Description

[Technical field]

[0001] The present invention relates to a cosmetic composition for enhancing exfoliation.

[0002] This application claims priority to Korean Patent Application No. 10-2018-0083069 filed on July 17, 2018, and Korean Patent Application No. 10-2019-0063704 filed on May 30, 2019, and the entire contents disclosed in the specification and drawings of the relevant applications are incorporated herein by reference. [Background technology]

[0003] The stratum corneum is the outermost layer of the skin and is composed of a lipid layer with a lamellar structure containing keratinocytes and ceramide. The stratum corneum protects against external substances and functions as a defense membrane that prevents moisture loss from the inside. A normal stratum corneum is composed of about 15 to 20 layers, and the decomposition of coniodesmosomes progresses due to the action of serine protease activated outside the stratum corneum, and eventually undergoes a process of complete detachment from the skin. This process takes about 15 to 20 days for normal cells. In addition to genetic diseases such as ichthyosis, aging, psoriasis, acne skin, etc., the period of the above keratin detachment process is longer than normal, and the stratum corneum shows a phenomenon of keratin stratification in which the stratum corneum becomes thicker.

[0004] It is known that stratum corneum is usually induced by factors such as a decrease in skin moisturizing power, a decrease in the expression or activity of serine proteases, ultraviolet rays, and a decrease in cell activity. In order to eliminate such stratum corneum, methods such as physical keratin peeling using scrub preparations, keratin peeling using chemical preparations, and physiological cell activity increase are usually used. Such keratin peeling methods aim to eliminate stratum corneum by artificially removing external keratinocytes and replacing the outermost stratum corneum with new keratinocytes from the inside, thereby solving appearance problems such as rough skin and dullness.

[0005] The elimination of stratum corneum stratification by promoting keratin peeling has additional effects such as removing fine lines and dark spots and improving rough skin, and is therefore used in a variety of products, with alpha hydroxy acid (AHA), beta hydroxy acid (BHA) and poly hydroxy acid (PHA) being the most commonly used. However, the action of AHA, BHA and PHA is to insert into the stratum corneum and release hydrogen ions to weaken the coniodesmosome bond, resulting in keratin peeling. When applied at low pH, they are easily inserted into the stratum corneum and have a high degree of hydrogen ion transfer, so a low pH is essential. Therefore, low pH can cause side effects such as stinging, itching and erythema, and research is being conducted to overcome these side effects. However, when the pH is adjusted to weakly acidic to neutral in order to alleviate the side effects caused by such skin irritation, the activity is reduced by more than 80%, and when the agent undergoes chemical transformation, the efficacy is offset, making it difficult to obtain the substantial benefits of exfoliation. Therefore, there is an increasing need for new exfoliants that exhibit exfoliation efficacy like AHA under acidic conditions even at weakly acidic or neutral pH.

[0006] Therefore, the inventors have confirmed in their previous research in Korean Patent Application No. 10-2017-0165268 that a composition containing serine and carnitine exhibits efficacy equal to or greater than that of AHA at the same concentration under neutral or weakly acidic conditions, and have confirmed that the composition causes relatively little or no irritation, and have provided it as an excellent new exfoliating material. [0008] The present invention aims to provide a cosmetic composition that maintains the accessibility of the keratin exfoliant to keratin binding proteins, thereby fully exerting the added efficacy of enhancing keratin exfoliation while causing no irritation.

[0009] Another object of the present invention is to solve the problem that the exfoliating effect of serine or carnitine, which has a superior effect to that of existing raw materials having exfoliating effect at a weakly acidic to neutral pH, is reduced when the serine or carnitine is actually applied in a cosmetic formulation.

[0010] More specifically, the present invention aims to provide a cosmetic composition for enhancing keratin exfoliation that contains serine or carnitine and is substantially free of moisturizing agent components having polyol groups or contains them at a certain concentration or less. [Means for solving the problem]

[0011] In order to achieve the above object, the present invention provides a cosmetic composition for enhancing exfoliation of skin, which contains an exfoliant and is free of a polyol moisturizer or contains a polyol moisturizer in an amount of 10 wt% or less based on the total weight of the composition.

