BRIEF DESCRIPTION OF DRAWINGS

[0007] Figure 1 Graphs detailing the results of the cleaning efficacy tests are depicted. DETAILED DESCRIPTION

[0008] As used herein, weight percent (wt.%), "percent by weight," "percent weight," %, and the like are synonyms that refer to the concentration of a substance as the weight of that substance divided by the total weight of the composition and multiplied by 100.

[0009] As used herein, the term "about" to modify the amount of an ingredient in the compositions of the present application or used in the methods of the present application refers to variation in the value that can occur, e.g., through typical measuring and liquid handling procedures used in preparing the use solutions in the real world; through inadvertent error when preparing the compositions or practicing the methods; through differences in the manufacture, source, or purity of the ingredients employed in making the compositions or practicing the methods; and the like. The term about also encompasses variations that occur due to different equilibration conditions for a composition that is produced from a particular initial mixture. Whether modified by the term "about" or not, the claims include equivalent values. The term "about" generally allows for variations of 5% in the recited percentage value. For example, if "about 10%" is used, the term "about" generally allows for variations of about 5% in the recited percentage value. Thus, "about 10%" can encompass + / - 0.5% variation.

[0010] It should be noted that, as used in this specification and the appended claims, the singular forms "a," "an," and "the" include plural referents unless the content clearly dictates otherwise. Thus, for example, reference to a composition containing "a compound" includes a mixture of two or more compounds. It should also be noted that the term "or" is generally employed in its sense including "and / or" unless the content clearly dictates otherwise.

[0011] For the purpose of brevity and clarity, any range of values stated in the present specification envisions all values within the range and is to be construed as supporting the requirement of claim limitations that recite any sub-range of values within the specified range. By way of illustrative example, a disclosure of from 1 to 5 in the present specification is to be interpreted to support a claim reciting any of the following ranges of values: 1-5; 1-4; 1-3; 1-2; 2-5; 2-4; 2-3; 3-5; 3-4; and 4-5.

[0012] As used herein, "cleaning" refers to the removal of organic or inorganic soils. In some examples, "cleaning" can refer to any process that aids in the removal of soils.

[0013] As used herein, the term "disinfecting" refers to the biological action of killing vegetative cells and reducing the population of microorganisms, including most recognized pathogenic microorganisms.

[0014] As used herein, the term "microorganism" refers to any non-cellular or unicellular (including colonial) organism. Microorganisms include all prokaryotes. Microorganisms include bacteria (including cyanobacteria), spores, lichens, fungi, protozoa, virioplankton, viroids, viruses, phages, and some algae. As used herein, the term "microbe" is synonymous with microorganism.

[0015] In the field of utilizing clean rooms, it is important to maintain a controlled environment, limiting or eliminating the growth of microorganisms, such as bacteria, yeast, or fungi. Additionally, it is important that these environments not only be free of microbial contamination, but also free of visible stains or soils. Thus, antimicrobial compounds must also be nearly foam-free when applied, and must leave minimal or no residue. Many commercially available antimicrobial agents rely on the use of quaternary ammonium disinfectants, which have high potency against Gram-positive bacteria, but reduced potency against Gram-negative bacteria, and little impact on yeast and fungi.

[0016] One alternative method of eliminating the growth of microorganisms is the use of amine-based antimicrobial compounds. Previous amine-based antimicrobial compositions have been limited by factors including limited microbial potency spectrum, poor cleaning performance, incompatibility with target surfaces, and / or foaming or residue formation. Additionally, some existing antimicrobial compositions do not achieve a desired 3-log reduction within a target time of 2, 5, or 10 minutes of contact with a target surface.

