Description

[Technical field]

[0001] The present disclosure relates to a liposome composition for cosmetics. [Background technology]

[0002] Liposomes are closed vesicles formed by a lipid bilayer containing lipids such as lecithin, and have an aqueous phase (internal aqueous phase) within the space of the closed vesicles. In addition, liposomes usually exist in a dispersed state in an aqueous solution (external aqueous phase) outside the closed vesicles.

[0003] In recent years, the application of liposomes containing medicinal ingredients or cosmetic ingredients in the internal aqueous phase to pharmaceutical compositions, cosmetics, and the like has been investigated.

[0004] For example, Patent Document 1 proposes a liposome composition that contains liposomes having an average particle size of 5 nm to 100 nm or less, which are contained in an internal aqueous phase that also contains a pharmaceutical ingredient. Furthermore, Patent Document 2 proposes a liposome composition for cosmetics that contains an oil or fat component in the internal aqueous phase and liposomes with an average particle size of 82 nm or less. Furthermore, Patent Document 3 proposes a liposome composition for cosmetics that contains a cosmetic ingredient in its internal aqueous phase and contains liposomes with an average particle size of 50 nm or less. Furthermore, Patent Document 4 proposes a cosmetic composition that contains a disk-shaped bicellar structure formed by a lipid bilayer membrane, instead of a liposome. Furthermore, Patent Document 5 proposes a cosmetic preparation containing vesicles in an external aqueous phase that contains ethanol. [Prior art documents] [Patent documents]

[0005] [Patent Document 1] Patent No. 6251385 [Patent Document 2] JP 2020-93992 A [Patent Document 3] Patent No. 5064717 [Patent Document 4] International Publication No. 2017 / 090740 [Patent Document 5] JP 2016-56198 A Summary of the Invention [Problem to be solved by the invention]

[0006] When liposomes encapsulating cosmetic ingredients are used in cosmetics, there is a demand for liposomes that can encapsulate a larger amount of cosmetic ingredients. The present inventors have now discovered that there is room for improvement in the encapsulation rate of cosmetic ingredients in the liposomes contained in the compositions proposed in Patent Documents 1 to 3, the bicellar structure contained in the composition proposed in Patent Document 4, and the vesicles contained in the composition proposed in Patent Document 5.

[0007] Furthermore, from the standpoint of the encapsulation rate of the cosmetic ingredient, it is desirable that the liposomes contained in the liposome composition contain a large proportion of liposomes having a single lamellar structure in which the lipid bilayer is one layer (unilamellar vesicles). The present inventors have further discovered that the compositions of Patent Documents 2, 3, and 5 have a small proportion of liposomes having a single lamellar structure, and therefore the encapsulation rate of the cosmetic ingredient is insufficient.

[0008] The present disclosure has been made in consideration of the above circumstances, and an object of the present disclosure is to provide a liposome composition for cosmetics, which contains liposomes with an improved encapsulation rate of a cosmetic ingredient. In the following embodiments, the components are not essential unless specifically stated otherwise. The same applies to the numerical values and their ranges, and they do not limit the present disclosure.

[0013] In the present disclosure, a numerical range indicated using "~" includes the numerical values before and after "~" as the minimum and maximum values, respectively. In the numerical ranges described in the present disclosure in stages, the upper or lower limit value described in one numerical range may be replaced with the upper or lower limit value of another numerical range described in stages. In addition, in the numerical ranges described in the present disclosure, the upper or lower limit value of the numerical range may be replaced with a value shown in the examples. In the present disclosure, each component may contain multiple types of corresponding compounds. When multiple substances corresponding to each component are present in the composition, the content of each component means the total content of the multiple substances present in the composition, unless otherwise specified.

[0014] In the present disclosure, a "liposome" is a closed endoplasmic reticulum formed by a lipid bilayer membrane. A structure with a single lipid bilayer is called a single lamellar structure, and a structure with multiple lipid bilayers is called a multilamellar structure. In this disclosure, "internal aqueous phase" refers to the solution present within closed endoplasmic reticulum. In this disclosure, "external aqueous phase" refers to the solution in which the liposomes are dispersed.

