Description

[0001] CROSS-REFERENCE TO RELATED APPLICATIONS

[0002] This application claims the benefit of U.S. Provisional Patent Application No. 63 / 422,700, filed on November 4, 2022, the entire contents of which are incorporated herein by reference. Technical Field

[0003] The technology described herein generally relates to automated diagnostic analyzers including components thereof. In some aspects, the technology more specifically relates to a multi-channel extraction drawer. Background Art

[0004] Diagnostic testing of biological samples helps the healthcare industry diagnose and treat diseases quickly and effectively. Clinical laboratories that perform such diagnostic testing already receive hundreds or thousands of samples per day, and the demand continues to increase. Automation of sample analysis helps with the challenge of managing such a large number of samples. Automated sample analysis is typically performed by automated analyzers, which are usually self-contained systems that perform multi-step processing on biological samples to obtain diagnostic results.

[0005] Several current automated clinical analyzers provide the user with a series of automated tests or assays that can be performed on a provided sample. Furthermore, when the samples arrive at the laboratory, they are often not ready for analysis. In order to prepare samples for testing with an automated analyzer, laboratory technicians typically transfer aliquots of the samples from primary containers received by the laboratory into secondary containers that can be used for analysis by the analyzer. Furthermore, the technician typically must know which tests are to be performed on the sample so that the technician can select a specific test reagent or diluent to pair with the sample. This can be time consuming and can result in operator error and the risk of contracting an infectious disease.

[0006] Pre-analytical systems have also emerged that are designed to help prepare samples for analysis and further free operators from the workflow between receiving the sample in the laboratory and the test results of the analyzer. However, many of these systems still require extensive technician involvement, such as: before the sample is loaded into the pre-analytical system; after the sample is prepared by the pre-analytical system; and after the analyzer completes the analysis.

[0007] For example, some pre-analytical systems can automatically transfer aliquots of a sample from a first container to a second container. However, such systems typically require a technician to manually match the identification codes of the first and second containers before loading them into the system, which can be time-consuming and prone to errors.

[0008] Additionally, many of these systems cannot be integrated with one or more analyzers, and conversely, the analyzers cannot be integrated with these systems. In this regard, a technician must be present to manually transfer the sample from the pre-analytical system to the analyzer and from the analyzer to a storage location once the analysis is complete. This requires a skilled labor force to perform menial tasks and can be distracting because the technician must always be aware of the sample's progress through the pre-analytical system and analyzer so that the technician is ready to transfer the sample when it is ready to minimize downtime.

[0009] In addition, current pre-analytical systems typically prepare samples at a different rate than the analyzer evaluates these samples. This makes the integration between the pre-analytical system and the analyzer more complex. In this regard, a technician may be required to continuously track samples prepared by the pre-analytical system until a full batch of samples has been accumulated for manual transfer to the analyzer. Alternatively, a technician may transfer a partial batch to the analyzer, which reduces the productivity of the analyzer.

[0010] Therefore, while current automated pre-analytical systems and analyzers benefit clinical laboratories, there is still room for better integration and automation of various systems. Summary of the invention

[0045] Return to reference Figure 2 , the auxiliary platform 4012 is disposed near the bottom of the analyzer 4000 and is located below the processing platform 4014. The auxiliary platform 4012 houses electronic components and waste storage. For example, the auxiliary platform 4012 may include a liquid waste storage 4002 that receives and holds all liquid waste, such as liquid waste from the extraction container 4026 during the extraction process and from the tank assembly 4050 during the emptying process. The auxiliary platform 4012 also includes one or more waste storages 4004 that are seated below each waste chute 4210 extending through the processing platform 4014, such as Figure 4AAs shown. For example, a single waste reservoir 4004 can be located below the waste chute 4210 and can collect all solid waste. In another example, two waste reservoirs 4004 can be used to collect used pipette tips and used amplification plates 4040, respectively. Each waste reservoir 4004 can include a sensing device for detecting waste levels. The sensing device can include, for example, an optical or ultrasonic sensor.

