Background Art
[0002] Korean ginseng has been used as a miraculous medicine in traditional Chinese medicine prescriptions since ancient times. Recently, as the efficacy and pharmacological effects of Korean ginseng have been proven pharmacologically and clinically, Korean ginseng products are widely favored as natural health functional foods beyond the concept of supplements that strengthen basic physical strength. From the perspective of traditional Chinese medicine, the efficacy of Korean ginseng is to replenish vitality, rescue from collapse, replenish blood, and restore the strength of the pulse. It also nourishes the heart, stabilizes the spirit, generates body fluids, quenches thirst, supplements the strength of the lungs, stops asthma, strengthens the gastrointestinal tract, stops diarrhea, eliminates toxins, and heals sores. Through modern medical pharmacological experiments, the efficacy of Korean ginseng has been clarified to enhance the non-specific resistance of the body to stress and have the function of maintaining the homeostasis of the body. In addition, its effects such as improving hypertension, enhancing insulin action, lowering blood sugar in diabetic mice, promoting RNA synthesis, protein synthesis, sugar and lipid metabolism in the liver of white mice, and anti-cancer effects have been clarified.
[0003] The consumption of Korean ginseng is mainly carried out in Southeast Asia such as China, Japan, and South Korea by region, and among different age groups, it is mostly used by middle-aged and elderly people. The reasons for such limited consumer groups of Korean ginseng include the bitterness of Korean ginseng and the earthy smell which is the unique fragrance of Korean ginseng. In particular, it is known that the fragrance of Korean ginseng has a very negative effect on Westerners. Therefore, there is a demand to develop a healthy food Korean ginseng product that suits the preferences of people of all ages and genders worldwide by alleviating the bitterness, earthy smell, etc. that are pointed out to be negative for taking Korean ginseng. However, it is not easy to mask the unique bitterness, and there is a problem that the content of ginsenoside, a functional ingredient, decreases in the processing process of removing bitterness.
[0004] Examples of the types of adsorbents include activated carbon, clay activated, acid clay, zeolite, diatomite, bentonite, etc. The activated carbon is one of the most commonly used adsorbents and is used for solvent recovery and removal of organic impurities. The acid clay is a clay with acidity, which is almost the same in appearance as bentonite, but has a higher silica content and stronger adsorption power. Also, the diatomite is a sediment composed of the remains of diatoms and is used for adsorbents, filters, abrasives, pharmaceuticals, purification of foods, etc.
[0005] Cyclodextrins are used in pharmaceuticals and food additives for purposes such as stabilizing incomplete ingredients, masking the taste and odor of components with off-odors, reducing side effects using inclusion compounds formed by inclusion reactions, prolonging drug efficacy, solubilizing poorly soluble components, and assisting dissolution. Cyclodextrin is a type of oligosaccharide composed of multiple glucopyranose repeating units and retains a round, ring-shaped three-dimensional molecular structure. Cyclodextrin is named α-cyclodextrin if it has 6 glucose residues, β-cyclodextrin if it has 7 glucose residues, and γ-cyclodextrin if it has 8 glucose residues, and the sizes of their respective cavities are different. Also, cyclodextrin has a unique donut-like structure, and in an aqueous solution, the inner surface of the cavity within the cyclodextrin molecule is hydrophobic, and the outer surface is hydrophilic, and it has the property of encapsulating a plurality of organic and inorganic compounds within its cavity.
[0006] Korean Patent Publication No. 2012-0110667 discloses a method for producing a ginseng beverage with reduced unique aroma and bitterness of Korean ginseng, and Korean Registered Patent No. 1194265 discloses a method for removing the bitterness of a concentrated solution of Korean ginseng or red ginseng using deep ocean water, but they are different from the method for producing a concentrated solution of ginseng products by double treatment with the composite adsorbent and composite cyclodextrin of the present invention.

Embodiments for Carrying Out the Invention
[0013] To achieve the object of the present invention, the present invention provides (1) After adding water to ginseng, extracting to produce a treatment liquid; (2) After immersing ginseng in the treatment liquid produced in step (1) and then taking it out and drying; (3) After adding water to the pulverized ginseng obtained by pulverizing the ginseng dried in step (2) and then extracting to produce a ginseng extract; (4) Mixing acid clay, activated carbon, and diatomaceous earth to produce a composite adsorbent; (5) Passing the ginseng extract produced in step (3) through the composite adsorbent produced in step (4) to produce a ginseng extract treated with the composite adsorbent; (6) Mixing α-cyclodextrin, β-cyclodextrin, and γ-cyclodextrin to produce a cyclodextrin mixed powder; (7) Adding the cyclodextrin mixed powder produced in step (6) to the ginseng extract treated with the composite adsorbent produced in step (5), stirring, standing, and filtering; (8) Concentrating the ginseng extract filtered in step (7). Provided is a method for producing a concentrated ginseng solution, characterized by including the above steps.
