AI Technical Summary
Conventional microneedles face challenges in efficiently injecting liquids intradermally due to skin swelling caused by wheal formation, which can lead to improper fluid distribution or leakage, especially when large volumes are administered.
The injection needle design features first and second protrusions with specific geometric arrangements and distances, including a cone-shaped first protrusion with an opening and a second protrusion without an opening, arranged to minimize skin swelling by allowing adequate space for skin bulge, thereby enhancing efficient liquid delivery.
The design effectively prevents skin swelling, ensuring efficient injection of liquids even with large volumes, reducing the risk of leakage and improving the overall injectability of the needle.

Abstract
Description
[Technical field]
[0001] The present invention relates to an injection needle. [Background technology]
[0002] In recent years, in the fields of medicine and beauty, intradermal administration of liquids such as medicines using injection needles equipped with fine needle-like protrusions, also called microneedles, has been attracting attention. With this injection needle, it is possible to inject liquid into the body by inserting the microneedle into a relatively shallow layer of the skin, such as the stratum corneum, and since the pain felt by the subject is significantly reduced compared to that felt by a normal syringe, it has attracted attention as a minimally invasive means of administering liquids.
[0003] Patent Document 1 describes a hub assembly including a columnar limiter from which a microneedle protrudes, and a stabilizing ring surrounding the microneedle and the limiter. The limiter is for controlling the insertion depth of the microneedle into the skin. The stabilizing ring is for suppressing deformation of the skin when the microneedle is inserted into the skin. The hub assembly has an attachment mechanism and is fixed to a drug delivery device via the attachment mechanism.
[0004] Patent Document 2 describes a microneedle sheet on which multiple minute needle peaks are formed and protrude from the surface. The document describes that the needle peaks include penetrating needle peaks in which through holes are formed from the front surface side to the back surface side, and dummy needle peaks in which no through holes are formed. [0005] [Patent Document 1] Special Publication No. 2009-516572 [Patent Document 2] International Publication No. 2021 / 177317 Summary of the Invention [Problem to be solved by the invention]
[0006] Intradermal injection has been attracting attention as an injection method that can efficiently take in antigens. It has been reported by the World Health Organization and others that intradermal injection can reduce the amount of vaccine required for one vaccination to one-fifth compared to intramuscular injection or subcutaneous injection. However, it is known that with conventional injection needles, it takes considerable training to master the technique of intradermal administration. Intradermal administration of liquids such as medicines using injection needles equipped with fine needle-like protrusions, also called microneedles, to facilitate intradermal administration has been attracting attention. However, with conventional microneedles, it may be difficult to properly inject the liquid into the skin due to the wheal that forms when the liquid such as medicine is injected into the skin. This point will be described in detail below.
[0007] When a microneedle is inserted into the skin and liquid is injected into the skin, the injected liquid causes the skin to swell and a wheal is formed (see FIG. 8(a)). If the injection needle has protrusions 82 other than the microneedle 81 for injecting liquid, such as the stabilizing ring in Patent Document 1 and the dummy pincushion in Patent Document 2, these protrusions 82 may suppress the swelling of the skin (see FIG. 8(b)). This may result in poor liquid injection, such as when the amount of liquid injected is large, making it difficult for the liquid to enter the body or causing liquid leakage. Thus, there is a risk that the injection of liquid may be difficult in Patent Documents 1 and 2.
[0008] The present invention relates to an injection needle that can efficiently inject liquid without causing the skin to swell due to the injected liquid when the liquid is injected into the skin with the injection needle. [Means for solving the problem]
[0009] The present invention provides an injection needle. In one embodiment, it is preferable to have fine first and second projections protruding from the sheet base. In one embodiment, the primary projections are preferably cone-shaped and have apertures at their tips. In one embodiment, the secondary projections preferably have no apertures at their tips. In one embodiment, in plan view, it is preferable to have a first region including one or more first projections, and a second region including two or more second projections and sandwiching or surrounding the first region. In one embodiment, the distance D1, which is the shortest distance between the first projection and the second projection, is preferably 2.0 mm or more and 10 mm or less. Effect of the Invention
[0010] According to the present invention, when liquid is injected into the skin with an injection needle, it is difficult to prevent the skin from swelling due to the injected liquid, and the liquid can be injected efficiently. [Brief description of the drawings]

[0012] The present invention will now be described with reference to preferred embodiments thereof. The injection needle 1 of the present invention has protrusions protruding from a sheet base 2. The sheet base 2 is a sheet-like portion that connects the protrusions. When the injection needle 1 is made by forming protrusions on a base sheet, the portion of the injection needle 1 other than the protrusions is the sheet base 2. The injection needle 1 typically has a flat sheet-like shape as a whole, and has a sheet base 2 and a plurality of protrusions protruding from one surface of the sheet base 2. The injection needle 1 typically has a projection arrangement region in which projections are arranged in a dispersed manner in a planar direction. The protrusion arrangement region typically includes a first region R1 and a second region R2, which will be described later.
