of the spectral absorbance curve reaches 90% of the integral of the UV spectrum from 290nm to 400nm. In addition to this requirement, there are regulatory proposals requiring "broad spectrum" sunscreens to meet a UVAI / UV ratio requirement of greater than or equal to 0.70. Therefore, the demand for broad spectrum sunscreens that can provide adequate protection from UV light and also meet the ever-changing regulatory standards remains unmet. Summary of the invention
[0008] The present disclosure provides new sunscreens that are believed to unexpectedly provide a UVAI / UV ratio requirement of greater than or equal to 0.70 while simultaneously maintaining a sun protection factor (SPF) of 40 and above.
[0009] It is difficult to obtain a desired UVAI / UV ratio while maintaining a relatively high SPF (e.g., ≥40) because increasing or maintaining a high SPF increases or maintains a large denominator, which results in a lower UVAI / UV ratio. The formula for obtaining the UVAI / UV ratio is as follows:
[0010] UVAI / UV=average absorbance 340nm to 400nm / average absorbance UVB (290nm to 320nm) + UVAII (320nm to 340nm) + UVAI (340nm to 400nm). In view of the above formula for obtaining the UVAI / UV ratio, the skilled person will likely recognize that reducing the UVB accessible range will increase the ratio, for example, by reducing the UVB accessible range, which simultaneously reduces the denominator of the above formula and thus increases the overall ratio. However, about 90% of erythema caused by sunlight is caused by wavelengths in the range of 290nm to 320nm (UVB). In addition, since SPF is a measure of the ability of a sunscreen to reduce erythema, it is an effective measure of the ability of a product to block UVB light (290nm to 320nm). Therefore, one of the simplest and most direct ways to increase the UVAI / UV ratio is to reduce the UVB accessible range, which will have the undesirable effect of simultaneously reducing the SPF value of the sunscreen.
[0011] The sunscreen compositions of the present disclosure unexpectedly achieve both a UVAI / UV ratio of greater than or equal to 0.70 and an SPF value of 40 and greater. In addition, the disclosed sunscreen compositions can have a UVAI / UV ratio of greater than or equal to 0.70 and an SPF value of 40 or greater, while achieving commercially desirable aesthetics. Therefore, in one aspect, the sunscreen compositions described herein comprise a metal oxide (e.g., zinc oxide), one or more SPF active ingredients (e.g., octinoxate and / or octisalate), and one or more SPF enhancing ingredients (e.g., photostabilizers (e.g., butyl octyl salicylate and / or ethylhexylmethoxycrylene) and / or antioxidants (e.g., plant extracts) (e.g., Pongamia chinensis extract). In another aspect, the resulting sunscreen comprises a metal oxide (e.g., zinc oxide), one or more SPF active ingredients (e.g., octinoxate and / or octylsalicylate), and one or more SPF enhancing ingredients (e.g., photostabilizers (e.g., butyl octylsalicylate and / or ethylhexyl methoxypolyacrylonitrile) and / or antioxidants (e.g., plant extracts) (e.g., Pongamia chinensis extract)), and has a UVAI / UV ratio requirement of greater than or equal to 0.70 (e.g., 0.7 to 0.8) and an SPF of 40 and above. [0145] Example
[0146] Example 1
[0147] Four non-limiting exemplary sunscreen compositions (Formulations 1 to 5) were prepared according to aspects of the present invention. The formulations of Formulations 1 to 5 are provided in Table 1 (below).
[0148] Table 1
[0149]
[0150] Example 2
[0151] Formulations 1 to 5 were evaluated to assess the SPF value, UVAI / UV ratio and critical wavelength of such sunscreen compositions. In order to calculate the critical wavelength of formulations 1 to 5, a sample of each of formulations 1 to 5 was placed in a polymethyl methacrylate (PMMA) plate suitable for UV transmittance measurement. Specifically, each corresponding sample was applied to the entire plate in a series of small dots and then spread evenly with a gloved finger. Spreading was performed for about 30 seconds with a very light spreading motion. The plate was then allowed to balance in the dark for 15 minutes before irradiation. The irradiation dose was equivalent to 800 J / m 2 The effective dose for erythema (i.e., 80 / J m 2 -efficient).
