Description

Title of the invention: COSMETIC COMPOSITION, USE OF THE COSMETIC COMPOSITION AND METHOD FOR MANUFACTURING THE COSMETIC COMPOSITION FIELD OF THE INVENTION

[0001] The present invention relates to a cosmetic composition comprising (a) iron oxides chosen from yellow, black and red iron oxides, (b) titanium oxide; (c) a mixture of niacinamide, tranexamic acid, hydroxyethylpiperazineethanesulfonic acid and (d) at least one UV filter. The present invention also relates to the use of the cosmetic composition for homogenizing the skin, treating hyperpigmentation and photoprotecting skin tones 1 to 6 (Fitzpatrick scale) and to the method of manufacturing the cosmetic composition. CONTEXT OF THE INVENTION

[0002] Widely used to protect the skin from sun damage, cosmetic sunscreen compositions constitute a large part of the market.

[0003] Due to the high demand for multi-benefit products, offering different benefits in the same product, consumers are always looking for products that provide high UV protection with a makeup effect and medium to high coverage and that adapt to various skin tones and undertones.

[0004] Among the scales for accessing the skin type of subjects, an example is the Fitzpatrick scale. In particular, a phototype 1 of said scale is associated with very light skin, red or blond hair, blue or green eyes and skin that always burns and / or never tans in the sun, phototype 5 is commonly associated with dark brown skin, brown or black hair, brown eyes and skin that browns easily and rarely burns when exposed to the sun, and phototype 6 is commonly associated with black skin, black hair, dark brown eyes and skin that does not burn when exposed to the sun.

[0005] Photoprotection of keratin materials, including skin and hair, is considered of great importance in order to protect against sun damage, sunburn, photoaging, as well as to reduce the risks of developing skin cancer caused by exposure to ultraviolet ("UV") rays. There are generally two types of UVA / UVB sunscreen cosmetic compositions used to accomplish photoprotection, namely inorganic UV filters and organic UV filters.

[0006] The degree of UV protection provided by a cosmetic composition is directly related to the quantity and type of UV filters it contains. In particular, cosmetic compositions must provide good protection against the sun's rays, measured by the Sun Protection Index (SP) value, a desirable balance between UVA and UVB protection, in particular a minimal UVA protection factor, while exhibiting a satisfactory sensory perception, such as a smooth, dry feel and a non-greasy sensation upon application.

[0007] Alongside photoprotection, consumers have a common interest and need for spot camouflage, reduction of spot intensity, skin homogenization, makeup and prevention.

[0008] However, in the sunscreen category, the compositions are mainly aimed at skin camouflage and prevention with tinted photoprotection formulas. In this sense, it is necessary to develop compositions that work on the three pathways of complexion management (skin homogenization, spot treatment and prevention against UVA / UVB and visible light) in a single product for their daily routine.

[0009] In other words, the inventors observed the need to simultaneously ensure the homogenization and camouflage of spots, the treatment of skin hyperpigmentation and the protection of the skin against UVA / UVB rays and visible light.

[0010] Thus, the objective of the invention is to provide a cosmetic composition by which the consumer can actually benefit from different perspectives of treating spots, achieving high clinical performance after periodic use in terms of size, intensity and reduction in the number of spots; providing protection against visible light; providing instant camouflage of marks and also providing photoprotection.

[0011] Among the disadvantages of the state of the art, besides the known challenges of developing a cosmetic composition with UV filters (chemical protection) and a high concentration of pigments (physical protection) due to the stability of the formulas and the good matching of skin tones with the tinted product, there is an additional challenge related to the market of providing a single stable routine product to homogenize and camouflage spots, treat skin hyperpigmentation and protect the skin from UVA / UVB rays and visible light.

[0012] Therefore, the inventors of the present invention have surprisingly succeeded in developing a stable cosmetic composition to cover the three main parameters for skin tones 1 to 6: i. homogenization of the skin and camouflage of spots, ii. treatment of the skin, and iii. sun protection / prevention by two complementary routes (protection UVB / UVA and visible light protection). Summary of the invention

[0013] The present invention relates to a cosmetic composition comprising (a) iron oxides chosen from yellow, black and red iron oxides, (b) titanium oxide; (c) a mixture of niacinamide, tranexamic acid, hydroxyethylpiperazineethanesulfonic acid and (d) at least one UV filter.

