Abstract
Description
[Technical field]
[0001] This application claims priority to Korean Patent Application No. 10-2020-0052911, filed on April 29, 2020, the entire specification of which is incorporated herein by reference.
[0002] The present invention relates to a recombinant protein in which an interferon β mutant having an amino acid residue substitution is fused with an antibody that binds to a specific antigen, a polynucleotide, an expression vector and a host cell, and an anti-cancer pharmaceutical composition containing the recombinant protein as an active ingredient. [Background technology]
[0003] Interferons (IFNs) are one of the major cytokines that play an important role in immunity and are known to have strong anticancer effects. Interferons are divided into type I (IFN-α and IFN-β), type II (IFN-γ), and type III (IFN-λ).
[0004] Type I interferons exhibit antiviral and antiproliferative effects, and also play an essential role in cancer immunosurveillance, which is the function of recognizing tumor-specific antigens and eliminating tumor cells.
[0005] Type I interferon signaling is the stepwise activation of multiple enzymes by the activity of interferon receptors, and in this case, transcription factors such as STAT1 and STAT2 also act. These signaling pathways activate the immune system and are essential for the anticancer effects of various anticancer drugs, such as chemotherapeutics such as anthracyclines, antibodies targeting growth factor receptors such as human epidermal growth factor receptor 2 (HER2) and epidermal growth factor receptor (EGFR), adjuvant administration, and oncolytic virotherapy. In particular, trastuzumab is an antibody cancer drug that targets HER2, and overexpression of MUC4 causes resistance to the drug. One of the signaling substances related to this resistance mechanism, pSTAT3, is known to have a polymerized form of two identical substances (pSTAT3 homodimer) and to be suppressed by pSTAT1. Therefore, when type I interferon, which activates pSTAT1, is combined with trastuzumab, an increased anticancer effect can be expected.
[0006] Interferon-β (IFN-β) has a stronger effect of suppressing cell growth than interferon-α (IFN-α). In particular, when used in combination with anticancer drugs, interferon-β has a very good antiproliferative activity range and synergistic effect. However, it is known that therapies using interferon have high cytotoxicity, and patients suffer from symptoms such as fever (80%), muscle pain (73%), headache (50%), fatigue (50%), and malaise (50%). In addition, interferon-β is a highly hydrophobic protein that tends to aggregate, resulting in reduced biological activity and productivity. In addition, it has a short half-life, which means it must be administered frequently. Therefore, in order to develop a therapeutic agent using interferon, it is necessary to minimize the limitations of interferon.
[0007] Thus, the present inventors have developed mutant interferon-β with improved productivity and reduced aggregation phenomenon by changing the physical properties of interferon through site-specific mutations. The interferon-β mutant can be used as a new anti-cancer drug that shows excellent anti-cancer effects when fused with an antibody cancer drug. A method for producing a recombinant protein comprising an antibody or a fragment thereof covalently bound directly or indirectly to an interferon-β mutant, comprising transforming a host cell so as to express a recombinant protein in which the 17th amino acid, cysteine, of the interferon-β mutant is mutated to another amino acid. The object of the present invention is to provide a method for increasing the stability of the recombinant protein during production or separation / purification.
[0013] Another object of the present invention is to provide use of the recombinant protein for producing an anticancer agent. Another aspect of the present invention aims to provide a method for treating cancer, comprising administering an effective amount of a composition containing the recombinant protein to an individual in need thereof. [Means for solving the problem]
[0014] According to one embodiment, the present invention provides an interferon beta (IFN-β) mutant in which the 17th amino acid residue is substituted with serine and the 27th amino acid residue is substituted with threonine; and a recombinant protein comprising an antibody or fragment thereof covalently linked directly or indirectly to the interferon beta mutant.
[0015] Interferon-β is a globular protein with five alpha-helices and a size of 22 kDa. It has been reported that it is effective in treating cancer, autoimmune disorders, viral infections, HIV-related diseases, hepatitis C, and rheumatoid arthritis due to various immunological activities such as antiviral activity, cell growth inhibition or antiproliferation activity, lymphocyte cytotoxicity enhancement activity, immunomodulatory activity, target cell differentiation induction or inhibition activity, macrophage activation activity, cytokine production increase activity, cytotoxic T cell effect increase activity, and natural killing cell increase activity. However, interferon-β is a highly hydrophobic protein that exhibits aggregation phenomenon, has low biological activity and productivity, and has a short half-life, so there are limitations to the use of interferon-β. Therefore, in the present invention, an interferon-β mutant with improved physical properties compared to wild-type interferon-β without mutation was developed by inducing site-directed mutagenesis in the interferon-β gene.