[0015] In this disclosure, "cholesterol" is a general term for sterols contained in animals, and "phytosterol" is a general term for sterols contained in plants.

[0016] In the present disclosure, the term "beauty ingredient" refers to an ingredient that acts on the body, such as the skin, and means an active ingredient used in cosmetics and the like. Moreover, the term "water-soluble" means that the solubility in 100 g of water at a liquid temperature of 22° C. and a pH of 7.0 is 0.1 g or more.

[0017] In the present disclosure, the term "average particle size" refers to the volume average particle size determined by dynamic light scattering. Examples of dynamic light scattering particle size measuring instruments include the Concentrated System Particle Size Analyzer FPAR-1000 (manufactured by Otsuka Electronics Co., Ltd.), Nanotrack UPA (manufactured by Nikkiso Co., Ltd.), and Dynamic Light Scattering Particle Size Distribution Measuring Instrument LB-550 (manufactured by Horiba, Ltd.). The average particle size is measured by diluting the liposome composition for cosmetics with pure water to 10 times its mass, at room temperature (for example, 25° C.). The "pure water" used herein is pure water obtained using an ultrapure water production apparatus manufactured by Merck Ltd.

[0018] In the present disclosure, "the ratio of the total number of liposomes having a single lamellar structure to the total number of liposomes contained in the liposome composition for cosmetics" is determined by observing the liposomes contained in the liposome composition for cosmetics in an ice-embedded state using a transmission electron microscope (TEM) and counting the total number of liposomes contained in the liposome composition for cosmetics and the total number of liposomes having a single lamellar structure. More specifically, first, a TEM image of the liposome composition for cosmetic use at a magnification of 50,000 times is obtained using a transmission electron microscope. The total number of liposomes present within a circle of radius 2 μm and the total number of liposomes having a single lamellar structure are counted at each of three arbitrary points on the TEM image to determine the "ratio of the total number of liposomes having a single lamellar structure to the total number of liposomes contained in the liposome composition for cosmetic use." In the present disclosure, the "ratio of the total number of liposomes having a single lamellar structure to the total number of liposomes contained in the liposome composition for cosmetics" is the average value of three points. In the present disclosure, the encapsulation rate of a cosmetic ingredient in a liposome can be evaluated by the "average particle size of the liposomes" and the "ratio of the total number of liposomes having a single lamellar structure to the total number of liposomes contained in the liposome composition for cosmetics."

[0019] (Liposome composition for cosmetics) The liposome composition for cosmetics of the present disclosure comprises polyoxyethylene phytosterol, lecithin, and at least one of cholesterol and a phytosterol other than the polyoxyethylene phytosterol, and comprises liposomes having an average particle size of 100 nm to 350 nm, water, ethanol, and a cosmetic ingredient, an inner aqueous phase that is encapsulated in the liposomes, and an outer aqueous phase that contains water and ethanol and disperses the liposomes.

[0020] According to the liposome composition for cosmetics of the present disclosure, it is possible to provide a liposome composition for cosmetics containing liposomes that have an excellent encapsulation rate of a cosmetic ingredient.

[0021] The reason why the above effect is achieved is presumed to be as follows, but is not limited to this. In the liposome composition for cosmetics of the present disclosure, the liposomes contain polyoxyethylene phytosterol, which inhibits the lipid bilayer membranes from approaching each other, and polyoxyethylene phytosterol contributes to the formation of liposomes, so it is presumed that the average particle size of the liposomes formed and the proportion of liposomes having a single lamellar structure are increased, thereby improving the encapsulation rate of the cosmetic ingredient.

