Description

[Technical Field]

[0001] The present invention relates to a skin penetration enhancer, a method for enhancing skin penetration, and a method for producing a cosmetic composition. [Background technology]

[0002] Royal jelly is known to exert beauty effects such as preventing skin aging and promoting moisturized and firm skin when applied to the skin (e.g., Patent Document 1). These beauty effects are thought to be due to fatty acids unique to royal jelly, particularly 10-hydroxy-2-decenoic acid. Therefore, in order to fully exert the beauty effects of royal jelly, the permeability of royal jelly, particularly the fatty acids unique to royal jelly, into the skin is important. Patent Document 2 discloses an external skin preparation containing royal jelly extract as a skin-activating ingredient, and describes that the external skin preparation stably maintains moisture on the skin for a long period of time, thereby increasing the permeability of the skin-activating ingredient into the skin. [0008] [Figure 1] FIG. 1 shows the results of imaging MS showing the skin permeability of 10-hydroxy-2-decenoic acid. [Figure 2] FIG. 2 shows the results of imaging MS showing the skin permeability of 10-hydroxydecanoic acid. DETAILED DESCRIPTION OF THE INVENTION

[0009] Hereinafter, embodiments of the present invention will be described in detail, but the present invention is not limited to the following embodiments.

[0010] The skin penetration enhancer for royal jelly of the present invention contains an acrylates / C10-30 alkyl acrylate crosspolymer and / or an ammonium acryloyldimethyltaurate / beheneth-25 methacrylate crosspolymer as an active ingredient.

[0011] The (acrylates / alkyl acrylate (C10-30)) crosspolymer is a copolymer of one or more monomers consisting of acrylic acid, methacrylic acid, or simple esters thereof and alkyl acrylate, crosslinked with an allyl ether of sucrose or an allyl ether of pentaerythritol. Specifically, the alkyl acrylate is an alkyl acrylate having an alkyl group having 10 to 30 carbon atoms. A commercially available (acrylates / alkyl acrylate (C10-30)) crosspolymer (e.g., Pemulen (trademark) TR-1) may be used as the (acrylates / alkyl acrylate (C10-30)) crosspolymer.

[0012] (Ammonium Acryloyldimethyltaurate / Beheneth-25 Methacrylate) Crosspolymer is a copolymer of ammonium acryloyldimethyltaurate and the methacrylic acid ester of Beheneth-25. Beheneth-25 is a polyethylene glycol ether of behenyl alcohol.

[0013] Acrylates / C10-30 alkyl acrylate crosspolymer and ammonium acryloyldimethyltaurate / beheneth-25 methacrylate crosspolymer are cosmetic ingredients known as thickeners and emulsion stabilizers, but their effect in promoting the penetration of royal jelly into the skin was not previously known.

[0014] "Enhancing the penetration of royal jelly into the skin" means that the components contained in royal jelly, particularly active ingredients with cosmetic effects, such as saturated fatty acids and unsaturated fatty acids, penetrate deeper into the stratum corneum of the skin compared to when a penetration enhancer is not included. Examples of saturated fatty acids include 10-hydroxydecanoic acid, 8-hydroxyoctanoic acid, 12-hydroxydodecanoic acid, 3,10-dihydroxydecanoic acid, and capric acid. Examples of unsaturated fatty acids include 10-hydroxy-2-decenoic acid, decenedioic acid, sebacic acid, and trans-2-decenoic acid. Preferably, the saturated fatty acid is 10-hydroxydecanoic acid, and the unsaturated fatty acid is 10-hydroxy-2-decenoic acid, decenedioic acid, and sebacic acid.

[0015] Enhanced penetration of royal jelly into the skin can be evaluated by analyzing the components contained in royal jelly, particularly components such as saturated fatty acids and unsaturated fatty acids, for example, by the method described in the Examples. Specifically, a composition containing royal jelly and a penetration enhancer is applied to the skin, and after a predetermined time (e.g., 30 minutes, 1 hour, or 2 hours), a stratum corneum sample is collected by a tape stripping test, and royal jelly attached to the tape is detected.