[0016] The interferon beta mutein is characterized in that the 17th cysteine (C) is replaced with serine (S) and the 27th arginine (R) is replaced with threonine (T), thereby containing glycosyl groups at the 80th and 25th amino acid residues. In this case, the interferon beta mutein may be represented by the amino acid sequence of SEQ ID NO: 1 or the nucleotide sequence encoding it, SEQ ID NO: 2. Such an interferon beta mutein contains all or a part of the amino acid sequence of wild-type interferon beta and has the activity of interferon beta.
[0017] The antibodies include monoclonal antibodies, polyclonal antibodies, multispecific antibodies (e.g., bispecific antibodies) and fragments thereof. An intact antibody is generally Y-shaped and consists of two long heavy chains (H) and two short light chains (L). Each heavy and light chain is linked to each other by sulfide bonds and is divided into a variable region (V), which is a site that reacts with an antigen, and a constant region (C), which is a site that expresses an effector function. The variable region contains complementarity determining regions (CDRs) that determine the structure of the variable region to enable the formation of specific bonds with antigens and regulate the antibody binding strength. The antibody fragment indicates a specific site capable of reacting with an antigen and exhibiting antigen-binding activity, and may be, for example, a Fab fragment (a fragment obtained by papain digestion), a Fab' fragment (a fragment obtained by pepsin digestion and partial reduction), a F(ab')2 fragment (a fragment obtained by pepsin digestion), a Facb (a fragment obtained by plasmin digestion), a Fd (a fragment obtained by pepsin digestion, partial reduction, and reaggregation), a scFv fragment (a fragment obtained by molecular biology techniques), etc. Preferably, the antibody is an antibody or a fragment thereof that recognizes a tumor-specific antigen, and can be used as a targeted therapeutic agent for cancer, etc. For example, the antibody may be trastuzumab, an antibody therapeutic agent that targets human epidermal growth factor receptor (HER-2), cetuximab, an antibody therapeutic agent that targets epidermal growth factor receptor (EGFR), or atezolizumab, an antibody therapeutic agent that targets PD-L1 expressed on the surface of cancer cells. In this case, trastuzumab may consist of a heavy chain having the amino acid sequence shown in SEQ ID NO: 3 and a light chain having the amino acid sequence shown in SEQ ID NO: 4, cetuximab may consist of a heavy chain having the amino acid sequence shown in SEQ ID NO: 5 and a light chain having the amino acid sequence shown in SEQ ID NO: 6, and atezolizumab may consist of a heavy chain having the amino acid sequence shown in SEQ ID NO: 7 and a light chain having the amino acid sequence shown in SEQ ID NO: 8.
[0018] The recombinant protein may be a fusion or complex formed by linking an interferon β mutant and an antibody or a fragment thereof via a peptide linker. The peptide linker refers to a molecule that links two or more separate substances to each other, which is a short fragment of an amino acid or an amino acid analog in which two or more amino acids or substances similar to amino acids are linked to each other via peptide bonds. In this case, glycine, serine, alanine, etc. are used as the main constituent amino acid, and a glycine-serine linker, glycine-serine-alanine linker, etc. may be used. Such a linker may be linked to the C-terminus of the heavy chain of the antibody or the C-terminus of the light chain of the antibody, or the N-terminus of the linker may be linked to the C-terminus of the light chain and the C-terminus of the heavy chain of the antibody. In this case, the N-terminus of the interferon β mutant may be linked to the C-terminus of the linker.
[0019] In another embodiment, the present invention provides a polynucleotide encoding the recombinant protein. A polynucleotide is a polymeric compound in which nucleotides, which are chemical monomers consisting of three elements: a base, a sugar, and a phosphate, are linked in a chain shape via numerous phosphate ester bonds. It is a polymer of deoxyribonucleotides or ribonucleotides that exists in a single-stranded or double-stranded form.
[0020] The polynucleotide may be an RNA genome sequence, DNA (gDNA or cDNA) and an RNA sequence transcribed therefrom, etc. In addition to the nucleotide sequence encoding the amino acid sequence of the recombinant protein, it also includes a sequence complementary to the sequence. Preferably, it may be a base sequence shown in SEQ ID NO: 9 encoding a recombinant protein in which an interferon β mutant and a heavy chain of trastuzumab are linked, a base sequence shown in SEQ ID NO: 10 encoding a recombinant protein in which an interferon β mutant and a heavy chain of cetuximab are linked, or a base sequence shown in SEQ ID NO: 11 encoding a recombinant protein in which an interferon β mutant and a heavy chain of atezolizumab are linked.