[0022] From the viewpoint of improving the encapsulation rate of a beauty ingredient, in the liposome composition for cosmetics, the ratio of the polyoxyethylene phytosterol content to the ethanol content (polyoxyethylene phytosterol content / ethanol content) is, by mass, preferably 0.001 to 1, more preferably 0.002 to 0.98, and even more preferably 0.03 to 0.97. In addition, by setting the ratio of the polyoxyethylene phytosterol content to the ethanol content within the above numerical range, it is possible to prevent the liposomes from aggregating over time, which would increase the particle size of the liposomes and cause the liposome composition for cosmetics to become suspended. Furthermore, by setting the ratio of the polyoxyethylene phytosterol content to the ethanol content within the above numerical range, it is possible to prevent aggregated liposomes from coalescing and becoming multilamellar, which would result in a decrease in the encapsulation rate of the beauty ingredient. The above-mentioned ability to inhibit aggregation of liposomes is hereinafter also referred to as temporal stability. The ratio of the polyoxyethylene phytosterol content to the ethanol content in the liposome composition for cosmetics is calculated by rounding off to the fourth decimal place.

[0023] In the liposome composition for cosmetics, the ratio of the polyoxyethylene phytosterol content to the cholesterol content (polyoxyethylene phytosterol content / cholesterol content) is preferably 2 or less, more preferably 1.7 or less, and even more preferably 1 or less, by mass. By setting the ratio of the polyoxyethylene phytosterol content to the cholesterol content in the liposome composition for cosmetics to 2 or less, on a mass basis, the proportion of liposomes having a single lamellar structure in the liposome composition for cosmetics is increased, and the encapsulation rate of the cosmetic ingredient is further improved. Furthermore, the lower limit of the ratio of the polyoxyethylene phytosterol content to the cholesterol content in the liposome composition for cosmetics is not particularly limited, but from the viewpoint of liposome formability, it is preferably 0.1 or more. The ratio of the polyoxyethylene phytosterol content to the cholesterol content in the liposome composition for cosmetics is calculated by rounding off to one decimal place.

[0024] (Liposomes) In the liposome composition for cosmetics of the present disclosure, the liposomes have an average particle size of 100 nm to 350 nm. By setting the average particle size of the liposomes to 100 nm or more, the encapsulation rate of the cosmetic ingredient is improved. In addition, by setting the average particle size of the liposomes to 100 nm or more, the proportion of liposomes having a single lamellar structure in the liposome composition for cosmetics tends to be increased. Liposomes with an average particle size of more than 350 nm have low stability over time, and tend to become multilamellar due to aggregation and coalescence over time, resulting in a decrease in the encapsulation rate of cosmetic ingredients. By making the average particle size of the liposomes 350 nm or less, the multilamellar formation can be suppressed. In addition, when the average particle size of the liposomes is more than 350 nm, the turbidity of the liposome composition for cosmetics produced tends to increase. By making the average particle size of the liposomes 350 nm or less, the increase in turbidity of the liposome composition for cosmetics can be suppressed, and the appearance of the liposome composition for cosmetics tends to be excellent. Furthermore, by making the average particle size of the liposomes 350 nm or less, the permeability of the liposome composition for cosmetics into the skin, etc. tends to be improved. From the viewpoint of improving the encapsulation rate of the cosmetic ingredient and stability over time, the average particle size of the liposomes is preferably 110 nm to 330 nm, more preferably 120 nm to 320 nm, and even more preferably 150 nm to 300 nm.

[0025] From the viewpoint of improving the encapsulation rate of the cosmetic ingredient, the liposomes are preferably liposomes having a single lamellar structure (also called unilamellar vesicles), and the ratio of the total number of liposomes having a single lamellar structure to the total number of liposomes contained in the liposome composition is preferably 50% or more, and more preferably 70% or more. The liposome composition may contain a liposome having a multilamellar structure (also called a multilamellar endoplasmic reticulum).

[0026] The liposome contains a polyoxyethylene phytosterol, lecithin, and at least one of cholesterol and a phytosterol other than the polyoxyethylene phytosterol. The evaporation mentioned above includes both intentional evaporation of a part or all of the organic solvent and water, and natural evaporation of a part or all of the organic solvent and water during the stirring and emulsification process.

