Description

Method for improving ginsenoside content of ginseng (PANAX GINSENG) containing silver colloidal solvent and ginseng composition with increased ginsenoside content

[0001] The present invention relates to a method for improving the ginsenoside content of ginseng including a silver colloidal solvent and a ginseng composition having increased ginsenoside content.

[0002] Ginseng (Panax ginseng) is a perennial shade-loving herbaceous plant belonging to the genus Panax in the family Araliaceae. It has long been used as an important medicinal ingredient in oriental medicine. Ginseng is generally classified as white ginseng and red ginseng depending on the processing method. White ginseng is raw ginseng dug up from the field and dried as is, while red ginseng is fresh ginseng that is steamed and dried. During the manufacturing process, various chemical changes such as saponin modification, amino acid change, and browning occur. Since ginseng contains saponin, it can be expected to have excellent pharmacological effects such as cancer prevention, inhibition of cancer cell growth, lowering blood pressure, protection of brain nerve cells, and improvement of learning ability.

[0003] Ginseng saponins are called ginsenosides, which are glycosides isolated from ginseng to distinguish them from other plant saponins. They are usually named ginsenoside-Rx, etc. (abbreviated as G-Rx). Here, "R" means Radix or Root. In addition, "x" is named in order from bottom to top according to the migration distance (Rf value) of the spot that appears on the TLC, such as o, a, b, c, d, e, f, g, h. The types of ginsenosides currently known to be contained in ginseng include Rg1, Rb1, Rg2, Rc, and Rd.

[0004] Korean Patent No. 10-0464901 discloses a bactericidal trace element fertilizer containing silver colloid particles, Korean Patent Publication No. 2016-0086149 discloses a composition containing a ginseng extract with increased ginsenoside content as an active ingredient, and Korean Patent No. 10-1603169 discloses a method for producing a ginseng extract with increased acidic polysaccharide and ginsenoside content. However, no method of using a silver colloid solvent to increase the content of ginsenoside according to the present invention has been disclosed.

[0005] Regarding the effects of Ag-NP (silver nano-particle) fertilization on plants, antibacterial action against soil pathogenic bacteria, plant growth promotion, and plant secondary metabolism enhancement effects have been reported in some plants, and the possibility of using silver colloidal solvents in agriculture has been reported, but there are not many examples of application methods.

[0006] The present invention aims to provide a method of utilizing a silver colloidal solvent to improve the ginseng ginsenoside content by analyzing the effect of applying a silver colloidal solvent having a different composition to ginseng on the growth and differentiation of ginseng and analyzing the change in the ginsenoside content in ginseng roots.

[0062] <Experimental Example 1> Growth-differentiation analysis

[0063] In order to investigate the effects of silver colloidal solvents Zeroxxe (Zx), Eunbi (En), Variant 1 (Silver Grass 1), Variant 2 (Silver Grass 2), and Variant 3 (Silver Grass 3) manufactured in Examples 1 to 5, 6-year-old ginseng plants grown in the open field were evenly fertilized twice at one-week intervals at a concentration of 10 ppm, and then growth information regarding crown diameter, stem length, leaf length, leaf width, number of leaves, root diameter, and root length was investigated.

[0064] Figures 1 to 7 show the results of comparative analysis of growth information of 6-year-old phosphoric acid plants according to treatment with silver colloid products manufactured in Examples 1 to 5.

[0065] As a result, the leaf tissue growth of 6-year-old ginseng was not significantly affected by the two-time application of 'silver' colloidal solvent (10 ppm) of Examples 1 to 5, but it was confirmed that there was a slight difference in the diameter and length of ginseng roots, but the difference was not statistically significant. However, it was found that the root length significantly increased in Example 1 (Zeroxxe (Zx)), Example 2 (Eunbi (En)), and Example 3 (Variant 1 (Eunjandi 1)).

[0066] <Experimental Example 2> Analysis of ginseng ginsenosides

[0067] After comparing and analyzing ginseng growth information, differences were observed in the length and diameter of ginseng roots. After treatment for 18 hours in a 10 ppm concentration solution of three types of silver colloids (En, Zx, V1), ginsenoside metabolism analysis was performed.