[0021] In another embodiment, the present invention provides an expression vector comprising the polynucleotide. An expression vector is a vector capable of expressing recombinant proteins including interferon β variants and antibodies or fragments thereof in a host cell, and refers to a gene product comprising a gene sequence and an expression control sequence operably linked to enable expression of a gene insert. The term "operably linked" refers to a functional linkage between a nucleic acid expression control sequence and a nucleic acid sequence encoding a target protein so as to perform a general function. The term "expression control sequence" refers to a DNA sequence that controls the expression of a polynucleotide sequence operably linked in a specific host cell. Such an expression vector can be produced using recombinant gene technology known in the art, and enzymes and the like commonly used in the art for site-specific DNA cleavage and ligation may be used.
[0022] The expression vector may contain a signal sequence or leader sequence for membrane targeting or secretion in addition to expression regulatory elements such as a promoter, an operator, a start codon, a stop codon, a polyadenylation signal, and an enhancer, and may be prepared in a variety of ways depending on the purpose. The expression vector may also contain a selection marker for selecting a host cell containing the expression vector, and in the case of a replicable expression vector, it may contain a replication origin and may self-replicate or be integrated into the host DNA. The expression vector may be any vector that is commonly used in the art for expressing foreign genes, and may be, for example, a plasmid vector, a cosmid vector, or a virus vector.
[0023] In another embodiment, the present invention provides a host cell transformed with the expression vector. A host cell may be selected to regulate the expression of the inserted sequence or to process the gene product in a preferred specific manner. Different host cells have characteristic and specific mechanisms of action for protein detoxification and post-detoxification processing and modification. A suitable cell line or host system may be selected to provide preferred modification and processing of the expressed heterologous protein. Expression in yeast can produce biologically active products. Expression in eukaryotic cells can increase the likelihood of folding of the peptide chains that make up the polypeptide or protein.
[0024] The host cell may be any host cell known in the art that can be used to continuously clone and express a vector while stabilizing the vector, and may be, for example, Escherichia coli such as E. coli JM109, E. coli BL21DE, E. coli DH5, E. coli RR1, E. coli LE392, E. coli B, E. coli X1776, E. coli W3110, etc., Agrobacterium genus strains such as Agrobacterium A4, bacilli such as Bacillus subtilis, Enterobacteriaceae such as Salmonella typhimurium and Serratia marcescens, various strains of the Pseudomonas genus, etc. Furthermore, when transfecting an expression vector into eukaryotic cells, yeast (Saccharomyces cerevisiae), insect cells, human cells (e.g., CHO (Chinese hamster ovary), W138, BHK, COS-7, 293, HepG2, 3T3, RIN, MDCK cell lines), etc. may be used as host cells.
[0025] The expression vector can be introduced using a transformation method commonly available in the art. The term "transformation" refers to an artificial and genetic change that occurs when an external DNA is introduced into a host cell, so that the DNA can replicate as a chromosomal factor in the host cell or by completing integration into the chromosome. The transformation methods include the CaCl2 precipitation method, the Hanahan method in which the efficiency of the CaCl2 method is increased by using a reducing agent called DMSO (dimethyl sulfoxide), electroporation, calcium phosphate precipitation, protoplast fusion, agitation using silicon carbide fiber, agrobacteria-mediated transformation, PEG-mediated transformation, dextran sulfate, lipofectamine, and desiccation / inhibition-mediated transformation.
[0026] In another embodiment of the present invention, there is provided an anti-cancer pharmaceutical composition comprising the recombinant protein as an active ingredient. The recombinant protein has a structure in which an interferon β mutant with improved physical properties is bound to an antibody or a fragment thereof, and it has been confirmed that it has excellent purification efficiency and bioactivity, and has immunomodulatory efficacy, anticancer efficacy, ADCC efficacy, etc. against various cancer cell lines. Therefore, a composition containing such a recombinant protein as an active ingredient exhibits an effect of suppressing the proliferation and activity of cancer cells, and can be used as an anticancer pharmaceutical composition.
[0027] The composition can be formulated by mixing with pharma- ceutical commonly used excipients, adjuvants, soothing agents, isotonicity agents, preservatives, and other pharma-ceutical commonly acceptable adjuvants, and formulating into a pharma-ceutical commonly acceptable formulation to produce a pharmaceutical preparation. The formulation may be in the form of an oral preparation such as a powder, granules, tablets, capsules, suspension, emulsion, syrup, aerosol, or in the form of an external preparation, suppository, and sterile injection solution. Solid preparations for oral administration may include tablets, pills, powders, granules, capsules, etc., and liquid preparations for oral administration include suspensions, liquid preparations, emulsions, syrups, etc. Preparations for parenteral administration include sterilized aqueous solutions, non-aqueous solvents, suspensions, emulsions, lyophilized preparations, and suppositories.