[0014] The present invention also relates to the use of the cosmetic composition for homogenizing the skin, treating hyperpigmentation and photoprotecting skin tones 1 to 6 (Fitzpatrick scale) and to the method of manufacturing the cosmetic composition.

[0015] Other features and advantages of the present invention will become apparent from the following more detailed description of desirable embodiments which illustrates, by way of example, the principles of the invention. Brief description of the drawings

[0060] In the context of the present invention, the iron oxides of the cosmetic composition for providing dark shades of the present invention are coated or uncoated iron oxides.

[0061] Examples of coated iron oxides that may be mentioned more particularly include aluminum stearoyl glutamate coated iron oxides sold, for example, under the reference NAI by the company Miyoshi Kasei. Other preferred examples of coated iron oxides include iron oxides coated with disodium stearoyl glutamate and aluminum hydroxide. Iron oxides such as CI 77491, CI 77492 and CI 77499 may be used in the present composition and are supplied by TITAN KOGYO.

[0062] For the purposes of the present invention, “coating” of a pigment generally designates the total or partial surface treatment of the pigment with a surfactant, absorbed, adsorbed or grafted onto said pigment.

[0063] The coating may represent from about 0.1 to about 20.0% by weight and in particular from about 0.5 to about 5.0% by weight, relative to the total weight of the coated pigment.

[0064] The coating can be carried out, for example, by adsorption of a liquid surfactant on the surface of the solid particles by simple mixing with agitation of the particles and said surfactant, optionally with heating, before incorporation of the particles into the other ingredients of the makeup or care composition.

[0065] The coating can be carried out, for example, by chemical reaction of a surfactant with the surface of the solid pigment particles and creation of a covalent bond between the surfactant and the particles. This method is notably described in US patent 4,578,266.

[0066] The chemical surface treatment may consist of diluting the surfactant in a volatile solvent, dispersing the pigments in this mixture, and then slowly evaporating the volatile solvent, so that the surfactant is deposited on the surface of the pigments.

[0067] The cosmetic composition of the present invention comprises pigments chosen from yellow, black and red iron oxides.

[0068] The cosmetic composition of the present invention comprises from about 1.0 to about 30.0 wt. %, from about 5.0 to about 20.0 wt. %, from about 6.0 to about 18.5 wt. %, from about 7.0 to about 17.0 wt. %, from about 10.0 to about 15.5 wt. %, including all intermediate ranges and sub-ranges, based on the total weight of the composition, of yellow, black and red iron oxides.

[0069] In addition, the cosmetic composition of the present invention may comprise additional pigments.

[0070] The term "pigments" is intended to designate white or colored particles, mineral or organic, which are intended to color and / or opacify the composition and / or the resulting deposit. These pigments may be white or colored, and mineral and / or organic.

[0071] The term "mineral pigment" is intended to mean any pigment that meets the definition in Ullmann's Encyclopedia in the chapter on inorganic pigments, with the exception of titanium dioxide pigment. Additional mineral pigments useful in the present invention include zirconium oxide or cerium oxide, and also zinc oxide, additional iron oxides (i.e., other than yellow, red, and black iron oxides) or chromium oxide, manganese violet, ultramarine blue, chromium hydrate, and ferric blue, and metal powders, for example, aluminum powder and copper powder. The following mineral pigments may also be used: Ta2O5, Ti3O5, Ti2O3, TiO, ZrO2 as a mixture with ZrO2, Nb2O5, CeO2, or ZnS. Titanium oxides

[0072] In the present cosmetic composition, the preferred titanium oxide is titanium dioxide (TiO2), and TiO2 is only a pigment in the present invention, not a UV filter.

[0073] As mentioned, the presence of titanium dioxide in cosmetic compositions is associated with the ashy appearance after application to the skin and the white effect, particularly in photographs. Nevertheless, it is important to differentiate titanium dioxide used as a pigment, herein referred to as “titanium dioxide pigment”, from titanium dioxide used as a UV filter, here called “titanium dioxide UV filter”.

[0074] Titanium dioxide can take several different forms, which have different properties. In particular, the main difference between pigment titanium dioxide and UV filter titanium dioxide is their particle size, with the pigment being larger than the UV filter. At small sizes and low concentrations, titanium dioxide appears as minimal or non-white residue, allowing for effective sunscreens that do not appear white.