[0016] The method for detecting royal jelly components that have penetrated into the stratum corneum is not particularly limited, and for example, they may be detected by imaging using a mass spectrometer as described in the examples of the present application.

[0017] (royal jelly) Royal jelly is a milky white gel-like substance produced by honeybee worker bees (3-12 days old) by mixing secretions from the hypopharyngeal and mandibular glands. The main physiologically active components in royal jelly include organic acids such as saturated and unsaturated fatty acids, which are unique to royal jelly, as well as proteins, amino acids, peptides, lipids, sugars, vitamin B, folic acid, nicotinic acid, pantothenic acid, and other vitamins, and various minerals.

[0018] In this specification, royal jelly may be, for example, fresh royal jelly or a processed royal jelly obtained by processing fresh royal jelly.

[0019] Fresh royal jelly can be obtained, for example, as a beekeeping product according to conventional methods. Royal jelly may be produced anywhere, including Japan, China, Brazil, European countries, Oceanian countries, and the United States. The type of honeybee used to harvest fresh royal jelly is not limited. Fresh royal jelly also includes frozen royal jelly, which is obtained by freezing and storing harvested royal jelly.

[0020] Examples of processed royal jelly include royal jelly concentrates or dilutions obtained by concentrating or diluting fresh royal jelly, dried royal jelly powder obtained by drying and powdering fresh royal jelly, royal jelly extracts (royal jelly extracts) obtained by extracting royal jelly with a solvent such as water or ethanol, and enzyme-treated royal jelly obtained by treating royal jelly with a protease. The processed royal jelly may also be one that has undergone multiple treatments. For example, royal jelly may be enzyme-treated royal jelly powder obtained by enzymatically hydrolyzing and powdering royal jelly.

[0021] Royal jelly concentrate can be obtained, for example, by removing water from fresh royal jelly, and royal jelly dilution can be obtained, for example, by adding water to fresh royal jelly.

[0022] Royal jelly powder can be obtained by powdering fresh royal jelly using methods known in the art, such as freeze-drying and spray-drying. Drying methods include natural drying (e.g., ventilation drying and sun drying), forced drying (using electricity or other heating), and freeze-drying, all of which are commonly used in food processing. Freeze-drying is preferred. The drying time is not particularly limited; for natural drying (e.g., ventilation or sun drying), it can be approximately 3 days, while for forced drying (e.g., using electricity or other heating), it can be approximately 1 to 3 days at approximately 50°C. It is generally preferable to dry the product to a moisture content of 10% by mass or less, preferably 5% by mass or less. If it is difficult to reduce the moisture content to 10% by mass or less, as with natural drying (e.g., ventilation or sun drying), a subsequent freeze-drying process to further reduce the moisture content may be used. Alternatively, royal jelly powder may be obtained by pulverizing the freeze-drying or spray-drying product using a mill (e.g., a pin mill, hammer mill, ball mill, or jet mill) after the dry-drying process.

[0023] Royal jelly extract (royal jelly extract) can be obtained by extracting raw royal jelly, royal jelly powder, or enzyme-treated royal jelly using, for example, a solvent such as water, ethanol, methanol, propanol, or acetone as an extraction solvent. The extraction time can be appropriately set depending on the form of royal jelly used as the raw material, the type and amount of solvent, the temperature and stirring conditions during extraction, etc. After extraction, solids may be removed by filtration, centrifugation, or the like. The extracted extract may be used as is, or the solvent may be removed from the extract and used as a concentrated liquid or dried powder. The royal jelly extract may be a royal jelly water extract extracted with water, or a royal jelly ethanol extract extracted with ethanol.

[0024] Enzyme-treated royal jelly can be obtained, for example, by treating raw royal jelly or royal jelly powder with a protease. The protease is preferably selected from the group consisting of enzymes with endopeptidase activity, enzymes with exopeptidase activity, and enzymes with both endopeptidase and exopeptidase activity. Peptidases that simultaneously possess both endopeptidase and exopeptidase activity are particularly preferred. Enzyme treatment using such peptidases can degrade proteins in a single step, offering the advantages of simple procedures and preventing the loss or significant reduction of the physiological activity of useful components contained in royal jelly.