[0028] The method of administering the composition varies depending on the patient's condition and body weight, the degree of the disease, the drug form, the administration route and period, but can be appropriately selected by those skilled in the art. However, for a preferable effect, it may be administered at 0.001 to 1,000 mg / kg per day. The administration may be administered once a day or in several divided doses. The administration route may be any method, and may be administered by oral, rectal or intravenous, intramuscular, subcutaneous, intrauterine dura or intracerebrovascular injection, for example.
[0029] In addition, in the present invention, it was confirmed that in the production, isolation and purification of the R27T mutant of interferon beta, C17 of interferon beta, i.e., the 17th amino acid, cysteine, plays an important role in the stability of the mutant, and as a result, if the 17th amino acid is changed to another amino acid, it will affect the production, isolation, purification, and even stability during storage and distribution.
[0030] Thus, the present invention provides (a) culturing a host cell transformed to express a recombinant protein comprising an antibody or fragment thereof covalently linked directly or indirectly to an interferon beta variant; and (b) isolating and purifying the recombinant protein comprising the antibody or fragment thereof covalently linked directly or indirectly to interferon-β from the cultured host cell or the culture. The present invention provides a method for increasing protein stability during the production or separation / purification of the recombinant protein.
[0031] In the method of the present invention, the interferon-β mutant is an R27T mutant of interferon-β, and C17 is further modified to another amino acid, resulting in the expression of a recombinant protein comprising an antibody or a fragment thereof covalently linked directly or indirectly to the C17N, R27T double mutant, where N represents any amino acid except cysteine. Preferably, the other amino acid to be modified may be serine.
[0032] The host cell expressing the recombinant protein comprising the antibody or fragment thereof covalently linked directly or indirectly to the mutant may also be a mammalian cell used for protein expression.Preferably, CHO cell line, W138, BHK, COS-7, 293, HepG2, 3T3, RIN and MDCK cell line may be used, and most preferably, the host cell in the present invention may be a CHO cell line.
[0033] The present invention also provides use of the recombinant protein for producing an anticancer agent. The present invention also provides a method for treating cancer, comprising administering to an individual in need thereof an effective amount of a composition comprising the recombinant protein.
[0034] The "effective amount" of the present invention refers to an amount that, when administered to an individual, shows an effect of improving, treating, preventing, detecting, or diagnosing cancer, or suppressing or reducing the disease, and the "individual" may be an animal, preferably a mammal, particularly an animal including a human, or may be a cell, tissue, organ, etc. derived from an animal. The individual may be a patient in need of the effect.
[0035] The "treatment" of the present invention comprehensively refers to improving cancer or symptoms of cancer, which may include curing, substantially preventing, or improving the condition of the disease, including, but not limited to, alleviating, curing, or preventing one or most symptoms resulting from cancer.
[0036] In this specification, the term "comprising" is used synonymously with "including" or "characterized by" and does not exclude additional components or method steps not specifically mentioned in the compositions or methods of the present invention. Additionally, the term "consisting of" means excluding additional elements, steps, or ingredients not otherwise stated. The term "essentially consisting of" means that the scope of a composition or method may include, in addition to the substances or steps stated, substances or steps that do not substantially affect its basic properties. Effect of the Invention
[0037] The recombinant protein in which the interferon β mutant and an antibody or a fragment thereof according to the present invention are combined may improve the physical properties of interferon β through site-specific mutation, and may increase the biological activity and purification efficiency, leading to increased productivity. Since such a recombinant protein expresses both the functions of interferon β and the properties of an antibody that binds to a specific antigen, it can be usefully used as a targeted therapeutic agent for cancer and the like. DETAILED DESCRIPTION OF THE PREFERRED EMBODIMENTS
[0039] The present invention will be described in more detail below with reference to the accompanying drawings. However, these descriptions are merely presented as examples to supplement understanding of the present invention, and the scope of the present invention is not limited by these exemplary descriptions.
[0040] 1. Materials and Methods 1-1. Fusion protein production design To establish cell lines transiently or stably expressing antibodies binding to interferon-β mutants, we used pCHO 1.0 (Life Technologies) as an expression vector. We used trastuzumab, cetuximab, and atezolizumab as antibodies, and used an interferon-β (IFN-β) mutant in which the 27th amino acid residue is replaced by threonine (IFN-β-R27T) or an additional 17th amino acid residue is replaced by serine (IFN-β-C17S-R27T).
