Abstract
Description
[Technical field]
[0001] cross reference This application claims the benefit of U.S. Provisional Application No. 62 / 845,119, filed May 8, 2019, and U.S. Provisional Application No. 62 / 884,588, filed August 8, 2019, both of which are incorporated herein by reference. [Background technology]
[0002] Janus kinases (JAKs) are a family of intracellular non-receptor tyrosine kinases that transduce cytokine-mediated signals through the JAK-STAT pathway. Four JAK family members, Janus kinase 1 (JAK1), Janus kinase 2 (JAK2), Janus kinase 3 (JAK3), and tyrosine kinase 2 (TYK2), are recognized as key components of cytokine-mediated actions. The important function of JAKs in cytokine signaling has implicated JAK inhibitors as potential therapeutics for a variety of diseases, including autoimmune and inflammatory diseases. [0087] JAK inhibitors Cytokines are important for host defense and immune regulation, but also play a major role in the immunopathogenesis of autoimmune diseases. Based on structure, different major families of cytokines can be recognized. The two main classes are the so-called type I and type II cytokine receptors. Type I receptors bind various interleukins (ILs), colony-stimulating factors, and hormones such as erythropoietin, prolactin, and growth hormone. Type II receptors bind interferons and IL-10-related cytokines. In contrast to other receptors whose intracellular domains encode kinases or other enzymatically active domains, the above receptors lack such elements. Instead, the cytoplasmic domains of type I and type II cytokine receptors bind members of a specific kinase family known as Janus kinases (JAKs), which include JAK1, JAK2, JAK3, and TYK2. Cytokine receptors pair with different JAKs, which are activated upon cytokine binding. As JAKs are phosphotransferases, they catalyze the transfer of phosphate from ATP to various substrates, such as cytokine receptors. This modification allows the recruitment of various signaling molecules, including members of the signal transducer and activator of transcription (STAT) family of DNA-binding proteins. STATs are another important JAK substrate. Phosphorylation of STATs promotes their nuclear accumulation and regulation of gene expression. In addition, studies with knockout mice support a critical and specific role for JAK signaling by type I / II cytokines but not other pathways. The important function of JAKs in cytokine signaling suggests the therapeutic potential of JAK inhibitors.

Protecting groups are used to block some or all of the reactive moieties and prevent such groups from participating in chemical reactions until the protecting group is removed. Each protecting group is preferably removable by a different means. Protecting groups that are cleaved under generally different reaction conditions fulfill the requirement of differential removal.
[0341] The protecting groups can be removed by acid, base, reducing conditions (e.g., hydrogenolysis), and / or oxidative conditions. Groups such as trityl, dimethoxytrityl, acetal, t-butyldimethylsilyl, etc. are acid labile and may be used to protect carboxy and hydroxy reactive moieties in the presence of Cbz groups, which are hydrogenolytically removable, and amino groups protected with Fmoc groups, which are base labile. Carboxylic acid and hydroxy reactive moieties may be blocked by acid labile groups such as t-butyl carbamate, or by base labile groups such as, but not limited to, methyl, ethyl, acetyl, etc., in the presence of amines blocked by carbamates, which are acid and base stable but hydrolytically removable.
[0342] Carboxylic acid and hydroxy reactive sites may further be blocked with hydrolytically removable protecting groups such as benzyl groups, while amine groups capable of hydrogen bonding with acids may be blocked with base-labile groups such as Fmoc. Carboxylic acid reactive sites may be protected by conversion to simple ester compounds as exemplified herein, including conversion to alkyl esters, or may be blocked with oxidatively removable protecting groups such as 2,4-dimethoxybenzyl, while coexisting amino groups may be blocked with fluoride-labile silyl carbamates.
[0343] Allyl blocking groups are useful in the presence of acid and base protecting groups because the acid blocking groups are stable and can be subsequently removed by metal or pi-acid catalysts. For example, allyl-blocked carboxylic acids can be reacted with Pd 0 -catalyzed deprotection. Another form of protecting group is a resin to which a compound or intermediate may be attached. As long as the residue is attached to the resin, the functional group is blocked and cannot react. Once released from the resin, the functional group becomes available for reaction.