[0013] The projections of the injection needle 1 preferably include a first projection 11 having a cone shape and an opening 11a at the tip, and a second projection 12 having no opening at the tip. This example is shown in Fig. 1. In the example shown in Fig. 1, the first projection 11 and the second projection 12 are fine projections, and preferably have a protruding height of 5000 µm or less. The pyramidal shape of the first projection 11 is typically an approximately conical shape, but may be a polygonal pyramid such as a quadrangular pyramid. The approximately conical shape includes, in addition to a conical shape, an eccentric cone in which the center and the tip of the projection are in different positions. The primary projections 11 are preferably minute projections having hollow portions 11b, so-called microneedles (see FIG. 3(a)). The second protrusion 12 may be hollow or solid. In the example shown in Fig. 1, the second protrusion 12 is solid (see Figs. 3(b) and (c)). By making the second protrusion 12 solid, when a liquid such as a medicine is injected from the injection needle 1, the amount of medicine remaining in the injection needle 1 can be reduced. As shown in FIG. 1, the second protrusion 12 preferably has a cone-shaped stimulation protrusion 32 and a cylindrical puncture depth control protrusion (hereinafter also referred to as the “control protrusion”) 33, whose tip surface serves as a puncture depth control section 40. It is preferable that the tip surface of the control projection 33 is flat, that is, a straight line extending in the horizontal direction, or a curved line convex toward the protruding direction of the control projection 33 . The cone shape of the stimulation protrusion 32 may be a substantially cone shape or a polygonal pyramid shape, similar to the first protrusion 11. The columnar shape of the control protrusion 33 may be a circular cylinder shape or a polygonal column shape such as a square column. The injection needle 1 preferably includes, as the second protrusion 12, a stimulation protrusion 32 having a cone shape or a control protrusion 33 having a cylinder shape, and more preferably includes both.
[0014] The injection needle 1 preferably has a first region R1 including one or more first projections 11 in plan view. The first region R1 is preferably located in the central region of the sheet base 2 in plan view. The central region is, for example, a circular region having a diameter that is one-third, preferably one-half, of the diameter of the sheet base 2. In the illustrated example, the center of the sheet base 2 and the center of the first region R1 coincide with each other, but the centers may be offset from each other.
[0015] The number of first projections 11 included in the first region R1 may be one or two or more. It is preferable that the multiple first projections 11 are arranged and collectively form any shape as a whole. It is preferable that the multiple first projections 11 are arranged, for example, in a triangular shape (see FIG. 2), a circular shape (see FIG. 6(a)), or a linear shape (see FIG. 6(d)). It is preferable that the multiple first protrusions 11 are arranged such that a virtual line C1 connecting the centers of the first protrusions 11 forms a ring in a plan view (see Figs. 2, 6(a) and 6(c)). Here, "ring" means a closed shape, and includes not only a circular shape but also a polygonal shape and the like. When the first protrusions 11 are arranged such that a virtual line C1 connecting the centers of the first protrusions 11 forms a ring, it is preferable that no first protrusions 11 that are entirely present within the region surrounded by the virtual line C1 are arranged. In the injection needle 1 shown in FIG. 2, the centers of the multiple first projections 11 are located on the same circle.
[0016] The injection needle 1 preferably has a second region R2 including two or more second projections 12. The second region preferably sandwiches or surrounds the first region R1. In the example shown in Figure 2, the second region R2 surrounds the first region R1. The second projections 12 included in the second region R2 are preferably arranged such that a virtual line connecting the centers of the second projections 12 forms a ring in a plan view. For example, the virtual line C2 connecting the centers of the second projections 12 may be a circular ring (see Figs. 4 and 6(a)), a square ring (see Fig. 6(b)), or a triangular ring (see Fig. 6(c)). In the injection needle 1 shown in Fig. 2, the centers of the multiple second projections 12 are located on the same circle. An example in which the second region R2 sandwiches the first region R1 is shown in Figure 6(d). When the second region R2 sandwiches the first region R1, it is preferable that the first region R1 does not extend beyond the second region R2.
[0017] Next, the first region R1 and the second region R2 will be described in detail. As shown in Figures 2 and 6(a), when multiple first protrusions 11 are concentrated in the central region of the seat base 2 in a planar view, the area inside the smallest circle surrounding the multiple first protrusions 11 is the first region R1. As shown in Figures 2 and 6(a), when multiple second protrusions 12 are arranged to surround a first region R1, the region between the smallest circle surrounding the multiple second protrusions 12 and the largest circle inscribed in the multiple second protrusions 12 is the second region R2. When a single first protrusion 11, instead of multiple first protrusions 11, is present in the central region of the seat base 2 when viewed in a plan view, the region inside the smallest circle surrounding the first protrusion 11 is defined as the first region R1.
[0018] When the multiple second protrusions 12 are arranged in a polygonal shape so as to surround the multiple first protrusions 11 present collectively in the central region, or when the multiple second protrusions 12 are arranged in a polygonal shape so as to surround a single first protrusion 11 present in the central region, the first region R1 may be an inner region of the smallest shape that is similar to the polygonal shape and surrounds the multiple first protrusions 11 (see Figures 6(b) and (c)). When the multiple secondary protrusions 12 are arranged in a polygonal shape, the second region R2 may be the region between the smallest line that is similar to the polygonal shape and surrounds the multiple secondary protrusions 12 and the largest line that is similar to the polygonal shape and inscribes the multiple secondary protrusions 12 in a planar view (see Figures 6(b) and (c)).
[0019] When a plurality of primary projections 11 are arranged in one direction, the inner region of the smallest rectangle that surrounds the plurality of primary projections 11 is the first region R1 (see FIG. 6(d)). When multiple second protrusions 12 are arranged to sandwich the first region R1 (see Figure 6 (d)), the area inside the smallest rectangle surrounding the second protrusions 12 arranged on each side of the first region R1 is the second region R2.
[0020] Since the injection needle 1 has the opening 11a in the first projection 11, it becomes a liquid injection tool that can be used for intradermal administration of liquid such as medicine. Typically, an opening 11a is formed at the tip of the first projection 11, and the hollow portion 11b of the first projection 11 communicates with the outside through the opening 11a. The opening 11a is a through hole that penetrates the base sheet forming the first projection 11 in the thickness direction, and is preferably located at the tip of the conical first projection 11. The hollow portion 11b of the first projection 11 functions as a reservoir or passage for the liquid discharged from the opening 11a to the outside. Here, the tip of the first projection 11 means a region on the tip side of the middle position of the projection height H1 from the surface of the sheet base 2 to the tip of the first projection 11. A specific example of this is shown in FIG. 3(a).