[0075] Pigment titanium dioxide is the most widely used white pigment due to its brightness and very high refractive index, in which it is surpassed by only a few other materials. The crystal size of pigment titanium dioxide is ideally above 220 nm, even reaching a micrometer scale, such as about 7,000 nm (measured by electron microscope) to optimize the maximum reflection of visible light. Pigment titanium dioxide is also an effective opacifier in powder form, where it is used as a pigment to provide whiteness and opacity to products such as paints, coatings, plastics, papers, inks, foods, medicines (i.e., pills and tablets) and most toothpastes. Pigment titanium dioxide is able to scatter light, which allows it to be used more for pigments.

[0076] Furthermore, nanoscale titanium dioxide, which should also be considered herein as a synonym for “UV filter titanium dioxide,” is found in the majority of physical sunscreens due to its high UV light absorption capabilities and resistance to fading under ultraviolet light. This advantage enhances its stability and ability to protect the skin from ultraviolet light. Nanoscale titanium dioxide particles (particle size less than 200 nm) are primarily used in sunscreen compositions because they scatter much less visible light than titanium dioxide pigments and can provide UV protection.

[0077] The cosmetic composition of the present invention comprises from about 1.0 to about 15.0 wt. %, from about 5.0 to about 18.0 wt. %, from about 6.0 to about 15.0 wt. %, from about 7.0 to about 12.0 wt. %, from about 8.0 to about 10.0 wt. %, including all intermediate ranges and sub-ranges, based on the total weight of the composition of titanium oxide pigments. [0154] Test 1 - Ex vivo sebum photoperoxidation test (Comparison of the antioxidant efficacy of the formula of inventive example 1 compared to untreated sebum (control)) I. Objective

[0155] The objective was to evaluate the antioxidant effect of the formula of inventive example 1 compared to untreated sebum alone, by UVA peroxidation of the sebum “ex vivo”.

[0156] The inventors verified that the formula of inventive example 1 was effective against untreated sebum, which is observed by an inhibition of sebum peroxidation of approximately 80% under our test conditions. II. Test conditions

[0157] A layer of sebum is taken from the forehead of a volunteer on two inert supports. Approximately 3 mg / cm2 of the active formula to be tested is applied to the first sample, without applying anything to the second sample (untreated sebum control).

[0158] The samples are exposed to UVA at 5 joules / cm2. Photo-oxidation of sebum leads to the formation of squalene peroxides, and the variation in peroxidation allows us to compare the effectiveness of the active formula compared to sebum alone (see [Fig.4]). III. Results

[0159] The results of the effectiveness of the formula of inventive example 1 compared to sebum alone are summarized in Table 1 below:

[0160] [Table 1] Table 1 - Effectiveness of the formula of example 1 compared to sebum alone UVA exposure 5 joules / cm2 SQOOH in pmoles / pg of SQ inhibition Control 940576 / 22 TRIAL 1 206 ± 8.7 39 + 2 84 ± 3% TRIAL 2 308 ± 13 40 + 0.9 IV. Review of results

[0161] The protection provided by the formula of inventive example 1 is effective, with an inhibition of approximately 80% compared to untreated sebum.

[0162] The degree of effectiveness can be explained by the combined effect of anti-UVA protection and antioxidant protection.

[0163] Test 2 - Study to evaluate pigment and anti-stain control

[0164] The protocol involved testing 5 shades on (i) all skin tones and (ii) all skin types over 8 weeks.

[0165] After the said 8 weeks, the results obtained revealed:

[0166] - with regard to complexion management, the inventors have observed results Surprisingly positive results related to clinical skin tone management for skin tones 1-6. Positive results show a reduction of (i) 22% in the number of spots; (ii) 46% in the intensity of spots; and (iii) 34% in the size of spots and a 35% improvement in skin radiance;

[0167] - regarding stain prevention, results were observed after 5 days, with SPF = 71.8 (6.7) + protection against visible light; and

[0168] - with regard to instant camouflage of marks on different phototypes, The inventors also observed improved results compared to prior art products.

[0169] The progress and improvements described above can be seen in [Fig.l] (number of spots), [Fig.2] (color intensity of the spot) and [Fig.3] (uniformity of the complexion).

[0170] Test 3 - Clinical study (Evaluation of the protective effects of four sunscreens on pigmentation induced by visible light compared to an untreated control area) Main objective:

[0171] Evaluate the protective effect of the inventive sunscreen composition to prevent visible light-induced pigmentation. Secondary objective:

[0172] Evaluate the local tolerance and overall tolerability of the test products based on the report of adverse effects. Methodology / Study Design:

[0173] This was a single-center, double-blind, randomized, controlled study with intra-individual comparisons.