[0014] In the method for producing a concentrated ginseng solution of the present invention, the treatment liquid in step (1) is preferably produced by adding water to ginseng at a ratio of 10 to 20 times (v / w) and extracting at 80 to 90°C for 1 to 3 hours. More preferably, it can be produced by adding water to ginseng at a ratio of 15 times (v / w) and extracting at 85°C for 2 hours.
[0015] The ginseng used for producing the treatment liquid is preferably one or more selected from the group consisting of fresh ginseng, red ginseng, dried ginseng, white ginseng, black ginseng, taegeuk ginseng, cheongsam ginseng, mountain-cultivated ginseng, camphor ginseng, cultured ginseng roots, new sprout ginseng, and wild ginseng. More preferably, it is black ginseng. Specifically, the black ginseng is produced by repeatedly steaming ginseng at 90 to 110°C for 1 to 3 hours, 3 to 4 times, and then drying at 45 to 55°C so that the moisture content is within 10 to 20% (v / w). More specifically, it can be produced by repeatedly steaming ginseng at 100°C for 2 hours, 3 times, and then drying at 50°C so that the moisture content is within 15% (v / w).
[0016] Also, when manufacturing the treatment liquid, it can be manufactured by further using vitamin tree and loofah. More specifically, water is added to a mixture of black ginseng, vitamin tree, and loofah at a weight ratio of 3.5 to 4.5:2.5 to 3.5:2.5 to 3.5 at 10 to 20 times (v / w), and extracted at 80 to 90 °C for 1 to 3 hours to manufacture the treatment liquid. More specifically, water is added to a mixture of black ginseng, vitamin tree, and loofah at a weight ratio of 4:3:3 at 15 times (v / w), and extracted at 85 °C for 2 hours to manufacture the treatment liquid. By pretreating Korean ginseng with the treatment liquid manufactured under the above conditions, while further effectively reducing the unique bitterness of Korean ginseng, it was possible to perform pretreatment so that the preference was excellent due to the taste and aroma of the treatment liquid.
[0017] Also, in the method for manufacturing the concentrated Korean ginseng liquid of the present invention, in the step (2), preferably, Korean ginseng is immersed in the treatment liquid at 25 to 35 °C for 25 to 35 minutes, then taken out and dried at 45 to 55 °C so that the moisture content becomes 10 to 20% (v / w). More preferably, Korean ginseng is immersed in the treatment liquid at 30 °C for 30 minutes, then taken out and dried at 50 °C so that the moisture content becomes 15% (v / w). By immersing Korean ginseng in the immersion liquid under the above conditions and then drying it, it was possible to improve the soft taste and throat-clearing effect.
[0018] The Korean ginseng in the step (2) is preferably also one or more Korean ginsengs selected from the group consisting of fresh ginseng, red ginseng, dried ginseng, white ginseng, black ginseng, taegeuk ginseng, cheong ginseng, mountain-grown ginseng, camphor ginseng, cultured mountain ginseng roots, new sprout ginseng, and wild ginseng. More preferably, it is red ginseng or black ginseng. Specifically, the red ginseng is manufactured by steaming Korean ginseng at 90 to 110 °C for 1 to 3 hours and then drying it at 45 to 55 °C so that the moisture content is within 10 to 20% (v / w). More specifically, it can be manufactured by steaming Korean ginseng at 100 °C for 2 hours and then drying it at 50 °C so that the moisture content is within 15% (v / w).
[0019] In addition, in the method for producing the concentrated ginseng extract of the present invention, the ginseng extract in the step (3) is preferably produced by adding water at 5 to 10 times (v / w) to the pulverized ginseng obtained by pulverizing dried ginseng and then extracting at 85 to 90°C for 1 to 3 hours. More preferably, it can be produced by adding water at 8 times (v / w) to the pulverized ginseng obtained by pulverizing dried ginseng and then extracting at 85 to 90°C for 2 hours.