[0017] Surprisingly, it has been discovered that the disclosed biocidal amine in combination with alkanolamine is effective against yeast in a short period of time. In addition to achieving the desired log reduction against yeast in the desired time, the disclosed formulations are effective against a range of microorganisms including gram-negative and gram-positive bacteria as well as yeast, exhibit minimal foaming and residue formation, and are compatible with a range of surfaces including glass, polymers, PVC, pharmaceutical terrazzo, epoxy, and stainless steel. The compound is also free of quaternary ammonium compounds. The disclosed compound is particularly useful for cleanroom applications with stringent requirements, but is generally useful for antimicrobial applications including healthcare as well as food and beverage applications.

Preferably, the chelating agent is a phosphorus-based chelating agent, such as a phosphonate.

[0033] In some examples, the phosphorus-based chelating agent is a phosphonate. The phosphonate can be added to the concentrate in an amount of about 0.01 wt% to about 2 wt%, about 0.01 wt% to about 1 wt%, about 0.01 wt% to about 0.75 wt%, about 0.01 wt% to about 0.5 wt%, about 0.01 wt% to about 0.1 wt%, about 0.1 wt% to about 2 wt%, about 0.5 wt% to about 2 wt%, about 0.75 wt% to about 2 wt%, or about 1 wt% to about 2 wt%.

[0034] Additional functional ingredient

[0035] In some embodiments, the antimicrobial composition can optionally include additional functional ingredients. Exemplary additional functional ingredients include thickening or gelling agents, co-solvents, additional foaming agents, chelating agents, or mixtures thereof.

[0036] Thickener or gelling agent

[0037] In some applications, it can be desirable for the present compositions to have a greater viscosity or to take the form of a thickened liquid or gel. Exemplary thickening agents include those that do not leave a contaminating residue on the surface of application. Exemplary thickening agents include natural gums such as xanthan gum. Cellulose polymers such as carboxymethyl cellulose can also be used in the present invention. [0041] Additional surfactant or foaming agent

[0042] While the present disclosure emphasizes the need for minimal foaming to reduce undesirable residue in cleanroom applications, in other applications, additional foaming can be a desirable characteristic. The formation of foam can increase the contact time on non-horizontal surfaces, allowing additional contact time for the active ingredients with the target microorganisms to achieve the desired log reduction.

[0043] pH

[0044] In some embodiments, the disclosed composition has a pH in the range of about 7 to about 14, about 8 to about 13, about 9 to about 12, or about 10 to about 12.

[0045] Absence of quaternary ammonium compound

[0046] In some embodiments, the disclosed composition does not contain a quaternary ammonium compound. Quaternary ammonium compounds are commonly used antimicrobial agents in cleaning solutions; however, some potential users prefer alternative antimicrobial agents, such as the disclosed composition. The reluctance to use quaternary ammonium compounds can be due to high manufacturing costs and low efficacy against microorganisms other than gram-positive bacteria.

[0047] Method of inactivating microorganisms

[0048] The present disclosure relates to methods of inactivating microorganisms, particularly bacteria and yeast, in a short amount of time using the disclosed antimicrobial compositions. In some embodiments, the disclosed antimicrobial compositions have a log reduction of greater than 4 logs of bacteria in 2 minutes of contact, a log reduction of greater than 5 logs of bacteria in 2 minutes of contact, a log reduction of 4 logs to 6 logs of bacteria in 2 minutes of contact, a log reduction of 4 logs to 7 logs of bacteria in 2 minutes of contact, a log reduction of 5.5 logs to 6.5 logs of bacteria in 2 minutes of contact, or a log reduction of 6 logs to 6.5 logs of bacteria in 2 minutes of contact at room temperature and without mechanical action. Exemplary test organisms include S. aureus, E. hirae, E. coli, and P. aeruginosa according to EN 13697.

[0049] In some embodiments, the disclosed antimicrobial composition has a 3 log or greater log reduction of yeast within 2 minutes of contact, a 2 log to 5 log log reduction of yeast within 2 minutes of contact, a 3 log to 4 log log reduction of yeast within 2 minutes of contact, or a 3.5 log to 4 log log reduction of yeast within 2 minutes of contact on a surface at room temperature without mechanical action according to EN 13697.