[0012] According to one aspect of the present invention, the exfoliant of the cosmetic composition for enhancing exfoliation of the present invention is one or more of serine and carnitine.

[0013] According to one aspect of the invention, the polyol moisturizer of the cosmetic composition for enhancing keratin exfoliation of the present invention is at least one selected from the group consisting of glycerine, dipropylene glycol, butylene glycol, glycereth-26, methyl gluceth-20, and pentylene glycol.

[0014] According to one embodiment of the present invention, the exfoliant in the cosmetic composition for enhancing exfoliation of keratin is contained in an amount of 0.001 to 20% by weight based on the total weight of the composition.

[0015] According to one aspect of the invention, the cosmetic composition for enhancing exfoliation of keratinocytes according to the invention is free of polyol moisturizers or contains less than 10% by weight of the polyol moisturizer relative to the total weight of the composition.

[0016] According to one embodiment of the present invention, the cosmetic composition for enhancing keratin exfoliation of the present invention has a pH of 5-8. Effect of the Invention

[0017] The present invention provides a cosmetic composition for exfoliating skin that contains serine or carnitine, which provides a superior exfoliating effect at a weakly acidic to neutral pH level without irritation compared to existing raw materials, and maintains the exfoliating effect of serine or carnitine without any reduction. [0019] In order to achieve the above object, the present invention provides a cosmetic composition for enhancing exfoliation of skin, which contains an exfoliant and does not contain a polyol moisturizer or contains a polyol moisturizer in an amount of 10% by weight or less based on the total weight of the composition. The present invention will be described in detail below with reference to the drawings.

[0020] The present invention aims to produce a commercially available cosmetic product that is endowed with a keratin exfoliation enhancing function by adding a keratin exfoliant to a cosmetic composition containing various additives such as surfactants, fragrances, preservatives, moisturizers, thickeners, and antioxidants. However, it was found that the efficiency of the keratin exfoliation enhancing function is significantly lower than expected depending on the concentration of the keratin exfoliant. During research into this issue, it was found that the cause of the decrease in keratin exfoliation function is the moisturizer components among various cosmetic additives, particularly moisturizers having polyol groups, which reduce the accessibility of the keratin exfoliating components to the keratin binding proteins, resulting in a decrease in the keratin exfoliation ability due to the lack of effect on the binding proteins between keratin. The present invention has been completed by proposing a solution to this problem.

[0021] The present invention provides a cosmetic composition that allows a keratin exfoliant, particularly serine or carnitine, to maintain its effect. More specifically, the present invention provides a serine or carnitine cosmetic that provides a superior keratin exfoliating effect at a weakly acidic to neutral pH without irritation compared to the efficacy of existing raw materials.

[0022] However, when this is applied to a formulation, the exfoliating effect of serine or carnitine is reduced, and the present invention has found that when serine or carnitine contains a polyol-containing moisturizer, such as one or more polyols such as glycerin, dipropylene glycol (DPG) and 1,3-butylene glycol (BG), at a specific concentration or more, the exfoliating effect is rapidly reduced. Therefore, the present invention provides serine or carnitine that is substantially free of polyol moisturizer components or does not significantly reduce the effect.

[0023] In one embodiment, the present invention provides a cosmetic composition for enhancing skin exfoliation, comprising an exfoliant and being substantially free of a polyol moisturizer.

[0024] As used herein, the term "substantially free" means containing less than 2% by weight, preferably less than 1% by weight, more preferably less than 0.5% by weight, even more preferably less than 0.3% by weight, even more preferably less than 0.1% by weight, and most preferably none at all, based on the total weight of the composition.

[0025] Meanwhile, moisturizers having polyol groups are as commonly used as purified water in cosmetic compositions, and in particular, in the case of exfoliating products, moisturizing is required more than usual, so it is an issue to provide a cosmetic composition for enhancing exfoliation that contains a polyol moisturizer.