[0046] In the illustrated embodiment, the processing platform 4014 includes six consumable drawer assemblies 4120. Each drawer assembly 4120 is a single-channel drawer. There are six drawers 4120 in total. In some cases, this drawer arrangement increases costs and needs to reduce costs. In some cases, this drawer arrangement is complicated for manufacturing or technical maintenance, and needs to reduce the complexity of manufacturing and technical maintenance. For example, each drawer 4120 includes its own extractor module 4125. For another example, each drawer 4120 includes its own latch feature. Therefore, it is necessary to reduce components, thereby reducing the cost associated with the drawer assembly. The drawer described herein may include any feature of the drawer 4120.

[0047] Figures 5A-5C 4130 and a corresponding extractor module 4135 according to the present disclosure. The dual channel drawer 4130 may include any of the features of the drawer 4120. The dual channel drawer 4130 may include advantageous features. The dual channel drawer 4130 may be more cost-effective. The dual channel drawer 4130 may hold the same number of consumables as two drawers 4120. The dual channel drawer 4130 may advantageously have only one extractor module 4135. The dual channel drawer 4130 may advantageously have only one set of telescoping slides to facilitate movement of the dual channel drawer 4130. The dual channel drawer 4130 may advantageously have only one retaining feature to retain consumables within the dual channel drawer 4130. Return to Reference Figure 4A, analyzer 4000 may include six consumable channels with six drawers 4120, six corresponding extractor modules 4125, and six corresponding latch mechanisms. Utilizing dual channel drawers 4130, the same analyzer 4000 may maintain six consumable channels, but utilize only three of the dual channel drawers 4130, three extractor modules 4135, and three corresponding latch mechanisms. Dual channel drawers 4130 may reduce the number of parts. Dual channel drawers 4130 may reduce costs. Dual channel drawers 4130 may reduce the complexity of manufacturing and technical maintenance. Dual channel drawers 4130 do not affect total throughput because the number of consumables is the same, and therefore the number of samples processed is the same. Dual channel drawers 4130 do not affect customer departure time because the number of consumables is the same, and therefore the number of samples processed is the same.

[0048] The dual channel drawer 4130 is configured to accommodate most of the consumables used in the assay workflow. The dual channel drawer 4130 may include a pipette tip station 4134. The dual channel drawer 4130 may include an extraction container station 4136. The dual channel drawer 4130 may include an amplification plate station 4138. The amplification plate station 4138 is configured to hold two amplification plates 4040. The dual channel drawer 4130 may be configured to hold the same types of consumables described herein. The dual channel drawer 4130 may hold twice as many consumables as the drawer 4120.

[0049] Stations 4134, 4136, 4138 may be arranged linearly. The arrangement may be from front to back, with the pipette tip station 4134 in the front, the extraction container station 4136 in the middle, and the amplification plate station 4138 in the back. The dual channel drawer 4130 may hold two pipette tip holders 4060 side by side in the front. The dual channel drawer 4130 may hold two extraction container holders 4020 side by side in the middle. The dual channel drawer 4130 may hold two amplification plates 4040 side by side in the back.

[0050] This arrangement can allow consumables to interface with other components of the analyzer 4000. The analyzer 4000 may include three of the dual channel drawers 4130. The dual channel drawer 4130 may be seated at the front of the analyzer 4000 between the two detectors 4260a, 4260b. The extraction container station 4136 may facilitate the extraction of polynucleotides from the sample. The extraction container station 4136 may be positioned relative to the extractor module 4135. Once the extraction is complete, the polynucleotides extracted in the extraction container 4026 are used to rehydrate the reagents in one of the amplification plates 4040 in the amplification plate station 4138, such as the master mixing reagent. The amplification plate 4040 may be prepared for amplification, such as PCR. The prepared amplification plate 4040 may be moved to appropriate readers 4260a, 4260b. The process may be achieved by using a multifunctional robot 4300, a plate sealer 4220, and an orbital mixer 4230. The dual channel drawer 4130 may include a visual indicator indicating the current state of the dual channel drawer 4130.