[0050] In some embodiments, the disclosed antimicrobial composition has a greater than 3 log log reduction of fungi within 5 minutes of contact on a surface without mechanical action, a greater than 3.5 log log reduction of fungi within 5 minutes of contact, a greater than 4 log log reduction of fungi within 5 minutes of contact at room temperature, a 3 log to 5 log log reduction of fungi within 5 minutes of contact at room temperature according to EN 13697.

[0051] Microbial strain

[0052] A variety of microorganisms were used in the testing to confirm the efficacy of the disclosed compositions. Among these microorganisms, both gram-positive and gram-negative bacterial strains were tested. The gram-positive bacterial strains used included Enterococcus hirae and Staphylococcus aureus. The gram-negative strains used included Escherichia coli and Pseudomonas aeruginosa. The yeast strain Candida albicans was also tested, as was the fungal strain Aspergillus brasiliensis. These strains are a sampling of potential species that can utilize the current compositions, and in no way encompass the antimicrobial efficacy of the current compositions. The test used was EN 13697.

[0053] Application

[0054] In some embodiments, the disclosed compositions are applied to hard surfaces located in research areas, manufacturing facilities, laboratories, clean rooms, hospitals, resorts, hotels and motels, commercial real estate, schools, and food or beverage manufacturers, among others. Amine-based antimicrobial agents are ideal for hard surfaces that are susceptible to microbial contamination or growth but do not have high soil content, as amines tend to break down and lose functionality upon exposure to soil. Thus, the present compositions are advantageous for use in clean rooms, pharmaceutical manufacturing facilities, laboratories, schools, resorts, hotels and motels, commercial real estate, hospitals, and other areas with minimal soil. Exemplary hard surfaces include counters, sinks, walls, drains, floors, ceilings, tables, carts, benches, transfer hatches, transfer pods, storage areas, cutting boards, knives, laboratory equipment, laboratory instruments, tanks, pipes, evaporators, membranes, heat exchangers, spray heads, nozzles, controls, tools, handles, remote controls, toilet handles, toilet seat covers, bedpan flushers, faucet handles, door knobs or handles, push plates, grab plates, toilet area grab bars, phone receivers, call buttons, tables, window curtains, restricted access barrier systems (RABS), clean room equipment garments, personal protective equipment, laboratory hoods, safety cabinets, laminar flow cabinets, clean room isolators, chair surfaces, chair arms, bed rails, drapes, room light switches, computer mice, computer keyboards, keyboard wrist rests, soap dispensers, and the like.

[0055] Example

[0056] Example 1

[0057] Example 1 tested various formula products for efficacy against various microorganisms, including bacteria and yeast. During the test, various undiluted formula products were contacted with multiple bacterial strains and the yeast strain Candida albicans under clean conditions at room temperature for a period of 5 minutes or 10 minutes according to test procedure EN 13697. To be considered having bactericidal activity, a formula product was required to exhibit at least a 4 log reduction after 5 minutes of exposure, up to 60 minutes of exposure. To be considered having a yeast killing activity, a formula product was required to exhibit at least a 3 log reduction after five minutes of exposure, up to 60 minutes of exposure. The test was performed on experimental formula products 1 listed in Table 1 and compared to a commercially available product, Klercide Low Residual Quaternary Ammonium Salt, from Ecolab. The results are shown in Table 1.

[0058] Experimental formulation 1

[0059]

[0060] Table 1

[0061]

[0062] The data shown in Table 1 indicate that experimental Formula Product 1 was able to achieve the desired 4 log reduction against bacteria after 5 minutes of exposure and the desired 3 log reduction against yeast after five minutes of contact.

[0063] Example 2

[0064] Example 2 tested the foaming performance of experimental Formula Product 1 over time. For this test, 100 ml of experimental Formula Product 1 was placed in a 250 ml graduated cylinder and shaken evenly and rhythmically by hand for 20 times. The height of the foam produced was measured on the graduated cylinder immediately after shaking the graduated cylinder, 1 minute after shaking the graduated cylinder, and five minutes after shaking the graduated cylinder. The results of this experiment are shown in Table 2.