[0174] The study consisted of a screening visit followed by six (6) assessment visits (Day 1, Day 2, Day 3, Day 4, Day 5 and Day 12). The study was conducted as follows:

[0175] A 3-week pre-screening period (within three (3) weeks prior to the baseline visit): Prior to any study procedure, subjects receive the necessary written and oral information, including the informed consent form. Eligibility was to be determined by physical examination and confirmation of all inclusion / non-inclusion criteria.

[0176] On Day 1, five (5) test areas were to be delineated on each side of the spine in the mid-back. Each test area consisted of an exposed portion (EZ) and a surrounding unexposed portion (NEZ). Two (2) duplicate reference colorimetric measurements were to be performed on both portions for each test area. The investigator was also to perform a clinical assessment of erythema and pigmentation for each test area and take standardized photographs.

[0177] Once the rest period was over, the five (5) test areas were exposed to a visible light dose of 144J / cm2.

[0178] From Day 2 to Day 4, the investigator was required to perform double baseline colorimetric measurements (for each test area and the surrounding skin), perform a clinical assessment of erythema and pigmentation for each test area.

[0179] Once the rest period was over, the five (5) test areas were exposed to a visible light dose of 144J / cm2.

[0180] On Day 4 only, a standardized photograph of the five (5) test areas was taken.

[0181] On Day 5 and Day 12 / End of study, the investigator was required to perform duplicates reference colorimetric measurements (for each test area and surrounding skin), perform a clinical assessment of erythema and pigmentation for each test area and take a standardized photograph for all five (5) test areas. Number of subjects (planned and analyzed):

[0182] Planned: 20; Shortlisted: 20; Engaged: 20; Completed: 20; and Analyzed: 20. Diagnosis and main inclusion criteria:

[0183] The subjects included in this study were healthy Caucasian subjects of both sexes aged 18 to 50 years, with skin type Illb to V according to the Fitzpatrick scale (1) and with an ITA° between -20 and 28° at pre-selection. Evaluation criteria: Main evaluation criterion:

[0184] Pigmentation intensity measured by the colorimetric angle of 1TTA° (between ZE and NEZ). Secondary evaluation criteria:

[0185] ❖ Pigmentation intensity measured by delta E between EZ and NEZ,

[0186] ❖ Colorimetric parameters (delta L*, delta a* and delta b*),

[0187] ❖ Clinical evaluation of pigmentation,

[0188] ❖ Clinical evaluation of erythema,

[0189] ❖ Visible light protection factors (VL-PF). Safety Assessments:

[0190] ❖ Local tolerance

[0191] ❖ General security: Adverse events reported continuously. Main statistical methods: Analysis of effects over time:

[0192] Data were analyzed using a mixed-effects model. This model included time, treatment, time*treatment interaction, and baseline as fixed effects.

[0193] Subject was added as a random effect. Comparisons between the post-baseline measurement point and baseline for each treatment were made regardless of interaction results using a Dunnett adjustment.

[0194] The degrees of freedom for the comparisons were approximated by the Kenward-Roger method. Analysis of treatment effects:

[0195] Change from baseline was analyzed using a mixed-effects model. This model included time, treatment, time*treatment interaction, and baseline as fixed effects. Subject was added as nested random effects. The comparison between the two treatments at each measurement point was performed regardless of the interaction outcome using a Tukey adjustment. Degrees of freedom for comparisons were approximated by the Kenward-Roger method. Summary of results:

[0196] The study was conducted as planned according to the protocol and no deviation from the protocol was observed. Results in terms of effectiveness:

[0197] Under the conditions of the present study, the analysis of the primary evaluation criterion (Delta ITA) led to the following conclusions:

[0198] ❖ Significant decreases in delta ITA° compared to the reference base have were observed from Day 2 for all test areas.

[0199] ❖ Significant differences were observed between all areas treated with tested and untreated formulations from Day 2.

[0200] Similar results were observed with the colorimetric parameter Delta L*. The same was true for Delta E although the results obtained were less pronounced.

[0201] Analysis of the clinical assessment of skin pigmentation corroborates the colorimetric measurements.

[0202] The highest protection factors were provided by the formula in the example 1 (39%). General conclusions:

[0203] In conclusion, under the study conditions, all four formulations tested were effective against visible light-induced pigmentation. The formula of Example 2 provided the best protective effects and its safety profile was excellent.