[0051] The dual channel drawer 4130 accommodates the extractor module 4135 within the housing 4132. The dual channel drawer 4130 may include one extractor module 4135. In some embodiments, the extractor module 4135 may process samples from two channels simultaneously. In some embodiments, the extractor module 4135 may process samples from two extraction container holders 4020 simultaneously. In some embodiments, the extractor module 4135 may process samples twice the number of samples of the extractor module 4125. The extractor module 4135 may move up and down relative to the two extraction container holders 4020 simultaneously. In some embodiments, the extractor module 4135 may process samples from two channels sequentially, for example, depending on when the sample is inserted into the extraction container 4026. In some embodiments, the extractor module 4135 may process samples from two extraction container holders 4020 sequentially. In some embodiments, the extractor module 4135 may process samples from two channels independently, for example, depending on when the sample is inserted into the extraction container 4026. In some embodiments, the extractor module 4135 can independently process samples from two extraction container holders 4020. In some embodiments, the pipette extracts polynucleotides from only one of the extraction container holders 4020. In some embodiments, the workflow does not process samples from both extraction container holders 4020. The software can be limited to process one extraction container holder 4020. The pipette can be limited to process one extraction container holder 4020. The instrument can be limited to process one extraction container holder 4020. The workflow timing can be limited to process one extraction container holder 4020.

[0052] The extractor module 4135 includes a magnet 4136. The magnet 4136 can be arranged to apply a magnetic field on opposite sides of the extraction container. The magnet 4136 provides a magnetic field for extracting polynucleotides from a sample. The extractor module 4135 is movable vertically. The extractor module 4135 can move the magnet 4136 and thereby move the corresponding magnetic field relative to the extraction container 4026 of the two extraction container holders 4020 located in the extraction container station 4136. The extractor module 4135 can be positioned below the extractor container station 4136. The extractor module 4135 can be selectively moved in the up and down directions along one or more guide rails 4137. The guide rail 4137 can be set on a side wall that separates the extraction container station 4136 and the amplification plate station 4138. As shown in FIG. Figure 5C 4130. As shown, the extractor module 4135 is in an extended position. The magnet 4136 can be near the extraction container 4026. The extractor module 4135 can be lowered below the extraction container 4026. The extractor module 4135 is housed in the dual channel drawer 4130. In some embodiments, the analyzer 4000 can include three and three extractor modules 4135 in the dual channel drawer 4130.

[0053] This movement of the magnet 4136 of the extractor module 4135 can facilitate the processing of the sample. To help separate the polynucleotides extracted from the sample, the polynucleotides can be bound to iron oxide ("FOX") particles located in the extraction container 4026. The particles allow the polynucleotides to be magnetically captured. This enables the polynucleotides to be separated from the remaining unwanted sample, which can be washed away from the eluent using a wash buffer located in the slot assembly 4050. To perform this separation, a magnetic field is applied to the extraction containers 4026 of the two extraction container holders 4020. The magnetic field is applied by using the extractor module 4135. The extractor module 4135 includes magnets 4136, which are positioned to ensure that each row of extraction containers 4026 is subjected to a magnetic field. In some embodiments, the magnets 4136 are disposed on both sides of the extraction container 4026. The magnets 4136 are selectively moved from a position completely below the extraction container 4026 to a position adjacent to the extraction container 4026. In some embodiments, the magnets 4136 can generally be located in one plane. Extraction container 4026 can intersect this plane in extraction mode. In other modes, extraction container 4026 can be completely below this plane. In some embodiments, magnet 4136 overlaps extraction container 4026. Extractor module 4135 applies a magnetic field to one side of extraction container 4026 that captures bound polynucleotides.

[0054] The dual channel drawer 4130 may include a hinged retaining feature 4131. The retaining feature 4131 may be a spring loaded plate. The retaining feature 4131 may be hingedly connected to the housing 4132. The hinge 4133 of the retaining feature 4131 may be positioned behind the extraction container station 4136. The hinge 4133 of the retaining feature 4131 may be positioned in front of the amplification plate station 4138.
