[0071] Constriction Cartridges for Use in Intracellular Payload Delivery Systems FIG. 1 illustrates an intracellular payload delivery system 100. In some embodiments, system 100 may share any one or more characteristics with any one of the systems described in PCT / US18 / 66295. Features of intracellular payload delivery systems and devices such as system 100 are described more fully in PCT / US18 / 66295. Briefly, an intracellular payload delivery system may enable delivery of a payload into a cell by flowing the cell through a constriction, such as a narrow microfluidic channel or narrow pore, thereby perturbing the cell's membrane and allowing the payload to enter the cell.
[0072] In some embodiments, a constriction, such as a narrow microfluidic channel or narrow pore, may be provided in a microfluidic chip or filter, which may be attached in fluid communication to an intracellular payload delivery system, such as system 100. In some embodiments, a microfluidic chip or filter (or any other element comprising one or more constrictions configured to perturb a membrane of a cell) may be provided with a constriction cartridge and fluidly connected to a system, such as system 100. A constriction cartridge may be any device configured to house an element comprising a constriction, such as a microfluidic chip or filter, and / or to facilitate fluid connection of an element (e.g., a chip or filter) to another portion of an intracellular payload delivery system, such as system 100.
[0073] As shown in FIG. 1, the system 100 may include a base plate 102, a housing 103, a hook 104, an input bag 106, a preparation vessel housing 110, an output bag tray area 111, an output bag 112, a display 120, and a cartridge 124. In some embodiments, these elements may share any one or more characteristics in common with the corresponding elements described with respect to FIG. 15A and associated figures and descriptions of PCT / US18 / 66295. In some embodiments, the cell suspension fluid may be held in the preparation vessel of the system 100 (e.g., stored inside the preparation vessel housing 110) and then flowed (e.g., under pressure) out of the preparation vessel through a constriction cartridge 124. Inside the constriction cartridge 124, the cell suspension fluid may be flowed through one or more elements (e.g., a microfluidic chip or a filter) having one or more constriction channels and / or pores. After flowing through one or more elements inside the constriction cartridge 124, the cell suspension fluid may then flow out of the constriction cartridge 124 and toward or into one or more downstream system components, including the output bag 112.
[0074] Described below are exemplary embodiments of improved constriction cartridges for use in systems for intracellular payload delivery, such as constriction cartridge 112 for use in system 100.
[0075] 2A-2C illustrate various views of a constriction cartridge 200 for use in a benchtop system for delivering a payload to a cell, according to some embodiments. In some embodiments, the constriction cartridge 200 is the same constriction cartridge 124 discussed above with reference to FIG. 1. FIG. 2A illustrates an exploded view of the constriction cartridge, according to some embodiments, FIG. 2B illustrates a cross-sectional view of the body of the constriction cartridge, according to some embodiments, and FIG. 2C illustrates a partial perspective view of the constriction cartridge, according to some embodiments.
[0076] In some embodiments, the constriction cartridge 200 may be any structure configured to contain or house one or more constriction-containing elements, such as a constriction filter (containing one or more constriction pores) or a constriction microfluidic chip (containing one or more constriction microfluidic channels). (A constriction filter according to some embodiments is disclosed in Application No. WO / 2017 / 041050A1, which is incorporated herein by reference in its entirety.) Note that in some embodiments, a constriction microfluidic channel or constriction pore may simply be referred to as a "constriction" or a "cell deformation constriction." A constriction-containing element may be any component, device, part, or equivalent having a channel, passage, or other opening (e.g., a constriction) having a diameter smaller than the cells of the cell suspension passing through the element, such that forcing cells through the opening under pressure causes a perturbation in the membrane of the cells as they are constricted by the opening. In some embodiments, the constriction cartridge 200 may include an integrated constriction filter or microfluidic channel configured to constrict cells, while in some embodiments the constriction cartridge 200 may be configured to house a distinct element (e.g., a chip or filter) that itself includes a constriction pore or constriction microfluidic channel. In either case, the constriction cartridge 200 may define a portion of a flow path of a system for delivering a payload to cells, such as the system 100, such that a cell suspension may flow from a preparation vessel of the system toward and into the constriction cartridge 200, and the cell suspension may then flow through, out of, and toward and into an output bag (or any other suitable downstream flow path component) of the system.
