Description

Composition for improving skin condition containing a peptide complex as an active ingredient

[0001] [Cross-citation with related applications]

[0002] This application claims the benefit of priority from Korean Patent Application No. 10-2023-0098514, filed July 27, 2023, the entire contents of which are incorporated herein by reference.

[0003] [Technical Field]

[0004] The present invention relates to the use of a peptide complex for improving skin condition.

[0005] The skin, an organ that covers all of the body's organs and systems, protects the body from external invasion and plays a crucial role in maintaining homeostasis by conserving moisture and temperature within the body. Skin aging occurs naturally with age, but it can also be triggered by external factors such as UV rays and environmental pollution.

[0006] Meanwhile, advanced glycation end products (AGEs) are formed by the non-enzymatic reaction between reducing sugars, such as glucose, and proteins under conditions of high blood sugar. Specifically, the carbonyl group of the reducing sugar reacts with the free amino group of the protein, lysine or arginine, to form the initial glycation product, the Schiff base. The reaction progresses further to form Amadori products, and these formed compounds continuously undergo condensation, rearrangement, oxidation, cracking, and crosslinking processes to form stable and irreversible compounds, called advanced glycation end products (AGEs).

[0007] Advanced glycation end products (AGEs) are difficult to break down in the body and are known to damage organs and cells if left unbroken. In skin tissue, AGEs bind to collagen and elastin, inhibiting their turnover. This leads to tissue hardening and loss of elasticity, contributing to wrinkle formation. In addition, it is known that when advanced glycation end products bind to RAGE (Receptor for Advanced Glycation Endproducts), which is an advanced glycation end product receptor on the surface of skin cells such as keratinocytes and fibroblasts, it induces skin cell damage by promoting the production of reactive oxygen species (ROS), inflammatory response, and activation of cell death signaling pathways (Chen Cy, et al., 2022, Advanced Glycation End Products in the Skin: Molecular Mechanisms, Methods of Measurement, and Inhibitory Pathways. Front. Med. 9:837222. doi: 10.3389 / fmed.2022.837222).

[0008] Synthetic pharmaceuticals developed to inhibit AGE formation include aminoguanidine and pyridoxamine (Diabetes 2002 Sep;51(9):2826-32). However, these substances are toxic with long-term administration, posing safety and efficacy challenges when applied to the skin. Furthermore, various technologies are being developed for natural extracts that exhibit AGE-inhibiting activity and can improve skin damage (WO 2020 / 071630). However, these various natural extracts lack satisfactory activity and efficacy, making them difficult to develop into final products.

[0009] Accordingly, there is a need for the development of safe and highly active active substances to improve skin damage that may be caused by various factors as well as advanced glycation end products and their precursors.

[0168] Experimental Example 2: Measurement of the expression levels of apoptosis regulators, MMPs, and RAGE in skin cells.

[0169] 1. Experimental method

[0170] To evaluate the skin damage improvement effect of the peptide complex manufactured in Manufacturing Example 1, the expression levels of apoptosis regulators, MMPs, and RAGE in skin cells were confirmed through Western blot.

[0171] Specifically, 2x10 human skin keratinocyte cell line HaCaT keratinocytes and NIH3T3 fibroblasts 5The cells were seeded in 6-well plates at a density of 10 cells / well and cultured for 24 hours. After that, the peptide complex prepared in Manufacturing Example 1 was pretreated to the cells at concentrations of 50 μM, 100 μM, and 200 μM for 1 hour. [0173] 2. Observation of the effect of inhibiting skin cell death

[0174] To confirm the effect of the peptide complex on inhibiting skin cell death, the expression levels of apoptosis regulatory factors were measured, and the measurement results are shown in Figs. 4a, 4b, 5a, 5b, 6a, and 6b. Cleaved PARP-1, cleaved caspase-3, and cleaved caspase-9 are characteristic marker proteins that can confirm apoptosis, Bax is an apoptosis mediating protein, and Bcl-2 is an anti-apoptotic protein.

[0175] First, the expression levels of cleaved PARP-1, cleaved caspase-3, cleaved caspase-9, and Bax, which are apoptosis regulatory factors induced by AGE-BSA treatment in HaCaT keratinocytes, were measured, and the measurement results are shown in Figs. 4a and 4b. As shown in the negative control group in Figs. 4a and 4b, the expression levels of cleaved caspase-3, cleaved caspase-9, and Bax increased by AGE-BSA treatment, and it was confirmed that the increased expression levels of the apoptosis regulatory proteins were decreased by peptide complex treatment.