[0025] Preferred examples of enzymes having endopeptidase activity include peptidases produced by Bacillus subtilis (trade names: Orientase 22BF, Nucleicin), peptidases produced by Bacillus licheniformis (trade name: Alcalase), peptidases produced by Bacillus stearothermophilus (trade name: Protease S), peptidases produced by Bacillus amyloliquefaciens (trade name: Neutrase), and peptidases produced by the genus Bacillus (trade name: Protamex).

[0026] Preferred examples of enzymes having exopeptidase activity include peptidases produced by Aspergillus oryzae (trade names: Umamizyme G, Promod 192P, Promod 194P, Sumizyme FLAP), peptidases produced by Aspergillus sojae (trade name: Sternzyme B15024), peptidases produced by the genus Aspergillus (trade name: Coclase P), and peptidases produced by Rhizopus oryzae (trade name: Peptidase R).

[0027] Preferred examples of enzymes having both exopeptidase activity and endopeptidase activity include peptidases produced by Aspergillus oryzae (trade names: Sumiteam LP-G, Proteax, Flavorzyme, Protease A), peptidases produced by Streptomyces griseus (trade name: Actinase AS), and peptidases produced by Aspergillus melleus (trade name: Protease P).

[0028] The reaction conditions (amount of protease used, reaction temperature, pH, reaction time, etc.) when treating raw royal jelly or royal jelly powder with a protease may be appropriately set depending on the type of protease used, etc.

[0029] The royal jelly is preferably an enzyme-treated royal jelly or a royal jelly extract, and particularly preferably an enzyme-treated royal jelly. The enzyme-treated royal jelly is preferably obtained by treating with an enzyme having both exopeptidase activity and endopeptidase activity.

[0030] Commercially available royal jelly may be used. Specific examples of royal jelly include Royal Jelly FD Powder (Nakahara Co., Ltd.), Royal Jelly Extract SF (Matsuura Pharmaceutical Co., Ltd.), Deproteinized Royal Jelly Powder F (Maruzen Pharmaceutical Co., Ltd.), and Deproteinized Royal Jelly Extract (API Co., Ltd.).

[0031] Royal jelly is not particularly limited as long as it has cosmetic effects such as anti-aging effects and moisturizing and firming skin, but preferably contains saturated fatty acids and / or unsaturated fatty acids, the saturated fatty acids preferably containing 10-hydroxy-2-decenoic acid, and the unsaturated fatty acids preferably containing at least one selected from the group consisting of 10-hydroxydecanoic acid, decenedioic acid, and sebacic acid. The penetration enhancer of the present invention can be used in external skin preparations, cosmetic compositions, etc. containing royal jelly.

[0032] (Cosmetic composition) The cosmetic composition of the present invention contains the skin penetration enhancer for royal jelly of the present invention and royal jelly.

[0033] The content of the (acrylates / alkyl acrylate (C10-30)) crosspolymer in the cosmetic composition may be an amount (effective amount) that is sufficient to exhibit the effect of promoting the penetration of royal jelly into the skin, and may be, for example, 0.01% by mass or more, 0.1% by mass or more, 0.3% by mass or more, 0.4% by mass or more, 0.5% by mass or more, 1% by mass or more, 2% by mass or more, 3% by mass or more, 5% by mass or more, or 20% by mass or less, 15% by mass or less, 10% by mass or less, 8% by mass or less, 5% by mass or less, 3% by mass or less, 2% by mass or less, 1% by mass or less, 0.5% by mass or less, relative to the total amount of the cosmetic composition.

[0034] The content of the (ammonium acryloyldimethyltaurate / beheneth-25 methacrylate) crosspolymer in the cosmetic composition may be an effective amount sufficient to enhance the skin penetration of royal jelly, and may be, for example, 0.1% by mass or more, 0.3% by mass or more, 0.5% by mass or more, 0.8% by mass or more, 1% by mass or more, 2% by mass or more, 3% by mass or more, or 5% by mass or more, relative to the total amount of the cosmetic composition, and may be 20% by mass or less, 15% by mass or less, 10% by mass or less, 8% by mass or less, 5% by mass or less, 3% by mass or less, 2% by mass or less, 1% by mass or less, or 0.5% by mass or less. When the cosmetic composition contains both the (acrylates / C10-30 alkyl acrylate) crosspolymer and the (ammonium acryloyldimethyltaurate / beheneth-25 methacrylate) crosspolymer, the total content thereof may be within the above range.