[0068] Specifically, among the 'silver' colloidal solvents of Examples 1 to 5, Example 1 (Zeroxxe (Zx)), Example 2 (Eunbi (En)), and Example 3 (Variant 1 (Eunjandi 1)) that showed differences in root length were selected, and quantitative analysis was conducted on a total of 10 well-known ginsenoside substances, including 5 substances (Rb1, Rb2, Rb3, Rc, Rd) from the protopanaxadiol (PPD; eg, Ra, Rb, Rc, Rd, Rg3, Rh2) series and 5 substances (Re, Rf, Rg1, Rg2, Rh) from the protopanaxatriol (PPT; eg, Re, Rf, Rg1, Rg2, Rh1) series.

[0069] Ginseng root samples treated with liquid nitrogen were finely ground and extracted and purified by adding 10 ml of 50% MeOH per 1 g. After extraction, the samples were analyzed using a high-performance liquid chromatography (HPLC) Agilent 110 series, Waters Symmetry C18 column at a flow rate of 1 ml / min under gradient conditions using a UV detector (203 nm).

[0070] Figure 8 shows the 10 major ginsenosides in the V1 treatment group and unknown substances A to N that increased compared to the control group, Figure 9 shows the 10 major ginsenosides in the En treatment group and unknown substances A to N that increased compared to the control group, and Figure 10 shows the 10 major ginsenosides in the Zx treatment group and unknown substances A to N that increased compared to the control group.

[0071] As a result of analyzing 10 major ginsenosides in the roots of 3-year-old ginseng grown in open fields treated with silver colloid in Examples 1 to 3, unknown metabolites that were different from the control group in addition to the 10 major ginsenosides were identified in the chromatogram results for the analysis of ginsenosides in the roots of 3-year-old ginseng treated with silver colloid in Examples 1 to 3.

[0072] Among the identified unknown metabolites (A to N), the substance corresponding to D was identified only in the Zx treatment and not in En and V1, and all A to N substances except D were found to be enhanced in the silver colloid treatment group.

[0073] Figure 11 shows the results of analyzing 10 types of ginsenosides in silver colloid-treated ginseng roots of Examples 1 to 3, Figure 12 shows the results of comparative analysis of protopanaxatriol (PPT) series ginsenosides, and Figure 13 shows the results of comparative analysis of protopanaxadiol (PPD) series ginsenosides.

[0074] As a result of the analysis, it was confirmed that Re, Rg1, Rf, Rb1, Rg2, Rh1, and Rc increased significantly (p<0.001) in the V1 treatment group, and that Rb2 and Rd increased in content at the P<0.05 level.

[0075] Within the En treatment group, Rg1, Rf, and Rc were significantly increased (P<0.001), and Rb1 and Rh1 were significantly increased (P<0.01). Re and Rg2 were significantly decreased (P<0.001).

[0076] Within the Zx treatment group, Rg1, Rf, Rb1, Rg2, Rh1, Rc, Rb2, and Rd were significantly increased (P<0.001), and Rb3 content increased at a level of P<0.01. Re content was confirmed to decrease at a level of P<0.01.

[0077] As shown in Figure 14, the total content of 10 types of ginsenoside was in the order of Zx > V1 > En, and since En was not statistically significant, it was finally confirmed that Zx and V1 treatment had a significant effect on promoting ginsenoside metabolism in ginseng roots.

[0078] In conclusion, it was confirmed that the presence or absence of fertilizer components (N, P, K) in silver colloidal solvent (10 ppm) and the presence or absence of silver particle stabilizer did not significantly affect the aerial growth of ginseng, but there was a slight difference in root growth.

[0079] In addition, the presence or absence of fertilizer components (N, P, K) in silver colloid solvent (10 ppm) did not significantly affect the increase in the total content of ginsenosides in ginseng root tissues, but the Rf, Rg1, and Rb1 substances were confirmed to be increased, and there was no significant difference between the conditions with or without stabilizers, confirming the efficacy of increasing the content of all 10 types of ginsenosides by silver grass (prototype) and Zeroxxe fertilization.

[0080]

[0081] According to the method using the silver colloid solvent of the present invention, it was confirmed that it is possible to increase the ginsenoside content with only two fertilizations at a low concentration of 10 ppm at the harvest time without having to fertilize consistently for six years.
