Abstract
The present application relates to the technical field of lutein extraction, and provides a green extraction method of lutein from marigold, which comprises the following steps: mixing marigold dry flower particles with deionized water to prepare slurry, and performing high-pressure homogenization treatment to obtain pretreated slurry; mixing the pretreated slurry with a deep eutectic solvent (DES) and performing countercurrent extraction under the protection of inert gas to obtain DES extraction liquid after filtration; preheating the DES extraction liquid and pumping it into a continuous flow fixed-bed enzyme reactor filled with immobilized lipase, and performing enzymolysis reaction under constant temperature to obtain an enzymolysis liquid; sequentially treating the enzymolysis liquid through a ceramic ultrafiltration membrane and a nanofiltration membrane; collecting the retentate of the nanofiltration membrane, adding preheated deionized water to the nanofiltration retentate, and performing low-temperature standing crystallization; and washing and vacuum freeze-drying the obtained crystals to obtain a lutein crystal product. Through the above technical scheme, the extraction rate of lutein is improved, and the problem of incomplete extraction caused by insufficient cell structure damage in the traditional extraction method is improved.