[0026] In another embodiment, the present invention provides a cosmetic composition for enhancing exfoliation of skin, comprising a keratin exfoliant and a polyol moisturizer. When the composition of the present invention comprises the polyol moisturizer, the content is from 0 to 10% by weight, preferably from 0 to 2% by weight, more preferably from 0 to 0.5% by weight. If the content of the polyol moisturizer is outside the above range, the exfoliation effect is significantly reduced, so the above content range is critical.

[0027] The term "keratin peeling" as used herein means that keratin accumulated on the stratum corneum of the skin is peeled off or removed from the stratum corneum of the skin. This concept includes not only the case where the keratin is removed by applying a physical force, but also the case where the keratin is peeled off from the stratum corneum of the skin only by treatment with a cosmetic composition without applying any other force.

[0028] According to one aspect of the present invention, the exfoliant of the cosmetic composition for enhancing exfoliation of the present invention may be one or more of serine and carnitine.

[0029] According to one aspect of the present invention, the keratin exfoliant in the cosmetic composition for enhancing keratin exfoliation of the present invention may be contained in an amount of 0.001 to 20% by weight, preferably 0.01 to 17% by weight, and more preferably 0.05 to 15% by weight, based on the total weight of the composition. If the content is less than 0.001% by weight, it is difficult to obtain the intended effect, and if it exceeds 20% by weight, the effect of increasing the content may be insignificant.

[0030] According to one aspect of the present invention, the polyol moisturizer of the cosmetic composition for enhancing keratin exfoliation of the present invention may be at least one selected from the group consisting of glycerin, dipropylene glycol (DPG), butylene glycol (BG), glycereth-26, methyl gluceth-20, and pentylene glycol. Specifically, it may be at least one selected from the group consisting of glycerin, dipropylene glycol, and 1,3-butylene glycol.

[0031] According to one aspect of the present invention, the cosmetic composition for enhancing keratin exfoliation of the present invention may contain no polyol moisturizer or may contain no more than 10% by weight, preferably no more than 5% by weight, more preferably no more than 2% by weight, even more preferably no more than 1% by weight, even more preferably no more than 0.5% by weight, even more preferably no more than 0.3% by weight, and even more preferably no more than 0.1% by weight. If the content is more than 10% by weight, it is difficult to obtain a sufficient keratin exfoliating effect of the intended keratin exfoliant.

[0032] According to one embodiment of the present invention, the cosmetic composition for enhancing keratin exfoliation of the present invention may have a pH of 5-8, preferably a pH of 5-7.5, and more preferably a pH of 5.5-7.

[0033] In addition to the above-mentioned components, the cosmetic composition for enhancing keratin exfoliation of the present invention may further contain components normally contained in cosmetics, selected as desired according to the cosmetic formulation or intended use, etc. Furthermore, the composition of each formulation may contain various bases and additives necessary and appropriate for the formulation of that formulation, and may be manufactured by containing known compounds such as preservatives, thickeners, surfactants, silicone polymers, oxidizing agents, organic solvents, antioxidants, emollients, colorants, and fragrances, within the range that does not reduce the effects of the formulation.

[0034] It was confirmed through a screening method using a pig skin exfoliation evaluation method and a human test method using a DHA staining method that the cosmetic composition according to the present invention exhibits excellent exfoliating effect among formulations containing a moisturizer having polyol.

[0035] Specifically, using a pig skin keratin peeling evaluation method, it was confirmed that serine or carnitine has activity in the neutral or slightly acidic pH range of 5-7 that is equal to or greater than the effect of gluconolactone (PHA) or lactic acid (AHA) at pH 4, and the effects were confirmed by combining different polyol moisturizer ingredients by material and concentration.

[0036] When the exfoliating effect of a cosmetic composition containing serine or carnitine and the polyol moisturizer components glycerin, dipropylene glycol, and 1,3-butylene glycol was evaluated using a pig skin exfoliation evaluation method, the exfoliating effect decreased sharply when the polyol moisturizer content was 0.1% by weight or more, and when it exceeded 10% by weight, the exfoliating effect was no longer observed, with the negative control group being water with 0% ability, which means that the exfoliating effect was not meaningfully observed even in human tests.