[0021] When the injection needle 1 is used for intradermal administration of a drug, the tip of the first projection 11 is inserted into the skin S, and the liquid drug L stored in the hollow portion 11b of the first projection 11 is injected into the skin S through the opening 11a at the tip (see FIG. 5). In this manner, the hollow portion 11b of the first projection 11 functions as a drug storage portion, typically when no drug is supplied from the outside. When a drug is supplied from the outside, for example, when a drug supply device (not shown) such as a syringe is used together with the injection needle 1, the hollow portion 11b functions as a passage for the drug. The drug supply device may be configured by disposing a sheet-like member on the back side of the injection needle 1 and forming a drug storage space between the back side of the injection needle 1 and the sheet-like member, and may supply the drug to the first protrusion 11 by pressing the sheet-like member (see JP 2020-096790 A
[0011] ).
[0022] In the injection needle 1, it is preferable that the distance D1 (see FIG. 4), which is the shortest distance between the first projection 11 and the second projection 12, is 2.0 mm or more and 10 mm or less. The advantages of the distance D1 being within the above-mentioned range are as follows. When the tip of the first projection 11 is inserted into the skin S and the drug L is injected into the skin S, the injected drug L causes the skin to swell and a wheal 9 is formed (see FIG. 5). At this time, by setting the distance D1 within the above-mentioned range, the second projection 12 can be prevented from suppressing the swelling of the skin. In other words, the skin is more likely to swell. This makes it easier for the drug L or other liquid to enter the body, even when a large amount is injected, and the liquid can be injected efficiently without leakage. The inventors have investigated the relationship between the amount of liquid V injected into the skin S and the radius R of the wheal that is formed, using the following method.
[0023] <Relationship between fluid volume and wheal radius> The skin of a Gottingen miniature pig (male, 6 weeks old, ventral part; Oriental Yeast Co., Ltd.) stored at -20°C was transferred to a refrigerator (4°C) the day before the test and thawed. The skin was placed on 5 to 7 Kimtowels, returned to room temperature, shaved with a shaver, and the surface was wiped with alcohol cotton before administration. Powdered rhodamine B (Sigma-Aldrich) was adjusted to 0.5 mg / mL with ultrapure water and filled into a microsyringe (Agilent). The microsyringe and the injection needle were connected to form an injection device. The injection device was then attached to a puncture device and air was removed. The amount of liquid in the microsyringe was set to 10, 25, 50, 100 or 250 μL, and excess liquid attached to the injection needle was removed by absorbing it with Kimwipes. The arrangement of the projections of the injection needle is shown in FIG. 10. The height H1 of the first projection 11 of the injection needle was 2.0 mm, the height H2 of the stimulation projection 32 was 1.4 mm, and the height H3 of the control projection 33 was 0.9 mm. The distance D1, which is the shortest distance between the first projection 31 and the second projection 12, was 2.2 mm. The radius D2 of the imaginary circle formed by the imaginary line C1 connecting the centers of the first projection 11 was 0.75 mm.
[0024] The needle was inserted perpendicularly into the skin using a puncture device, and an appropriate amount of rhodamine B aqueous solution was administered intradermally. After administration, the needle was removed, and the outline of the wheal (an area that appears whiter and raised than the surrounding area) was immediately marked with a marker pen, and the long axis length A and short axis length B of the wheal were measured using a vernier caliper. Figure 11 shows a schematic plan view of the wheal. In Figure 11, reference numeral 91 indicates the outline of the wheal, and reference numeral 92 indicates the fluid inside the wheal. Each dose was administered three times. The average value of the long axis length A and the short axis length B divided by 2 was used as the radius R of the wheal, and a logarithmic approximation curve was calculated using Microsoft Excel spreadsheet software. The results are shown in Figure 12. Details of the puncture device will be described later.
[0025] In the injection needle 1, it is preferable to provide a predetermined gap between the first protrusion 11 and the second protrusion 12 so that the swelling of the skin is less likely to be suppressed by the second protrusion 12. By providing a predetermined gap between the first protrusion 11 and the second protrusion 12, when the drug L is injected into the skin S using the injection needle 1, a space is secured for the skin to swell around the first protrusion 11, more specifically, between the first protrusion 11 and the second protrusion 12. Then, the swelling of the skin is less likely to be suppressed by the second protrusion 12, and the liquid can be injected efficiently. In order to ensure the predetermined distance between the first projection 11 and the second projection 12, the distance D1 is preferably 2.0 mm or more. For example, as shown in Fig. 12, when a medicine is injected into the skin S by the injection needle 1, a wheal is formed rapidly even with a small amount of injected liquid. Therefore, by setting the distance D1 to 2.0 mm or more, the second projection 12 is less likely to inhibit the formation of the initial wheal 9, and the injectability of the liquid can be improved.
[0026] From the viewpoint of inhibiting the swelling of the wheal 9 and suppressing a decrease in the injectability of the liquid, the distance D1 is preferably at least half, more preferably at least two-thirds, of the wheal radius R of the wheal 9 formed when the drug L is injected into the skin S by the injection needle 1, and is even more preferably larger than the wheal radius R. The wheal radius R here can be calculated by the following formula (1). In formula (1), V is the amount of liquid injected into the skin S by the injection needle 1. The injected liquid is the liquid used in the above <Relationship between the amount of liquid and the radius of the wheal>. In formula (1), ln represents the natural logarithm. R = 0.5406ln(V) + 0.9483 (1)
[0027] The reason why the distance D1 is preferably equal to or greater than the above-mentioned ratio to the wheal radius R calculated by the following formula (1), particularly, greater than the wheal radius R, will be described below. For example, when the injection needle 1 having only one first protrusion 11 is used to inject the drug L into the skin S, the first protrusion 11 inserted into the skin S will be located at the center of the wheal 9. Therefore, by making the distance D1 greater than the wheal radius R of the wheal 9, the second protrusion 12 is less likely to inhibit the formation of a wheal when the drug is injected, and therefore, the second protrusion 12 can be effectively prevented from inhibiting the injection of the drug. Furthermore, when the drug L is injected into the skin S using an injection needle 1 having a plurality of first protrusions 11, a wheal is formed centered on each of the first protrusions 11 inserted into the skin S. Therefore, by making the distance D1 for each first protrusion 11 greater than the wheal radius R of the wheal formed by each first protrusion 11, the second protrusions 12 are less likely to inhibit the formation of a wheal when the drug is injected, and thus it is possible to effectively prevent inhibition of the injection of the drug.