[0077] As shown, the constriction cartridge 200 may comprise a body 202, a cap 203, a removable cover 204, an inlet 208, an outlet 210, ports 209 and 211, a branch channel 212, an O-ring cavity 213, an input channel 220, and an output channel 222. The constriction cartridge 200 may be configured to flow a cell suspension (and / or buffer fluid or other fluid) into and through a constriction containing element 206, which may be a constriction microfluidic chip having multiple constriction microfluidic channels and / or a constriction filter having multiple constriction openings or pores. In either case, the constriction containing element 206 may have individual constriction containing element inlets 216 for fluid flow into the individual elements and individual constriction cartridge outlets 218 for fluid flow out of the individual elements. The constriction cartridge 200 may be configured to receive one or more constriction-containing elements 206 and hold them in place using one or more grooves or ridges and / or by using frictional forces applied by one or more deformable components, such as O-rings, when the removable cover 204 is in place on the cartridge 200.
[0078] 2A-2C, the constriction cartridge 200 is disposed on the cartridge body 202 and includes a constriction cartridge inlet 208 and a constriction cartridge outlet 210 that define a beginning and an end of a flow path for the cell suspension and / or buffer fluid flowing through the constriction cartridge 200. In some embodiments, the inlet 208 and the outlet 210 may include any one or more connection mechanisms suitable for creating and securing a fluid connection between the inlet or outlet and another fluid-transporting component of a system in which the cartridge 200 is used, for example, the connection mechanisms may include threaded connections and / or Luer type connections.
[0079] As shown, the inlet 208 and the outlet 210 may be disposed on the same side and / or face of the body 202. In some embodiments, the inlet 208 and / or the outlet 210 may define a tubular protrusion from the body 202 extending outward and away from the center of the body 202. In some embodiments, the inlet 208 and the outlet 210 may be interchangeable with one another, i.e., the cartridge 200 may be reversible with respect to the function and / or orientation of the inlet 208 and the outlet 210, depending on user preference.
[0080] As an alternative or in addition to being optionally reversible with respect to the inlet 208 and outlet 210, the cartridge 200 may in some embodiments be optionally reversible with respect to the pair of ports 209 and 211 and the pair of inlets / outlets 208 and 210. As shown in FIGS. 2A-2C, the ports 209 and 211 may extend from the body 202 in a direction outward and away from the center of the body 202 and in a direction opposite that of the inlets 208 and outlets 210, opposite the direction in which the inlets / outlets 208 and 210 extend. In some embodiments, the body 202 may be symmetrical along three different axes that are all mutually perpendicular, such that each end of the body 202 has two ports, one on each side of the centerline of the body 202. For example, manufacturing of the body 202 by molding may be made more efficient by making the body 202 symmetrical in this way, even when it is meant that the body will have four ports, and even when only two ports are to be used in the application of the body 202.
[0081] As shown in FIG. 2B, ports 209 and 211 may be blocked by a cap 203, which may be attached to the ports by adhesive, a threaded connection, a luer lock connector, or the like. By capping ports 209 and 211, a flow path may be defined between two other ports, namely inlet 208 and outlet 210. In some other embodiments, other ports may be capped to define different flow paths through body 202 (e.g., capping inlet 208 and outlet 210 to define a flow path between port 209 and port 211, capping inlet 208 and port 211 to define a flow path between outlet 210 and port 209, or capping outlet 210 and port 209 to define a flow path between inlet 208 and port 211). By capping two of the four ports on body 202, a flow path may thus be created that begins and ends at a single port. In some alternative embodiments, body 202 may be constructed with only two ports, such that a flow path may be defined between the two ports without having to cap or otherwise block any other ports, and channels 220 and 222 (discussed in further detail below) may terminate before the ends of body 202 opposite inlet 208 and outlet 210. Although different configurations are possible as discussed above, the present description will proceed with respect to the exemplary arrangement shown in Figures 2A-2C, in which ports 209 and 211 are blocked by cap 203, thereby defining a flow path from inlet 208 to outlet 210.
[0082] As described above, the flow path through the cartridge 200 may be defined to begin at the inlet 208 and end at the outlet 210. Between the inlet 208 and the outlet 210, the flow path through the cartridge 200 may pass through one or more constriction-containing elements (e.g., microfluidic chips or filters), such that fluid flowing into the inlet 208 must flow through one or more constriction-containing elements before the fluid flows out of the outlet 210. In some embodiments, multiple constriction-containing elements within the cartridge 200 may be arranged in series such that the flow path through the cartridge 200 may be a single linear path. In some embodiments, multiple constriction-containing elements within the cartridge 200 may be arranged in parallel such that the flow path within the cartridge 200 may diverge into multiple parallel compartments as the fluid progresses through the constriction cartridge 200, and then reconverge before flowing out of the constriction cartridge 200. In some embodiments, three or more constriction-containing elements may be arranged within the cartridge 200 such that one or more of the elements are in series with another of the constriction-containing elements and one or more of the elements are in parallel with another of the constriction-containing elements.