[0152] After irradiation, SPF-290S is used to determine the average transmittance value of each wavelength in the UV spectrum (290 nanometers to 400 nanometers). The transmittance value is measured at intervals of 1 nanometer. The measurement of the spectrum irradiation of each wavelength transmission by the PMMA plate coated with 15 microliters of glycerol is carried out 5 times, and is stored in the memory of the computer together with the average value of 5 measurements using SPF-290S, to obtain a reference reading. Subsequently, the PMMA plate coated with the test product is repeated for 5 times to measure, and the average value is calculated and stored so that the computer can then calculate the ratio of the average reference reading under each wavelength to the average test preparation reading, to calculate the average test preparation transmittance. The average transmittance value is then converted into an average absorbance value.
[0153] Three separate PMMA plates were used. Each plate was scanned five times at different locations, yielding a total of 15 measurements. The critical wavelength value was determined as the wavelength at which the integration of the spectral absorbance curve reaches 90% of the UV spectrum integral from 290 nm to 400 nm. An average critical wavelength of 370 nm or greater was classified as broad spectrum protection.
[0154] The static sun protection factor ("SPF") of the above formulations 1 to 5 was measured in accordance with FDA, 21 CFR Sec. 201.327, subpart (i), SPF Test Procedure, Sunscreen Drug Products for Over-the-Counter Human Use, Final Monograph, Federal Register, Vol. 76, No. 117, June 17, 2011, the contents of which are incorporated herein by reference in their entirety. Specifically, a series of five UV doses expressed in Joules / square meter were provided to 10 participants, increasing in increments of 25%. This formed the basis for the unprotected MEDu. The participants were asked to avoid additional UV exposure.
[0155] Participants were evaluated approximately 16 to 24 hours after the MEDu dose was completed to determine the response and unprotected minimum erythema dose ("MEDu") for each participant. MEDu is the amount of erythema effective energy. Subsequently, on day 2, a UV dose was applied and the protected MED ("MEDp") of the participant was determined according to the process of FDA, 21 CFR Sec, 201.327, subpart 4 (iii) and FDA, 21 CFR Sec, 201.327, subpart 5 (iii), the contents of which are incorporated herein by reference in their entirety. Participants were then evaluated approximately 16 to 24 hours after the completion of the UV dose on day 2. The MED of all sites (both protected and unprotected) receiving the UV dose was assessed. The SPF values for all of Formulations 1 through 5 were calculated based on FDA standards using FDA, 21 CFR Sec 201.327, first (i) subpart (6) by dividing the MEDp value produced in the sunscreen-protected area by the MEDu value produced in the unprotected test area, using the following calculation for each participant:
[0156] MEDip / MEDiu=SPFi value
[0157] Data from 10 participants were used to calculate the label SPF values for the test formulations (i.e., formulations 1 to 5). The mean SPF value and standard deviation for the participants were also calculated.
[0158] A summary of the UVAI / UV ratios, SPF values, and critical wavelengths for Formulations 1 through 5 is provided in Table 2 (below).
[0159] Table 2
[0160]
[0161] As is clear from the table above, the addition of SPF enhancing ingredients increases the UVAI / UV ratio to over 0.7, increases the SPF to over 40, and increases the critical wavelength to over 370. Non-limiting exemplary compositions comprising certain combinations of SPF enhancing ingredients and UV filters unexpectedly achieve the desired SPF while meeting the required UVAI / UV ratio. In addition, Formulations 1 and 2 achieve improvements in SPF values and UVAI / UV ratios while having a reduced total amount of UV filters compared to market-based formulations, which is unexpected.
[0162] Example 3
[0163] The qualitative aspects of formulations 1 to 5 from Example 1 were evaluated by a clinical trial using eight panelists. A summary of the qualitative aspects and scoring methods for formulations 1 to 5 is provided in Tables 3 to 5. For Tables 3 and 4, higher values are assigned to formulations that exhibit more evaluated characteristics, while lower values are assigned to formulations that exhibit fewer evaluated characteristics. The viscosity in Table 4 was measured using a viscometer.

[0170] The above scores are based on the selection of formulations relative to the market-based formulation described in Example 2. For example, eight (out of eight) panelists preferred Formulation 3 relative to the market-based formulation, and six (out of eight) panelists preferred Formulation 4 relative to the market-based formulation.
[0171] Example 4
[0172] Five non-limiting exemplary sunscreen compositions (Formulations 6 to 10) were prepared according to aspects of the present disclosure. The formulations of such sunscreen compositions are provided in Table 6 (below).