[0050] When the organic solvent and water are intentionally evaporated, the evaporation method is not particularly limited, but examples thereof include a method of heating the organic solvent and water, a method of leaving the liposome composition for use in cosmetics to stand, a method of stirring the liposome composition for use in cosmetics, and a method of performing vacuum degassing.

[0051] The method for encapsulating the cosmetic ingredient in the liposome is not particularly limited, and can be carried out by using an aqueous phase composition in which the cosmetic ingredient is dissolved during emulsification.

[0052] The obtained liposome composition for cosmetics may be subjected to dialysis, filtration, extrusion treatment, etc., which can make the average particle size of the liposomes contained therein uniform. The extrusion treatment is a method in which the liposome composition for cosmetics is passed through a filter having fine holes to apply physical shearing force and atomize the composition. When passing the liposome composition for cosmetics, the liposome composition for cosmetics and the filter are kept at a temperature equal to or higher than the phase transition temperature of lecithin, thereby enabling rapid atomization.

[0053] (Application) In one embodiment, the liposome composition for cosmetics of the present disclosure is a cosmetic. The form of the cosmetic is not particularly limited, and may be liquid, jelly, gel, cream, solid such as a stick, or semi-solid. Examples of the cosmetic include skin care cosmetics (skin lotion, emulsion, serum, sunscreen (sunscreen agent), etc.), body cosmetics (body lotion, etc.), scalp cosmetics, packs, etc., but are not limited thereto.

[0054] All publications, patent applications, and standards mentioned in this specification are herein incorporated by reference to the same extent as if each individual publication, patent application, and standard was specifically and individually indicated to be incorporated by reference. [0079] <<Encapsulation rate evaluation-1 (proportion of liposomes with single lamellar structure)>> The liposomes contained in the liposome compositions for cosmetics produced in the Examples and Comparative Examples were embedded in ice and TEM images were taken at a magnification of 50,000 times using a transmission electron microscope.

[0080] The total number of liposomes present within a circle of radius 2 μm and the total number of liposomes having a single lamellar structure were counted at three arbitrary points of the TEM image, and the average ratio of the total number of liposomes having a single lamellar structure to the total number of liposomes contained in the liposome composition for cosmetics was confirmed and evaluated based on the following evaluation criteria. The evaluation results are shown in Tables 1 to 3. TEM images of Example 1-2, Example 3-1, Example 3-2, Comparative Example 1-2, and Comparative Example 1-5 are shown in Figures 1 to 5. The total number of liposomes and the total number of liposomes having a single lamellar structure are indicated by circles shown in Figures 1 to 5, which represent an arbitrary circle where the total number of liposomes and the total number of liposomes having a single lamellar structure were counted. (Evaluation Criteria) A: The proportion of liposomes having a single lamellar structure was 70% or more. B: The proportion of liposomes having a single lamellar structure was 50% or more and less than 70%. C: The proportion of liposomes having a single lamellar structure was 30% or more and less than 50%. D: The proportion of liposomes having a single lamellar structure was less than 30%.

[0081] <<Encapsulation rate evaluation-2 (average particle size of liposomes)>> The liposome compositions for cosmetics produced in the Examples and Comparative Examples were diluted 10 times by mass with pure water, and the volume average particle size of the liposomes contained therein was measured by dynamic light scattering using a concentrated particle size analyzer FPAR-1000 (manufactured by Otsuka Electronics Co., Ltd.). The volume average particle size was measured within 20 to 24 hours after the production of the liposome compositions for cosmetics. The temperature of the measurement environment was 25°C. The measurement results are shown in Tables 1 to 3.