[0041] The linker and the interferon-β mutant R27T or C17S-R27T were cloned so that they could be linked to the heavy chain site of the antibody. The final heavy chain gene was then obtained by inserting the restriction enzyme AvrII (CCTAGG) cleavage site and Bstz17I (GTATAC) cleavage site (Thermo Scientific, USA) at the 3' and 5' ends of the entire gene, respectively. The final light chain gene was obtained by inserting the restriction enzyme EcoRV (GATATC) cleavage site and PacI (TTAATTAA) at the 3' and 5' ends of the antibody light chain, respectively. The final heavy and light chain genes were inserted into the pCHO 1.0 vector.
[0042] 1-2. Expression of fusion protein constructs in mammalian cells Trastuzumab, trastuzumab-interferon β-R27T and trastuzumab-interferon β-C17S-R27T expression vectors were transduced into CHO-S cells (Thermo Scientific) using FreeStyleTM MAX reagent (Thermo Scientific). Cetuximab and atezolizumab antibodies were transduced in the same manner. The mixture of FreeStyleTM MAX reagent-DNA complex and OptiPROTM SFM (Thermo Scientific) was added to CHO-S cells in a flask and cultured under humidified conditions at 8% CO2 atmospheric pressure. After 48 hours of transduction, cell lines stably expressing the fusion proteins were selected. Cells were sorted by secondary selection with puromycin 10-50 μg / mL and MTX 100-1,000 nM. The selected cells were cultured with glucose for 5 or 7 days to express the fusion protein at 37°C in a humidified 8% CO2 atmosphere at 130 rpm.
[0043] 1-3. Purification of fusion proteins The fusion protein expressed in CHO-S cells was purified by affinity chromatography. The CHO-S culture medium was passed through a column packed with Protein A Mabselect sure (Cytiva), and the purified protein was obtained by sequentially adding equilibration buffer, wash buffer, and elution buffer.
[0044] 1-4. Protein aggregation analysis Aggregation analysis (FPLC-SEC) was performed to measure the physical properties. A packed size exclusion chromatography column, HiLoad 16 / 600 Superdex 200 pg, was connected to an AKTA purifier (Cytiva). After setting the maximum pressure to 0.3 MPa, the sample purified by affinity chromatography was adjusted to a concentration of 1 mg / mL and injected to about 50% of the length of the sample loop. The protein aggregation rate was confirmed by checking the UV (280 nm) peak.
[0045] 1-5. Cell lines and culture conditions Human breast cancer cell lines (MCF-7, MDA-MB-231, HCC-1954, MDA-MB-453) and human gastric cancer cell lines (SNU1, SNU620, Hs746T, MKN45) were purchased from the Korea Cell Line Bank (KCLB, Seoul, South Korea). Human gastric cancer cell line NCI-N87 was purchased from the American Cell Line Bank (ATCC, Manassas, VA, USA). U937 cell line (iLite type I IFN assay ready cells, ATCC#CRL1593.2) was purchased from Euro Diagnostica AB (Malmo, Sweden).
[0046] MCF-7, MDA-MB-231, HCC-1954, MDA-MB453, NCI-N87, SNU1, SNU620, Hs746T, MKN45 and U937 cell lines were cultured in RPMI-1640 (HyClone, USA) medium containing 10% FBS (HyClone, USA), 100 units / mL penicillin and 100 μg / mL mycin, while Hs746T cell line was cultured in DMEM (HyClone, USA) medium containing 10% FBS (HyClone, USA), 100 units / mL penicillin and 100 μg / mL streptomycin. All cell lines were cultured at 37 °C in a humidified 5% CO2 atmosphere.
[0047] 1-6. Analysis of HER2 recognition ability by flow cytometer MCF-7, MDA-MB-231, and NCI-N87 cell lines were collected in EP tubes, and then incubated with blocking buffer at 4°C for 1 hour and washed twice with PBS. Then, 1 μg of trastuzumab, trastuzumab-IFNβ-R27T, and trastuzumab-IFNβ-C17S-R27T were incubated and washed three times with PBS. Then, FITC-conjugated anti-human IgG secondary antibody was incubated and washed three times. The cells reacted with the antibody were analyzed using a CytoFlex (Beckman Coulter, USA) flow cytometer.