[0028] From the viewpoint of more effectively preventing the second projections 12 from suppressing the swelling of the skin, the distance D1 is preferably 2.05 mm or more, more preferably 2.1 mm or more. More specifically, when the amount of liquid V injected into the skin S by the injection needle 1 is less than 10 μL, the distance D1 is preferably 2.0 mm or more, more preferably 2.1 mm or more. When the amount of liquid V is 10 μL or more, the distance D1 is preferably 2.1 mm or more, more preferably 2.2 mm or more.
[0029] From the viewpoint of making the injection needle 1 smaller to improve its ease of handling and effectively suppressing the stretching of the skin by the stimulation protrusions 32, the distance D1 is preferably 10 mm or less, more preferably 7 mm or less, and even more preferably 4 mm or less. The immune induction effect can be promoted by piercing the skin with the stimulating protrusion 32, and in order for this effect to be prominent, it is preferable that the injection point by the injection needle 1, i.e., the point where the first protrusion 11 is inserted, and the point where the stimulating protrusion 32 is inserted are close to each other, and specifically, it is preferable that the upper limit value of the distance D1 is within the range described above. Here, the distance between the first protrusion 11 and the second protrusion 12 means the distance between the apex of the first protrusion 11 and the apex of the second protrusion 12. When the second protrusion 12 is the control protrusion 33, the distance between the first protrusion 11 and the second protrusion 12 means the distance between the apex of the first protrusion 11 and the center of the control protrusion 33 in a plan view. As in the example shown in FIG. 1, when the first region R1 includes a plurality of primary projections 11, it is preferable that the distance D1 for each of the plurality of primary projections 11 is within the above-mentioned range.
[0030] The injection needle 1 preferably has, between the first region R1 and the second region R2, a projection-free region R3 that does not include the first projection 11 and the second projection 12. Specific examples of this are shown in Figure 4 and Figures 6(a) to (d). The non-protrusion region R3 is a region in which the distance D1 is within the above-mentioned range for each of the first protrusions 11 included in the first region R1. The non-protrusion region R3 is a region having a predetermined width, and in the example shown in FIG. 4, the width of the non-protrusion region R3 is the same as the distance D1. It is preferable that the non-protrusion region R3 is a region that does not include at least the tips of the first protrusions 11 and the second protrusions. The first protrusions 11 and the second protrusions 12 may rise sharply or gently from the seat base 2, and the non-protrusion region R3 may include the base end of the first protrusions 11 or the second protrusions 12 when the first protrusions 11 or the second protrusions 12 rise gently from the seat base 2, for example. When the first protrusion 11 or the second protrusion 12 rises gently from the seat base 2, the distance between the first protrusion 11 and the second protrusion 12 at a height position that is 80% of the height H1 of the first protrusion 11 may be defined as the width of the protrusion non-existence region R3. When the first protrusion 11 or the second protrusion 12 rises sharply from the seat base 2, the distance between the base ends of the first protrusion 11 and the second protrusion 12 may be defined as the width of the protrusion non-existence region R3.
[0031] It is preferable that the non-protrusion region R3 surrounds the first region R1 in a plan view of the seat base 2. It is also preferable that the non-protrusion region R3 has a predetermined width and continuously surrounds the periphery of the first region R1. Specific examples of this are shown in Figure 4 and Figures 6(a) to (c). The protrusion-free region R3 may sandwich the first region R1 in a plan view of the seat base 2. A specific example of this is shown in Fig. 6(d). By providing the injection needle 1 with the non-protrusion region R3, when liquid is injected into the skin with the injection needle 1, the skin can rise without restriction, making it possible to inject the liquid more efficiently. The planar shape of the protrusion-free region R3 is not particularly limited, but since wheals occur in a circular shape, it is preferably annular, and more preferably circular. For example, it may be a circular ring (see Fig. 4 and Fig. 6(a)), a rectangular ring (see Fig. 6(b)), a triangular ring (see Fig. 6(c)), etc.
[0032] In the injection needle 1, the second protrusion 12 preferably has a stimulating protrusion 32 and a control protrusion 33. Since the second protrusion 12 has the stimulating protrusion 32, when the injection needle 1 is pressed against the skin and the first protrusion 11 is inserted into the skin from the tip side, the stimulating protrusion 32 also penetrates the skin. By inserting the stimulating protrusion 32 into the skin, it is possible to prevent the skin from stretching, and therefore it is possible to improve the puncture property. Furthermore, by inserting the stimulating protrusion 32 into the skin, the stimulating protrusion 32 stimulates the skin and exerts a blood flow promoting effect, and it is also possible to promote the immune induction effect. Therefore, according to the injection needle 1 having this configuration, the first protrusion 11 can easily inject liquid into the skin, and the stimulating protrusion 32 can promote the blood flow and promote the immune induction effect. The presence or absence of a blood flow promoting effect can be determined by various known methods, for example, by determining whether or not a flare reaction occurs in the skin, or by visualizing the blood flow distribution using a laser blood flow meter.