[0082] <<Evaluation of stability over time>> The liposome compositions for cosmetics produced in Examples 1-5 to 1-7, Examples 2-1 to 2-5, Examples 3-1 to 3-2, and Comparative Example 1-4 were allowed to stand in a 50° C. environment for one week. The liposome composition for cosmetics after standing was diluted 10 times by mass with pure water, and the volume average particle size of the liposomes contained in the liposome composition for cosmetics was measured by the method described above. The ratio of the volume average particle diameter of the liposomes contained in the liposome composition for cosmetic preparations immediately after production to the volume average particle diameter of the liposomes contained in the liposome composition for cosmetic preparations immediately after production (volume average particle diameter of the liposomes contained in the liposome composition for cosmetic preparations after being left to stand / volume average particle diameter of the liposomes contained in the liposome composition for cosmetic preparations immediately after production) was determined, and the temporal stability of the liposome composition for cosmetic preparations was evaluated based on the following evaluation criteria. The evaluation results are shown in Tables 1 to 3. In addition, in Comparative Examples 1-2 and 1-3, the proportion of liposomes having a single lamellar structure was small and the encapsulation rate of the cosmetic ingredient was insufficient, so evaluation of stability over time was not performed, and the evaluation results are indicated as "-" in Table 1. In addition, in Comparative Example 1-5, since the evaluation of the encapsulation rate was insufficient, the evaluation of the stability over time was not performed, and the evaluation result in Table 1 is marked with "-". (Evaluation Criteria) A: The volume average particle size ratio was less than 1.1. B: The ratio of the volume average particle size was 1.1 or more and less than 1.4. C: The volume average particle size ratio was 1.4 or more.

[0083] [Table 1]

[0084] [Table 2]

[0085] [Table 3]

[0086] The results in Table 1 show that the liposome compositions obtained in Examples 1-1 to 1-7 have a higher proportion of liposomes having a single lamellar structure and are superior in encapsulation rate of cosmetic ingredients compared to the liposome compositions obtained in Comparative Examples 1-2 to 1-3 that do not contain at least one of polyoxyethylene phytosterol and ethanol. Furthermore, the results in Table 1 show that the liposome compositions obtained in Examples 1-1 to 1-7 have a higher proportion of liposomes having a single lamellar structure and are superior in encapsulation rate of cosmetic ingredients compared to the liposome composition obtained in Comparative Example 1-5, which does not contain at least one of cholesterol and phytosterols other than polyoxyethylene phytosterol and contains liposomes with an average particle diameter of less than 100 nm. Moreover, it is clear that the liposomes contained in the liposome compositions obtained in Examples 1-1 to 1-7 have a large average particle size and are excellent in the encapsulation rate of the cosmetic ingredient. In Comparative Examples 1-2 to 1-3, the content of the cosmetic ingredient is the same as in Examples 1-1 to 1-7; however, since the proportion of liposomes having a single lamellar structure contained in the obtained liposome composition is small, most of the cosmetic ingredient is contained in the external aqueous phase rather than the internal aqueous phase. The liposome compositions obtained in Examples 1-1 to 1-7 are found to have superior stability over time compared to the liposome composition obtained in Comparative Example 1-4, which does not contain polyoxyethylene phytosterol and contains liposomes with an average particle size of more than 350 nm. Incidentally, it is presumed that the liposome formation was low in Comparative Example 1-1 because ethanol was not contained.

[0087] The results in Tables 2 and 3 show that the liposome compositions obtained in Examples 2-1 to 2-5 and Examples 3-1 to 3-2 have a high proportion of liposomes having a single lamellar structure and are excellent in encapsulation rate of cosmetic ingredients. Moreover, it is clear that the liposomes contained in the liposome compositions obtained in Examples 2-1 to 2-5 and Examples 3-1 to 3-2 have a large average particle size and are excellent in encapsulation rate of the cosmetic ingredient. Furthermore, it is understood that the liposome compositions obtained in Examples 2-1 to 2-5 and Examples 3-1 to 3-2 have excellent stability over time.

[0088] 1 to 5, it can be seen that the liposome compositions obtained in the Examples have a higher ratio of liposomes having a single lamellar structure and are superior in encapsulation rate of cosmetic ingredients compared to the liposome compositions obtained in the Comparative Examples. Also, it can be seen from Fig. 5 that a large amount of disk particles, rather than liposomes, is contained.
