Description

TECHNICAL FIELD

[0001] The present invention relates to a soothing acne patch and a method of preparing the same, more specifically, a hydrophilic colloid; hydrogenated hydrocarbon; sodium carboxymethyl cellulose; mineral oil; polyethylene glycol 600; polyethylene polypropylene glycol; carboxylic acid; melaleuca alternifolia (tea tree) leaf oil; and aloe vera leaf extract; and it is about an acne patch containing active ingredients. BACKGROUND

[0002] Acne is a common skin disease caused by clogging of skin pores with sebum and dead skin cells. Acne is most common in adolescence, but it can also occur in adults and can occur on the face, neck, chest, and back. Acne can be roughly divided into whiteheads, red bumps, and deeper cysts. It varies in severity from mild to severe, and in severe cases, it can cause mental stress in addition to skin disease. Therefore, various methods have been used to treat acne. Topical drugs, oral drugs, and skin care methods are commonly used. Among them, the patch product is a topical drug that is attracting attention. The acne patch is made of a hydrophilic colloid and an acrylic resin, and they have both moisture permeability and adhesion. When the user applies the patch to the acne area, it can promote the elution of sebum, pus, and the like, and prevent side effects such as pain and pigmentation. In addition to concealment and simple coverage, the patch has a drug delivery system (DDS) function. The acne patch contains various active ingredients such as salicylic acid, retinol, vitamins, hydroquinone, ginsenosides, etc., which can enhance the therapeutic effect.

[0003] However, the conventional acne patch product still has drawbacks. First, there is a problem of low adhesion and shape retention of the patch, so the patch is easily peeled off or wrinkled. In addition, it also has problems such as short shelf life of the patch, secondary irritation after the patch is removed, etc. Finally, there are limitations such as portability and concealment, which are somewhat low in ease of use.

[0004] Therefore, the inventors of the present invention have confirmed that the present invention has excellent effects of epidermal adhesion, shape stability, biodegradability, and stability, etc.

[0051] Figure 1 A picture showing a product consisting of a pimple patch in a rectangular, circular, or T-zone shape according to one embodiment is shown.

[0052] Hereinafter, the composition of the present application and its effects will be explained in more detail through specific embodiments and comparative examples. However, the present embodiments are intended to describe the present application in more detail, and the scope of the present application is not limited to these embodiments.

[0053] Example 1: Manufacture of a single-layer pimple patch of the present application

[0054] The water gel adhesive 38 wt, hydrogenated hydrocarbon 30 wt, sodium carboxymethyl cellulose 21.2 wt, mineral oil 9.45 wt, polyethylene glycol 600 0.7 wt%, polyethylene-polypropylene glycol 0.2 wt, carboxymethyl acid 0.2 wt, Melaleuca alternifolia (tea tree) leaf oil 0.2 wt, aloe vera leaf extract 0.05 wt% were added to a mixer to prepare a single layer acne patch composition as shown in the following figure [Table 1].

[0055] [Table 1]

[0056]

[0057] The composition was heated at 80°C for 30 minutes and melted. Then the molten composition was injected into a circular mold and cooled to produce a circular patch. The thickness of the patch was 0.5 mm and the diameter was 15 mm.

[0058] By evaluating the physical properties such as adhesion, shape retention, and moisture permeability of the manufactured patch, it showed excellent properties.

[0059] Example 2. Manufacture of a multi-layer structure acne patch of the present invention

[0060] The same composition as in Example 1 was prepared, but was divided into three layers.

[0061] - Inner ingredient release control layer composition: water gel adhesive 20 Wt, hydrogenated hydrocarbon 15 Wt%, sodium carboxymethyl cellulose 10 Wt%, sodium carboxymethyl cellulose 0.2 Wt%, tea tree leaf oil 0.2 Wt%, aloe extract 0.05 Wt%.

[0062] - Middle moisturizing layer composition: polyethylene glycol 600 40% by weight, polyethylene-polypropylene glycol 20% by weight.

[0063] - Outer adhesive layer composition: mineral oil 60% by weight.

[0064] After melting the three compositions at 80°C, they were stacked into a concentric multi-layer structure, injected into a circular mold, and cooled to produce a multi-layer structure patch.