[0179] Example 6
[0180] Seven compositions (Formulations 16 to 22) were prepared according to aspects of the present invention. Formulations 16 to 22 were evaluated according to the procedure described in Example 2 to determine SPF values and UVAI / UV ratios. The change in SPF value (ΔSPF) relative to Formulation 16 used as a control was calculated. X-制剂16 ). A summary of the formulations of Formulations 16 to 22 and the SPF values and UVAI / UV ratios for each corresponding composition is provided in Table 8 (below).
[0181] Table 8
[0182]
[0183]
[0184] As seen in Table 8, certain combinations of SPF active ingredients, metal oxides, photostabilizers, and / or antioxidants achieve significant increases in UVAI / UV ratios while simultaneously achieving the desired SPF values. In addition, the significant increase in SPF values achieved by Formulation 22 with the increase in UVAI / UV ratio is unexpected in light of Formulation 17, which suggests that the addition of butyl octyl salicylate to octyl octyl ester and zinc oxide reduces the SPF value and UVAI / UV ratio of the sunscreen composition; in light of Formulation 20, which suggests that the addition of ethylhexyl methoxy polyacrylonitrile and Pongamia extract to octyl octyl ester and zinc oxide reduces the SPF value and UVAI / UV ratio of the sunscreen composition; and in light of Formulation 21, which suggests that the addition of butyl octyl salicylate and ethylhexyl methoxy polyacrylonitrile to octyl octyl ester and zinc oxide reduces the SPF value and UVAI / UV ratio of the sunscreen composition. Formulation 22 was further evaluated by in vivo testing and was determined to have an SPF value of 52.
[0185] Formulation 22 and the market-based formulation from Example 1 were evaluated by an expert panel to rate the whiteness of these formulations. Formulation 22 showed similar whiteness to the market-based formulation, which was unexpected because Formulation 22 contained approximately 22% more zinc oxide than the market-based formulation.
[0186] Example 7
[0187] Eight compositions (Formulations 23 to 30) were prepared according to aspects of the present invention. Formulations 23 to 30 were evaluated according to the process described in Example 2 to determine the SPF value and UVAI / UV ratio. The formulas of Formulations 23 to 30 and the summary of the SPF value and UVAI / UV ratio of each corresponding composition are provided in Table 9.
[0188] Table 9
[0189]
[0190]
[0191]
[0192] As seen in Table 9 (above), certain combinations of SPF active ingredients, metal oxides, photostabilizers, and / or antioxidants achieve significant increases in UVAI / UV ratios while simultaneously achieving desired SPF values. The significant increases in SPF values and UVAI / UV ratios achieved by certain sunscreen compositions disclosed in Table 9 (e.g., but not limited to, Formulations 27 and 30) are unexpected and surprising.
[0193] Formulation 30 and the market-based formulation from Example 1 were evaluated by an expert panel to assess the whiteness of these formulations. Formulation 30 showed similar whiteness to the market-based formulation, which was unexpected because Formulation 30 contained approximately 22% more zinc oxide than the market-based formulation. Formulation 30 was evaluated according to the in vivo test and was determined to have an SPF of 51.
[0194] Example 8
[0195] A composition (Formulation 31) was prepared according to aspects of the present invention. Formulation 31 was evaluated according to the process described in Example 2 to determine the SPF value and UVAI / UV ratio. The formula of Formulation 31 and a summary of the SPF value and UVAI / UV ratio of Formulation 31 are provided in Table 10.
[0196] Table 10
[0197]
[0198]
[0199] Example 9
[0200] A composition (Formulation 32) was prepared according to aspects of the present invention. The formulation of Formulation 32 is provided in Table 11.
[0201] Table 11
[0202]
[0203]
[0204] Formulation 32 was evaluated by an expert panel in comparison to the market-based formulation from Example 1 to assess the whiteness of Formulation 32. The expert panel applied samples of the respective compositions and evaluated the whiteness of the respective samples after three rubs during application, after twelve rubs during application, at the end of application of the sample, instantaneous whiteness, and whiteness after five minutes. Table 12 includes a summary of the evaluation of Formulation 32 and the market-based formulation.
[0205] Table 12
[0206]
[0207] As seen in Table 12, Formulation 32 exhibits lower whiteness than the commercially based formulation. This is unexpected because Formulation 32 has approximately 22% more zinc oxide than the commercially based formulation.