[0033] Since the second protrusion 12 has the control protrusion 33, when the first protrusion 11 and the stimulation protrusion 32 are inserted into the skin from the tip side, the puncture depth control unit 40 of the control protrusion 33 comes into contact with the surface of the skin. This stops the first protrusion 11 and the stimulation protrusion 32 from penetrating, and prevents the first protrusion 11 and the stimulation protrusion 32 from penetrating deeper into the skin. In other words, the puncture depth control unit 40 functions as a stopper that limits the penetration depth of the first protrusion 11 and the stimulation protrusion 32. That is, the injection needle 1 is capable of intradermally administering a drug to any depth in the skin and effectively promoting blood flow by making it possible to control the penetration depth of the first protrusion 11 and the stimulation protrusion 32 into the skin. Therefore, by adjusting the penetration depth of the first protrusion 11 according to the type of drug, etc., it is possible to maximize the effect of the drug. In addition, by adjusting the penetration depth of the stimulation protrusion 32 according to the desired degree of blood flow promotion, it is possible to effectively produce the blood flow promotion effect.
[0034] In the injection needle 1, the first protrusion 11 is preferably higher than the second protrusion 12. By doing so, the second protrusion 12 comes into contact with the skin after the first protrusion 11 is inserted into the skin. Therefore, the first protrusion can be inserted into the skin efficiently, and the second protrusion 12 can be prevented from stretching the skin. After that, the first protrusion 11 can be smoothly inserted to any depth. Typically, the second protrusion 12 has a stimulation protrusion 32 and a control protrusion 33, and it is preferable that the first protrusion 11 is higher than each of the stimulation protrusion 32 and the control protrusion 33. In order to significantly reduce the pain associated with intradermal administration or to reliably inject the drug into the skin S from the first projection 11, the difference H1-H3 between the protrusion height H1 of the first projection 11 and the protrusion height H3 of the control projection 33 is preferably 100 μm or more, and more preferably 300 μm or more. Moreover, the difference H1-H3 is preferably 4995 μm or less, and more preferably 3000 μm or less, from the viewpoint of not damaging the skin more than necessary.
[0035] From the viewpoint of significantly reducing the pain associated with the blood flow promoting effect of the stimulation protrusions 32 and enabling the stimulation protrusions to be sufficiently inserted into elastic skin, it is preferable that the protruding height H2 of the stimulation protrusions 32 be greater than or equal to the protruding height H3 of the control protrusions 33, i.e., H2≧H3, and it is even more preferable that H2>H3. From the same viewpoint, the difference H2-H3 between the protruding height H2 of the stimulation protrusion 32 and the protruding height H3 of the control protrusion 33 is preferably 0 μm or more, more preferably 100 μm or more, and further preferably 400 μm or more. Moreover, the difference H2-H3 is preferably 4995 μm or less, and more preferably 3000 μm or less, from the viewpoint of not damaging the skin more than necessary.
[0036] From the viewpoint of ensuring a more reliable blood flow promoting effect, ensuring reliable puncturing of the first protrusions 11, and efficient injection, it is preferable that the first protrusions 11 are taller than the stimulating protrusions 32. More specifically, the difference H1-H2 between the protruding height H1 of the first protrusions 11 and the protruding height H2 of the stimulating protrusions 32 is preferably 10 μm or more, and more preferably 100 μm or more. Moreover, from the viewpoint of easily achieving both the ease of injecting the liquid and the blood flow promoting effect, the difference H1-H2 is preferably 4995 μm or less, and more preferably 3000 μm or less.
[0037] The protrusion height H1 of the first protrusion 11 is preferably 105 μm or more, and more preferably 150 μm or more, so that the first protrusion 11 can be successfully pierced even if the skin is deformed when pressed against the skin, improving puncture performance and reliably injecting the medicine into the skin S. Moreover, from the viewpoint of minimal invasiveness, the protrusion height H1 of the primary projections 11 is preferably 5000 μm or less, more preferably 4000 μm or less, and further preferably 3000 μm or less.
[0038] The protruding height H2 of the stimulating projections 32 is preferably 5 μm or more, and more preferably 20 μm or more, from the viewpoint of stimulating the skin by pressing against the skin and promoting blood flow. Moreover, from the viewpoint of minimal invasiveness, the protruding height H2 of the stimulating projection 32 is preferably 5000 μm or less, more preferably 4000 μm or less, and further preferably 3000 μm or less. The protrusion height H3 of the control protrusion 33 is preferably 5 μm or more, and more preferably 10 μm or more, from the viewpoint of making the skin as non-invasive as possible and improving puncture properties by increasing the distance between the base material and the skin. From the viewpoint of improving the puncture ability of the primary projections 11, the projection height H3 of the control projections is preferably 4000 μm or less, more preferably 3000 μm or less, and even more preferably 2000 μm or less.
[0039] In the injection needle 1 shown in FIG. 1, when the smallest imaginary circle M1 (see FIG. 4) including the centers of all the first projections 11 included in the first region R1 is assumed, it is preferable that the radius D2 (see FIG. 4) of the imaginary circle M1 is smaller than the distance D1 (see FIG. 4), which is the shortest distance among the distances between the first projections 11 and the second projections 12. When the center of the first projection 11 is located on the circumference of the imaginary circle M1, the center of the first projection 11 is included in the imaginary circle M1. By setting the relationship between the radius D and the distance D1 as described above, when the liquid is injected into the skin by the injection needle 1, the formation of the wheal 9 is more unlikely to be inhibited, and the liquid can be injected more efficiently. From the viewpoint of achieving this effect more significantly, the ratio D1 / D2 of the radius D2 to the distance D1 is preferably 1.0 or more, more preferably 1.5 or more. From the viewpoint of dispersing the injection sites for the formed wheals and making the injection needle 1 small, the ratio D1 / D2 is preferably 20 or less, and more preferably 10 or less.