[0065] The manufactured multi-layer patch simultaneously showed excellent drug release control, moisture permeability, and adhesion.

[0066] Example 3. Manufacture of an acne patch using a non-woven fabric support

[0067] Example 1 was prepared by adding a non-woven fabric support to the composition.

[0068] After applying the composition of Example 1 to the non-woven fabric support, it was heat cured at 85°C for 30 minutes to adhere to the non-woven fabric and cooled.

[0069] By nonwoven support, the manufactured patch shows better shape retention.

[0070] It has been confirmed that the acne patch manufactured by the embodiment of the present application exhibits excellent performance and can be effectively used for acne skin care and treatment.

[0071] Example 1 is the simplest single-layer structure, Example 2 is a multi-layer structure separating the functions of each layer, and Example 3 is a structure using nonwoven support to improve shape retention.

[0072] Experimental Example 1. Efficacy evaluation

[0073] The following experiments were conducted to evaluate the clinical efficacy of the hydrocolloid preparation in treating acne according to the present application.

[0074] Human application test: A randomized, double-blind, controlled clinical trial was conducted for 8 weeks on 60 patients with moderate to severe acne. In the test group (30 people), the hydrocolloid preparation according to the present application was applied to the acne lesion site, and in the control group (30 people), a commercially available acne treatment agent (benzoyl peroxide 5% gel) was applied to the entire face. Since the total number of acne and the number of inflammatory lesions were measured during biweekly visits, the test group showed a 65% and 72% average decrease in the total number of acne and the number of inflammatory lesions, respectively, after 8 weeks, showing a better therapeutic effect than the control group (58% and 62% reduction). No adverse events were observed during the trial.

[0075] The mechanism of action was analyzed by identifying the antibacterial, anti-inflammatory, and moisturizing effects of human keratinocytes and skin-derived macrophages treated with aloe vera, tea tree oil, and polyethylene glycol, which are the main components of the preparation according to the present application. Aloe vera and tea tree oil were shown to inhibit acne bacteria and suppress the production of inflammatory cytokines, while polyethylene glycol was found to help moisturize the skin by increasing the water content in the cells. In addition, when the three ingredients were used together, a synergistic effect was confirmed by showing higher efficacy compared to each single treatment.

[0076] Physicochemical characterization by evaluating the adhesion, skin adhesion, and active ingredient release behavior of the preparation according to the present application, it was found that it exhibited excellent skin adhesion and durability according to the properties of the hydrophilic colloid, and the main active ingredients were gradually released for more than 12 hours. It was thus confirmed that a long-term continuous effect on the acne lesion area can be expected.

[0077] The patient availability evaluation, which is a usability and satisfaction survey conducted on patients in the clinical application test, showed that 83% of the patients were satisfied with the ease of use of the product and 73% of the patients thought that the effect was improved compared to the existing treatment method. In particular, it can be used freely day and night and is evaluated to have good treatment continuity since it adheres continuously over time.

[0078] As described above, it is confirmed that the hydrocolloid preparation for treating acne according to the present application has an overall effect of antibacterial, anti-inflammatory, and moisturizing, and can show excellent therapeutic effects by continuously acting on the acne lesion area. In addition, it is considered to have a high degree of convenience and preference compared to the existing full application formulation, and thus it can be recognized as a progressive treatment for acne.

[0079] Experimental Example 2. Formulation evaluation

[0080] The following experiment was conducted to compare and evaluate the hydrocolloid adhesive preparation for treating acne according to the present application with the existing full skin application formulation.

[0081] The application site and adhesion force evaluation applied the formulation according to the present application and a commercially available benzoyl peroxide 5% gel product to the lesion site and the entire face of 10 patients with moderate to severe acne, respectively, and observed the residual amount after 6 hours. As a result, the formulation according to the present application was adhered only to the acne lesion site, just like when it was initially applied, whereas the benzoyl peroxide gel was mostly peeled off. From this, it is confirmed that the formulation according to the present application has selective but excellent adhesion to the acne lesion.

[0082] Experimental Example 3. Evaluation of active ingredient release behavior

[0083] Since the amount of tea tree oil and polyethylene glycol released over time in the formulation according to the present application was measured, it showed a continuous behavior of rapid release within the first 3 hours and gradually up to 12 hours thereafter. On the other hand, in the benzoyl peroxide gel, most of the active ingredients were released within the first 2 hours, and it is expected that the effect will dissipate in a short time.