[0176] In addition, the expression levels of cleaved caspase-3, cleaved caspase-9, and Bcl-2, which are apoptosis regulatory factors whose expression levels were changed by MGO treatment in HaCaT keratinocytes, were measured, and the measurement results are shown in Figures 5a and 5b. As shown in the negative control group of Figures 5a and 5b, the expression level of cleaved caspase-3 increased by MGO treatment, the expression level of cleaved caspase-9 was not changed, and the expression level of Bcl-2, an anti-apoptotic factor, decreased. However, as shown in the experimental group, the expression levels of cleaved caspase-3 and caspase-9 decreased by treatment with the peptide complex. In addition, the expression level of Bcl-2, which was decreased by MGO treatment, was increased by treatment with the peptide complex.

[0177] The expression levels of cleaved PARP-1, cleaved caspase-3, cleaved caspase-9, Bax, and Bcl-2, which are apoptosis regulatory factors whose expression levels are changed by AGE-BSA treatment in NIH3T3 fibroblasts, were measured, and the measurement results are shown in Fig. 6a and Fig. 6b. As shown in the negative control group in Fig. 6a and Fig. 6b, the expression levels of cleaved PARP-1, cleaved caspase-3, cleaved caspase-9, and Bax increased by AGE-BSA treatment, while the expression level of Bcl-2, an anti-apoptotic factor, decreased. However, as shown in the experimental group, the expression levels of cleaved PARP-1, cleaved caspase-3, and Bax, which were increased by AGE-BSA treatment, were decreased by peptide complex treatment. In the case of Bcl-2, the expression level of Bcl-2, which was reduced in the negative control group, increased in a concentration-dependent manner by treatment with the peptide complex.

[0178] In summary, it was confirmed that the peptide complex of the present invention inhibits the expression of apoptosis-promoting factors induced by AGE-BSA or MGO in HaCaT keratinocytes and NIH3T3 fibroblast cells and promotes the expression of anti-apoptotic factors, thereby inhibiting cell death and thereby improving skin cell damage.

[0179] 3. Observation of the effect of inhibiting MMP expression

[0180] Matrix Metalloproteinases (MMPs) are proteolytic enzymes that specifically target collagen. MMP-1 and MMP-2 can degrade collagen, thereby reducing skin tissue elasticity and contributing to wrinkle formation. Therefore, we examined whether treating skin cells with the peptide complex of the present invention reduced the expression level of MMPs, and the results are shown in Figures 7a, 7b, 8a, 8b, 9a, and 9b.

[0181] First, we confirmed the expression levels of MMP-1 and MMP-2 induced by AGE-BSA treatment in HaCaT keratinocytes and the changes in the expression levels of MMP-1 and MMP-2 upon peptide complex treatment, and the measurement results are shown in Figures 7a and 7b. As shown in Figures 7a and 7b, the expression levels of MMP-1 and MMP-2 increased in the negative control group upon AGE-BSA treatment. However, as shown in the experimental group, the protein expression levels of MMP-1 and MMP-2 decreased upon treatment with the peptide complex.

[0182] In addition, we confirmed the increased expression levels of MMP-1 and MMP-2 by MGO treatment in HaCaT keratinocytes and the change in the expression levels of MMP-1 and MMP-2 when treated with a peptide complex, and the measurement results are shown in Figures 8a and 8b. As shown in Figures 8a and 8b, the expression levels of MMP-1 and MMP-2 increased by MGO treatment in the negative control group. However, as shown in the experimental group, when treated with a peptide complex, the protein expression levels of MMP-1 and MMP-2 decreased compared to the negative control group.

[0183] The expression level of MMP-1 increased by AGE-BSA treatment in NIH3T3 fibroblasts and the change in the expression level of MMP-1 when treated with a peptide complex were confirmed, and the measurement results are shown in Figures 9a and 9b. As shown in Figures 9a and 9b, the expression level of MMP-1 increased by treatment with AGE-BSA in the negative control group. However, as shown in the experimental group, when treated with the peptide complex, the protein expression level of MMP-1 decreased compared to the negative control group.

[0184] In summary, it was confirmed that the peptide complex of the present invention inhibits the expression of MMP-1 and MMP-2 increased by AGE-BSA or MGO treatment in HaCaT keratinocytes and NIH3T3 fibroblasts, thereby improving skin damage such as enhancing the elasticity of skin tissue and preventing wrinkle formation by reducing the degradation of collagen.

[0185] 4. Observation of the effect of suppressing RAGE expression

[0186] RAGE (Receptor for Advanced Glycation Endproducts) is a transmembrane receptor protein present on the cell surface to which advanced glycation end products can bind. It is known that when advanced glycation end products bind to RAGE in skin cells such as keratinocytes and fibroblasts, it can induce skin cell damage by inducing ROS production, inflammatory response, and activation of apoptotic signaling pathways within the skin cells. When the peptide complex of the present invention was treated to skin cells, it was confirmed whether the expression level of RAGE was reduced, and the results are shown in Figs. 10a, 10b, 11a, 11b, 12a, and 12b.