[0083] As shown in FIG. 2B, the body 202 may include channels, namely, an input channel 220 and an output channel 222, each defined through the interior volume of the body 202. The input channel 220 and the output channel 222 may each be fluidly connected to a plurality of branch channels 212 that are defined through the interior volume of the body 202 and join and / or intersect the channels at a perpendicular angle, for example. In the example shown in FIG. 2B, the branch channel 212 extends from the top side of the body 202 to the bottom side of the body 202 and intersects with the input and output channels at the center of the body. The branch channels 212 may, in some embodiments, allow for fluid flow between the input and output channels and the constriction-containing element 206. In some embodiments, the branch channels 212 may be positioned in such a manner that they will align with the constriction-containing element inlet 216 and / or the constriction-containing element outlet 218 when the constriction-containing element 206 is installed in the cartridge 200. It should be noted that branch channels 212 may in some embodiments extend in only a single direction from the input or output channels, or in some embodiments may extend in multiple different directions from the input or output channels. In the example shown in Figures 2A-2C, branch channels 212 may extend in opposing directions upward and downward through body 202, and away from the input and output channels in opposing directions, such that fluid may flow into / from different constriction-containing elements, which may be located on opposing sides (e.g., top and bottom) of body 202.
[0084] The branch channel 212 may also be aligned along a shared axis with a respective one of the cavities 213 (or a respective pair of cavities 213, i.e., one on the top side of the body 202 and one on the bottom side of the body 202), each of which may be formed as a recessed cavity on the surface of the body 202. The cavity 213 may be configured to receive an O-ring such that the O-ring seats inside the cavity 213 between the body 202 and one of the constriction-containing elements 206, allowing a fluid-tight seal to be formed between the body 202 and the constriction-containing element, such that fluid may flow through the cavity and through the center of the O-ring, thereby flowing between the body and the constriction-containing element without leaking out of the flow path defined in part by the O-ring. In some embodiments, other sealing options aside from or in addition to an O-ring may also be used to create a seal for the fluid connection between the constriction cartridge and the constriction-containing element, for example, overmolding, chemical bonding, and / or mechanical interlocking may be used.
[0085] Thus, in some embodiments, fluid enters cartridge 200 through inlet 208, flows from inlet 208 into and through input channel 220, flows from input channel 220 into and through one of the branch channels 212 that intersect with input channel 220, flows from one of the branch channels 212 into and through one of the cavities 213 aligned with the branch channel, flows from one of the cavities 213 into one of the constriction-containing element inlets 216, flows from the constriction-containing element inlet 216 through the constriction-containing element 206. 2, may flow out of the constriction-containing element 206 through its constriction-containing element outlet 218, from the constriction-containing element outlet 218 into and through another of the cavities 213 aligned with the branch channel 212 that intersects with the output channel 222, from that second one of the cavities 213 into and through the aligned one of the branch channels 212, from that second one of the branch channels 212 into and through the output channel 222, from the output channel 222 to and through the outlet 210, and exit the cartridge 200. Thus, in brief, a fluid such as a buffer fluid or a cell suspension may flow into the constriction cartridge 200 and may be passed through one or more constriction-containing elements before flowing out of the constriction cartridge 200.
[0086] In some embodiments, the constriction cartridge 200 may be configured to be capable of receiving an empty surrogate element in place of a functional constriction-containing element, which may not contain any channels or pores or may otherwise be configured to not allow flow through the portion of the constriction cartridge 200 that stores the surrogate element. By using an empty surrogate element, the constriction cartridge 200 may cause fluid flow through a small number of constriction-containing elements at a time, or through only one constriction-containing element at a time, so that the system does not have to be used at full capacity of constriction-containing elements all the time.
[0087] 2A-2C, the stenosis cartridge 200 may include a removable cover 204, which may be an element configured to be placed alongside one or more stenosis-containing elements 206 and to urge the one or more stenosis-containing elements 206 towards the cartridge body 202 and / or otherwise hold the one or more stenosis-containing elements 206 in place. In some embodiments, the removable cover 204 may be configured to apply an inward force to the stenosis-containing elements 206, urging them towards the cartridge body 202, using one or more springs or other compressible or deformable components, such as rubber O-rings as discussed elsewhere herein. In some embodiments, the removable cover 204 may be configured to press flush against one or more surfaces of the stenosis-containing elements 206. In some embodiments, the removable cover 204 may serve to ensure that the constriction-containing elements 206 do not delaminate under the pressure of fluid being forced through them, and by pressing the cover 204 against one side of the constriction-containing elements 206 under force, the constriction-containing elements 206 may be prevented from delaminating.
[0088] In some embodiments, one or more of the removable covers 204 may be attached to one or more other components of the cartridge 200 by a sliding connection, a threaded connection, a hinge connection, a tab and slot connection, a locking mechanism, by one or more screws, by one or more cams, or in any other suitable manner such that the cover can be removed, for example, to replace the constriction-containing element 206.