[0035] The content of royal jelly in the cosmetic composition is not particularly limited, as long as it is within a range in which royal jelly can exert its cosmetic effects, and may be, for example, 0.1% by mass or more, 0.5% by mass or more, 1% by mass or more, 1.5% by mass or more, 2% by mass or more, 3% by mass or more, 5% by mass or more, 8% by mass or more, 10% by mass or more, 15% by mass or more, or 20% by mass or less, 15% by mass or less, 10% by mass or less, 8% by mass or less, 5% by mass or less, 3% by mass or less, 2% by mass or less, 1% by mass or less, 0.5% by mass or less, relative to the total amount of the cosmetic composition.

[0036] The cosmetic composition may be in any form, such as a solid, liquid, or paste. The cosmetic may be a medicated cosmetic (i.e., a quasi-drug). The cosmetic composition includes any cosmetic composition that can be applied to the skin, mucous membranes, body hair, head hair, scalp, nails, teeth, facial skin, lips, etc. of animals (particularly humans). Of these, from the viewpoint of promoting the penetration of royal jelly, a cosmetic composition that can be applied to the skin of the entire body, such as the face, scalp, lips, etc., is preferred.

[0037] The cosmetic composition may contain, in addition to the penetration enhancer of the present invention and royal jelly, cosmetically acceptable ingredients such as whitening agents, moisturizers, antioxidants, oily ingredients, UV absorbers, surfactants, thickeners, alcohols, powder ingredients, coloring materials, aqueous ingredients, water, and various skin nutrients.

[0039] The method for producing the cosmetic composition is not particularly limited and can be any known method. For example, the cosmetic composition for the above purpose can be obtained by mixing the penetration enhancer and royal jelly with an intermediate product or a final product in the cosmetic production process.

[0040] (Method for promoting royal jelly penetration into the skin) In one embodiment, a method for enhancing the penetration of royal jelly into the skin comprises incorporating a penetration enhancer of the present invention into a cosmetic composition containing royal jelly. By incorporating the penetration enhancer of the present invention into a cosmetic composition containing royal jelly, when the resulting cosmetic composition is applied to the skin, the presence of the penetration enhancer of the present invention enhances the penetration of the royal jelly contained in the cosmetic composition into the skin more than a cosmetic composition not containing the penetration enhancer of the present invention.

[0041] In another embodiment, a method for enhancing the penetration of royal jelly into the skin comprises applying the cosmetic composition of the present invention to the skin. By applying the cosmetic composition of the present invention to the skin, the presence of the penetration enhancer of the present invention enhances the penetration of royal jelly contained in the cosmetic composition into the skin more than a cosmetic composition not containing the penetration enhancer of the present invention. [Example]

[0042] In order to specifically explain the present invention, examples are shown below, but the present invention is not limited to the following examples.

[0043] (Preparation of enzyme-treated royal jelly extract) The enzyme-treated royal jelly extract was prepared from raw royal jelly according to the method described in WO2020 / 196065 A1. Specifically, 474 g of raw royal jelly was mixed with 400 ml of ion-exchanged water and stirred until homogeneous to prepare a diluted royal jelly solution. The pH of the diluted royal jelly solution was adjusted to 7.8 by adding 2N NaOH aqueous solution. Next, a solution of 4.75 g of Proteax (Amano Enzyme Inc.), which has both endopeptidase and exopeptidase activity, dissolved in 20 mL of ion-exchanged water was added to the diluted royal jelly solution, and further ion-exchanged water was added to bring the total volume to 980 g. The reaction mixture was stirred with a propeller at 50°C (in a thermostatic water bath) for 2 hours to hydrolyze the mixture. The temperature of the thermostatic water bath was then raised to 80°C and the mixture was stirred for 15 minutes to inactivate the protease. The resulting enzyme-treated royal jelly solution was filtered through a filter with a particle pore size of 3 μm to obtain an enzyme-treated royal jelly solution. Butylene glycol, pentylene glycol, phenoxyethanol, and purified water were added to the enzyme-treated royal jelly solution so that the concentration of the enzyme-treated royal jelly solution became 2.5 wt % to obtain an enzyme-treated royal jelly extract.

