Abstract
Description
Cosmetic composition of solubilized formulation containing bio-derived isosorbide and fermented ceramide
[0001] This application relates to a cosmetic composition comprising a solubilized formulation containing bio-derived isosorbide and fermented ceramide. This application claims priority to Republic of Korea Patent Application No. 10-2023-0097033, filed with the Korean Intellectual Property Office on July 25, 2023, the disclosure of which is incorporated herein by reference.
[0002] Ceramides are the main lipids that make up the lamellar structure of intercellular lipids in the skin, and are known to play an essential role in the skin's permeability barrier function. A decrease in ceramides in the skin can lead to decreased moisture content, increased moisture loss, and stratum corneum exfoliation, which can lead to a damaged or unhealthy skin barrier. These symptoms can be improved by supplying ceramides to the skin, and ceramides are utilized as effective ingredients for improving skin condition and enhancing moisturizing effects. However, ceramides are poorly soluble substances with a high tendency to crystallize. Despite their excellent efficacy, as mentioned above, their application in cosmetics is difficult due to formulation stability issues. Specifically, gelation, a phenomenon in which ceramides precipitate, frequently occurs during the formulation process. In addition, because its solubility in purified water, the most basic cosmetic ingredient, is very low, its use has been limited to cosmetic preparations with a certain degree of viscosity, such as lotions and creams, and its application to transparent, soluble cosmetics, such as toners and skins, is limited.
[0003] Meanwhile, cosmetic solubilizing formulations can be categorized into oil-in-water, water-in-oil, and aqueous dispersions. While aqueous dispersions contain only polyols and ionic components that readily dissolve in water, water-in-oil formulations encapsulate aqueous components in the oil phase, and oil-in-water formulations encapsulate oil components in the aqueous phase. These formulations often present challenges, such as stability over time. Consequently, many cosmetic compositions for solubilizing products utilize aqueous dispersions, meeting customer needs as products without stability issues over time. However, these solubilizing compositions, which consist solely of aqueous components, have the disadvantage of low affinity and adsorption with the skin's lipid layer. Therefore, many researchers have been developing technologies for encapsulating oil components in solubilizing formulations in the cosmetics and pharmaceutical industries. The most representative methods include chemical and physical methods. Chemical methods include a method that completely solubilizes oil components using a large amount of surfactant, but this method can cause skin irritation due to the large amount of surfactant, and can cause skin diseases such as atopy by reducing the skin barrier or skin flora.
[0004] Under this technological background, there is a need for the development of a solubilized formulation that includes ceramide, a soluble ingredient, but is transparent and has improved usability while maintaining formulation stability (Korean Patent Publication No. 2023-0050156), but this is still insufficient.
[0005] The present inventors have conducted extensive research to solve the problems of the prior art and have confirmed that by using isosorbide, the solubility of ceramide in purified water can be increased, and a transparent type of soluble cosmetic such as a toner can be manufactured, and that it can provide the user with an excellent feeling of use and an improved moisturizing sensation, and have completed the present invention based on this.
[0006] Accordingly, one aspect is to provide a cosmetic composition in the form of a solubilized formulation comprising ceramide and isosorbide.
[0007] Other purposes and advantages of this application will be further clarified by the detailed description below, taken in conjunction with the appended claims and drawings. Any details not described herein will be readily apparent and inferred by those skilled in the technical field of this application or similar technical fields, and therefore, their description will be omitted.
[0008] Each description and embodiment disclosed in this application may also be applied to each other description and embodiment. That is, all combinations of the various elements disclosed in this application fall within the scope of this application. Furthermore, the scope of this application is not limited by the specific descriptions described below.
[0009] One aspect is an oil-in-water (O / W) type cosmetic composition comprising an oil phase comprising ceramide; and an aqueous phase comprising isosorbide.
[0010] A cosmetic composition is provided, wherein the isosorbide is included in an amount of 6 to 43 wt% based on the total weight of the cosmetic composition.
[0011] As used herein, the term "isosorbide" refers to an anhydrous sugar alcohol in the form of a diol obtained by dehydration of sorbitol, a sugar alcohol having six carbon atoms. The isosorbide may be obtained naturally or artificially. For example, if it is obtained naturally, isosorbide can be synthesized through a dehydration reaction via an intermediate product of sorbitol by hydrogenating D-glucose from corn starch under a catalyst, but is not limited thereto.