[0040] In the injection needle 1, it is also preferable that the shortest distance D3 (see FIG. 4) among the distances between the first projections 11 in the first region R1 is shorter than the distance D1. When liquid is injected into the skin using the injection needle 1, the formation of the wheal 9 is less likely to be inhibited, and the liquid can be injected more efficiently. To make this effect more pronounced, the ratio D1 / D3 of the distance D3 to the distance D1 is preferably 1.0 or more, more preferably 1.5 or more. From the viewpoint of dispersing the injection sites for the formed wheals and making the injection needle 1 small, the ratio D1 / D3 is preferably 20 or less, and more preferably 10 or less.
[0041] In the injection needle 1, from the viewpoint of balancing the ease of puncturing and injecting liquid and the effect of promoting blood flow, the ratio of the number of second protrusions 12 to the number of first protrusions 11 (number of second protrusions 12 / number of first protrusions 11) is preferably 2.4 or more, more preferably 3.0 or more, and even more preferably 3.5 or more. From the viewpoint of minimal invasiveness, the number is preferably 10 or less, more preferably 8 or less, and even more preferably 6 or less.
[0042] In order to enable the injection needle 1 to efficiently inject liquid, the first region R1 of the injection needle 1 includes preferably two or more first projections 11, and more preferably three or more first projections 11. Moreover, from the viewpoint of enabling good injection of liquid from all of the primary projections 11, the number of projections is preferably 20 or less, more preferably 15 or less, and even more preferably 10 or less.
[0043] In the injection needle 1, the second region R2 may include the first projection 11, but preferably does not include the first projection 11 (see Figs. 4 and 6). Since the second region R2 does not include the first projection 11, only the first region R1 in the injection needle 1 is a region into which liquid can be injected, so that the position of the wheal can be limited to the first region, and it is possible to prevent the formation of the wheal from being inhibited in other positions.
[0044] The injection needle 1 may have, as the second protrusion 12, a composite second protrusion 52 in which the puncture depth control unit 40 is integrally formed around the stimulating protrusion 32. This example is shown in FIG. 7. In the example shown in FIG. 7, the puncture depth control unit 40 is integrally molded on the outer periphery of the base side portion of the stimulating protrusion 32, thereby forming the composite second protrusion 52. More specifically, an enlarged diameter portion 41 is formed on the base side portion, and a step portion that protrudes outward in the radial direction of the stimulating protrusion 32 is formed on the upper end of the enlarged diameter portion 41. The step portion serves as the puncture depth control unit 40. When the first protrusion 11 and the composite second protrusion 52 are inserted into the skin from the tip side, the puncture depth control unit 40 comes into contact with the surface of the skin, stopping the first protrusion 11 and the composite second protrusion 52 from penetrating deeper into the skin, preventing the first protrusion 11 and the composite second protrusion 52 from penetrating deeper into the skin. In the composite second protrusion 52, the puncture depth control unit 40 is integrally formed around the stimulating protrusion 32, so that the composite second protrusion 52 can be more reliably prevented from penetrating deeper into the skin.
[0045] Next, the constituent materials of the injection needle 1 will be described. The injection needle 1 preferably contains a thermoplastic resin from the viewpoints of material handleability, strength and processability of the injection needle, and hardness of the first projections 11 and the second projections 12, facilitating injection of liquid and improving the blood flow promoting effect. The injection needle is more preferably formed from a base sheet containing a thermoplastic resin. The thermoplastic resin may include one or more selected from polyolefin, polyester, polyamide, polyamideimide, polyetheretherketone, polyetherimide, polyvinyl chloride, acrylic resin, polystyrene resin, and the like. The polyolefin may include one or more selected from polypropylene, polyethylene, and the like. The polyester may include one or more selected from polyethylene terephthalate, polyfatty acid ester, polylactic acid, polycaprolactone, polybutylene succinate, and the like. The polyamide may include one or more selected from nylon and the like. From the viewpoint of biodegradability, it is preferable that the composition contains a polyfatty acid ester. Specifically, the polyfatty acid ester may include one or more selected from polylactic acid and polyglycolic acid.
[0046] The proportion of the mass of the thermoplastic resin contained in the injection needle 1 to the total mass of the injection needle 1 is preferably 50% or more, more preferably 70% or more, and even more preferably 90% or more, from the viewpoint of improving the moldability and dimensional stability of the injection needle. The mass ratio of the thermoplastic resin is preferably 100% or less, more preferably 98% or less, and even more preferably 96% or less, from the viewpoint of imparting various effects to the injection needle by adding, for example, a functional agent. Here, as various functional agents, antibacterial agents, bactericides, moisturizing agents, flow improvers, antistatic agents, colorants, etc. can be used.
[0047] The drug to be injected into the skin by the injection needle 1 can be appropriately selected depending on the application of the injection needle 1. The injection needle 1 can be used for subcutaneous administration as well as intradermal administration of drugs. The drug injected into the skin by the injection needle 1 may be an intradermal drug or a subcutaneous drug. An intradermal drug means a drug whose recommended administration method is intradermal administration. A subcutaneous drug means a drug whose recommended administration method is subcutaneous administration. Since the injection needle 1 can easily administer the drug intradermally, the drug injected into the skin by the injection needle 1 is preferably an intradermal drug, and more preferably an intradermal vaccine drug. In particular, when an intradermal vaccine drug is used, the drug can be reliably administered intradermally, and antigen recognition by the immune system can be efficiently strengthened compared to subcutaneous administration, so that the effect of the vaccine is expected to be improved. The skin has, in order from the body surface side, the epidermis, dermis, and subcutaneous tissue, and the dermis has a relatively large number of immune cells. Therefore, when an intradermal vaccine drug is administered intradermally by the injection needle 1, it is preferable to administer it into the dermis from the viewpoint of enhancing the effect of the vaccine.