[0344] In general, blocking / protecting groups may be selected from:

[0348] Pharmaceutical Compositions and Methods of Administration The JAK inhibitors described herein are administered to a subject in a biologically compatible form suitable for local administration to treat or prevent a disease, disorder, or condition. Administration of the JAK inhibitors as described herein may be in a pharmacological form that includes a therapeutically effective amount of the JAK inhibitor alone or in combination with a pharma- ceutically acceptable carrier.
[0349] In some embodiments, the compounds described herein are administered as pure chemicals. In other embodiments, the compounds described herein are combined with a pharma- ceutically suitable or acceptable carrier (also referred to herein as a pharma- ceutically suitable (or acceptable) excipient, a physiologically suitable (or acceptable) excipient, or a physiologically suitable (or acceptable) carrier) selected based on the selected route of administration and standard pharmaceutical practice, e.g., as described in Remington: The Science and Practice of Pharmacy (Gennaro, 21st Ed. Mack Pub. Co., Easton, PA (2005)).

A carrier (or excipient) is acceptable or suitable if it is compatible with the other ingredients of the composition and not deleterious to the recipient (subject) of the composition.
[0351] In some embodiments, there is a pharmaceutical composition comprising a pharma- ceutically acceptable carrier and a compound of Formula (I'), (I), (Ia'), (Ia), (Ib), (II'), (II), (IIa), (III), (IIIa), or (IV) or a pharma- ceutically acceptable salt or solvate. In some embodiments, there is a pharmaceutical composition comprising a pharma- ceutically acceptable carrier and a compound of Formula (I) or a pharma- ceutically acceptable salt or solvate. In some embodiments, there is a pharmaceutical composition comprising a pharma- ceutically acceptable carrier and a compound of Formula (Ia) or a pharma- ceutically acceptable salt or solvate. In some embodiments, there is a pharmaceutical composition comprising a pharma- ceutically acceptable carrier and a compound of Formula (Ib) or a pharma- ceutically acceptable salt or solvate. In some embodiments, there is a pharmaceutical composition comprising a pharma- ceutically acceptable carrier and a compound of Formula (II) or a pharma- ceutically acceptable salt or solvate. In some embodiments, there is a pharmaceutical composition comprising a pharma- ceutically acceptable carrier and a compound of Formula (IIa) or a pharma- ceutically acceptable salt or solvate.
[0352] Another embodiment is a pharmaceutical composition consisting essentially of a pharmaceutically acceptable carrier and a compound of Formula (I'), (I), (Ia'), (Ia), (Ib), (II'), (II), (IIa), (III), (IIIa), or (IV) or a pharmaceutically acceptable salt or solvate. In some embodiments, a pharmaceutical composition consisting essentially of a pharmaceutically acceptable carrier and a compound of Formula (I) or a pharmaceutically acceptable salt or solvate. In some embodiments, a pharmaceutical composition consisting essentially of a pharmaceutically acceptable carrier and a compound of Formula (Ia) or a pharmaceutically acceptable salt or solvate. In some embodiments, a pharmaceutical composition consisting essentially of a pharmaceutically acceptable carrier and a compound of Formula (II) or a pharmaceutically acceptable salt or solvate. In some embodiments, there is a pharmaceutical composition consisting essentially of a pharma- ceutically acceptable carrier and a compound of Formula (IIa), or a pharma- ceutically acceptable salt or solvate.
[0353] In certain embodiments, the compounds described herein are substantially pure in that they contain less than about 5%, less than about 1%, or less than about 0.1% of other small organic molecules, e.g., contaminating intermediates or by-products produced during one or more of the steps of a synthetic method.
[0354] These formulations include those suitable for oral, topical, buccal, parenteral (eg, subcutaneous, intramuscular, intradermal, or intravenous) or aerosol administration.
