Description

A composition for improving the skin of companion animals, comprising reishi mushroom mycelia and a fermented product thereof

[0001] The present invention relates to a composition for improving the skin or hair condition of a companion animal, comprising reishi mushroom mycelia or a marine lactic acid bacteria fermentation product of the mycelia culture.

[0002]

[0003] Most pets, including dogs and cats, have thinner dermis and are covered with long hair compared to humans. They lack sweat pores and have a slightly alkaline skin pH of 8-9. Furthermore, because pets' skin is covered with hair, it has limited contact with the outside air, making rashes and inflammation more likely to develop and difficult to detect. This increases the risk of skin diseases, which significantly impact the pet's health. Furthermore, pet skin harbors various bacteria, such as mites and demodex mites, which can damage not only the skin but also the hair, leading to hair-related problems like hair loss and matting.

[0004] Reishi (Ganoderma lucidum) is a mushroom native to various regions, including Korea, China, Japan, Greece, the United States, South America, and Australia. It is often called "Lucid Nurse Mushroom" or "Reishi Mushroom." It has been used in traditional Chinese and East Asian medicine for centuries and is incorporated into various health supplements and foods for health and therapeutic purposes. Reishi helps strengthen the immune system, reduces inflammation in the body, and is rich in antioxidants, protecting cells from free radicals and delaying aging. It can also be used to treat various inflammatory diseases.

[0005] Among the various effects of edible mushrooms including Reishi mushrooms, the function related to skin whitening is known through Korean Patent Publication No. 2010-0130109 and Korean Patent Publication No. 2005-0034833, and in particular, Korean Patent Publication No. 2013-0050751 relates to 'Novel compound derived from Reishi mushrooms and application thereof', and describes a cosmetic composition for skin whitening containing ganadomanandiol, an inhibitor of melanin biosynthesis and tyrosinase activity from the fruiting body of Reishi mushrooms, as an active ingredient.

[0006] However, the use of reishi mushroom mycelia culture for skin protection of companion animals has not yet been reported, and as reishi mushrooms are expensive raw materials, research and development of technologies including additional processes such as fermentation to further enhance their effectiveness is required.

[0007] Accordingly, the inventors of the present invention have completed the present invention by including an additional process to improve the function of the effective ingredients of triterpenoids such as ganadomanandiol contained in the mycelia of the reishi mushroom, and to utilize it for the purpose of improving the skin of companion animals.

[0008] Accordingly, the purpose of the present invention is to provide a composition for skin regeneration of a companion animal comprising a culture of Reishi mushroom mycelia or a lactic acid bacteria fermentation product of the culture.

[0009] Another object of the present invention is to provide a pharmaceutical composition for preventing or treating atopy or allergy in companion animals, comprising a culture of Reishi mushroom mycelia or a fermented product of the culture using lactic acid bacteria.

[0010] Another object of the present invention is to provide a pharmaceutical composition for preventing and treating inflammatory skin diseases in companion animals, comprising a culture of Reishi mushroom mycelia or a lactic acid bacteria fermentation product of the culture.

[0011] Another object of the present invention is to provide an antibacterial or antifungal composition for companion animals comprising a culture of Reishi mushroom mycelia or a lactic acid fermented product of the culture.

[0012] Another object of the present invention is to provide a composition for improving the coat quality of companion animals, comprising a culture of Reishi mushroom mycelia or a fermented product of the culture with lactic acid bacteria.

[0013] In order to achieve the above purpose, the present invention provides a composition for skin regeneration of a companion animal comprising a culture of Reishi mushroom mycelia or a fermented product of the culture with lactic acid bacteria.

[0059] The mycelia of Reishi mushroom were isolated and cultured aseptically on PDA (containing 15 g of agar, 4 g of potato extract, and 20 g of glucose per 1 L of culture medium) medium from Reishi No. 2, which was provided by the Mushroom Division of the National Institute of Horticultural and Herbal Science, Rural Development Administration.

[0060] 1-2) Preparation of Reishi mushroom mycelia culture

[0061] To prepare a liquid culture medium for increasing the content of ganadomanandiol in the mycelia of Reishi mushroom, 100 ml of PDB medium (potato dextrose broth, containing 4 g of potato extract and 20 g of glucose per 1 L of culture medium) was added to a 250 ml Erlenmeyer flask to concentrations of 1, 5, 10, and 15% (v / v), respectively, and the Reishi mushroom mycelia of 1-1 were aseptically cut into 5 mm pieces, and 10 pieces of 100 mg of mycelia were placed in each 250 ml Erlenmeyer flask, followed by shaking culture at 28°C and 100 rpm for 5 days.

[0062] 1-3) Production of fermented reishi mushroom mycelia

[0063] Latilactobacillus sakei subsp. Sakei (M21F010) was added to the above reishi mushroom mycelia culture and further cultured.

[0064] The strain was confirmed to be capable of growth under conditions of temperature between 20°C and 35°C, salinity between 0% and 7%, and pH 4 to pH 10, and in this experimental example, it was cultured under conditions of 30°C, 1% salinity, and pH 5.

[0065] 1-4) Preparation of Reishi mushroom mycelia extract

[0066] The above-mentioned cultured Ganoderma lucidum mycelia were separated from the culture medium and dried at 60°C for 3 days. 50 ml of 70% ethanol was added to 0.2 g of the dried sample, and the mixture was refluxed and extracted twice at 85°C for 2 hours. Only the extract was filtered using a filter paper. The obtained extract was concentrated under reduced pressure to obtain 50 mg of Ganoderma lucidum mycelia extract.