[0037] In addition, the exfoliating effect of a cosmetic composition containing serine or carnitine and polyol moisturizer components glycerin, dipropylene glycol and 1,3-butylene glycol was evaluated in a human body test using DHA staining. As a result, when the polyol moisturizer content was 10% or 50%, the number of days for keratin turnover (TT100) was not significantly different from 20 days without application, but it was confirmed that the cosmetic composition for promoting exfoliation of the present invention reduced the number of days for keratin turnover to 17 days or less, preferably 14 days, compared to 20 days without application.

[0038] Therefore, the present invention provides a keratin exfoliant, serine or carnitine, which is substantially free of polyol moisturizer components or does not significantly reduce its effect, since its activity is reduced to an insignificant level by the polyol moisturizers glycerin, dipropylene glycol, and 1,3-butylene glycol. In other words, the keratin exfoliant composition used in the present invention includes serine or carnitine, and is substantially free of one or more of the polyol moisturizers glycerin, dipropylene glycol, and 1,3-butylene glycol, or is selected within a range that does not significantly reduce its effect.

[0039] The present invention will be described in more detail with reference to the following examples. However, these are merely for the purpose of explaining the present invention in more detail, and the scope of the present invention is not limited thereto.

[0040] 1. Combination configuration For the purpose of experiments to confirm the effects of the present invention, combinations of serine or carnitine and moisturizing ingredients having a polyol group were prepared as shown in Table 1 below.

[0041] [Table 1]

[0042] Example 1. Evaluation of porcine skin keratin peeling Existing clinical evaluation methods take a long time, and it is difficult to conduct experiments for all the various concentrations and pH conditions of various raw materials, and it is also difficult to conduct experiments for all the compositions that can be combined from each raw material. Therefore, there is an increasing need for a non-clinical keratin peeling evaluation method.

[0043] Therefore, in the present invention, the keratin peeling evaluation method mentioned in Journal of the European Academy of Dermatology and Venereology, 2014, 28, 415-423 was used. In detail, pig skin was biopsied and the sample to be tested was added. Then, the number of keratin that fell off was measured with a cell counter, and the results were diagrammed with a sample containing only water as the negative control group (0%) and 10% gluconolactone (pH 4) as the positive control group (100%).

[0044] (1) Glycerin was added to a 5% serine solution in various combinations (combinations A and B) as shown in Table 2 below, and the experimental results are shown in Figure 1.

[0045] [Table 2]

[0046] The experimental results showed that the efficacy of serine was rapidly reduced by the addition of glycerin, and when the concentration was over 10%, it reached the range of 0% or less where no efficacy was observed. This confirmed that the efficacy of serine decreased with increasing glycerin concentration, and that glycerin must be used at 10% or less to be effective as an exfoliant (Figure 1).

[0047] (2) A 5% carnitine solution was mixed with glycerin in various combinations (combinations E and F) as shown in Table 3 below, and the results of the experiment on exfoliating pig skin are shown in Figure 2.

[0048] [Table 3]

[0049] The experimental results showed that the exfoliating effect decreased sharply when the glycerin content was 5% or more, and when it exceeded 10% by weight, the exfoliating effect was reduced to below that of water (the negative control group), confirming that it had no exfoliating effect (Figure 2).

[0050] (3) The results of the compositions in which glycerin, dipropylene glycol, and 1,3-butylene glycol were each 10% and 12% with 5% serine (Table 4; combinations A, B, C, and D) are shown in Figure 3. In addition, the experimental results for combination D, in which DPG was further combined at various concentrations with a 5% serine solution, are shown in Figure 4.

[0051] [Table 4]

[0052] The experimental results confirmed that the exfoliating effect was reduced when dipropylene glycol and 1,3-butylene glycol were included, as was the case with glycerin, and that when dipropylene glycol and 1,3-butylene glycol were combined at more than 10%, the degree of exfoliating activity was reduced to below that of the negative control group (water), and no significant difference was observed compared to the non-application group. Therefore, it was further confirmed that one or more of glycerin, dipropylene glycol, and 1,3-butylene glycol, which are moisturizers having polyol groups, in combination with serine, exhibit exfoliating effect at a concentration of 10% or less (Figures 3 and 4).