[0020] In addition, in the method for producing the concentrated ginseng extract of the present invention, the composite adsorbent in the step (4) is preferably produced by mixing acid clay, activated carbon, and diatomaceous earth at a weight ratio of 1.8 to 2.2:0.8 to 1.2:0.8 to 1.2. More preferably, it can be produced by mixing acid clay, activated carbon, and diatomaceous earth at a weight ratio of 2:1:1.
[0021] In addition, in the method for producing the concentrated ginseng extract of the present invention, when passing the ginseng extract in the step (5) through the composite adsorbent to produce a composite adsorbent-treated ginseng extract, specifically, the ginseng extract is passed through the composite adsorbent at a flow rate (bed volume) of 12 to 16 l / hr and a flow velocity (space velocity) of 9 to 13 cm 3 / hr. More specifically, the ginseng extract can be passed through the composite adsorbent at a flow rate (bed volume) of 14 l / hr and a flow velocity (space velocity) of 11 cm 3 / hr. By treating with the adsorbent under the above conditions, it was possible to effectively reduce the off-flavor, off-odor, and bitterness of the ginseng extract while preventing the reduction of ginsenosides in the ginseng extract. However, when the adsorbent treatment conditions are outside the above range, there is a problem that the active ingredients of ginseng are adsorbed by the adsorbent, resulting in a decrease in the ginsenoside content, or the bitterness reduction effect is negligible.
[0022] Also, in the method for producing the concentrated Korean ginseng liquid of the present invention, the cyclodextrin mixed powder in the step (6) is preferably produced by mixing α-cyclodextrin, β-cyclodextrin, and γ-cyclodextrin at a weight ratio of 1.5 to 2.5:4.5 to 5.5:2.5 to 3.5, and more preferably, α-cyclodextrin, β-cyclodextrin, and γ-cyclodextrin can be produced by mixing at a weight ratio of 2:5:3. By using α-cyclodextrin, β-cyclodextrin, and γ-cyclodextrin in a specific ratio in combination as described above, the bitterness of Korean ginseng can be effectively reduced while further enhancing the palatability.
[0023] Also, in the method for producing the concentrated Korean ginseng liquid of the present invention, in the step (7), preferably, 3 to 7% (w / v) of cyclodextrin mixed powder is added to the Korean ginseng extract treated with the composite adsorbent, stirred, then allowed to stand at 4 to 7°C for 10 to 14 hours, filtered, and more preferably, 5% (w / v) of cyclodextrin mixed powder is added to the Korean ginseng extract treated with the composite adsorbent, stirred, then allowed to stand at 4 to 7°C for 12 hours, and filtered. As described above, by treating with cyclodextrin, the harmful components and bitterness in the concentrated Korean ginseng liquid can be effectively reduced without reducing the content of ginsenosides in the concentrated Korean ginseng liquid.
[0024] Also, in the method for producing the concentrated Korean ginseng liquid of the present invention, the concentration in the step (8) is preferably concentrated at 50 to 60°C to 60 to 70 brix for the Korean ginseng extract, and more preferably, the Korean ginseng extract can be concentrated at 55°C and 65 brix.
[0025] The method for producing the concentrated Korean ginseng liquid of the present invention is more specifically, (1) A step of adding water to Korean ginseng at 10 to 20 times (v / w), and then extracting at 80 to 90°C for 1 to 3 hours to produce a treatment liquid; (2) A step of immersing Korean ginseng in the treatment liquid produced in the step (1) at 25 to 35°C for 25 to 35 minutes, then taking it out, and drying at 45 to 55°C so that the moisture content becomes 10 to 20% (v / w); (3) Adding water at 5 - 10 times (v / w) to the pulverized ginseng product obtained by pulverizing the ginseng dried in the previous step (2), and then extracting at 85 - 90 °C for 1 - 3 hours to produce a ginseng extract; (4) Mixing acidic clay, activated carbon, and diatomaceous earth in a weight ratio of 1.8 - 2.2:0.8 - 1.2:0.8 - 1.2 to produce a composite adsorbent; (5) Passing the ginseng extract produced in the previous step (3) through the composite adsorbent produced in the previous step (4) to produce a ginseng extract treated with the composite adsorbent; (6) Mixing α - cyclodextrin, β - cyclodextrin, and γ - cyclodextrin in a weight ratio of 1.5 - 