[0065] Table 2

[0066]

[0067] The results shown in Table 2 indicate that some foam formation occurred immediately after application. These results also indicate that the foam decreased rapidly over time. These results are desirable for cleanroom applications because some foam formation helps to support cleaning, but excess foam formation can lead to residue or smudging.

[0068] Example 3

[0069] Example 3 tested the residue characteristics of various formula products on stainless steel surfaces as a result of spraying and wiping. When experimental Formula Product was sprayed on stainless steel and allowed to evaporate, a slight residue was left behind.

[0070]

[0071] When sprayed on glass, or when wiped on glass or stainless steel and allowed to evaporate, no residue was left behind.

[0072] Example 4

[0073] Example 4 tested the cleaning characteristics of various formula products. Along with experimental Formula Product 1, two commercial formula products were tested, Klercide Neutral Detergent and Anios Clean Surf, both of which are commercially available from Articon Group. These formula products were tested with tap water as a control.

[0074] The test was done using the Gardner Rub Test (Gardner device model 494; DIN-ASTM-515; Erichsen GmbH & Co. KG). For the test, white PVC test coupons were coated with 2 grams of soil (IPP 83 / 21, available from WFK in Krefeld, Germany). 12.5 ml of test detergent or tap water control was applied to the test strip, and another 12.5 ml of test detergent or control was added to the sponge on the device, for a total of 25 ml per test cycle. The test strip was then rubbed back and forth over the surface of the coated test strip using a polyester flat sponge for 100 cycles. After the test was complete, the test strips were removed from the machine, rinsed, dried, and their degree of cleaning was evaluated and judged by measuring the whiteness of the test strips. The whiteness of the test coupons was measured with a Minolta colorimeter CR-200, and the percentage of soil removed was recorded. If the test coupon was completely clean (all soil was removed), the test had a measurement of 100%. Seven replicate tests were done for each cleaning solution. The results of the experiment are shown in the percent cleaning in Tables 3 and Figure 1

[0075] Table 3

[0076]

[0077]

[0078] The results shown in Tables 3 and Figure 1 show that the average cleaning efficacy of the experimental formula products was 85.55%. The highest average efficacy was observed with the commercial formula product Klercide Neutral Detergent, which had an average efficacy of 87.48%. The lowest average efficacy of the Anios cleaning surfactant was 68.10%, while the average cleaning efficacy of the control was 64.32%. This data indicates that the disclosed compositions have higher or comparable cleaning efficacy than other commercially available cleaning products.

[0079] Example 5

[0080] Examples 1-4 tested the performance of experimental formula product 1 under low soiling conditions. Example 5 compared the performance of experimental formula product 1 under low soiling conditions and high soiling conditions.

[0081] Table 4 shows the results of testing experimental formula product 1 under low and high soiling conditions. A total of eighteen tests were conducted. Experimental formula product 1 was evaluated as a bactericide, a yeasticide, and a virucide against enveloped viruses under low soiling conditions. Experimental formula product 1 was tested in each of these three functions in suspension tests, surface tests without mechanical action, and surface tests with mechanical action. For each of these variables, the contact time was measured in minutes. The time period in Table 4 represents the number of minutes that experimental formula product 1 was required to satisfactorily function as a bactericide, a yeasticide, and a virucide according to the requirements of each of the three tests. The same procedure was conducted under high soiling conditions.

[0082] Table 4

[0083]

[0084] The results in Table 4 show that experimental formula product 1 provides substantially the same cleaning under low and high soiling conditions. For example, experimental formula product 1 adequately functions as a bactericide under low and high soiling conditions for the same time period. This is advantageous because experimental formula product 1 can be used as a bactericide, a yeasticide, and a virucide in both low and high soiling applications, which increases the applicability of the formula product.