[0053] (4) The results for the compositions containing 5% carnitine, 10% glycerin, 10% dipropylene glycol and 12% 1,3-butylene glycol, respectively (combinations E, F, G and H), are shown in FIG.

[0054] [Table 5]

[0055] The experimental results showed that, in the case of carnitine, when glycerin, dipropylene glycol, or 1,3-butylene glycol was included, the exfoliating activity of the skin was reduced to an insignificant level in human trials (Figure 5).

[0056] (5) Furthermore, the results for the combination (combination I) in which a mixture of moisturizing agents (glycerin:DPG:1,3-BG=1:1:1) was added to a 5% serine solution are shown in Figure 6.

[0057] This confirmed that the exfoliating efficacy of the exfoliant was reduced in a similar manner when glycerin, dipropylene glycol, and 1,3-butylene glycol were mixed together compared to when they were used individually. In other words, even when a mixture of moisturizers with polyol groups was included, exfoliating efficacy was observed at a concentration of 10% or less (Figure 6).

[0058] Therefore, when one or more moisturizers having a polyol group selected from glycerin, dipropylene glycol, and 1,3-butylene glycol are included in the range of 10% by weight or less in combination with serine or carnitine, they exhibit keratin exfoliating effects. Therefore, when such moisturizer components are substantially not included or are used at a specific concentration or less, preferably 10% by weight or less, a significant keratin exfoliating effect is achieved.

[0059] (6) In addition, the concentration of serine was changed to 0.5% serine experimental group and 5% serine experimental group. The results of comparing the reduction in exfoliation efficacy depending on the concentration of glycerin are shown in Figure 7, with the case where serine was used alone being converted into the positive control group (100%).

[0060] As a result of the experiment, it was found that the level of exfoliating efficacy decreased in a similar manner with increasing glycerin concentration in both the 0.5% serine experimental group (dashed line in FIG. 7) and the 5% serine experimental group (solid line in FIG. 7), and it was confirmed that the concentration of the moisturizing ingredient having a polyol group in the composition plays a major role in decreasing the exfoliating efficacy, regardless of the concentration of serine or carnitine. This means that it is preferable that serine or carnitine is substantially free of moisturizing ingredients having a polyol group or contains a specific concentration, preferably 10% or less, of the moisturizing ingredient having a polyol group.

[0061] Taking the above results together, it was confirmed that the exfoliating ability of serine and carnitine depends on the concentration of moisturizers having polyol groups, namely glycerin, dipropylene glycol and 1,3-butylene glycol, and therefore, when moisturizer components having polyol groups are not substantially used or are used at a concentration of 10% or less, a noticeable effect is achieved by the user.

[0062] (7) In a combination of carnitine (5%) and glycerin, an experiment was conducted to measure the relative comparative value of the keratin exfoliation ability by dividing the concentration of glycerin into smaller amounts. The results are shown in Table 6 and FIG. 8 below.

[0063] [Table 6]

[0064] The experimental results showed that when the amount of glycerin was doubled from 1% to 2% by weight, the exfoliating ability remained almost the same, but when the amount of glycerin was increased to 3% by weight, the exfoliating ability was significantly decreased. In addition, when the amount of glycerin was increased five-fold from 0.1% to 0.5% by weight, the exfoliating ability actually increased, but when the amount of glycerin was increased to 1% by weight, the exfoliating ability significantly decreased, confirming the critical significance of the content range.

[0065] (8) In a combination of serine (5%) and glycerin, an experiment was conducted to measure the relative comparative value of the exfoliating ability by further dividing the concentration of glycerin. The results are shown in Table 7 and FIG. 9 below.