[0084] Evaluation of patient compliance and moisturizing effect 30 patients with acne used the formulation according to the present application, benzoyl peroxide gel, and Cleanderm Perfectin solution, respectively, for 2 weeks, and then a survey was conducted. As a result, 87% of the users of the formulation according to the present application expressed that it is convenient to apply it only at a specific time of the day. In addition, 67% reported that the treatment selectively moisturizes the acne area without tightening the skin compared to the full application formulation.

[0085] In summary, the hydrocolloid adhesive formulation of the present application is selectively applied only to the acne lesion site, unlike the existing full skin application formulation, and can be expected to have a long-lasting effect through excellent adhesion and continuous release of active ingredients. In addition, it has improved functionality in terms of patient convenience and selective moisturization compared to the existing formulation.

[0086] Experimental Example 4. Evaluation of Physical Properties

[0087] The following tests were conducted to evaluate the physical properties of the hydrocolloid adhesive formulation for treating acne according to the present application.

[0088] Thickness Measurement The thickness of the formulation according to the present application was measured on 6 samples, and the result was 309 ± 2 μm, satisfying the target specification of 310 ± 30 μm.

[0089] Adhesion Measurement After the formulation according to the present application was attached to a stainless steel plate, the peeling force required was measured by peeling at an angle of 180°. The average adhesion of 6 samples was 1083 ± 28 gf / 12 mm, satisfying the target specification of 150 gf / 12 mm or more.

[0090] Tensile Strength Measurement The formulation according to the present application was cut to a certain size, and the tensile strength was measured using a universal testing machine. The average tensile strength of 6 samples was 0.80 ± 0.03 Kgf / 10 mm, satisfying the target specification of 0.7 Kgf / 10 mm or more.

[0091] Absorbance Measurement The formulation according to the present application was cut to a certain size, and then immersed in a saline solution for 24 hours to measure the weight gain rate. As a result, the average absorption amount was 3.96 ± 0.07 g / 10 cm2·24 hrs, satisfying the target specification of 0.1 g / 10 cm2·24 hrs.

[0092] Experimental Example 5. Measurement of Moisture Permeability

[0093] The moisture permeability of the formulation according to the present application was evaluated using a measuring cup method. The average moisture permeability of 6 samples was 0.29 ± 0.01 g / 10 cm2·24 hrs, satisfying the target specification of 0.05 g / 10 cm2·24 hrs or more.

[0094] From the above results, it was confirmed that the hydrocolloid adhesive formulation according to the present application has appropriate thickness, excellent adhesion and tensile strength, and appropriate water absorption and moisture permeability levels. In this way, it is expected to stably adhere to the acne lesion site and allow continuous release of active ingredients.

[0095] Experimental Example 6. Evaluation of Durability

[0096] The following experiment was conducted to evaluate the lifespan of the acne patch according to the present application.

[0097] Specimen preparation: The acne patch was prepared in the same manner as in Example 1.

[0098] Test method: The manufactured patch was attached to the arm of 20 subjects, and the patch adhesion was visually observed and scored at 2-hour intervals for 6 to 24 hours. The durability score was as follows:

[0099] 5 points - the patch was completely attached

[0100] 4 points - the edges of the patch were slightly raised

[0101] 3 points - 25% of the patch area was away from the patch.

[0102] 2 points - 50% of the patch area was away from the patch

[0103] 1 point - the patch was completely closed

[0104] Results: The acne patch of the present invention showed an average high durability score of 4.2 points after 24 hours, confirming its excellent durability.

[0105] Experimental Example 7. Evaluation of skin adhesion

[0106] The following experiment was conducted to evaluate the skin adhesion.

[0107] Specimen preparation: The acne patch was prepared in the same manner as in Example 1.

[0108] Test method: After attaching the patch of the present invention to an artificial skin model having similar properties to human skin, the adhesion was measured by a 180-degree peeling test.

[0109] Results: The skin adhesion of the patch of the present invention was 120 gf / 25 mm, having excellent adhesion.

[0110] Experimental Example 8. Evaluation of shape stability

[0111] The following experiment was conducted to evaluate the shape stability.