[0355] Typical pharmaceutical compositions are used in the form of pharmaceutical preparations, such as solid, semi-solid, or liquid forms, which contain one or more of the disclosed compounds as active ingredients mixed with organic or inorganic carriers or excipients suitable for in vitro, enteral, or parenteral use. In some embodiments, the active ingredient is usually formulated with a non-toxic, pharma- ceutically acceptable carrier for, for example, tablets, pellets, capsules, suppositories, solutions, emulsions, suspensions, and other forms suitable for use. The active subject compound is included in the pharmaceutical composition in an amount sufficient to exert the desired effect depending on the disease process or condition.
[0356] In some embodiments, the JAK inhibitors described herein are administered to a subject in a biologically compatible form suitable for topical administration to treat or prevent a skin disease, disorder, or condition. "Biologically compatible form suitable for topical administration" refers to a form of the JAK inhibitor to be administered in which any toxic effects outweigh the therapeutic benefits of the inhibitor. Administration of the JAK inhibitor as described herein may be in a pharmacological form that includes a therapeutically effective amount of the JAK inhibitor alone or in combination with a pharma- ceutical acceptable carrier.
[0357] Topical administration of JAK inhibitors can be provided in the form of aerosol, semi-solid pharmaceutical composition, powder, or solution. The term "semi-solid composition" refers to ointments, creams, salves, jellies, or other pharmaceutical compositions with substantially similar consistency suitable for application to the skin.Examples of semi-solid compositions are provided in Chapter 17 of The Theory and Practice of Industrial Pharmacy, Lachman, Lieberman and Kanig, published by Lea and Febiger (1970), and Chapter 67 of Remington's Pharmaceutical Sciences, 15th Edition, published by Mack Publishing Company (1975).
[0358] Dermal or skin patches are another method of transdermal delivery of the therapeutic or pharmaceutical compositions described herein. Patches can provide an absorption enhancer, such as DMSO, to increase absorption of the compound. Patches may include those that control the rate of drug delivery to the skin. Patches may provide various administration systems, including reservoir systems or monolithic systems, respectively. Reservoir designs may have, for example, four layers: an adhesive layer that directly contacts the skin, a control membrane that controls the diffusion of the drug molecules, a reservoir of drug molecules, and a water-resistant backing. Such designs deliver a certain amount of drug over a specific period of time, and the delivery rate must be below the saturation limit of various skin types. For example, monolithic designs generally have only three layers: an adhesive layer, a polymer matrix that contains the compound, and a water-tight backing. This design provides a saturating amount of drug to the skin. Delivery is thereby controlled by the skin. When the amount of drug falls below the saturation amount in the patch, the delivery rate decreases.
[0359] In one embodiment, the topical composition may be in the form of a hydrogel, for example based on polyacrylic acid or polyacrylamide, in the form of an ointment containing polyethylene glycol (PEG) as a carrier, for example the standard ointment DAB8 (50% PEG300, 50% PEG1500), or in the form of an emulsion, in particular a microemulsion based on water-in-oil or oil-in-water, optionally with the addition of liposomes. Suitable penetration enhancers (entraining agents) include sulfoxide derivatives such as dimethyl sulfoxide (DMSO) or decyl methyl sulfoxide (decyl-MSO), and transcutol (diethylene glycol monoethyl ether) or cyclodextrins, as well as pyrrolidones, such as 2-pyrrolidone, N-methyl-2-pyrrolidone, 2-pyrrolidone-5-carboxylic acid, or biodegradable N-(2-hydroxyethyl)-2-pyrrolidone and their fatty acid esters, urea derivatives such as dodecylurea, 1,3-didodecylurea, and 1,3-diphenylurea, and terpenes, such as D-limonene, menthone, a-terpinol, carbol, limonene oxide, or 1,8-cineole.
[0360] Ointments, pastes, creams, and gels may further contain excipients such as starch, tragacanth, cellulose derivatives, polyethylene glycol, silicone, bentonite, silicic acid, and talc, or mixtures thereof. Powders and sprays may further contain excipients such as lactose, talc, silicic acid, aluminum hydroxide, calcium silicate, and polyamide powder, or mixtures of these substances. The solution of nanocrystalline antimicrobial metals can be converted into an aerosol or spray by any of the known means routinely used to make aerosol formulations. Typically, such methods include pressurizing a container of the solution with, or providing a means for pressurization, a carrier gas, usually inert, and passing the pressurized gas through a small orifice. In addition, sprays may contain conventional propellants such as chlorofluorohydrocarbons, and volatile unsubstituted hydrocarbons such as butane and propane.