[0012] As used herein, the term "ceramide" refers to an essential component for the formation of the structure or function of the stratum corneum as an intercellular lipid that fills the space between keratinocytes with N-acylsphingosine. The ceramide may include not only natural ceramide but also a substance that acts as a skin barrier, such as natural ceramide, and the ceramide may be at least one selected from the group consisting of, for example, ceramide EOP, ceramide NS, ceramide NP, ceramide AS, ceramide AP, ceramide EOS, and ceramide NDS. As the content of the ceramide increases, the skin moisturizing effect of the cosmetic composition increases and the skin may become smoother.
[0013] In one specific embodiment, the cosmetic composition may be in the form of an emulsion formulation.
[0014] As used herein, the term "emulsion" refers to a type of colloid formed by combining two immiscible liquids, for example, an oil phase and an aqueous phase, and may be used interchangeably with the term emulsion. The emulsion formulation may be an oil-in-water formulation composed of an aqueous dispersed continuous phase and an oily dispersed discontinuous phase, or a water-in-oil formulation composed of an oily dispersed continuous phase and a water-dispersed discontinuous phase. A solvent, a solubilizer, or an emulsifier may be used as a carrier component for the emulsion formulation, and may include, for example, water, ethanol, isopropanol, ethyl carbonate, ethyl acetate, benzyl alcohol, benzyl benzoate, propylene glycol, 1,3-butylglycol oil, aliphatic esters, polyethylene glycol, or fatty acid esters of sorbitan.
[0015] In one specific example, the isosorbide may be included in an amount of 6 to 43 wt% based on the total weight of the cosmetic composition. If the content of isosorbide is too low outside the above range, the ceramides in the composition cannot be completely dissolved, resulting in layer separation between the oil phase and the water phase. Conversely, if the content of isosorbide is excessively high, layer separation may also occur as described above. For example, the isosorbide may be included in an amount of 6 to 43 wt%, 6 to 40 wt%, 6 to 35 wt%, 6 to 30 wt%, 6 to 25 wt%, 6 to 20 wt%, 6 to 15 wt%, 6 to 10 wt%, 8 to 43 wt%, 8 to 40 wt%, 8 to 35 wt%, 8 to 30 wt%, 8 to 25 wt%, 8 to 20 wt%, 8 to 15 wt%, 8 to 10 wt%, 10 to 43 wt%, 10 to 40 wt%, 10 to 35 wt%, 10 to 30 wt%, 10 to 25 wt%, 10 to 20 wt%, or 10 to 15 wt% based on the total weight of the cosmetic composition, but is not limited thereto.
[0016] According to one embodiment, a cosmetic composition in the form of an oil-in-water emulsion was prepared by preparing an oil phase comprising ceramide and an aqueous phase comprising isosorbide, and then adding and mixing the oil phase to the aqueous phase. It was confirmed that the cosmetic composition contained isosorbide, and thus the layers of the aqueous phase and the oil phase did not separate, and crystallized ceramide did not precipitate.
[0017] In one specific example, the ceramide may be included in an amount of 2 to 8 wt% based on the total weight of the cosmetic composition. If the content of ceramide is too low outside the above range, the skin moisturizing effect may not be sufficient, and conversely, if the content of ceramide is excessively high, a gelling phenomenon in which crystallized ceramide is precipitated within the formulation may occur. For example, the ceramide may be included in an amount of 2 to 8 wt%, 2 to 7 wt%, 2 to 6 wt%, 3 to 8 wt%, 3 to 7 wt%, or 3 to 6 wt% based on the total weight of the cosmetic composition, but is not limited thereto, and the range may be appropriately changed within a range in which effective efficacy can be exhibited within the cosmetic composition.
[0018] In one specific example, the cosmetic composition may be diluted by further adding purified water and may have a solubilized formulation.
[0019] The term "solubilization formulation" used herein may refer to an oil-in-water (O / W) type composition in which the solubility of an oily ingredient is enhanced. As an example, the cosmetic composition of the solubilization formulation may be provided in the form of a transparent solubilization cosmetic, such as a toner, essence, serum, pack, ampoule, or immersion sheet mask. The solubilization formulation has the advantage of being much lighter and more refreshing, and providing an immediate moisturizing feeling. However, since ceramide is a representative poorly soluble substance, there are many limitations in producing a cosmetic of the solubilization formulation, and the gelling phenomenon in which ceramide crystallizes and precipitates acts as a factor that lowers the stability of the solubilization agent during the formulation process.
[0020] The term "solubilization" as used herein refers to the ability of a small amount of an oily component to be transparently dissolved in a large amount of an aqueous component, and for the purpose of the invention, may indicate the degree of solubility of ceramide in purified water. The solubilized formulation or formulation with increased solubility may be prepared by, but is not limited to, a method of pre-mixing a solubilizing agent and a substance to be solubilized and diluting the mixture by adding a dispersion medium, or a method of dissolving a solubilizing agent in a dispersion medium and adding a substance to be solubilized thereto.