[0089] In some embodiments, a sliding connection, such as that shown in Figures 2A-2C, may allow the cover 204 to slide laterally along the cartridge 202 (in the direction of flow into and out of the cartridge shown in Figures 2A-2C) such that an edge of the cover 204 may slide over a corresponding edge, protrusion, groove, or tooth of the body 204. In some embodiments, a removable cover configured to slide onto and off the body of the constriction cartridge may, in some embodiments, be configured to completely surround the constriction cartridge body, thereby avoiding the need for interlocking grooves or teeth or the like.
[0090] Such a sliding connection may be removed with little lateral force (e.g., force in the direction of the sliding), but may provide greater strength in a direction perpendicular to the sliding direction and extending away from the side of the body 202 against which the one or more stenosis-containing elements 206 are located. Thus, a cover having a sliding connection may be easily removed manually, but may nevertheless provide greater durability under pressure than other connection mechanisms that may be used to manually assemble a stenosis cartridge, such as attaching a cover by threaded components. In some embodiments, in addition to or as an alternative to one or more removable covers, a stenosis cartridge may be configured to securely store one or more stenosis-containing elements without the use of a removable cover.
[0091] In some embodiments, cartridge 200 may be less than or equal to 25 mm, 50 mm, 75 mm, 100 mm, 125 mm, 150 mm, or 200 mm in length (e.g., in a direction extending parallel to channels 220 and 222). In some embodiments, cartridge 200 may be greater than or equal to 25 mm, 50 mm, 75 mm, 100 mm, 125 mm, 150 mm, or 200 mm in length.
[0092] In some embodiments, cartridge 200 may be less than or equal to 10 mm, 20 mm, 30 mm, 40 mm, 50 mm, or 60 mm in width (e.g., in a direction extending perpendicular to and between channels 220 and 222). In some embodiments, cartridge 200 may be greater than or equal to 10 mm, 20 mm, 30 mm, 40 mm, 50 mm, or 60 mm in width.

In some embodiments, the cartridge 200 may be greater than or equal to 5 mm, 10 mm, 15 mm, 20 mm, 25 mm, or 30 mm in height.
[0094] In some embodiments, one or more of the constriction-containing elements 206 may be less than or equal to 20 mm, 30 mm, 40 mm, 50 mm, or 60 mm in length (e.g., in a direction extending from the inlet 216 toward the outlet 218). In some embodiments, one or more of the constriction-containing elements 206 may be greater than or equal to 20 mm, 30 mm, 40 mm, 50 mm, or 60 mm in length.
[0095] In some embodiments, one or more of the constriction-containing elements 206 may be less than or equal to 20 mm, 30 mm, 40 mm, 50 mm, or 60 mm in width (e.g., in a direction perpendicular to channels 220 and 222 in the arrangement shown in FIG. 2A). In some embodiments, one or more of the constriction-containing elements 206 may be greater than or equal to 20 mm, 30 mm, 40 mm, 50 mm, or 60 mm in width.
[0096] In some embodiments, one or more of the constriction-containing elements 206 may be less than or equal to 0.1 mm, 0.5 mm, 1 mm, 2 mm, 3 mm, or 5 mm in thickness (e.g., in a direction extending from one cover 204 to the other in the arrangement shown in FIG. 2A). [0097] In some embodiments, the constriction (e.g., constricted channel or constricted pore) of a constriction-containing element (e.g., one of elements 206) may be less than or equal to 0.25 μm, 0.5 μm, 1 μm, 5 μm, 10 μm, 20 μm, or 50 μm in width. In some embodiments, the constriction (e.g., constricted channel or constricted pore) of a constriction-containing element (e.g., one of elements 206) may be greater than or equal to 0.25 μm, 0.5 μm, 1 μm, 5 μm, 10 μm, 20 μm, or 50 μm in width.

In some embodiments, the constriction (e.g., constriction channel or constriction pore) of a constriction-containing element (e.g., one of elements 206) may be greater than or equal to 1 μm, 5 μm, 10 μm, 20 μm, 30 μm, 40 μm, 50 μm, or 100 μm in length.
[0099] In some embodiments, the constriction (e.g., constricted channel or constricted pore) of a constriction-containing element (e.g., one of elements 206) may be less than or equal to 10 μm, 15 μm, 20 μm, 50 μm, 80 μm, 100 μm, or 200 μm in depth. In some embodiments, the constriction (e.g., constricted channel or constricted pore) of a constriction-containing element (e.g., one of elements 206) may be greater than or equal to 10 μm, 15 μm, 20 μm, 50 μm, 80 μm, 100 μm, or 200 μm in depth.