[0048] The drug may include one or more selected from, for example, vaccines for preventing infectious diseases such as hepatitis A, hepatitis B, hepatitis C, influenza, COVID-19, tuberculosis, rabies, polio, chickenpox, rubella, measles, tetanus, and shingles; vaccines for treating chronic hepatitis B, tuberculosis, rabies, malignant neoplasms, and shingles; analgesics for cancer patients; insulin; biological products; gene therapy drugs; injectables, and skin application preparations. Since the first protrusion 11 of the injection needle 1 punctures the skin, the injection needle 1 can be applied to pharmacologically active substances that require subcutaneous injection in addition to pharmacologically active substances used for conventional transdermal administration. The skin S into which the drug L is injected by the injection needle 1 may be human skin or may be the skin of an animal other than a human.
[0049] The present invention includes an injection device in which an injection needle is connected to a drug supplying device filled with a drug. The above-mentioned injection needle 1 of the present invention can be used as the injection needle of the injection device of the present invention. For example, a syringe, a tube, an electric injector, etc. can be used as the drug supplying device. The drug supplying device may have a drug storage section that is filled with a drug in advance. According to the injection device of the present invention, the formation of the wheal 9 is less likely to be inhibited when the drug is injected, so the drug can be injected efficiently. Furthermore, since the injection device of the present invention can reliably administer the drug intradermally, the effect of the intradermal drug can be further improved by using a drug supplying device filled with an intradermal drug as the drug supplying device.
[0050] The present invention includes a kit including an injection needle, a drug supplying device filled with a drug, and a drug. The injection needle 1 of the present invention described above can be used as the injection needle included in the kit of the present invention. The drug supplying device can be the same as the drug supplying device related to the injection device of the present invention described above. The drug can be the same as the drug used in the injection needle 1 of the present invention described above. The kit may include a puncturing device having a structure that can impart speed to the injection device described above in a direction perpendicular to the administration site. The puncturing device is a tool used to puncture the skin S with the injection needle, and has a spring or the like as a structure for imparting speed for puncturing the injection needle toward the skin. In addition, "speed" means the speed at which the first protrusion 11 of the injection needle 1 punctures the skin. According to the kit of the present invention, the formation of the wheal 9 is less likely to be inhibited when the drug is injected, so that the drug can be injected efficiently.
[0051] The puncture device has, for example, a holding section for holding the injection needle 1, a guide mechanism for regulating the moving direction of the injection needle 1 so that the first protrusion 11 of the injection needle 1 pierces the skin substantially perpendicularly, and a drive mechanism for moving the injection needle 1 along the guide mechanism. The drive mechanism preferably includes a drive means such as a spring for moving the injection needle 1 while controlling the puncture speed of the first protrusion 11 of the injection needle 1 to a predetermined speed. The drive mechanism is preferably configured so that the drive means can be operated manually.
[0052] The holding part is, for example, a part of an adaptor member capable of connecting the injection needle 1 and a drug supply device to which the injection needle 1 is attached. The holding part can be formed, for example, at one end of the adaptor member. In a state in which the injection needle 1 and the drug supply device are connected via the adaptor member, it is preferable that the drug can be supplied from the drug supply device to the first protrusion 11 of the injection needle 1. There is no particular limitation on the means for attaching the injection needle 1 to the holding part, and any means such as fitting, screwing, fusion, and adhesion can be used. In addition, the holding part and the injection needle 1 each have an engagement part that engages with each other, and the injection needle 1 may be attached to the holding part by engaging the engagement parts. The other end of the adaptor member is preferably capable of attaching the drug supply device. For example, the other end of the adaptor member is preferably capable of inserting the tip opening of a syringe. The guide mechanism includes, for example, a guide hole provided in a side wall of a cylindrical main body that constitutes the main part of the puncture device, and an injection tool holder having a guide protrusion inserted into the guide hole. The guide hole has an opening shape that extends along the axial direction of the main body. The injection tool holder holds an injection tool formed by connecting an injection needle 1 and a medicine feeder via an adapter member. By inserting the guide protrusion of the injection tool holder into the guide hole, the movement direction of the injection tool holder is restricted to the axial direction of the main body. The injection tool holder is disposed inside the main body. The driving means of the driving mechanism is preferably adapted to move the syringe holder. When the driving means is a spring, the puncture device preferably has a stopper mechanism for maintaining the spring in a contracted state. The stopper mechanism preferably maintains the spring in a contracted state by fixing the position of the guide protrusion.
[0053] A method for injecting a drug into the skin using a lancing device is described. First, a syringe, which is a drug supply device, is filled with a drug. Specifically, with the tip opening of the syringe immersed in the drug, a plunger inserted into the syringe is pulled up to aspirate the drug into the syringe. An injection needle 1 is attached to the syringe filled with the drug via an adapter member to form an injection device. The plunger of the syringe is pressed down to fill the first protrusion 11 of the injection needle 1 with the drug. The injection device is held by an injection device holder and attached to a puncture device. The guide protrusion of the injection device holder is pushed up along the guide hole to contract the spring of the drive mechanism that biases the injection device holder. The guide protrusion is fixed by a stopper mechanism so that the contracted state of the spring is maintained (hereinafter, this state is also referred to as a "standby state"). The first protrusion 11 of the injection needle 1 in the puncture device in the standby state is brought close to the skin to which the drug is to be administered. At this time, it is preferable that the axial direction of the main body of the puncture device is perpendicular to the skin. Then, the fixation of the guide protrusion is released. Then, the injection needle 1 is urged toward the skin, and the first protrusion 11 pierces the skin. Then, the plunger of the syringe is pressed down to inject the medicine into the skin.
[0054] Although the present invention has been described based on the preferred embodiment, the present invention is not limited to the above-described embodiment and can be modified as appropriate. For example, in the example shown in FIG. 3, the second projection 12 is solid, but the second projection 12 may be hollow. Also, in the example shown in FIG. 7, the composite second projection 52 is solid, but the composite second projection 52 may be hollow. In the injection needle 1, it is preferable that the projections other than the first projection 11 are solid. By doing so, it is possible to prevent the liquid to be discharged from the opening 11a of the first projection 11 from being accumulated inside the other projections, so that the liquid can be injected efficiently.
