[0009] Research Period: July 1, 2024 - September 23, 2024

[0010] Stem cells are undifferentiated cells that can be obtained from various tissues of embryos and adults, and have characteristics such as self-renewal, differentiation, and immortality. Stem cells can be divided into embryonic stem cells and adult stem cells depending on the tissue of origin. While therapeutic experiments using embryonic stem cells are difficult due to ethical issues and the possibility of tumor formation, adult stem cells have the advantage of being easily obtained from various tissues, and active research is being conducted to apply them to the treatment of various diseases.

[0011] Among stem cells, multipotent mesenchymal stem cells (MSCs) are a component of the tissue matrix of all adult organs, likely located in perivascular regions, where they play a crucial role in tissue homeostasis, surveillance, repair, and remodeling. Typical immunophenotypic profiles of MSCs include the expression of the surface markers CD44, CD73, CD90, and CD105, and the absence of CD34, CD45, CD14, and HLA-DR. MSCs possess multilineage differentiation potential into osteocytes, adipocytes, and chondrocytes.

[0012] Mesenchymal stem cells are known to suppress inflammation, induce the production of regulatory T cells (Treg), or induce the death of immune cells involved in apoptosis, and various therapeutic agents using them are being actively developed.

[0013] The properties of mesenchymal stem cells are influenced by biological, biochemical, and biophysical factors both in vivo and in vitro, driven by interactions between cells, the extracellular matrix (ECM), and soluble bioactive factors. Mesenchymal stem cells interact with surrounding tissues and cells in a three-dimensional space, modulating the ECM to promote angiogenesis, produce anti-inflammatory molecules, prevent cell death (anti-apoptotic effects), and modulate the immune system.

[0014] Accordingly, recent research has been actively conducted to enhance the lineage differentiation and function of mesenchymal stem cells by controlling biological, biochemical, and / or biophysical factors to improve the therapeutic potential of mesenchymal stem cells. One known method is the technique of priming stem cells using proinflammatory mediators. Representative priming techniques include (a) MSC priming using inflammatory cytokines or mediators, (b) MSC priming using hypoxia, (c) MSC priming using pharmacological drugs and chemicals, (d) MSC priming using biomaterials and other culture conditions, and (e) MSC priming with other molecules.

[0015] One object of the present invention is to provide a priming composition capable of enhancing the immunomodulatory activity or regenerative activity of stem cells.

[0016] Another object of the present invention is to provide stem cells or a culture solution derived from the stem cells with enhanced immunomodulatory activity or regenerative activity through treatment with the above-described composition.

[0017] Another object of the present invention is to provide various applications of stem cells or stem cell-derived culture fluids having enhanced immunomodulatory activity or regenerative activity through treatment with the above-described composition.

[0018] However, the technical problems to be solved by the present invention are not limited to the problems mentioned above, and other problems not mentioned can be clearly understood by those skilled in the art from the description below.

[0019] Hereinafter, various embodiments described herein will be described with reference to the drawings. In the following description, various specific details, such as specific configurations, compositions, and processes, are set forth to provide a thorough understanding of the present invention. However, certain embodiments may be practiced without one or more of these specific details, or in conjunction with other known methods and configurations. In other instances, well-known processes and manufacturing techniques have not been described in specific detail so as not to unnecessarily obscure the present invention. Reference throughout this specification to "one embodiment" or "an embodiment" means that a particular feature, configuration, composition, or characteristic described in connection with the embodiment is included in one or more embodiments of the present invention. Thus, the appearances of "in one embodiment" or "an embodiment" in various places throughout this specification do not necessarily refer to the same embodiment of the present invention. [0113]

[0114] [Preparation Example 1] Culturing of mesenchymal stem cells

[0115] Umbilical cords were obtained from healthy fetuses of healthy mothers. The umbilical cords were washed twice with DPBS, once with 83% ethanol, and then twice with PBS. After removing all blood vessels within the umbilical cord tissue, the cord tissue was minced into 1–2 mm fragments. Liberase (Roche) was added to the fragmented umbilical cord tissue samples for 90 min at 37°C according to the manufacturer's protocol to prepare tissue lysates. Liberase was then inactivated by treating the samples with MEM alpha (Gibco) nutrient medium containing 10% (v / v) fetal bovine serum and 1% (v / v) penicillin-streptomycin (P / S) (Hyclone, Cytiva). The above tissue lysate was filtered through a 100 μm Cell Strainer (pluriSelect), and the filtrate was centrifuged at 1,000 g for 5 minutes. The supernatant except for the cell pellet was then removed, and the nutrient medium was added to the cell pellet and resuspended by pipetting. The viability of the cells contained in the cell suspension was measured using a Trypan blue solution. The cells were seeded in a culture vessel, the nutrient medium was added, and the cells were cultured at 5% CO2 and 37 °C. Thereafter, the medium was replaced every 3 days, and primary culture was performed for 10 days. After 10 days, the morphology of the cultured cells was observed through an optical microscope, and the morphology of mesenchymal stem cells was observed. Flow cytometry analysis was performed using the Stemflow Human MSC Analysis Kit (BD Bioscience) according to the manufacturer's protocol, confirming that the primary culture of umbilical cord-derived mesenchymal stem cells was completed. After washing the identified cells with DPBS, TrypLE Express (no phenol red) (Gibco) solution was added and incubated in an incubator at 37°C for 5 minutes to detach the cells from the culture vessel. The TrypLE solution was neutralized by adding the above nutrient medium to the sample, and centrifuged at 300 g for 5 minutes. The supernatant excluding the cell pellet was then removed, and the cell pellet was resuspended by pipetting with the nutrient medium. The viability of the cells contained in the cell suspension was measured using a trypan blue solution. The cells were inoculated into a new culture dish, nutrient medium was added, and the cells were cultured at 5% CO2 and 37°C, and subcultures were performed at 3-day intervals.