[0100] In some embodiments, one or more of the input channel 220 and the output channel 222 may be less than or equal to 25 mm, 50 mm, 75 mm, 100 mm, 125 mm, 150 mm, or 200 mm in length. [0104] In some embodiments, cartridge 200 may be configured to be capable of being used with systems that push fluid through the cartridge at pressures less than or equal to 1, 5, 10, 25, 50, 75, 100, 125, 150, or 200 PSI. In some embodiments, cartridge 200 may be configured to be capable of being used with systems that push fluid through the cartridge at pressures greater than or equal to 1, 5, 10, 25, 50, 75, 100, 125, 150, or 200 PSI. In some embodiments, cartridge 200 may be configured to be used with constriction-containing elements (e.g., constriction-containing elements) that can individually (e.g., "per tip") provide a processing capacity of less than or equal to 50, 100, 150, 200, 250, 300, or 400 mL of red blood cell suspension per minute, respectively. [0105] In some embodiments, cartridge 200 may be configured for use with constriction-containing elements (e.g., constriction-containing elements) that may individually (e.g., "per chip") provide a processing capacity of peripheral blood mononuclear cell suspension less than or equal to 25, 50, 75, 100, 125, 150, or 200 mL per minute, respectively. In some embodiments, cartridge 200 may be configured for use with constriction-containing elements (e.g., constriction-containing elements) that may individually (e.g., "per chip") provide a processing capacity of peripheral blood mononuclear cell suspension greater than or equal to 25, 50, 75, 100, 125, 150, or 200 mL per minute, respectively.
[0106] In some embodiments, the cartridge 200 may have an overall fluid handling capacity (e.g., including all constriction-containing elements contained within the cartridge 200) that is less than or equal to 0.25, 0.5, 1, 1.5, 2, 2.5, or 5 L / min. In some embodiments, the cartridge 200 may have an overall fluid handling capacity (e.g., including all constriction-containing elements contained within the cartridge 200) that is greater than or equal to 0.25, 0.5, 1, 1.5, 2, 2.5, or 5 L / min.
[0107] In some embodiments, cartridge 200 may be configured to store a plurality of stenosis-containing elements, such as stenosis-containing element 206. In some embodiments, cartridge 206 may be configured to store fewer than or equal to 2, 5, 10, or 20 stenosis-containing elements. In some embodiments, cartridge 206 may be configured to store fewer than or equal to 2, 5, 10, or 20 stenosis-containing elements per side. In some embodiments, cartridge 206 may be configured to store more than or equal to 2, 5, 10, or 20 stenosis-containing elements. In some embodiments, cartridge 206 may be configured to store more than or equal to 2, 5, 10, or 20 stenosis-containing elements per side.
[0108] In some embodiments, the cartridge 200 may comprise one or more components made from metal, plastic, polymer, and / or glass. In some embodiments, the cartridge 200 may comprise one or more components made from polycarbonate, polypropylene, and / or polymethylmethacrylate.
[0109] 3A-3C illustrate various views of a constriction cartridge 300 for use in a benchtop system for delivering a payload to a cell, according to some embodiments. In some embodiments, the constriction cartridge 300 is the same constriction cartridge 124 discussed above with reference to FIG. 1. FIG. 3A illustrates an exploded view of the constriction cartridge, according to some embodiments, FIG. 3B illustrates a partial cross-sectional view of the body of the constriction cartridge, according to some embodiments, and FIG. 3C illustrates a partial perspective view of the constriction cartridge, according to some embodiments.
[0110] In some embodiments, cartridge 300 may share any one or more characteristics in common with cartridge 200. In some embodiments, one or more components of cartridge 300 may share any one or more characteristics in common with a corresponding component or components of cartridge 200. In some embodiments, as discussed below, cartridge 300 may differ from cartridge 200 in that while cartridge 200 may include inlet and outlet ports formed as part of the cartridge body and fluidically connected to internal channels formed within the cartridge body, cartridge 300 may instead include inlet and outlet ports formed as part of a removable cap of the cartridge (rather than the body) and configured to fluidically connect to internal chambers formed within the cartridge body when the cap is positioned on the cartridge body in an assembled position. Thus, while a fluid pathway within cartridge 200 may be defined by a single cartridge component (e.g., the cartridge body) in conjunction with one or more constriction-containing elements, a fluid pathway within cartridge 300 may be defined by three separate cartridge components (e.g., the cartridge body and two removable covers) in conjunction with one or more constriction-containing elements.