[0048] 1-7. Analysis of STAT1 protein phosphorylation by Western blot NCI-N87 cells were treated with IFN-β (5–200 pg / mL), IFNβ-C17S-R27T (5–200 pg / mL), or trastuzumab-IFNβ-C17S-R27T (20–2,000 pg / mL), and then lysed using RIPA lysis buffer (containing 150 mM NaCl, 1% tryptone X-100, 1% sodium dioxycholate, 0.1% SDS, 50 mM Tris-HCl [pH 7.5], and 2 mM EDTA) supplemented with a protease inhibitor mixture (Roche) and a phosphatase inhibitor mixture (Roche). Lysed cells were incubated at 4°C for 20 min and centrifuged at 14,000 g for 10 min. The supernatant was collected and protein concentration was measured using a BCA protein assay kit (Thermo Scientific) according to the manufacturer's instructions. 20 μg of protein was separated by 10% SDS-PAGE and transferred to a PVDF membrane (BioRad). The PVDF membrane was blocked with TBS-Tween 20 buffer (containing 5% BSA) and incubated with primary antibodies overnight at 4°C. It was then incubated with secondary antibodies for 1 hour at room temperature. The primary antibodies used here were anti-pSTAT1, anti-STAT1 (Cell Signaling Technology) or anti-β-actin (Santa Cruz Biotechnology), and the secondary antibodies were anti-mouse IgG-HRP (ThermoFisher Scientific) or anti-rabbit IgG-HRP (ThermoFisher Scientific). Protein-bound HRP was visualized with Clarity Western ECL substrate (Bio-Rad).
[0049] 1-8. Measurement of antibody-dependent cellular cytotoxicity Target cells, HCC1954 or MDA-MB453 cells, were treated with control IgG (0.0001-100 nM), trastuzumab (0.0001-100 nM), IFN-β (0.000001-1 nM), or trastuzumab-IFNβ-C17S-R27T (0.0001-100 nM) and co-cultured with effector cells to measure antibody-dependent cellular cytotoxicity. Effector cells were natural killer (NK) cell line NK-92MI-CD16a, which artificially overexpressed CD16a to induce antibody-dependent cellular cytotoxicity, and reacted with the drug for 4 hours at an effector cell to target cell ratio (E:T ratio) of 8:1. Cytotoxicity of target cells was measured using a non-radioactive cytotoxicity assay measuring lactate dehydrogenase (LDH) release (Promega, USA) according to the manufacturer's instructions.
[0050] 1-9. Measurement of antiproliferative activity Antiproliferative activity was measured using a water-soluble tetrazolium (WST) colorimetric assay (Ez-Cytox; Daeil Lab Service). NCI-N87, SNU1, SNU620, Hs746T, and MKN45 cells were plated in 96-well plates and treated with trastuzumab, T-DM1, IFNβ-R27T, or trastuzumab-IFNβ-C17S-R27T at various concentrations (0.0001 to 100 nM) for 72 h. Then, 10 ul of WST was added to each well and the cells were incubated for 4 h. The absorbance at 450 nm was measured using a microplate reader (TECAN).
[0051] 1-10. Measurement of antiviral activity The antiviral activity of trastuzumab-IFNβ, trastuzumab-IFNβ-R27T, and trastuzumab-IFNβ-C17S-R27T was measured using a cytopathic effect assay (CPE assay). To measure the cytopathic effect, A549 cells were plated in a 96-well plate and treated with each drug at different concentrations (0.78125-100 IU / mL). 22 hours after drug treatment, 1000 TCID50 / mL of EMCV was added. After 22 hours of co-culture, the cytopathic effect was measured by the absorbance value obtained using a WST colorimetric assay.
[0052] 1-11. Drug delivery to tumors Six-week-old female nude mice were subcutaneously injected with the HER2-positive gastric cancer cell line NCI-N87 to perform xenografts. To track the drugs, a fluorescent labeling agent (CF750) was attached to the IgG-IFNβ-R27T, trastuzumab-IFNβ-R27T, IgG-IFNβ-C17S-R27T, or trastuzumab-IFNβ-C17S-R27T drugs. The size of the formed tumors was approximately 100 mm. 3 When the mice reached the age of 10, three mice were randomly assigned to each group and IgG-IFNβ-R27T, trastuzumab-IFNβ-R27T, IgG-IFNβ-C17S-R27T, or trastuzumab-IFNβ-C17S-R27T were administered at 100 μg / mice. 24 hours after drug administration, the mice were anesthetized and the biodistribution of the drug was observed using a small animal optical bioimager (Ami HTX, Spectral Instruments Imaging). In addition, the tumor, liver, kidney, spleen, lung, and intestine of the mice with cancer xenografts were isolated for ex vivo analysis to observe the drug distribution between tissues. The experimental results were analyzed and statistically processed using Student's t-test.