[0067] Example 2. Confirmation of the content of effective ingredients in fermented reishi mushroom mycelia.

[0068] The content of ganadomananediol, an active ingredient contained in the fermented mycelia of the reishi mushroom manufactured above, was compared with that of the reishi mushroom mycelia and strain culture medium. The quantification of ganadomananediol was performed using HPLC (SPD-20A UV / VIS Deteor, LC-20AT Pumping System, Manual Injection Value 20ul Loop, Shimadzu Corp., Japan) equipped with a Phenomenex Luna C18UV(2) column (4.6 x 250 mm, 5 um, USA). The mobile phase was acetonitrile and 0.2% acetic acid, and the flow rate was 0.8 ml / min, and the measurement was performed at 245 nm with a UV (ultraviolet-visible) detector. In addition, ChemFace ®(China) was used as a standard material to perform quantitative analysis of ganadomananediol in Ganoderma lucidum mycelia. The ganadomananediol content in Ganoderma lucidum mycelia was analyzed according to the concentration of lacquer tree extract added to the Ganoderma lucidum culture medium, and the results are shown in Table 1 below.

[0069] Experimental group Ganadomannane diol content (mg) Reishi mushroom mycelia + Latilactobacillus sakei subsp. Sakei (M21F010) strain 3.45 ± 0.17 Reishi mushroom mycelia (Comparative Example 1) 1.63 ± 0.08 Latilactobacillus sakei subsp. Sakei (M21F010) strain culture medium (Comparative Example 2) 0

[0070] As shown in the table above, it was confirmed that the content of ganadomanandiol increased in the fermented product of Reishi mushroom mycelia compared to the mycelia or strain culture solution.

[0071]

[0072] Experimental Example 1. Confirmation of the antioxidant (anti-inflammatory) effect of fermented Reishi mushroom mycelia.

[0073] To confirm the antioxidant effect of the fermented Ganoderma lucidum mycelia, a DPPH inhibitory activity test was conducted. To compare the activity, 1,1-diphenyl-2-picrylhydrazyl (DPPH) solution was added to the Ganoderma lucidum mycelia (Comparative Example 1), the Latilactobacillus sakei culture medium containing no Ganoderma lucidum mycelia (Comparative Example 2), and the fermented Ganoderma lucidum mycelia of the present invention, stirred, allowed to stand for 10 minutes or more, and the absorbance was measured.

[0074] As a result, as shown in Fig. 1, in the case of fermented Reishi mushroom mycelia, the DPPH inhibitory activity was higher than in other experimental groups, and it was confirmed that this increased in a concentration-dependent manner.

[0075] Experimental Example 2. Confirmation of the skin regeneration effect of fermented reishi mushroom mycelia.

[0076] 2-1) Measurement of elastase inhibitory activity of Reishi mushroom mycelia culture

[0077] To confirm the skin regeneration and hair quality improvement effects of fermented Reishi mushroom mycelia, the inhibitory activity of elastase, an enzyme that promotes the decomposition of elastin, a protein that is a component of skin and hair, was measured. To compare the activity, Reishi mushroom mycelia (Comparative Example 1) and Latilactobacillus sakei cultures containing no Reishi mushroom mycelia (Comparative Example 2) were treated with elastase using the same method, and the results were compared.

[0078] As a result, as shown in Fig. 2, the elastase inhibitory activity of the fermented Reishi mushroom mycelia was higher than that of the other experimental groups. From this, it can be confirmed that the fermented Reishi mushroom mycelia has a concentration-dependent skin regeneration and hair quality improvement effect.

[0079] 2-2) Measurement of tyrosinase inhibitory activity of reishi mushroom mycelia culture

[0080] To confirm the skin damage recovery effect of fermented Reishi mushroom mycelia, the inhibitory activity of tyrosinase was measured. The DOPA chrome produced as a result of the action of tyrosinase was measured by colorimetry to confirm the change in activity. To compare the activity with each experimental group, Reishi mushroom mycelia (Comparative Example 1) and Latilactobacillus sakei culture solution without Reishi mushroom mycelia (Comparative Example 2) were treated to skin cells using the same method, and the results were compared.

[0081] As a result, as shown in Figure 3, the fermented Reishi mushroom mycelia exhibited higher tyrosinase inhibitory activity than the other experimental groups. From this, it was confirmed that the fermented Reishi mushroom mycelia exhibited a concentration-dependent recovery effect on skin damage.

[0082] Experimental Example 2. Confirmation of the antibacterial effect of fermented Reishi mushroom mycelia.

[0083] In order to confirm the antibacterial effect of the fermented Reishi mushroom mycelia, the antibacterial activity against four types of fungi (Aspergillus flavus, Aspergillus niger, Candida albicans, and Saccharomyces cerevisiae) that mainly cause skin diseases on the skin of companion animals was measured. In order to compare the activity of the fermented Reishi mushroom mycelia with the above four types of fungi, the Reishi mushroom mycelia and the Latilactobacillus sakei culture medium without the Reishi mushroom mycelia were treated in the same manner as above, and the results were compared as shown in Table 2 below.
