Description

Technical Field

[0001] The invention belongs to the technical field of separation crystallization and relates to a method for preparing a stable crystal form of daptomycin. Background Art

[0002] Daptomycin (English name Daptomycin, CAS: 103060-53-3, molecular formula: C 72 H 101 N 17 O 26 , molecular weight: 1620.67g / mol), daptomycin contains a cyclic decapeptide formed by ten amino acids (3-alanine-S-lactone, 3-methyl-L-glutamyl, D-seryl, L-threonyl, L-aspartylglycyl, D-alanyl, L-aspartyl, L-ornithine, glycyl), and also contains a decanoyl and three amino acids (L-asparagyl, L-asparagine, L-tryptophanyl) connected to the side chain, containing a total of 13 amino acids.

[0003]

[0004] Daptomycin has a complex structure and is difficult to chemically synthesize. It is usually fermented by Streptomyces (S. roseosporus). In 1961, Falcao and Dailia isolated and screened S. roseosporus from soil samples from the Ararat Mountains in Turkey and obtained it by fermentation. The molecular structure of daptomycin is relatively complex. It has both hydrophilic carboxyl and amino groups, as well as hydrophobic benzene rings, saturated alkane chains and other groups, which give it both hydrophilic and lipophilic properties. Daptomycin is a water-soluble antibiotic, and the pH value of its aqueous solution is generally around 3.5; when the pH value is maintained at 3.5-9.0, daptomycin can be fused in a good state. At room temperature, the daptomycin aqueous solution can remain relatively stable for more than a week. However, if the pH value changes more extreme (the pH value is greater than 9.0 or less than 2.0), daptomycin will denature and this denaturation cannot be reversed. Daptomycin is relatively less soluble in acetone and chloroform than in other organic solvents and is almost insoluble in acetonitrile.

[0005] Obtaining a stable crystalline form of daptomycin has significant patent value in the field of drug development and formulation. As a glycopeptide antibiotic, the efficacy of daptomycin is affected by the stability, solubility and bioavailability of the crystal form. By obtaining a stable crystal form, its chemical stability can be effectively improved to avoid degradation or quality loss during storage and transportation; at the same time, solubility and bioavailability are improved, thereby improving the therapeutic effect of the drug. The optimization of the stable crystal form also provides a basis for the dosage form innovation of daptomycin, especially in oral preparations and injection formulations, where better dosage accuracy and drug efficacy persistence can be achieved through crystal form control. In addition, the stable crystal form technology involved in the present invention can not only be applied to daptomycin, but also provides a valuable reference for the crystal form improvement of other glycopeptide antibiotics, and has broad application prospects. [0100] (2) Crystalline hygroscopicity

[0101] The stability of the daptomycin crystals prepared in Example 1, Comparative Example 1 and Comparative Example 2 at room temperature was compared. About 1.5 g of daptomycin sample was added to a culture dish dried to constant weight and placed in an environment with relative humidity (Relative Humidity) of RH43% (K 2 CO 3 aqueous solution), RH67%(CuCl 2 Aqueous solution), RH98% (saturated K 2 SO 4 ) was placed in a closed desiccator at room temperature for 72 h, and the weight change during the process was recorded. The results are shown in Table 1.

[0102] The experimental results are shown in Table 1:

[0103] The crystals not prepared by the present patent (Comparative Example 1) gained 3.2% in an environment of RH43% and were hygroscopic but less hygroscopic; when the humidity was 76%, they gained 8.4% and were hygroscopic; when the relative humidity was 98%, they gained 25.2% and were extremely hygroscopic. The crystals not prepared by the present patent (Comparative Example 2) gained 4.2% in an environment of RH43% and were hygroscopic but less hygroscopic; when the humidity was 76%, they gained 9.7% and were hygroscopic; when the relative humidity was 98%, they gained 26.3% and were extremely hygroscopic. The crystals prepared by the present invention (Example 1) gained 0.1% in an environment of RH43%, gained 0.95% in an environment of humidity of 76%, and gained 4.9% at a relative humidity of 98%, were hygroscopic but less hygroscopic.

[0104] The above data were obtained by adding about 1.5 grams of daptomycin sample to a culture dish dried to constant weight, placing it in a high-temperature oven at 50°C for 12 hours, recording the weight every hour, and observing the weight change of the entire experiment. The final results recorded are shown in Table 1.

[0109] The purity results of the daptomycin crystals prepared in Example 1, Comparative Example 1, and Comparative Example 2 after being placed in a drying dish at 90°C and a humidity of 43% for 6h, 12h, 24h, and 48h were determined by spectrophotometry. The results are shown in Table 2, and the results show that the stability of the daptomycin crystals obtained in Example 1 is significantly better than that of the crystals in Comparative Examples 1 and 2.

[0112] The present invention provides a method and idea for preparing a stable crystalline form of daptomycin. There are many methods and approaches to implement the technical solution. The above is only a preferred embodiment of the present invention. It should be pointed out that for ordinary technicians in this technical field, several improvements and modifications can be made without departing from the principle of the present invention. These improvements and modifications should also be regarded as the scope of protection of the present invention. All components not specified in this embodiment can be implemented by existing technologies.