[0116]

[0117] [Example 1] Priming of mesenchymal stem cells using agmatine

[0118] The umbilical cord-derived mesenchymal stem cells prepared in the above Preparation Example 1 were cultured in a medium containing agmatine. The agmatine was prepared by dissolving agmatine sulfate (Sigma-Aldrich) in distilled water and adding it to the medium so that the final concentration of agmatine in the medium was 100, 200, or 400 μM. The medium used for primary culture aimed at promoting cell activity was MEM alpha (Gibco) medium supplemented with 10% (v / v) FBS (Gibco) and 1% (v / v) penicillin / streptomycin (Hyclone, Cytiva) or 0.5% (v / v) gentamicin (Gibco). The medium used for secondary culture aimed at obtaining cell culture fluid was DMEM 1X (low-glucose, no phenol red) (Gibco) and MEM alpha (Gibco) medium supplemented with 2 mM L-glutamine (Gibco) and 1% (v / v) penicillin / streptomycin (Hyclone, Cytiva) or 0.5% (v / v) gentamicin (Gibco). However, as a negative control, an equal volume of distilled water was added to the medium.

[0119] Specifically, 5 ml of the primary culture medium containing agmatine as described above was dispensed into a T25 flask, and the umbilical cord-derived mesenchymal stem cells prepared in Preparation Example 1 were subcultured to passage number 5 to 6 (Cell passage 5 to 6) to 2,500 to 3,500 cells / cm 2 The stem cells were seeded into the flask at a density of 10 and cultured for 2 days under conditions of 5% CO2 and 37°C. Each medium was collected from the cultured stem cells in a 15 ml tube and centrifuged at 850 g for 10 minutes. The stem cells from which the medium was collected were washed once with 3 mL DPBS (Dulbecco's Phosphate Buffered Saline), and 5 mL of secondary culture medium was added to the cells and cultured for an additional 2 days under conditions of 5% CO2 and 37°C. Thereafter, each medium was collected from the cultured stem cells in a 15 ml tube and centrifuged at 850 g for 10 minutes. The supernatant excluding the precipitated material was obtained from each centrifuged tube and stored at -80°C until analysis. Total RNA from each cell was extracted from the stem cells from which the medium was collected using the GeneJET RNA Purification Kit (Thermo Scientific) according to the manufacturer's protocol, and stored at -80°C until analysis.

[0120]

[0121] [Example 2] Priming of mesenchymal stem cells using punicalagin

[0122] The umbilical cord-derived mesenchymal stem cells prepared in the above Preparation Example 1 were cultured in a medium containing punicalagin. The punicalagin was prepared by dissolving punicalagin (Sigma-Aldrich) in 0.25% (v / v) dimethyl sulfoxide (DMSO) and adding the solution to the medium so that the final concentration of punicalagin in the medium was 1, 5, or 10 nM. The experiment was performed in the same manner as in Example 1, but a solution containing punicalagin was added instead of agmatine to the primary culture medium and the secondary culture medium. However, as a negative control, an equal amount of 0.25% (v / v) DMSO was added to the medium.

[0123]

[0124] [Example 3] Priming of mesenchymal stem cells using agmatine and punicalagin

[0125] The umbilical cord-derived mesenchymal stem cells prepared in the above Preparation Example 1 were cultured in a medium containing both agmatine and punicalagin. The experiment was performed in the same manner as in Example 1, but a solution of agmatine sulfate (Sigma-Aldrich) dissolved in distilled water and a solution of punicalagin (Sigma-Aldrich) dissolved in 0.25% (v / v) DMSO (dimethyl sulfoxide) were added to the primary culture medium and the secondary culture medium, respectively, so that the final concentration of agmatine contained in each medium was 100, 200, or 400 μM, and the final concentration of punicalagin was 1, 5, or 10 nM.

[0126]

[0127] [Experimental Example 1] Evaluation of the functional enhancement effect of stem cells on immune-regulatory activity and regenerative activity.