[0361] The carrier may further include other pharma- ceutically acceptable excipients to modify or maintain the pH, osmolarity, viscosity, clarity, color, sterility, stability, dissolution rate, or odor of the formulation. The anti-skin aging composition may further include antioxidants, sunscreens, natural retinoids (e.g., retinol), and other additives commonly found in skin treatment compositions.
[0362] In some embodiments for preparing solid compositions such as tablets, the primary active ingredient is mixed with a pharmaceutical carrier, such as conventional tablet ingredients such as corn starch, lactose, sucrose, sorbitol, talc, stearic acid, magnesium stearate, dicalcium phosphate, or gums, and other pharmaceutical diluents, such as water, to form a solid preformulation composition containing a homogenous mixture of the disclosed compound or a non-toxic pharma- ceutically acceptable salt thereof. When such a preformulation composition is referred to as homogenous, it is meant that the active ingredient is evenly dispersed throughout the composition such that the composition may be readily subdivided into equally effective unit dosage forms, such as tablets, pills, and capsules.
[0363] In solid dosage forms for oral administration (capsules, tablets, pills, dragees, powders, granules, etc.), the subject compositions may be formulated with one or more pharma- ceutically acceptable carriers, such as sodium citrate or dicalcium phosphate, and / or one or more of the following: (1) fillers or extenders, such as starch, cellulose, microcrystalline cellulose, silicided microcrystalline cellulose, lactose, sucrose, glucose, mannitol, and / or silicic acid; (2) saccharides, such as carboxymethylcellulose, hypromellose, alginates, gelatin, polyvinylpyrrolidone, sucrose, and / or acacia. The composition may be mixed with any of the following: a binder, (3) a diluent such as glycerin, (4) a disintegrant such as crospovidone, croscarmellose sodium, sodium starch glycolate, agar-agar, calcium carbonate, potato or tapioca starch, alginic acid, certain silicates, and sodium carbonate, (5) a solution retardant such as paraffin, (6) an absorption promoter such as a quaternary ammonium compound, (7) a wetting agent such as docusate sodium, cetyl alcohol, and glycerol monostearate, (8) an absorbent such as kaolin and bentonite clay, (9) a lubricant such as talc, calcium stearate, magnesium stearate, solid polyethylene glycol, sodium lauryl sulfate, and mixtures thereof, and (10) a coloring agent. In the case of capsules, tablets, and pills, the composition may include a buffer in some embodiments. In some embodiments, solid compositions of a similar type are also employed as fillers in soft and hard-filled gelatin capsules using such excipients as lactose or milk sugar, as well as high molecular weight polyethylene glycols and the like.
[0364] In some embodiments, tablets are made by compression or molding, optionally with one or more accessory ingredients. In some embodiments, compressed tablets are prepared using binders (e.g., gelatin or hydroxypropylmethylcellulose), lubricants, inert diluents, preservatives, disintegrants (e.g., sodium starch glycolate or cross-linked sodium carboxymethylcellulose), surface active agents, or dispersants. In some embodiments, molded tablets are made by molding a mixture of the subject composition moistened with an inert liquid diluent in a suitable machine. In some embodiments, tablets and other solid dosage forms, such as dragees, capsules, pills, and granules, are scored or prepared with coatings and shells, such as enteric coatings and other coatings.
[0365] Compositions for inhalation or insufflation include solutions and suspensions in pharma- ceutically acceptable aqueous or organic solvents, or mixtures thereof, and powders.Liquid dosage forms for oral administration include pharma-ceutically acceptable emulsions, microemulsions, solutions, suspensions, syrups, and elixirs. In addition to the subject compositions, in some embodiments, liquid dosage forms contain inert diluents such as water or other solvents, solubilizers, and emulsifiers, such as ethyl alcohol, isopropyl alcohol, ethyl carbonate, ethyl acetate, benzyl alcohol, benzyl benzoate, propylene glycol, 1,3-butylene glycol, oils (especially cottonseed oil, peanut oil, corn oil, germ oil, olive oil, castor oil, and sesame oil), glycerol, tetrahydrofuryl alcohol, polyethylene glycol, and fatty acid esters of sorbitan, cyclodextrin, and mixtures thereof.