[0111] As shown, the constriction cartridge 300 may comprise a body 302, a removable cover 304, an inlet 308, an outlet 310, a branch channel 312, an O-ring cavity 313, an input chamber 320, an output chamber 222, a raised edge 324, a groove 326, and a body O-ring 328. The constriction cartridge 300 may be configured to flow a cell suspension (and / or buffer fluid or other fluid) into and through a constriction containing element 306, which may be a constriction microfluidic chip having multiple constriction microfluidic channels and / or a constriction filter having multiple constriction openings or pores. In either case, the constriction containing element 306 may have individual constriction containing element inlets 316 for fluid flow into the individual elements and individual constriction cartridge outlets 318 for fluid flow out of the individual elements. The constriction cartridge 300 may be configured to receive one or more constriction-containing elements 306 and hold them in place using one or more grooves or ridges and / or by using frictional forces applied by one or more deformable components, such as O-rings, when the removable cover 304 is in place on the cartridge 300. In some embodiments, the components of the constriction cartridge 300 and / or the constriction-containing elements 306 may share any one or more characteristics in common with the corresponding components discussed above with respect to Figures 2A-2C.
[0112] The body 302 of the cartridge 300 may share any one or more characteristics in common with the body 202 of the cartridge 200, but instead of (or in addition to) internal input and output channels, the body 302 may have an input chamber 320 (as shown in FIGS. 3A and 3B) and an output chamber 322 (as shown in FIG. 3C). The input chamber 320 and the output chamber 322 may each be chambers formed within the interior volume of the body 302 and open to one side of the body 302. The openings of the chambers to the outside of the body 302 may be elongated openings as shown in FIGS. 3A-3C. As shown in FIG. 3B, the input chamber 320 may extend from its elongated opening on one side of the body 302 toward the center of the body 302, but may be separated from the output chamber 322 by a wall or partition portion within the body 302 so that the two chambers do not abut. In some embodiments, the input chamber 320 and the output chamber 322 may serve a purpose similar to the input channel 220 and the output channel 222 discussed above, in that they may direct the flow of fluid into and out of the branch channel 312.
[0113] In some embodiments, one or more of the input chamber 320 and the output chamber 322 may be less than or equal to 10 mm, 15 mm, 20 mm, 25 mm, 50 mm, 75 mm, 100 mm, 125 mm, 150 mm, or 195 mm in length (e.g., in a direction extending along a surface of a side of the cartridge 300 and in an elongated dimension of an opening formed in the side of the cartridge 300). In some embodiments, one or more of the input chamber 320 and the output chamber 322 may be greater than or equal to 10 mm, 15 mm, 20 mm, 25 mm, 50 mm, 75 mm, 100 mm, 125 mm, 150 mm, or 195 mm in length.
[0114] In some embodiments, one or more of the input chamber 320 and the output chamber 322 may be less than or equal to 1 mm, 2.5 mm, 5 mm, 10 mm, 15 mm, 20 mm, or 25 mm in width (e.g., in a direction extending along a surface of a side of the cartridge 300 and perpendicular to an elongation dimension of an opening formed in the side of the cartridge 300). In some embodiments, one or more of the input chamber 320 and the output chamber 322 may be greater than or equal to 1 mm, 2.5 mm, 5 mm, 10 mm, 15 mm, 20 mm, or 25 mm in width.
[0115] In some embodiments, one or more of the input chamber 320 and the output chamber 322 may be less than or equal to 2.5 mm, 5 mm, 10 mm, 15 mm, 20 mm, 25 mm, 30 mm, 40 mm, 50 mm, or 55 mm in depth (e.g., in a direction extending from an opening on a side of the cartridge 300 toward the interior of the cartridge 300). In some embodiments, one or more of the input chamber 320 and the output chamber 322 may be greater than or equal to 2.5 mm, 5 mm, 10 mm, 15 mm, 20 mm, 25 mm, 30 mm, 40 mm, 50 mm, or 55 mm in depth.
[0116] The cartridge 320 may include a removable cover 304, which may serve at least two functions. First, the removable cover 304 may serve to hold the constriction-containing element 306 in place relative to the body 302 as the removable cover is slid over the constriction-containing element 306 in a manner similar to the removable cover 204 discussed above. In some embodiments, the removable cover 304 may be configured to press flush against one or more surfaces of the constriction-containing element 306. In some embodiments, the removable cover 304 may serve to ensure that the constriction-containing element 306 does not delaminate under the pressure of fluid being forced therethrough, and by pressing the cover 304 against one side of the constriction-containing element 306 under force, the constriction-containing element 306 may be prevented from delaminating.
[0117] In some embodiments, one or more of the removable covers 304 may be attached to one or more other components of the cartridge 300 by a sliding connection, a threaded connection, a hinge connection, a tab and slot connection, a locking mechanism, by one or more screws, by one or more cams, or in any other suitable manner such that the cover can be removed, for example, to replace the constriction-containing element 306. In some embodiments, the removable cover 304 may be molded to fit closely around the body 302, for example, by sliding over and over all or a portion of the body 302.