[0066] [Table 7]

[0067] As a result of the experiment, it was confirmed that when the amount of glycerin was doubled from 1% to 2% by weight, the exfoliating ability remained almost the same, but when the amount of glycerin was increased to 3% by weight, the exfoliating ability significantly decreased. In addition, when the amount of glycerin was increased five-fold from 0.1% to 0.5% by weight, the exfoliating ability actually increased, but when the amount of glycerin was increased to 1% by weight, the exfoliating ability significantly decreased, confirming the critical significance of the content range.

[0068] (9) In the case of carnitine (0.5%), in order to confirm how the inhibition of exfoliation ability due to the combination with polyol changes compared to the existing concentration of 5%, an experiment was conducted to measure the relative comparison value of the exfoliation ability by dividing the concentration of glycerin. The results are shown in Table 8 and FIG. 10 below.

[0069] [Table 8]

[0070] As a result of the experiment, when the glycerin content was 0.5 wt%, it showed excellent exfoliating ability comparable to that when no glycerin was added, but when the glycerin content was increased to 1 wt%, the exfoliating ability significantly decreased, confirming the critical significance of the content range.

[0071] (10) In the case of serine (0.5%), in order to confirm how the inhibition of exfoliating ability due to the combination of polyols changes compared to the existing concentration of 5%, an experiment was conducted to measure the relative comparison value of the exfoliating ability by subdividing the concentration of glycerin. The results are shown in Table 9 and FIG. 11 below.

[0072] [Table 9]

[0073] As a result of the experiment, when the glycerin content was 0.5 wt%, it showed excellent exfoliating ability comparable to that of a product without glycerin, but when the glycerin content was increased to 1 wt%, the exfoliating ability significantly decreased, confirming the critical significance of the content range.

[0074] Example 2. Method for evaluating the promotion of keratin turnover using the DHA staining method The DHA staining method for evaluating the turnover rate of stratum corneum was performed on 10 healthy males aged 20-40 years old using the method mentioned in Dermatology 1993, 186, 133-137. Before applying the sample, the color of the inside of the lower and upper arms of the subjects was measured with a Chroma Meter, and then about 0.4 ml of 10% dihydroxy acetone (DHA) was applied to a 1.5 x 1.5 cm gauze and left on the inside of the upper and lower arms for 8 hours. After 48 hours, the color of the area stained brown by DHA was measured and compared with the color difference before the sample was applied. The sample was then applied twice a day, and the degree of decolorization was measured with a Chroma Meter every day, and the time it took for the skin to return to its original color was measured. TT (Turnover Time) 100% means the time it takes for the existing stratum corneum to be 100% replaced by new stratum corneum, and in this experiment, the time it takes for the skin pigmented by DHA to return to 100% of its original state was shown by relative comparison analysis using regression analysis. The results of the negative control group were calculated on the 20th day, and the relative values were compared.

[0075] The results for the non-treated group were calculated on the 20th day, and the visual changes that appeared 10 days after application are shown in Figure 12. When visually distinguished, there was no significant difference between the non-treated group and the 10% or 50% glycerin concentration, and it was confirmed that when glycerin was used alone, effects were observed at both 0.5% and 5% concentrations.

[0076] When this was converted into a quantitative value using a Chromameter, it was as shown in Figure 13. When 5% glycerin was used, it was confirmed that TT100% decreased by about 6 days from 20 days without application to 14 days without application, and when the glycerin concentration was 10% or 50%, there was no significant change compared to the non-application group.

[0077] None of the combination groups showed any side effects such as erythema, itchiness, or pain after the experimental period. This confirmed that the inhibition of exfoliation in the concentration range selected by the pig skin exfoliation evaluation method was also confirmed in human experiments. In conclusion, it was confirmed that the exfoliation effect of serine or carnitine can be achieved by using no or a specific concentration of 10% or less of moisturizing agents having polyol groups, such as glycerin, dipropylene glycol, and 1,3-butylene glycol, without any substantial use.

[0078] Although a preferred embodiment of the present invention has been described above, this is merely one embodiment embodying the technical idea of the present invention, and any modifications or alterations that embody the technical idea of the present invention should be construed as falling within the scope of the present invention.