[0053] 1-12. Xenograft animal experiments Five-week-old female nude mice were subcutaneously injected with the HER2-positive breast cancer cell line, HCC1954. The tumors formed were approximately 100 mm in size.3 When tumors reached 100%, five mice were randomly assigned to receive vehicle, trastuzumab-IFNβ-R27T, or trastuzumab-IFNβ-C17S-R27T at the appropriate dose, three times a week for three weeks (day 0). Tumors were measured twice a week using calipers and were calculated as (length * width) 2 ) / 2=Volume (mm 3 The experimental results were statistically processed using two-way ANOVA analysis.
[0054] 2. Results 2-1. Comparison of purification efficiency of fusion proteins Trastuzumab-IFNβ-C17S-R27T, trastuzumab-IFNβ-R27T and trastuzumab-IFNβ were expressed and purified under the same conditions using cell lines expressing trastuzumab-IFNβ. The transduced CHO-S cell line was cultured at 37 °C and 8% CO2 for 5 days. The expression concentration of the fusion protein in the cell culture medium was measured using Cedex-bio (Roche), and then purified by affinity chromatography using the AKTA instrument system and protein A beads.
[0055] As a result, as shown in Figures 2a to 2d, the purification efficiency of trastuzumab-IFNβ-C17S-R27T was approximately 92%, which was significantly improved compared to trastuzumab-IFNβ (52%) or trastuzumab-IFNβ-R27T (54%). It was also confirmed that the purification efficiency of the fusion protein bound to IFNβ-C17S-R27T was superior when cetuximab and atezolizumab antibodies were applied.
[0056] 2-2. Comparison of protein aggregation ratios of fusion proteins The problem of protein aggregation is closely related to the activity of the substance and may induce immunogenicity in patients, so it is an important consideration in the development of therapeutic drugs. To analyze protein aggregation, purified fusion proteins were comparatively analyzed using the FPLC-SEC method.
[0057] As a result, as shown in Figures 3a to 3c, the protein aggregation rate of trastuzumab-IFNβ-C17S-R27T was approximately 8%, which was significantly reduced compared to 40% trastuzumab-IFNβ-R27T or 85% trastuzumab-IFNβ. It was also confirmed that when cetuximab antibody and atezolizumab antibody were applied, the protein aggregation rate was significantly reduced when IFNβ-C17S-R27T was bound.
[0058] 2-3. Comparison of trastuzumab-IFNβ-C17S-R27T's ability to recognize HER2 antigen on the cell surface The HER2 antigen recognition ability of trastuzumab-IFNβ-C17S-R27T was compared with that of trastuzumab-IFNβ-R27T and trastuzumab. Each substance was reacted with NCI-N87 cells with high HER2 expression or MCF-7 and MDA-MB-231 cells with low HER2 expression, and the HER2 antigen recognition ability on the cell surface was analyzed by shifting the fluorescence intensity peak using a flow cytometer.
[0059] As a result, as shown in Figure 4a and Figure 4b, the untreated group was the control group, and the peak did not shift, but the groups treated with trastuzumab, trastuzumab-IFNβ-R27T, and trastuzumab-IFNβ-C17S-R27T shifted the fluorescence intensity peak according to the level of HER2 expression. In MCF-7 and MDA-MB-231 cell lines with low HER2 expression, the fluorescence intensity of all three substances increased slightly, but in NCI-N87 cell line with high HER2 expression, the fluorescence intensity increased significantly. In particular, the fact that there was no difference in the fluorescence peaks of the three substances confirmed that the HER2 antigen recognition ability of trastuzumab-IFNβ-C17S-R27T was no different from that of trastuzumab.
[0060] 2-4. IFN bioactivity of trastuzumab-IFNβ-C17S-R27T Interferon activity occurs through the phosphorylation of STAT, a sub-signal transduction pathway that binds to type I IFN receptors. To confirm the interferon activity of trastuzumab-IFNβ-C17S-R27T, NCI-N87 cells were treated with IFNβ-C17S-R27T and trastuzumab-IFNβ-C17S-R27T at different concentrations (0.004-40 pM) for 1 hour, and STAT1 phosphorylation and the expression of STAT1 and β-actin were confirmed by Western blotting.
[0061] As a result, as shown in Figure 5, the STAT1 phosphorylation signal was increased depending on the treatment concentration of trastuzumab-IFNβ-C17S-R27T, and the activity level of trastuzumab-IFNβ-C17S-R27T was shown to be similar to that of IFNβ-C17S-R27T.