[0118] In some embodiments, a sliding connection, such as the connection shown in Figures 3A-3C, may allow the cover 304 to slide laterally along the cartridge 302 (in the direction of flow in and out of the ports on the sliding cover 304 shown in Figures 3A-3C). As shown in Figures 3A and 3C, the removable cover 304 may be molded to have an open side facing the inlet / outlet such that the open side secures the element 306 in place and may slide over a portion of the body 302 to form a fluid connection with an input or output chamber in the body 302 that opens toward the cover.
[0119] Such a sliding connection may be removed with little lateral force (e.g., force in the direction of the sliding), but may provide greater strength in a direction perpendicular to the sliding direction and extending away from the side of the body 302 against which the one or more stenosis-containing elements 306 are located.
[0120] Second, the removable cover 304 may serve to fluidly connect the input / output chambers in the body 302 to other components of a system for delivering a payload to a cell, using inlets 308 and outlets 310, each of which may be configured in one of the removable covers 304. As shown, the inlets 308 and outlets 310 may extend in opposing directions from one another when the cover 304 is placed on the cartridge 300, and each may extend in a direction perpendicular to the side of the body 302 from which they extend, and aligned with the direction along which the inlets 320 and outlets 322 extend into the body 302. In some embodiments, the inlets 308 and outlets 310 may include any one or more connection mechanisms suitable for creating and securing a fluid connection between the inlets or outlets and another fluid-transporting component of a system in which the cartridge 200 is used, for example, the connection mechanisms may include threaded connections and / or luer-type connections.
[0121] In some embodiments, the inlet 308 and the outlet 310 may be interchangeable with one another, i.e., the cartridge 300 may be reversible with respect to the function and / or orientation of the inlet 308 and the outlet 310, depending on user preference. In some embodiments, the cover 304 may be configured to fit onto both sides of the body 302 or onto only one side of the body 302.
[0122] When the removable cover 304 is slid into place on the body 302, a fluid connection may be formed between the removable cover and a chamber within the body. Thus, while the inlet 208 and outlet 210 in the cartridge 200 were included as part of the cartridge body 202, the inlet 308 and outlet 310 may be included as part of the removable cover 304 such that fluid may enter the cartridge 300 through a flow path in one of the removable covers 304, then flow into the body 302, and then subsequently exit the cartridge 300 through a flow path in the other of the removable covers 304.
[0123] The fluid connection between the removable cover and the chamber within the body may, in some embodiments, be sealed by a body O-ring 328, which may seat in a groove 326 formed in a raised edge 324 formed on and / or around one or more edges of the body 302. As shown, the raised edge 324 and groove 326 may hold the body O-ring 328 in place and urge the body O-ring 328 against an inner surface of the removable cap 304 such that when the removable cap 304 is slid onto the body 302, a seal is formed and fluid in the flow passage inside the removable cap 304 may flow into the chamber inside the body 302 without leaking or otherwise escaping.
[0124] In addition to securing the body O-ring 328 in place against the inner surface of the removable cap 304, the raised edges 326 may additionally serve to hold one or more of the constriction-containing elements 306 in place. In some embodiments, the two raised edges 326 may be spaced apart from one another, for example along opposing edges of the surface of the body 302, such that the constriction-containing element 306 may be seated within a recess formed between the raised edges 326, and thus the constriction-containing element 306 is prevented from sliding or moving laterally about the branch channel 312, and is properly aligned therewith (e.g., the branch channel 312 is aligned with a respective constriction-containing element inlet 316 and constriction-containing element outlet 318).
[0125] 3A, the constriction-containing element 306 may include an inlet 316 and an outlet 318, which may share any one or more characteristics in common with the inlet 216 and outlet 218 described above with respect to FIGS. 2A-2C. The inlet 316 and outlet 318 may be configured to align with the branch channel 312 and O-ring cavity 313, which may share any one or more characteristics in common with the branch channel 212 and O-ring cavity 213 described above with respect to FIGS. 2A-2C. For example, the constriction-containing element 306 may be pressed by the removable cover 306 towards the body 302, thereby being pressed into one or more O-rings housed within the respective cavities 313, thereby forming a sealed flow path between the constriction-containing element 306 and the branch channel 312, which is aligned with the cavity 313 in the body 302.
[0126] The branch channel 312 may differ from the branch channel 212 as discussed above with reference to Figures 2A-2C in that, in some embodiments, the branch channel 312 may intersect with an input chamber 320 or an output chamber 322 rather than with an input or output channel. In some embodiments, the branch channel 312 may intersect with an input or output chamber at an angle perpendicular to the surface of the chamber, as shown, for example, in Figure 3B. In some embodiments, the axis of one of the branch channels 312 may be perpendicular to both the inner surface of the input / output chamber and the direction along which the elongated opening of the input / output chamber extends along the surface of the body 302.