[0187] First, we examined the expression levels of RAGE induced by AGE-BSA treatment in HaCaT keratinocytes and the changes in RAGE expression levels induced by the peptide complex. As shown in Figures 10a and 10b, RAGE expression levels increased in the negative control group following AGE-BSA treatment. However, as shown in the experimental group, treatment with the peptide complex resulted in a concentration-dependent decrease in RAGE protein expression levels.

[0188] Furthermore, we examined the expression levels of RAGE induced by MGO treatment in HaCaT keratinocytes and the changes in RAGE expression levels induced by the peptide complex. As shown in Figures 11a and 11b, RAGE expression levels increased in the negative control group due to MGO treatment. However, as shown in the experimental group, when treated with the peptide complex, RAGE protein expression levels decreased compared to the negative control group at peptide complex concentrations of 100 μM or higher.

[0189] We examined the expression levels of RAGE induced by AGE-BSA treatment in NIH3T3 fibroblasts and the changes in RAGE expression levels induced by the peptide complex. As shown in Figures 12a and 12b, RAGE expression levels increased in the negative control group following AGE-BSA treatment. However, as shown in the experimental group, RAGE expression levels decreased compared to the negative control group when treated with the peptide complex.

[0190] In summary, we confirmed that the increased expression level of RAGE induced by AGE-BSA or MGO in HaCaT keratinocytes and NIH3T3 fibroblast cells, which are skin cells, was reduced by treatment with the peptide complex of the present invention. Through this, we confirmed that the peptide complex can improve skin damage by reducing the expression level of RAGE increased by AGE-BSA and MGO, thereby reducing the binding of advanced glycation end-products and suppressing ROS production and the activation of inflammatory response and apoptosis signaling pathways.

[0191] Experimental Example 3: Confirming the expression level of glyoxalase-1, which inhibits the production of advanced glycation end products.

[0192] Active dicarbonyl compounds, such as glyoxal, methylglyoxal, and 3-deoxyglucosone, can crosslink with proteins to form advanced glycation end products (AGEs). Glyoxalase-1 is a detoxifying enzyme that converts methylglyoxal to lactic acid, thereby eliminating it.

[0193] The effect of the peptide complex prepared in Example 1 on the expression level of glyoxalase-1 in HaCaT keratinocytes was evaluated using RT-PCR. First, human HaCaT keratinocytes were cultured at 2x10 5 The cells were seeded in 6-well plates at a density of 10 cells / well and cultured for 24 hours. Afterwards, the cells were pretreated with the peptide complex prepared in Manufacturing Example 1 at concentrations of 50 μM, 100 μM, and 200 μM for 1 hour. The pretreated cells were treated with 200 μM MGO and cultured in a CO2 incubator at 37°C for 24 hours. After washing twice with PBS, easy-BLUE TM Total RNA was isolated using (iNtRON, Cat. No.: 17061, Korea). After quantifying the isolated total RNA, cDNA was synthesized using an RT kit (enzynomics, Cat. No.: RT200, Korea), and PCR was performed using a PCR kit (enzynomics, Cat. No.: P581T, Korea). The primers used are shown in Table 4 below.

[0195] As a result, as shown in Figures 13a and 13b, the expression level of glyoxalase-1 decreased in the negative control group due to MGO treatment. However, as shown in the experimental group, it was confirmed that the expression level of glyoxalase-1 increased when the peptide complex was treated. Therefore, it was confirmed that the peptide complex can improve skin damage by inhibiting the production of advanced glycation end products by increasing the expression of glyoxalase-1, which was decreased by MGO treatment, and thereby enabling the decomposition of methylglyoxal, a precursor of advanced glycation end products.

[0196] In summary, these results indicate that treatment of skin cells with the peptide complex of the present invention reduces the expression of inflammatory response factors induced by advanced glycation end products (AGEs), exhibiting an anti-inflammatory effect, and inhibiting cell death by reducing the expression of apoptotic factors. In addition, it can be seen that the expression of collagenase is reduced, thereby enhancing skin tissue elasticity and preventing wrinkle formation. In addition, it was confirmed that treatment with the peptide complex of the present invention reduces the expression of RAGE, which is an AGE receptor, thereby reducing the binding of AGEs, thereby inhibiting ROS production and the activation of inflammatory response and apoptotic signaling pathways, thereby improving skin damage. It was confirmed that the peptide complex of the present invention can improve skin damage by inhibiting the production of AGEs by increasing the expression level of glyoxalase-1, which was reduced by MGO, and decomposing methylglyoxal, a precursor of AGEs.

[0197] Although representative embodiments of the present invention have been described above as examples, the scope of the present invention is not limited to the specific embodiments described above, and those skilled in the art will be able to make appropriate changes within the scope described in the claims of the present application.