2.5:4.5 - 5.5:2.5 - 3.5 to produce a cyclodextrin mixed powder; (7) Adding 3 - 7% (w / v) of the cyclodextrin mixed powder produced in the previous step (6) to the ginseng extract treated with the composite adsorbent produced in the previous step (5), stirring, then standing at 4 - 7 °C for 10 - 14 hours, and filtering; (8) Concentrating the ginseng extract filtered in the previous step (7) at 50 - 60 °C to 60 - 70 brix. The method includes the above steps. More specifically, (1) Adding water at 15 times (v / w) to ginseng, and then extracting at 85 °C for 2 hours to produce a treatment liquid; (2) Immersing ginseng in the treatment liquid produced in the previous step (1) at 30 °C for 30 minutes, then taking it out and drying at 50 °C until the water content reaches 15% (v / w); (3) Adding water at 8 times (v / w) to the pulverized ginseng product obtained by pulverizing the ginseng dried in the previous step (2), and then extracting at 85 - 90 °C for 2 hours to produce a ginseng extract; (4) Mixing acidic clay, activated carbon, and diatomaceous earth in a weight ratio of 2:1:1 to produce a composite adsorbent; (5) Passing the ginseng extract produced in the previous step (3) through the composite adsorbent produced in the previous step (4) to produce a ginseng extract treated with the composite adsorbent; (6) Mixing α-cyclodextrin, β-cyclodextrin and γ-cyclodextrin in a weight ratio of 2:5:3 to produce a cyclodextrin mixed powder; (7) Adding 5% (w / v) of the cyclodextrin mixed powder produced in step (6) to the composite adsorbent-treated Korean ginseng extract produced in step (5), stirring, standing at 4 - 7°C for 12 hours, and filtering; (8) Concentrating the Korean ginseng extract filtered in step (7) at 55°C and 65 brix. It may include the above steps.
[0026] The present invention also provides a concentrated Korean ginseng solution produced by the above method.

[0040] Materials and Methods
[0041] 1) Preparation of Composite Adsorbent Column For the production of the composite adsorbent column, an acid clay, activated carbon, and diatomaceous earth were used to produce the composite adsorbent column (Figure 1). Acid clay is mainly composed of hydrated aluminum silicate obtained by purifying montmorillonite-based clay minerals. After adding 500 mL of water to 1 g of acid clay and suspending it, the pH of the filtered liquid is 3.0 - 5.0, and the desirable acidity is 4.5. The moisture content is 15% or less, but the desirable condition is 10% or less. Activated carbon is obtained by carbonizing carbon-containing substances such as large sawdust, wood chips, plant fibers of coconut husks, lignite, or petroleum and then activating them. Desirably, it is activated carbon made from coconut husks. Put 0.5 g of activated carbon into a test tube, heat it with a direct flame while blowing air into the test tube mouth, and pass the gas generated by burning without ignition through a potassium hydroxide test solution to cause turbidity. Add 10 mL of a light methylene blue test solution and 2 drops of light hydrochloric acid to 0.1 g of activated carbon powder, shake well and mix, and the filtered liquid through quantitative filter paper must not be colorless. Diatomite is a substance composed of diatoms and the protective membranes (diatom shells) of single-celled plants.

[0043] 2) Taste pattern analysis via electronic tongue (E-tongue) analysis Red ginseng concentrate without bitterness control (RG-C), red ginseng concentrate treated with an adsorbent (RG-B), and red ginseng concentrate treated with cyclodextrin after adsorbent treatment (RG-A) were each diluted 100 times with distilled water and then measured for 120 seconds for each sample using an electronic tongue (100 Ml of the diluted solution), and repeated 5 times.
[0044] The electronic tongue has seven types of sensors (AHS - sour taste, CTS - salty taste, NMS - umami, PKS - sweet taste, ANS - bitter taste, CPS, SCS), and CPS and SCS were used in the standard. The seven sensors did not measure the respective chemical components, but sensed the overall taste of the analyzed red ginseng concentrate and converted the respective sensor sensitivities into taste scores with a range of 0 - 10 for display. The taste component pattern analysis of each sample was performed using the discriminant function analysis (DFA) with the Alpha MOS program AlphaSoft 17 ver. (Alpha MOS, Toulouse, France).