[0366] In some embodiments, suspensions contain a subject composition as well as a suspending agent such as, for example, ethoxylated isostearyl alcohol, polyoxyethylene sorbitol and sorbitan esters, microcrystalline cellulose, aluminum metahydroxide, bentonite, agar, tragacanth, and mixtures thereof.
[0367] In some embodiments, powders and sprays contain the subject composition as well as excipients such as lactose, talc, silicic acid, aluminum hydroxide, calcium silicate, and polyamide powder, or mixtures of these substances. In some embodiments, sprays further contain conventional propellants such as chlorofluorohydrocarbons and volatile unsubstituted hydrocarbons, such as butane and propane.
[0368] Alternatively, the compositions and compounds disclosed herein are administered by aerosol. This administration is accomplished by preparing an aerosol aqueous solution, liposomal preparation, or solid particles containing the compound. In some embodiments, a non-aqueous (e.g., fluorocarbon propellant) suspension is used. In some embodiments, a sonic nebulizer is used, which causes the compound contained in the subject composition to break down. This is because it minimizes the shearing of the drug. Typically, an aerosol aqueous solution is made by formulating an aqueous solution or suspension of the subject composition together with conventional pharma- ceutically acceptable carriers and stabilizers. Carriers and stabilizers vary according to the requirements of the particular subject composition, but typically include non-ionic surfactants (Tween, Pluronics, or polyethylene glycol), innocuous proteins such as serum albumin, sorbitan esters, amino acids such as oleic acid, lecithin, glycine, buffers, salts, sugars, or sugar alcohols. Aerosols are typically prepared from isotonic solutions.
[0369] Pharmaceutical compositions suitable for parenteral administration may comprise the subject composition in combination with one or more pharma- ceutically acceptable sterile isotonic aqueous or non-aqueous solutions, dispersions, suspensions, or emulsions, or sterile powders to be reconstituted immediately before use into sterile injectable solutions or dispersions, which in some embodiments contain antioxidants, buffers, bacteriostats, solutes that render the formulation isotonic with the blood of the intended recipient, suspending agents, or thickening agents.
[0370] Examples of suitable aqueous and non-aqueous carriers that can be used in the pharmaceutical compositions include water, ethanol, polyols (such as glycerol, propylene glycol, polyethylene glycol, etc.) and suitable mixtures thereof, vegetable oils such as olive oil, and injectable organic esters such as ethyl oleate and cyclodextrin. Proper fluidity is maintained, for example, by the use of coating materials such as lecithin, by maintaining the required particle size in the case of dispersions, and by the use of surfactants.
[0371] The dosage of a composition comprising at least one compound described herein will vary depending on the condition of the patient (eg, human), ie, the stage of the disease, general health, age, and other factors.
[0372] The pharmaceutical composition is administered in a manner appropriate for the disease to be treated (or prevented). The appropriate dosage, as well as the appropriate duration and frequency of administration, will be determined by factors such as the patient's condition, the type and severity of the patient's disease, the specific form of the active ingredient, and the method of administration. In general, the appropriate dosage and treatment regimen provides the composition in an amount sufficient to provide therapeutic and / or prophylactic benefits (e.g., improved clinical outcomes such as more frequent complete or partial remissions, longer disease-free and / or overall survival, or reduced severity of symptoms). Optimal dosages are generally determined using experimental models and / or clinical trials. In some embodiments, optimal dosages depend on the patient's body type, weight, or blood volume.
[0373] Oral administration is generally performed at about 1.0 mg to about 1000 mg per day, 1 to 4 times a day.
[0374] The dose may be repeated depending on the pharmacokinetic parameters of the dosage formulation and the route of administration used.