[0127] It should be noted that the branch channels 312 may in some embodiments extend in only a single direction from the input or output chamber, or in some embodiments may extend in multiple different directions from the input or output chamber. In the example shown in Figures 3A-3C, the branch channels 312 may extend in opposing directions through the body 302 and away from the input and output chambers in opposing directions such that fluid may flow to / from different constriction-containing elements, which may be located on opposing sides of the body 302.
[0128] Thus, in some embodiments, fluid enters cartridge 300 through inlet 308 in removable cap 304, flows from inlet 308 into and through input chamber 320, flows from input chamber 320 into and through one of the branch channels 312 that intersect with input chamber 320, flows from one of the branch channels 312 into and through one of the cavities 313 aligned with the branch channel, flows from one of the cavities 313 into one of the constriction-containing element inlets 316, flows from the constriction-containing element inlet 316 through the constriction-containing element 306, , flow out of the constriction-containing element 306 through its constriction-containing element outlet 318, flow from the constriction-containing element outlet 318 into and through another of the cavities 313 aligned with the branch channel 312 that intersects with the output chamber 322, flow from that second one of the cavities 313 into and through the aligned one of the branch channels 312, flow from that second one of the branch channels 312 into and through the output chamber 322, flow from the output chamber 322 through the outlet 310 of the other removable cap 304, and may exit the cartridge 300. Thus, in brief, a fluid such as a buffer fluid or a cell suspension may flow into the constriction cartridge 300 and may be passed through one or more constriction-containing elements before flowing out of the constriction cartridge 300.
[0129] In some embodiments, the constriction cartridge 300 may be configured to be capable of receiving an empty surrogate element in place of a functional constriction-containing element, which may not contain any channels or pores or may otherwise be configured to not allow flow through the portion of the constriction cartridge 300 that stores the surrogate element. By using an empty surrogate element, the constriction cartridge 300 may cause fluid flow through a small number of constriction-containing elements at a time, or through only one constriction-containing element at a time, so that the system does not have to be used at full capacity of constriction-containing elements all the time.
[0130] (Example) FIG. 4 shows data from a study evaluating how volumetric scaling is performed for the passage of red blood cells through a chip contained within a cartridge constructed according to the cartridge properties disclosed herein.
[0131] In the depicted example, human whole blood was first leukoreduced and washed using the LOVO Cell Processing System prior to the stenosis process. The blood contained approximately 4×10 9 The blood was concentrated to 1000 red blood cells. The prepared blood was then passed through one or more constriction-containing tips mounted in one of a variety of different cartridges. The blood was passed through a separate cartridge at a pressure of 60 PSI with 10 μg / mL of ALEXA FLUOR 647 ovalbumin conjugate (far-red fluorescent protein). Following passage of the blood through the separate cartridges, a small-scale manual wash at 8000 RCF was performed. The cells in the blood were stained with ANNEXIN V ALEXA FLUOR 488 READY FLOW CONJUGATE. Flow cytometry was performed to measure persistence, ALEXA FLUOR 647 delivery percentage (to assess the percentage of cells successfully delivered with the fluorescent protein), delivered mean fluorescent intensity (MFI) (to assess the number of fluorescent proteins delivered to each cell), and ANNEXIN The V ALEXA FLUOR 488+ percentage (to assess the percentage of cells in which the complex was normally attached to the cell membrane, which can be taken as an indication of cell age) was assessed.
[0132] The same tip (SQZ Wier tip with a footprint of 11.5 mm x 21 mm) was used in the various different steps performed. In each cartridge configuration, the cartridge was tested to withstand up to 75 PSI. The number of tips used in the various cartridge configurations across the different steps was one tip, four tips, or eight tips. For steps with one SQZ Weir tip, a clamshell cartridge was used, for steps with four SQZ Weir tips, a four-sided cartridge with one tip per side was used, and for steps with eight SQZ Weir tips, a double-sided cartridge holding four tips per side was used, configured according to the embodiment shown in Figures 2A-2C above. As shown in the graph in Figure 4, an internal control was also performed, and delivery and conjugation percentages were measured for cells that were not passed through any constriction.
[0133] Use of the 8-tip configuration with the cartridge types depicted in Figures 2A-2C improved volumetric flow rates (throughput in mL) and reduced run times, as shown in the table of Figure 4. There was little impact on performance of the different cartridge configurations in terms of total delivery percentage or total ANNEXIN V ALEXA FLUOR 488+ percentage, as shown in the graph of Figure 4.
