AI Technical Summary
Current methods for producing pomegranate juice are hindered by issues such as sediment formation, turbidity, and discoloration, especially during storage and temperature changes, which existing techniques like gelatin use and ultrafiltration fail to effectively address.
The method involves adding the rubisco enzyme to pomegranate juice, causing hydrolyzable and condensed tannins to form a precipitate, which is then removed by filtration, resulting in clear and sediment-free juice.
This approach effectively eliminates tannin-induced turbidity, sediment, and astringency, reduces the clarification process from 16 hours to 1 hour, and eliminates the need for gelatin and graining equipment, making the process more efficient and cost-effective.

Abstract
Description
[0001] CLEAR AND SEDIMENT-FREE POMEGRANATE JUICE PRODUCTION METHOD
[0002] Technical Field
[0003] The invention relates to a method for producing clear and sediment-free pomegranate juice containing rubisco enzyme.
[0004] Prior art
[0005] In the literature, the production of high quality pomegranate juice is one of the most challenging issues for the juice industry. Although many fruit juices can be produced with the desired quality, unfortunately, to date, no method has been found to eliminate all defects for pomegranate juice. Although the processes applied during the production of pomegranate juice provide instant “high quality pomegranate juice production”, high sediment formation, turbidity and discoloration often occur in pomegranate juices, especially during storage and exposure to temperature differences. Pomegranate juice producers generally use “gelatin use” and “ultrafiltration” to remove defects in pomegranate juice. However, neither gelatin nor ultrafiltration is successful in this regard.
[0006] In the technical field, solutions for preventing turbidity in beverages have been identified. Accordingly, the invention EP1511835 relates to a method for preventing or reducing turbidity in beverages. The invention discloses a solution for an improved method for preventing or reducing cloudiness in beverages comprising the use of rubisco, which provides sequence information of a genomic DNA, cDNA as well as protein sequences. Furthermore, the invention discloses a method for removing turbidity formation using an “endoprotease and auxiliary enzyme”. Here, in the invention, the rubisco enzyme is not used for clarification. It is reported that it can only be a promoter specific to the gene encoding the polypeptide. The invention does not provide for the formation of a precipitate with rubisco and removal of the precipitate from pomegranate juice by filtration.
[0007] In conclusion, all the problems mentioned above have made it necessary to make an innovation in the relevant technical field. Objectives of the Invention
[0008] The present invention relates to a method of producing clear and sediment-free pomegranate juice in order to eliminate the above-mentioned disadvantages and to bring new advantages to the relevant technical field.
[0009] One object of the invention is to produce a clear and sediment-free pomegranate juice in which the effect of tannins, which cause turbidity, sediment and astringency in pomegranate juice as a result of the addition of rubisco, is eliminated.
[0010] Brief Description of the Invention
[0011] In order to fulfill all the aforementioned objects and those which will arise from the detailed description below, the present invention is a method of producing clear and sediment-free pomegranate juice comprising the addition of rubisco enzyme (12). The invention comprises a first step (10) of adding rubisco enzyme (12) to pomegranate juice (16) in a production tank (14), wherein hydrolyzable and condensed tannins (18) in the pomegranate juice form a precipitate with the rubisco enzyme (20); removing the precipitate (24) at the bottom of the tank (26) from the pomegranate juice (26) by filtration (28); removing the precipitate (24) at the bottom of the tank without moving the pomegranate juice (30) and performing standard juice filtration process (32) in a second stage (22) following the first stage processes.
[0012] Thus, as a result of the addition of rubisco, clear and sediment-free pomegranate juice can be produced in which the effect of tannins causing turbidity, sediment and astringency in pomegranate juice is eliminated. As the advantages of the invention, since tannins (hydrolyzable and condensed tannins), which are the substrate of turbidity, sediment formation and astringency, will be removed from pomegranate juice with rubisco, these defects of pomegranate juice are eliminated; the clarification process, which generally takes 16 hours, is reduced to a short period of 1 hour; consumer concerns about whether the gelatin frequently used in the clarification of pomegranate juice is halal or not due to its source are eliminated; and there is no need for graining equipment frequently used in pomegranate juice production. It is also very easy to apply the invention to existing technologies. “Rubisco” will be used instead of the currently used clarification auxiliaries. Rubisco is present in all organisms from prokaryotes (photosynthetic and chemoautotrophic bacteria, cyanobacteria and archaea) to eukaryotes (various algae and higher plants). It is therefore cheaply available due to its availability in multiple sources. In addition, thanks to the invention, the graining process, which is frequently used in current pomegranate juice production, will not be necessary with the invention. In this case, the additional cost that the industrialist will incur for the graining equipment will be eliminated and the significant amount of energy spent for the graining process will be saved. In addition, with the adoption of this clarification method by the industry, rubisco pomegranate juice can be produced and used by the producer himself. If the proposed clarification method is adopted by the industry, there is no increase or problem with the cost of the method.
[0013] One possible embodiment of the invention is characterized in that after the rubisco enzyme is added to the pomegranate juice, it is incubated at a temperature between 1°C and 10°C and for a period of 30 minutes to 150 minutes. Thus, at the end of this incubation, all tannin- induced precipitate is formed during storage.
[0014] Brief Description of the figures
[0015] Figure 1 shows a schematic representation of the flow chart of the inventive clear and sediment-free pomegranate juice production method.
[0016] Detailed Description of the Invention
[0017] In this detailed description, the inventive clear and sediment-free pomegranate juice production method is explained only with examples that will not have any limiting effect for a better understanding of the subject matter.
[0018] Figure 1 shows schematically the flow chart of the inventive clear and sediment-free pomegranate juice production method. The clear and sediment-free pomegranate juice production method is a method comprising the addition of rubisco enzyme (ribulose-1,5- bisphosphate carboxylase / oxygenase) (12). The inventive clear and sediment-free pomegranate juice production method comprises the following process steps;
[0019] - in a first step (10), the rubisco enzyme (12) is added (16) to the pomegranate juice in a production tank (14).
[0020] - hydrolyzable and condensed tannins (18) in the pomegranate juice are precipitated (20) by the rubisco enzyme. - in a second stage (22) following the first stage operations, the precipitate (24) at the bottom of the tank formed in the first stage (10) is removed from the pomegranate juice (26) by filtration (28).
[0021] - Without moving the precipitate (24) at the bottom of the tank, the pomegranate juice is removed (30) and standard juice filtration is performed (32).
[0022] In one embodiment of the invention, after adding rubisco enzyme to the pomegranate juice, incubation is carried out at a temperature between 1°C and 10°C, preferably at 2°C and for a period between 30 minutes and 150 minutes, preferably for 1 hour.