[0044] (Preparation of cosmetic composition of formulation example 1) The enzyme-treated royal jelly extract, acrylates / C10-30 alkyl acrylate crosspolymer as a penetration enhancer, and other ingredients listed in Table 1 were mixed in a predetermined ratio to prepare a cosmetic composition containing the enzyme-treated royal jelly extract of Formulation Example 1. The composition of the resulting Formulation Example 1 is shown in Table 1.

[0045] [Table 1]

[0046] (Preparation of cosmetic composition of formulation example 2) The enzyme-treated royal jelly extract, ammonium acryloyldimethyltaurate / beheneth-25 methacrylate crosspolymer as a penetration enhancer, and other ingredients listed in Table 2 were mixed in the prescribed proportions and stirred at 3,000 rpm for 10 minutes using a homogenizer to prepare the cosmetic composition of Formulation Example 2. The composition of the resulting Formulation Example 2 is shown in Table 2.

[0047] [Table 2]

[0048] (Preparation of Comparative Cosmetic Composition) A comparative cosmetic composition was prepared by mixing the enzyme-treated royal jelly extract and the other ingredients listed in Table 3 in the prescribed proportions. The composition of the obtained comparative example is shown in Table 3.

[0049] [Table 3]

[0050] (Skin penetration test) The skin permeability of the cosmetic compositions of Formulation Example 1, Formulation Example 2, and Comparative Example was evaluated by the following method. First, samples were prepared using a tape stripping test. The forearm of one volunteer was washed and left at room temperature for 15 minutes. The cosmetic compositions of Formulation Example 1, Formulation Example 2, and Comparative Example were applied at 50 mg / cm. 2 6.25cm of forearm 2 The composition was applied to a square of skin and left to stand for 2 hours. The applied area was washed and left to stand for 15 minutes. A 2 cm diameter circular tape (D-Squame sampling discs (CUDERM)) was then applied to the applied skin area, pressure was applied for 1 minute, the circular tape was then peeled off, and the center of the removed circular tape was cut to a width of 5 mm to serve as an imaging mass spectrometry (imaging MS) sample. Similarly, the circular tape was applied to the applied skin area and peeled off nine more times, and a total of 10 imaging MS samples were obtained by cutting the center of the circular tape to a width of 5 mm. Ten samples were also prepared in the same manner as a control, using skin before application of the test cosmetic composition.

[0051] Imaging MS was performed using a DESI Xevo G2 XS QTof (Waters). The amounts of 10-hydroxy-2-decenoic acid and 10-hydroxydecanoic acid contained in royal jelly were quantified by detecting the m / z 185.1172 and m / z 187.1328 ions.

[0052] Figures 1 and 2 show the results of imaging MS analysis demonstrating the skin permeability of 10-hydroxy-2-decenoic acid and 10-hydroxydecanoic acid, respectively. Figures 1 and 2 show that, while no skin permeation of 10-hydroxy-2-decenoic acid or 10-hydroxydecanoic acid was observed in the control and comparative cosmetic compositions, 10-hydroxy-2-decenoic acid and 10-hydroxydecanoic acid were detected up to the sixth tape in the cosmetic composition of Formulation Example 1. Similarly, 10-hydroxy-2-decenoic acid and 10-hydroxydecanoic acid were detected up to the fifth tape in the cosmetic composition of Formulation Example 2. These results demonstrate that the addition of an acrylates / C10-30 alkyl acrylate crosspolymer or an ammonium acryloyldimethyltaurate / beheneth-25 methacrylate crosspolymer can enhance the skin permeation of royal jelly.