[0128] In order to confirm whether the functions related to the immunoregulatory activity and regenerative activity of mesenchymal stem cells were enhanced by treatment with agmatine and / or punicalagin in Examples 1, 2 and 3 above, changes in the expression levels of factors related to immunosuppression, anti-inflammation and regeneration were measured from RNA derived from stem cells.

[0129] Specifically, after thawing the RNA samples prepared in Examples 1, 2, and 3, the RNA concentration of each sample was measured using a spectrophotometer (NanoDrop One, Thermo fisher scientific), an Epoch Microplate Spectrophotometer (BioTek), and a Take3 Micro-volume plate (BioTek) according to the manufacturer's protocol.

[0130] Based on the measured RNA concentration value, 1 μg of each RNA sample was sampled, and cDNA of each sample was synthesized according to the manufacturer's protocol using Maxime RT PreMix Kit (Intron) and Alpha Cycler 1 (PCR max). Next, qRT-PCR was performed on the prepared cDNA samples according to the manufacturer's protocol using 2X qPCRBIO SyGreen Blue Mix Lo-ROX (PCR Biosystems), LightCycler 96 System (Roche), and QuantStudio6 Flex (Thermo fisher scientific). The primers used here were purchased from Cosmogenetech (KR), and the information on the primers for each gene is shown in Table 1 below. The qRT-PCR results for IL-10, IL-1RA, Galectin-1, Galectin-3, TSG-6, and VEGF-A mRNA genes according to each treatment were normalized to the GAPDH expression level, and the relative mRNA expression levels were graphically presented in Figures 1 to 6. The statistical significance level for each sample group compared to the control group (the average value of mRNA expression levels in the DW treatment group and DMSO treatment group) was indicated in each figure using an unpaired two-tailed t-test. [0132] As shown in Figures 1 to 6, when mesenchymal stem cells were treated with agmatine or punicalagin according to the present invention, the expression levels of IL-10, IL-1RA, Galectin-1, Galectin-3, and TSG-6 mRNA, which are factors related to immunosuppression and anti-inflammation, were significantly increased compared to the control group treated with only the solvent, and the expression level of VEGF-A mRNA, a representative factor related to regeneration, was also significantly increased compared to the control group treated with only the solvent. In addition, when mesenchymal stem cells were treated with agmatine and punicalagin together, the expression levels of the above factors were confirmed to increase further.

[0133] Through this, it was found that agmatine, punicalagin, or a mixture thereof according to the present invention has a priming effect that enhances the immunosuppressive ability or regenerative activity of stem cells.

[0134]

[0135] [Experimental Example 2] Evaluation of the effect of enhancing the immune-regulatory activity of stem cells.

[0136] In order to confirm whether the function related to the anti-inflammatory activity of mesenchymal stem cells was enhanced by treatment with agmatine and / or punicalagin in the above Example 3, the supernatant obtained after secondary culture of stem cells was subjected to BD OptEIA Human IL-6 ELISA Set (Cat# 555220, BD Biosciences), R&D Human FGF basic DuoSet (Cat# DY233, R&D) and Epoch Microplate Spectrophotometer (BioTek), Multiskan TM The changes in the contents of IL-6 and FGF-2 cytokines in the cell cultures obtained in Examples 1, 2, and 3 were analyzed using a GO Microplate Spectrophotometer (Thermo Scientific) according to the manufacturer's protocol, and the results are shown in Figs. 7 and 8. The statistical significance level for each sample group compared to the control group (the average value of IL-6 contents in the DW treatment group and the DMSO treatment group) was indicated in the figure using an unpaired two-tailed t-test. (* p≤0.05; ** p≤0.01; *** p≤0.001)

[0137] As shown in Fig. 7, when agmatine and punicalagin according to the present invention were combined and treated with mesenchymal stem cells, it was confirmed that the expression level of IL-6, a representative inflammatory cytokine, was significantly reduced compared to the control group.

[0138] In addition, as shown in Fig. 8, when agmatine and punicalagin according to the present invention were treated to mesenchymal stem cells, it was confirmed that the expression level of FGF-2, a representative regenerative factor, was significantly increased compared to the control group treated with only the solvent.

[0139] Through this, it was found that agmatine, punicalagin, or a mixture thereof according to the present invention has a priming effect that enhances the anti-inflammatory function of stem cells.

[0140]

[0141] Through the above experiments, it was found that priming stem cells with agmatine, punicalagin, or a mixture thereof according to the present invention activates the immune-regulatory and regenerative activities of the stem cells themselves, thereby enhancing the utility of the stem cells as a cell therapeutic agent. In addition, it was found that the priming treatment increases the expression levels of immune-suppressive and anti-inflammatory factors and regenerative factors contained in the stem cell culture medium, and thus the stem cell culture medium, the conditioned medium from which cells have been removed, or the secretome can be utilized as a therapeutic agent for various diseases.