[0045] 3) Measurement of ginsenoside content The content analysis of ginsenosides was performed by HPLC (High Performance Liquid Chromatography) equipped with a UVD (Ultra Visible Detector), and the analysis conditions are as shown in Table 2 below, and the content calculation formula is as shown in the following formula. Ginsenoside content (mg / g) = S × (a × b) / sample collection amount (g) S: Individual ginsenoside concentration (mg / ml) in the test solution a: Total volume (ml) of the test solution b: Dilution factor

Table 2
[0047] 4) Sensory evaluation Twenty adults were made to ingest each processed red ginseng concentrate to evaluate the degree of preference. In the preference evaluation, for the items of sweet taste, sour taste, bitter taste, salty taste and spicy taste, the higher the intensity, the higher the score was given. Also, for the overall preference degree, the higher the taste evaluation, the higher the score was given.
[0048] Example 1. Results of electronic tongue analysis
[0049] The results of principal component analysis (PCA) for comparing the patterns of the red ginseng concentrated liquid flavors of Production Example 1 (RG-A), Comparative Example 1 (RG-C), and Comparative Example 2 (RG-B) are as shown in Fig. 3. As a result of principal component analysis, the red ginseng concentrated liquids of RG-C, RG-B, and RG-A were shown differently, indicating that there were relative differences in taste. The distance between RG-A and RG-B, and between RG-B and RG-C was relatively close, but the distance between RG-A and RG-C was far apart.
[0050] As a result of discriminant factorial analysis (DFA), it was shown that there were differences between RG-C, RG-A, and RG-B (Fig. 4). The distance values for each red ginseng concentrated liquid are as shown in Table 3 below. A large distance means that there are many taste differences between them, and a small distance means that they are more similar to each other.
[0051]
Table 3
[0052] In addition, as a result of quantifying the sensitivity of each sensor for the three samples in the electronic tongue analysis results for relative comparison, RG-C was generally relatively higher than RG-B and RG-A in terms of bitterness, sourness, sweetness, saltiness, umami, etc. In the comparison between RG-B and RG-A, RG-A with the adsorbent + cyclodextrin double treatment showed better taste results in terms of taste such as bitterness and sweetness than RG-B with the single adsorbent treatment (Fig. 5).
[0053] Example 2. Ginsenoside analysis results
[0054] The ginsenoside analysis results of the red ginseng concentrated liquid in Production Example 1 and the comparative examples are as shown in Table 4 below. As a result, the total content of the five ginsenosides in the red ginseng concentrated liquid of Production Example 1 was shown to be even higher than that of the red ginseng concentrated liquid of the comparative examples.

[0056] Example 3. Selection of Composite Adsorbent Types
[0057] After treating the red ginseng extract with different types of composite adsorbent columns and then concentrating the red ginseng concentrate (without cyclodextrin treatment), the results of the sensory evaluation using the concentrated red ginseng concentrate are as shown in Table 5 below. As a result, the red ginseng concentrate treated with the composite adsorbent column obtained by mixing acidic clay, activated carbon, and diatomaceous earth in a weight ratio of 2:1:1 had the lowest bitterness and the highest overall preference.
[0058]
Table 5
[0059] Example 4. Selection of Cyclodextrin Types
[0060] After treating the red ginseng extract with different types of cyclodextrins (α, β, γ) and then concentrating the red ginseng concentrate (without adsorbent treatment), the sensory evaluation was compared. As a result, the red ginseng concentrate treated with the cyclodextrin mixed powder obtained by mixing α-cyclodextrin, β-cyclodextrin, and γ-cyclodextrin in a ratio of 2:5:3 had high sweetness, low bitterness, and the highest preference.
[0061]
Table 6
[0062] Example 5. Comparison by Composite Treatment
[0063] The ratio of the adsorbent and cyclodextrin optimized through Examples 3 and 4 was determined, and a sensory evaluation of the red ginseng extract with the combined treatment of the adsorbent and cyclodextrin was carried out. As a result, compared with the treatment with the adsorbent alone or cyclodextrin alone, the sweetness of the red ginseng concentrate was further increased, and the sourness and bitterness were decreased by the combined treatment of the adsorbent + cyclodextrin, confirming that the preference for the red ginseng concentrate could be further improved.

Table 7
[0065] Example 6. Comparison by treatment liquid materials
[0066] The results of the sensory evaluation using the red ginseng concentrates of Production Example 1 and Production Example 3 are as shown in Table 8 below. As a result, it was confirmed that Production Example 3, which used black ginseng, vitamin tree, and loofah in the production of the treatment liquid, had a higher preference while further reducing the bitterness and sourness compared to the red ginseng concentrate of Production Example 1.