[0021] According to one embodiment, the cosmetic composition can include a high content of ceramide while maintaining formulation stability and transparency by including isosorbide together with the useful ingredient ceramide, and can also provide the user with an excellent feeling of use and an enhanced moisturizing sensation. Therefore, the cosmetic composition according to one embodiment has increased solubility of ceramide in purified water, and can be applied to the manufacture of transparent, solubilized cosmetics such as toners.
[0022] In one specific embodiment, the ceramide may be included in an amount of 0.01 to 0.04 wt% based on the total weight of the cosmetic composition. For example, the ceramide may be included in an amount of 0.01 to 0.04 wt%, 0.01 to 0.035 wt%, 0.01 to 0.03 wt%, 0.01 to 0.025 wt%, 0.01 to 0.02 wt%, 0.01 to 0.015 wt%, 0.02 to 0.04 wt%, 0.02 to 0.035 wt%, 0.02 to 0.03 wt%, 0.02 to 0.025 wt%, 0.03 to 0.04 wt%, or 0.03 to 0.035 wt% based on the total weight of the cosmetic composition. If the ceramide content is too low outside the above range, the skin moisturizing effect may not be sufficient, and conversely, if the ceramide content is high, there is a concern that transparency may be reduced and formulation stability may be reduced.
[0023] In one specific example, the isosorbide may be included in an amount of 0.04 to 0.2 wt% based on the weight of the cosmetic composition. For example, the isosorbide may be included in an amount of 0.04 to 0.2 wt%, 0.04 to 0.15 wt%, 0.04 to 0.1 wt%, 0.1 to 0.2 wt%, 0.1 to 0.15 wt%, or 0.15 to 0.2 wt% based on the weight of the cosmetic composition. If the content of isosorbide is too low outside the above range, the ceramide in the composition may not be completely dissolved, resulting in crystallization of the ceramide, which may cause problems with transparency and formulation stability. Conversely, if the content of isosorbide is excessively high, the ceramide may precipitate.
[0024] In one specific example, a cosmetic composition may be provided comprising 0.015 to 0.03 wt% of ceramide and 0.052 to 0.16 wt% of isosorbide based on the total weight of the cosmetic composition. If the content of isosorbide is too low or too high relative to ceramide outside the above range, the effect of promoting the solubility of ceramide in purified water may be inhibited, thereby reducing formulation stability and transparency, which may result in a deterioration of the user's feeling of use.
[0025] In one specific example, the cosmetic composition may be prepared by diluting an oil-in-water emulsifier containing an aqueous phase containing isosorbide and an emulsifier, and an oil phase containing ceramide, with purified water.
[0026] As used herein, the term “emulsifier” may mean a substance that can form a stable emulsion of two liquids that do not mix with each other. [0027] In one specific embodiment, the emulsifier may be included in an amount of 0.001 to 0.1 wt% based on the total weight of the cosmetic composition. For example, the emulsifier may be included in an amount of 0.001 to 0.1 wt%, 0.001 to 0.05 wt%, 0.001 to 0.01 wt%, 0.001 to 0.005 wt%, 0.005 to 0.1 wt%, 0.005 to 0.05 wt%, 0.005 to 0.01 wt%, 0.01 to 0.1 wt%, or 0.01 to 0.05 wt% based on the total weight of the cosmetic composition, but is not limited thereto.

[0062] A cosmetic composition in the form of a solubilized formulation containing isosorbide and ceramide was prepared. Specifically, an oil-in-water (O / W) type emulsifying raw material was prepared by mixing an aqueous phase and an oil phase, and then an appropriate amount of purified water was added thereto to prepare a cosmetic composition in the form of a solubilized formulation.
[0063] The components and contents of each of the water and oil phases are as shown in Table 1 below. In addition, the isosorbide (NOVASORB®) was obtained from Samyang Innochem and used (purity 99.8%, pH 5.6).
[0064] [Table 1]
[0065]
[0066] Specifically, an appropriate amount of purified water and a polyol moisturizer were added to isosorbide, stirred while heating to 75℃, and Ethylenediaminetetraacetic Acid, trisodium Salt (EDTA 3Na) was mixed and dissolved therein to prepare an aqueous phase. Meanwhile, oil, lecithin, ceramide, and a preservative were mixed, stirred, and dissolved while heating to separately prepare an oil phase. Thereafter, the prepared oil phase was added to the aqueous phase, emulsified using a homogenizer at 4,500 rpm for 5 minutes, and homogenized three times under the condition of 1,000 bar using a microfluidizer, thereby preparing an oil-in-water emulsified raw material containing ceramide and isosorbide.