[0055] In the example shown in Fig. 1, the injection needle 1 has a flat sheet-like shape as a whole, but the overall shape of the injection needle 1 is not limited to this. The injection needle 1 may be formed integrally with an adapter member. For example, the first and second protrusions 11, 12 may protrude from one surface side of a plate-like member, and a cylindrical part that functions as an adapter member may be provided on the peripheral part of the other surface side by bonding by adhesion or integral molding. In this case, the plate-like part is the sheet base 2.

EXAMPLES
[0064] The present invention will be described in more detail below with reference to examples, but the present invention is not limited to such examples.
[0065] Example 1 An injection needle having the same configuration and projection arrangement as the injection needle 1 shown in Figs. 1 and 2 was manufactured. The dimensions of each projection of the injection needle, the distance D1 which is the shortest distance between the first projection and the second projection, the radius D2 of the smallest imaginary circle including the centers of all the first projections included in the first region, the shortest distance D3 between the first projections in the first region, and the ratio of the mass of the thermoplastic resin to the total mass of the injection needle are as shown in Table 1. In the injection needle of Example 1, the planar shape of the projection-free region is a circular ring shape surrounding the first region. The injection needle was formed by performing projection formation processing using a processed needle to which ultrasonic vibration was applied to a base sheet made of 100% polylactic acid, which is a thermoplastic resin.
[0066] [Table 1]
[0067] Example 2 An injection needle was manufactured having the same configuration as the injection needle 1A shown in Fig. 1 and the same arrangement as in Fig. 6(d). The other elements were as shown in Table 1 and the other elements were the same as in Example 1. In the injection needle of Example 2, the non-protrusion region has a rectangular shape in a plan view, and the non-protrusion region sandwiches the first region. Comparative Example 1 An injection needle having a first protrusion 11, a stimulating protrusion 32 and a control protrusion 33 was manufactured (see FIG. 9(a)). The elements were as shown in Table 1, and the protrusions 11, 32 and 33 were arranged as shown in FIG. 9(a) in the same manner as in Example 1. The injection needle of Comparative Example 1 does not have a protrusion-free region. Comparative Example 2 An injection needle having only the first protrusion 11 was manufactured (see FIG. 9(b)). Each element was as shown in Table 1. It is the same as Example 1 except that it does not have the stimulation protrusion 32 and the control protrusion 33. The injection needle of Comparative Example 2 does not have the second region and the protrusion-free region.
[0068] [Administration of drug solution] Using the injection needles of Examples 1 and 2 and Comparative Examples 1 and 2, drug solutions were administered into the skin of Gottingen miniature pigs in the following manner. The skin of a Gottingen miniature pig (male, 6 weeks old, ventral part; Oriental Yeast Co., Ltd.) stored at -20°C was transferred to a refrigerator (4°C) the day before the test and thawed. The skin was placed on 5 to 7 Kimtowels, shaved with a shaver, and the surface was wiped with alcohol cotton before administration. Powdered rhodamine B (Sigma-Aldrich) was adjusted to 0.5 mg / mL with ultrapure water and filled into a microsyringe (Agilent). The microsyringe was set in a puncture device, and the injection needles of Examples 1 and 2 and Comparative Examples 1 and 2 were attached, respectively, and air was removed. The liquid volume in the microsyringe was set to 100 μL, and excess liquid adhering to the injection needle was removed by absorbing it with Kimwipes. The projection of the injection needle was punctured vertically into the puncture target, and 100 μL of rhodamine B aqueous solution was injected.
[0069] After administering the drug solution to the skin of a Gottingen miniature pig by the method described in [Administration of drug solution] above, the needles of Examples 1 and 2 and Comparative Examples 1 and 2 were evaluated for puncture property and administration property according to the following evaluation criteria. The results are shown in Table 1. [Evaluation criteria for puncture resistance] ○: Meets both (1) and (2) below. (1) When the first protrusion is inserted, it can be inserted to a depth sufficient to allow injection. (2) When the first protrusion is punctured, it does not penetrate too deeply, but can be punctured to a depth that causes a wheal to form when the drug solution is injected. ×: Either or both of (1) and (2) above are not satisfied. [Evaluation criteria for administration] ○: Meets both (1) and (2) below. (1) Immediately after administration of the drug solution, the formation of a wheal was visually confirmed, and the formation of the wheal was not inhibited by the secondary projections. (2) After administration, the surface of the skin at the injection site and the surface of the injection needle are wiped with a paper towel whose weight has been measured in advance, and the change in weight of the paper towel before and after wiping is 5 mg or less. ×: Either or both of (1) and (2) above are not satisfied.
[0070] As shown in Table 1, the injection needles of Examples 1 and 2 were evaluated as "good" for both puncture property and administration property, which shows that the first projection can be easily punctured and wheal formation is unlikely to be inhibited during injection. As shown in Table 1, the injection needle of Comparative Example 1 was evaluated as "good" for puncture property, but was evaluated as "poor" for administration property. As shown in Table 1, the injection needle of Comparative Example 2 was evaluated as "poor" for both puncture property and administration property. In particular, with regard to puncture performance, when the injection needle of Comparative Example 2 punctured the first protrusion, the excised skin stretched, making it difficult to puncture the first protrusion properly or to inject the drug solution to the desired depth because the first protrusion was inserted too deeply. Also, with regard to administration performance, the first protrusion was unable to puncture the skin, causing the drug solution to leak, or the first protrusion was inserted too deeply, causing the drug solution to be unable to be injected to the desired depth, resulting in no wheal formation. Therefore, it can be seen that the injection needle of the present invention is unlikely to suppress the swelling of the skin when injecting liquid, and can inject liquid efficiently.