[0375] It is particularly convenient to formulate compositions in dosage unit forms for ease of administration and uniformity of dosage. Dosage unit forms as used herein refer to physically discrete units suitable as unitary dosages for the mammalian subject to be treated, each unit containing a predetermined amount of active compound calculated to produce the desired therapeutic effect in association with the required pharmaceutical carrier. The specifications for dosage unit forms are dictated by and directly dependent on (a) the inherent characteristics of the JAK inhibitor and the particular therapeutic effect to be achieved, and (b) the inherent limitations in the art of compounding such active compounds for the treatment of individual sensitivities. A specific dosage may be easily calculated by those skilled in the art according to, for example, the approximate body weight or body surface area of the patient, or the volume of the body space to be occupied. The dosage is also calculated depending on the particular route of administration selected. Further refinement of the calculations required to determine the appropriate dosage for treatment is routinely performed by those skilled in the art. Such calculations may be performed by those skilled in the art without undue experimentation, taking into account the activity of the JAK inhibitors disclosed herein in the assay preparation of target cells. The exact dosage is determined in conjunction with standard dose-response testing. It will be understood that the amount of composition actually administered will be determined by the medical practitioner taking into account the relevant circumstances, including the disease being treated, the choice of composition to be administered, the individual age, weight, and response of the patient, the severity of the patient's symptoms, and the chosen route of administration.
[0376] The toxicity and therapeutic efficacy of such JAK inhibitors are e.g. 50 (a dose lethal to 50% of the population) and ED 50The dose that is therapeutically effective in 50% of the population can be determined by standard pharmaceutical procedures in cell cultures or experimental animals. The dose ratio between toxic and therapeutic effects is the therapeutic index, which is the LD 50 / ED 50 The therapeutic index can be expressed as a ratio of 0.01 to 0.02. JAK inhibitors that exhibit a large therapeutic index are preferred. JAK inhibitors that exhibit toxic side effects may be used, but care must be taken to design a delivery system that targets such inhibitors to the site of affected tissue in order to reduce side effects by minimizing potential damage to uninfected cells.
[0377] Data from cell culture assays and animal studies can be used to formulate a range of dosages for use in humans. Such doses of JAK inhibitors can be used to achieve ED with little or no toxicity. 50 The dose may vary within this range depending on the dosage form and route of administration utilized. For any JAK inhibitor used in the methods described herein, the therapeutically effective dose can be estimated initially from cell culture assays. The IC 50 Dosages may be formulated in animal models to achieve a circulating plasma concentration range (the concentration of the JAK inhibitor that achieves half-maximal inhibition of symptoms) that includes. Such information can be used to more accurately determine useful doses in humans. Plasma concentrations can be measured, for example, by high performance liquid chromatography. EXAMPLES

[0463] Example 9: JAK / TYK2 Assay 10 mM test compound stocks or 1 mM control compound stocks (tofocitinib, ruxolitinib, or staurosporine) in DMSO were diluted to 0.4 mM in DMSO. 3-fold serial dilutions were then performed in DMSO to generate 10 different concentrations of compound. The assay was performed in a 384-well white plate. 0.5 μL of 40x compound DMSO solutions at different concentrations were mixed with 10 μL of 2x enzyme prepared in reaction buffer (20 mM HEPES, 10 mM MgCl2, 0.01% Tween, 1 mM DTT, pH 7.5). 10 μL of 2x substrate mix prepared in reaction buffer was then added to start the reaction. All the solution was allowed to drop to the bottom of the plate by briefly swirling. The final concentrations of test compounds in the reaction mixtures were 10000, 3333, 1111, 370, 123, 41.2, 13.7, 4.57, 1.52, and 0.51 nM. The concentration of control compounds was 10 times lower. The enzyme reaction was carried out at 25°C for 1-2 hours. The reaction was stopped by adding 10 μL of KinaseGlo reagent to generate a luminescent signal, which was measured using Envision. The luminescent signal was inversely correlated with kinase activity. Reaction mixtures that did not contain enzyme were negative controls. Compound-free reaction mixtures were positive controls. The final concentrations of enzyme and substrate, as well as incubation times, are summarized in the table below.