[0067] Thereafter, the above-mentioned water-in-oil emulsifying raw material was diluted to 0.5% with purified water to prepare a cosmetic composition in the form of a solubilized formulation having the ingredients and contents of Table 2.

[0085] Experimental Example 1. Stability of the solubilized formulation with or without isosorbide
[0086] In this experimental example, the effect of isosorbide on the stability of solubilized formulations was investigated. To this end, the stability of cosmetic compositions manufactured using the oil-in-water emulsifier raw materials of Example 1 and Comparative Example 1 as solubilized formulations was visually evaluated.
[0087] Fig. 1 shows the results of visual inspection of the cosmetic composition of Example 1, and Fig. 2 shows the results of visual inspection of the cosmetic composition of Comparative Example 1 that does not contain isosorbide. As a result, as shown in Figs. 1 and 2, in the cosmetic composition according to one embodiment, ceramide crystals were not precipitated, and thus, it was confirmed that it can be applied as a solubilized formulation. On the other hand, in the case of Comparative Example 1 that does not contain isosorbide, the phenomenon of ceramide, which is a poorly soluble substance, crystallizing in the oil-in-water emulsifying raw material progressed, and therefore, it was confirmed that there is a limit to applying it as a solubilized formulation in the cosmetic composition.
[0088]
[0089] Experimental Example 2. Confirmation of Formulation Stability According to Isosorbide Content
[0090] In this experimental example, the effect of the isosorbide content level on the formulation stability of a cosmetic composition was examined. To this end, the formation of a solubilized formulation and the transparency or turbidity level of the cosmetic compositions of Examples 1 to 5 and Comparative Examples 2 to 4 having different isosorbide contents were evaluated. Here, the transparency or turbidity level was evaluated by transferring each cosmetic composition to a transparent container, placing the transparent container on a plate printed with letters or lines, and visually checking the letters or lines reflected through the manufactured cosmetic composition.
[0091] Figures 3 to 6 are the results of visual inspection of the cosmetic compositions of Examples 1 to 3 and Example 5, and Figures 7 to 9 are the results of visual inspection of the cosmetic compositions of Comparative Examples 2 to 4 containing isosorbide having various contents.
[0092] As a result, as shown in FIGS. 3 to 6, it was confirmed that the cosmetic composition according to one embodiment is a highly transparent solubilizing agent, and that the stability of the formulation is maintained even when a high content of ceramide is included. On the other hand, as shown in FIGS. 7 to 9, when the content of isosorbide is excessively low or high, a layer separation phenomenon occurs, or, as in Comparative Example 1, ceramide crystallizes, resulting in a decrease in formulation stability or a significant decrease in transparency, which limits its application to products such as skin, toner, and lotion. The above experimental results indicate that the stability and transparency level as a solubilizing formulation can be achieved by isosorbide in a specific content range.

[0094] Experimental Example 3. Usability Evaluation
[0095] In this experimental example, the effect of isosorbide on improving the usability of a cosmetic composition was investigated. To this end, a total of 15 adults were selected as test subjects, and the cosmetic composition (skin) shown in Table 7 below was applied to the back of the hand. The usability was measured using a 5-point preference scale (5: very good, 4: good, 3: average, 2: bad, 1: very bad) for a total of 5 items: moisture, spreadability, oiliness, stickiness, and residual feeling. The average value for each item was calculated, and the results are shown in Table 8 below.
[0096] [Table 7]
[0097]
[0098] [Table 8]
[0099]
[0100] As a result, as shown in Table 8 above, the ceramide skin containing isosorbide had improved moisture, spreadability, and residual feeling compared to the skin not containing isosorbide, while the stickiness that has a negative impact on the user was reduced. In particular, it was confirmed that the moisture and spreadability that provide a light, refreshing, and immediate moisturizing feeling, which are characteristics of a solubilized formulation, were significantly improved. The above experimental results indicate that the cosmetic composition according to one embodiment can improve the usability and usability as a moisturizing cosmetic composition by including isosorbide.
[0101] In summary of the above results, a cosmetic composition of a solubilized formulation according to one embodiment including isosorbide can contain a high content of ceramide while maintaining formulation stability and high transparency, and can provide a user with an excellent feeling of use and an enhanced feeling of moisture retention, thereby improving its utility as a cosmetic composition such as a skin, and in particular, as a moisturizing cosmetic.
