Abstract
Description
Technical Field
[0001] The present invention relates to a heteroaryl derivative compound and its medical use. Specifically, the present invention relates to a heteroaryl derivative compound having epidermal growth factor receptor (EGFR) inhibitory activity. Background Art
[0002] Protein kinases are molecular switches involved in signal transduction pathways, and the transition between the active and inactive states of target proteins in cells should be smoothly controlled by kinases. If the transition between the active and inactive states is abnormally controlled, the signal transduction in the cell will be overactivated or inactivated, thereby inducing uncontrolled cell division and proliferation. In particular, abnormal activation caused by mutation, amplification and / or overexpression of protein kinase genes leads to the occurrence and progression of various tumors, or plays a vital role in the occurrence of various diseases such as inflammatory diseases, degenerative brain diseases, autoimmune diseases, etc.
[0003] Epidermal growth factor receptor (EGFR) is a receptor tyrosine kinase of the ErbB family. It is abnormally activated in many epithelial tumors, including non-small cell lung cancer (NSCLC), breast cancer, glioma, head and neck squamous cell carcinoma, colorectal cancer, rectal adenocarcinoma, head and neck cancer, gastric cancer and prostate cancer. It is known that the activation of EGFR-tyrosine kinase can lead to sustained cell proliferation, invasion of surrounding tissues, distant metastasis, angiogenesis and increased cell survival.
[0004] Meanwhile, it is known that EGFR mutations (i.e., EGFR Del19 or EGFR L858R) are the main causes of non-small cell lung cancer and head and neck cancer, and Iressa and Tarceva, which are therapeutic drugs for them, have been developed and used in clinical practice. However, when patients use these drugs, acquired resistance is observed, i.e., secondary mutations of EGFR are caused according to the drug structure, and it is also found that this resistance is the main cause of actual resistance. When the first-generation EGFR inhibitors are applied for an average of about 10 months, acquired resistance, i.e., the T790M mutation located in the EGFR kinase gatekeeper, occurs, so the first-generation EGFR inhibitors do not show efficacy. In other words, the occurrence of EGFR Del19 / T790M or EGFR L858R / T790M double mutations makes existing therapeutic drugs unable to exert their efficacy. Therefore, the third-generation EGFR-TKI targeted drug osimertinib has been developed, which shows high responsiveness to drug resistance caused by EGFR T790M mutation, but there are also reports of drug resistance (Clin Cancer Res, 2015, 17: 21). EGFR C797S mutation is considered to be one of the main mechanisms leading to osimertinib resistance. It is reported that about 40% of clinical trial patients have EGFR C797S mutation (Nature Medicine, 2015, 21: 560-562).
[0005] There is a growing but unmet need for novel compounds that can treat EGFR-related diseases by modulating EGFR activity, particularly C797S mutations, such as EGFR Del19 / T790M / C797S or EGFR L858R / T790M / C797S. Summary of the invention Technical issues

[0237] <Experimental Example 1> Evaluation of EGFR C797S enzyme inhibitory activity
[0238] The compounds of the present invention were reacted with purified human epidermal growth factor receptor (EGFR) (d746-750 / T790M / C797S) (668-end, SignalChem) enzyme and specific substrate, and the enzyme inhibitory activity was evaluated as follows. The reaction buffer consisted of 40mM Tris-HCl (pH 7.4), 20mM MgCl2, 0.5mg / mL BSA and 50μM DTT, and the reactions of all test substances were carried out in the reaction buffer. The compounds were diluted continuously from 10mM DMSO stock solution in 12 gradients, and the enzyme activity was measured when the final compound concentration was 10, 3, 1, 0.3, 0.1, 0.03, 0.01, 0.003, 0.001, 0.0003, 0.0001 and 0μM. In this test, human epidermal growth factor receptor (d746-750 / T790M / C797S) (3ng) enzyme was reacted with purified ATP (5μM) and enzyme substrate (0.2μg) at 25°C for 1 hour, and then the enzyme activity was determined using the in vitro ADP-GloTM kinase assay (Promega). The enzyme activity reaction solution, ADP-Glo reaction solution and enzyme activity detection solution were reacted in a ratio of 2:2:1, and the enzyme activity inhibition was determined by luminescence. Based on the enzyme activity luminescence of the solvent control group that was not treated with the compound, the degree of inhibition of the enzyme activity by the treatment concentration of each compound was calculated, and the concentration of each compound that inhibited the enzyme activity by 50% was determined as IC 50(nM) values were calculated using GraphPad Prism 8.4.3 (GraphPad Software, San Diego). The results are summarized in Table 3 below.
[0239] [Table 3] (A:IC 50 <50nM; B: 50nM≤IC 50 <500nM; C: 500nM≤IC 50 <5000nM; D: 5000nM≤IC 50 )

The compound was diluted with dimethyl sulfoxide (3-fold dilution, a total of 12 concentrations), and 1 μL was injected for each concentration to make a final concentration of 0.2nM to 5μM. After 72 hours of treatment with the compound, the living cells were reacted in a carbon dioxide incubator at 37°C for 72 hours, and then reacted at room temperature for 10 minutes using Cell Titer-Glo cell viability luminescent reagent (Promega), and the luminescence intensity was measured using a microplate reader (SynergyNeo2, Biotek). The calculated result value is the cell growth rate (%) compared with the control group. GI was calculated using the GraphPadPrism version 8.4.3 (GraphPad Software, San Diego) program 50 The results are summarized in Table 4 below.
[0243] [Table 4] (A:GI 50 <500nM; B: 500nM≤GI 50 <5000nM; C: 5000nM≤GI 50 )