[0062] 2-5. Antibody-dependent cellular cytotoxicity of trastuzumab-IFNβ-C17S-R27T Antibody-dependent cellular cytotoxicity is one of the main anticancer mechanisms of trastuzumab: when the antibody binds to target cells, cancer cell killing occurs through the activation of NK cells that have Fc gamma receptors that can bind to the Fc domain of trastuzumab. Trastuzumab-IFNβ-C17S-R27T contains trastuzumab, so it may have antibody-dependent cellular cytotoxicity like trastuzumab. Target cells, HER2-positive cancer cells (HCC1954, MDA-MB453), were treated with control IgG, trastuzumab, IFN-β, and trastuzumab-IFNβ-C17S-R27T at various concentrations, and co-cultured with effector cells, NK cells (NK-92MI-CD16a), at an effector to target cell ratio of 8:1, and antibody-dependent cellular cytotoxicity was confirmed.
[0063] As a result, as shown in Figure 6, control IgG and IFN-β induced little antibody-dependent cellular cytotoxicity, whereas trastuzumab targeting HER2 and trastuzumab-IFNβ-C17S-R27T containing it induced the same level of antibody-dependent cellular cytotoxicity. It was confirmed that the antibody-dependent cellular cytotoxicity effect induced by trastuzumab was fully maintained by trastuzumab-IFNβ-C17S-R27T.
[0064] 2-6. HER2 expression level in cancer cells and the inhibitory effect of trastuzumab-IFNβ-C17S-R27T on cancer cell proliferation IFN-β has the effect of directly suppressing the proliferation of cancer cells through receptor binding. The antiproliferative activity of trastuzumab-IFNβ-C17S-R27T was evaluated in gastric cancer cell lines with various levels of HER2 expression. As a result, as shown in Figures 7a and 7b, trastuzumab and the trastuzumab-based antibody-drug conjugate T-DM1 showed significant antiproliferative activity only in NCI-N87 cells with high HER2 expression, whereas trastuzumab-IFNβ-C17S-R27T showed significant antiproliferative activity not only in cell lines with high HER2 expression, but also in gastric cancer cell lines with intermediate (SNU1, SNU620) and low (Hs746T, MKN45) HER2 expression.
[0065] 2-7. Antiviral activity of trastuzumab-IFNβ-C17S-R27T The most common analytical method for evaluating the activity of IFN-containing substances is to measure the antiviral activity, which protects cells from the cytopathic effect (CPE) of a specific virus. Therefore, the IFN activity of trastuzumab-IFNβ, trastuzumab-IFNβ-R27T, and trastuzumab-IFNβ-C17S-R27T was quantitatively measured and compared using the CPE assay. As a result, as shown in Figures 8a and 8b, the antiviral activity of trastuzumab-IFNβ-C17S-R27T was approximately three times more effective than that of trastuzumab-IFNβ-R27T, and trastuzumab-IFNβ showed almost no antiviral activity within the concentration range tested.
[0066] 2-8. Tumor targeting ability of trastuzumab-IFNβ-C17S-R27T The extent to which each fusion protein trafficked in a tumor-specific manner was comparatively analyzed in an animal model. As a result, as shown in Figures 9a and 9b, a significant tumor targeting effect was observed in the trastuzumab fusion protein group compared to the IgG fusion protein group. In particular, trastuzumab-IFNβ-R27T showed a two-fold higher tumor targeting effect than IgG-IFNβ-R27T, while trastuzumab-IFNβ-C17S-R27T showed a four-fold higher tumor targeting effect than IgG-IFNβ-C17S-R27T. In addition, it was confirmed that the rate of tumor migration of trastuzumab-IFNβ-C17S-R27T was higher than that of trastuzumab-IFNβ-R27T compared to other organs.
[0067] 2-9. In vivo anticancer efficacy of trastuzumab-IFNβ-C17S-R27T To confirm the anti-cancer efficacy of the fusion protein, a tumor xenograft model was constructed, each substance was administered, and the tumor size was measured. As a result, as shown in Figure 10, a significant tumor suppression effect was observed in all groups treated with 10 mpk trastuzumab-IFNβ-R27T or 1, 3, and 10 mpk trastuzumab-IFNβ-C17S-R27T compared to vehicle. Trastuzumab-IFNβ-C17S-R27T reduced tumor volume in a concentration-dependent manner, and complete remission (1 / 5) was observed in some mice at 10 mpk. In addition, it was confirmed that trastuzumab-IFNβ-C17S-R27T had a better tumor suppression effect than trastuzumab-IFNβ-R27T at the same concentration (10 mpk). [Industrial Applicability]