[0112] Specimen preparation: Single-layer patches and multi-layer structure patches were prepared using the methods of Example 1 and Example 2, respectively.

[0113] Test method: The prepared patches were placed in a condition of accelerated aging at 40°C and 80% relative humidity for 4 weeks, and the change in the shape of the patch was periodically observed.

[0114] Results: The single-layer patch began to deform after 3 weeks. On the other hand, even after 4 weeks, the multi-layer patch did not show shape deformation, confirming excellent shape stability.

[0115] Experimental Example 9. Evaluation of biodegradability

[0116] The following experiment was conducted to evaluate the biodegradability.

[0117] Preparation of the sample: The patch containing the nonwoven fabric carrier was prepared in the same manner as in Example 3.

[0118] Test method: The prepared patch was buried in a soil medium for 24 weeks, and then the biodegradability was evaluated by measuring the weight loss rate periodically.

[0119] Results: The weight loss rate of the patch of the present application was about 75% after 24 weeks, and it had excellent biodegradability.

[0120] Experimental Example 10. Evaluation of skin soothing effect

[0121] The following experiment was conducted to evaluate the soothing effect on the skin.

[0122] Preparation of the sample: The acne patch was prepared in the same manner as in Example 1.

[0123] Test method: The artificial patch was applied to 20 patients with acne-prone skin, and their skin conditions were observed after 2 weeks. The skin soothing effect was evaluated by visual observation and measurement of the erythema index.

[0124] As a control group, the experimental group using a conventional acne patch not containing natural ingredients was separately manipulated.

[0125] Results: In the experimental group using the patch of the present application, it was observed that the erythema of the skin after removal of the patch was significantly reduced, and the erythema index was decreased by 45% compared to the control group, confirming the excellent skin soothing effect.

[0126] Through the above examples and experimental examples, it was confirmed that the acne patch of the present application has excellent life, adhesion, shape stability, biodegradability, and skin soothing effect.

[0127] Therefore, the single-layer structure acne patch shown in Example 1 of the present application exhibited excellent adhesion, durability, and skin soothing effect, but was inferior in terms of adjusting the release of the acne-improving ingredient and moisturizing power to Example 2.

[0128] The multi-layer acne patch manufactured as shown in Example 2 of the present application also exhibited the best overall performance in terms of adjusting the release of the acne-improving ingredient and moisturizing power.

[0129] The analysis of the ingredient release profile of the patch prepared by including the inner side ingredient release control layer composition in the inner side ingredient release control layer showed that it had a mild and sustained drug release behavior for 48 hours after the initial rapid release. This seems to mean that it has better drug release control compared to the existing single-layer patch.

[0130] The moisture permeability and moisturizing power were very good with a moisture permeability of 59% for the entire patch due to the intermediate moisturizing layer. In addition, the moisturizing power between the patch and the skin remained 90% or more even after 24 hours, showing excellent long-term moisturizing effects.

[0131] The adhesion of the outer adhesive layer to the patch skin was 125 gf / 25 mm, showing a similar excellent level to the single-layer patch of Example 1.

[0132] In terms of durability and shape stability, the multi-layer patch prepared in Example 2 showed a very high durability score of 4.5 points on average after 24 hours, and no shape deformation was observed even under accelerated aging for 4 weeks, confirming excellent conformational stability.

[0133] When applied to patients with acne skin, the redness index was reduced by 52% compared to the control group, improving the skin soothing effect more than the single-layer patch of Example 1.

[0134] Thus, the multi-layer structure acne patch of Example 2 showed the most excellent overall performance, such as drug release control, high moisture permeability and moisturizing power, excellent adhesion and durability, shape stability, and excellent skin soothing effect.

[0135] Compared to Example 2, the nonwoven fabric support acne patch of Example 3 of the present application showed better adhesion and shape stability, while the results in terms of drug release, moisture permeability, moisturizing power, etc. were not as good as Example 2.

[0136] In conclusion, it can be judged that the components of Examples 1 to 3 of the present application have excellent effects as acne patch products when manufactured, and more preferably, the multi-layer acne patch (Example 2) showed the most excellent overall performance, making it the most suitable as a patch for acne treatment and management.
