Abstract
Description
Technical Field
[0001] The invention belongs to the field of organic chemical synthesis and medicinal chemistry, and specifically relates to a cyclic dinucleotide analog containing a triazole structural unit, and a preparation method and application thereof. Background Art
[0002] Cyclic dinucleotides are a class of cyclic compounds composed of two purine nucleotides connected by two phosphodiester bonds. As second messengers, they participate in the regulation of various important physiological processes of bacteria and play an important role in the nucleic acid immunity mechanism of higher organisms. At the same time, they are directly related to various human disease states. With the deepening of research, people have also found that cyclic dinucleotides are also important immune transmitters for bacterial antiviral immunity. The latest research also confirms that there is an evolutionary connection between the antiviral immune signaling mechanism of microorganisms and higher organisms using cyclic dinucleotides as second messengers.
[0003] There are two types of natural cyclic dinucleotides that have been discovered: one is cyclic dinucleotides derived from microorganisms, in which two purine nucleotides are connected by 3 , -5 , Phosphodiester bonds form a ring, including 3 , , 3 , -Cyclic diguanylate (3 , , 3 , -c-di-GMP), 3 , , 3 , -cyclic di-AMP (3 , , 3 , -c-di-AMP) and 3 , , 3 , -cyclic GMP (3 , , 3 , -cGAMP), such cyclic dinucleotides can be used as second messengers to make molecules ubiquitous in bacteria and participate in a variety of physiological processes, including cell differentiation, the transition from motile state to biofilm state, the production of pathogenic factors, the formation of biofilm, etc. One type is 2 , , 3 , -cyclic GMP (2 , , 3 , -cGAMP), whose two purine nucleotide bases are linked by a 2 , -5 , - and a 3 , -5 , -Two phosphodiester bonds are connected to form a ring. This cyclic dinucleotide is the endogenous ligand of STING. It binds to the interferon gene stimulator STING, activates the cascade reaction of downstream signals, and ultimately induces the production of type I interferon and other cytokines, initiating an immune response.
[0004] In view of this, the design and development of cyclic dinucleotide (CDNS) analogs is of great significance. Summary of the invention

[0105] Example 10: Study on the biofilm inhibition activity of compounds
[0106] Three strains of bacteria were used: Escherichia coli, Pseudomonas aeruginosa, and Staphylococcus aureus
[0107] Dilute the bacteria to 5 × 10 5Colony forming units / unit milliliter (CFU / mL), or OD595nm of about 0.4 to 0.6. Add 75 μL of the above-diluted bacterial dilution to each well of a 96-well plate, dissolve the compound in 25 μL of sterile saline, and add the compound to the wells according to the gradient concentration of 1.25mM, 625μM, 312μM, 156μM, 78μM, 39μM, 19μM, and 10μM. There are 3 replicates in each group. After incubation at 37°C overnight for 18h, aspirate the bacterial solution and wash 3 times with PBS. (Note: Be careful not to puncture the biofilm during the washing process) Turn the plate upside down on paper, dry for 30min, add 150μL of crystal violet and let it stand for 15min. Remove the crystal violet, wash with PBS to remove the residual crystal violet, and dry for 30min. Add 30% ethanol and incubate in a 37°C incubator for 30min to dissolve the biofilm. Take 100 μL and add it to a new 96-well plate, and measure the absorbance at 595 nm using an ELISA reader.
