Description
70378-02 5-6-5 / 6-BISARYL COMPOUNDS AS FLT3 INHIBITORS CROSS-REFERENCE TO RELATED APPLICATION
[0001] This application claims priority to U.S. provisional patent application no.63 / 542,619, which was filed October 5, 2023, and which is hereby incorporated by reference in its entirety TECHNICAL FIELD
[0002] The present disclosure relates to 5-6-5 / 6-bisaryl compounds that are selective inhibitors of Fms-related tyrosine kinase 3 (FLT3), pharmaceutical compositions comprising the same, and their use in treating cancer and inflammatory diseases. BACKGROUND
[0003] This section introduces aspects that may help facilitate a better understanding of the disclosure. Accordingly, these statements are to be read in this light and are not to be construed as admissions about what is or is not prior art.
[0004] Kinase inhibitors are increasingly being used for diverse therapeutic interventions. One of the factors that accounts for dose-limiting toxicities of current kinase inhibitors, such as Fms- related tyrosine kinase 3 (FLT3) inhibitors, is target promiscuity, especially targets critical for essential functions. For example, the FLT3 receptor is similar to essential receptor tyrosine kinases (RTKs), such as kinase insert domain receptor (KDR, also known as VEGFR2), fibroblast growth receptor (FGFR), platelet-derived growth factor receptor (PDGFR), proto-oncogene receptor tyrosine kinase (c-KIT), and rearranged during transfection (RET). It has been challenging to develop an inhibitor that selectively inhibits FLT3 without co-inhibiting these related kinases. It is well known that targeting cardiovascular (KDR, FGFR1 / 2, and PDGFR ^ / ^) and / or kidney (RET) related kinases can result in low compound tolerability and increased side effects. Additionally, because both FLT3 and c-KIT are important for myeloid cells, the co- inhibition of FLT3 and c-KIT is synthetically lethal and has been shown to result in myelosuppression.
[0005] FLT3 activation can also lead to a block in myeloid differentiation or apoptosis. FLT3 is overexpressed in over 90% of acute myeloid leukemia (AML) or acute lymphoblastic leukemia (ALL) patients (Blood, 1993, 82, 1110-119). In addition to overexpression of wild-type FLT3 in hematologic malignancies, about 30% of AML patients harbor mutated FLT3 (activating kinase domain mutation or internal tandem duplication, ITD, in the juxtamembrane domain), and these 1 70378-02 patients typically have a poorer prognosis than those harboring wild-type FLT3. Many FLT3 kinase mutations that are insensitive to one or more of clinically trialed FLT3 inhibitors have been observed in AML patients, including FLT3-D835Y / V / H (type II inhibitor, such as quizartinib, resistant), gatekeeper mutation F691I / L (gilteritinib resistant), and K429E (crenolanib resistant) (Cancer Discovery, 2019, 9, 1050–1063; Blood Adv. 2022, 6, 2144–2155; Biomedicines, 2020, 8, 245). Some of these FLT3 inhibitors have been moderately successful in increasing survival rates of patients with AML, but often, dose-limiting toxicities hinder efficacy, especially in elderly patients, vide supra (CA Cancer J. Clin., 2022, 72, 7-33).
[0006] In addition to the need for a new FLT3 inhibitor that could overcome on-target drug resistance (such as FLT3-ITD-F691L, which is resistant to standard-of-care gilteritinib), there is also a need for a selective FLT3 inhibitor that avoids critical anti-targets, such as KDR, FGFR1 / 2, PDGFR ^ / ^, c-KIT, and RET. Such selective FLT3 inhibitors could find applications in maintenance therapy, which requires chronic dosing, and / or extend FLT3 inhibitor therapy to elderly patients with cardiovascular or renal co-morbidities.
[0007] Therefore, there is an unmet need for a potent and selective FLT3 inhibitor compound which avoids inhibition of anti-targets, such as c-KIT, KDR, FGFR1 / 2, PDGFR ^ / ^, and RET. It is an object of the present disclosure to provide such a compound. This and other objects and advantages, as well as inventive features, will be apparent from the detailed description. SUMMARY

[0041] For the purposes of promoting an understanding of the principles of the present disclosure, reference will now be made to the embodiments illustrated in the drawings, and specific language will be used to describe the same. It will nevertheless be understood that no limitation of the scope of the claimed invention is thereby intended.
[0042] The term "FLT3 inhibitors" refers to a compound that inhibits Fms-related tyrosine kinase 3 (FLT3, also called Fms-related receptor tyrosine kinase 3; Fms-like tyrosine kinase 3; Receptor- Type Tyrosine-Protein Kinase FLT3; Stem Cell Tyrosine Kinase 1; FL Cytokine Receptor; CD135; CD135 Antigen; STK1; FLK2; FLK-2; EC 2.7.10.1; Growth Factor Receptor Tyrosine Kinase Type III; Fetal Liver Kinase 2; Fetal Liver Kinase-2).
[0043] The present disclosure is predicated, at least in part, on the discovery that the imidazo[1,2- b] pyridazine moiety, found in many approved and experimental drugs, binds to the hinge region of kinases. Substitutions at positions 2, 3, 6, 7, and 8 dictate kinase selectivity and potency (Eur. J. Med. Chem., 2021, 226, 113867). It is reported that fusing kinase hinge binders, such as isoquinolines, napthyridines, and imidazo[1,2-b]pyridazine, with alkynyl pyridines, are highly potent FLT3 inhibitors. But, these compounds and FDA-approved FLT3 inhibitors, such as 11 70378-02 gilteritinib, quizartinib, and midostauri, kinase inhibitors, such as sorafenib (KDR inhibitor) or ponatinib (ABL1 inhibitor) which are also FLT3 inhibitors, and other FLT3 inhibitors, such as crenolanib, tandutinib, FF-10101, and tuspetinib (see Fig.1) also potently inhibit anti-targets such as KDR, c-KIT, FGFR1 / 2, PDGFR ^ / ^, and RET. FLT3 has also been reported as a potential target for chronic pain, and for non-oncology applications, a high therapeutic ratio is usually required.
[0044] In view of the above, the present disclosure provides 5-6-5 / 6-bisaryl compounds comprising substituted imidazo[1,2-b] pyridazine core that are selective FLT3 inhibitors. The compounds can be substituted at the C6 position with a moiety such as a substituted morpholine or a substituted piperidine to obtain potent and selective FLT3 inhibition with low off-target activity.
[0045] Thus, provided is a compound of formula (I), or a pharmaceutically acceptable salt, hydrate, tautomer, or optical isomer thereof: (I) wherein the compound of formula (I) comprises: (i) B, a 5-6 bi-aryl moiety represented by a structure: wherein each X is independentlyor CRb, wherein Rais selected from H, C1-C6alkyl, C3-C8 cycloalkyl, hetero C2-C6 alkyl, C1-C6 alkylcarbonyl, arylcarbonyl, hetero C2-C6 alkylcarbonyl, amide, and sulfonamide; and Rbis selected from H, cyano, halo, C1-C6alkyl, trifluoro C1-C6 alkyl, halo C1-C6 alkyl, dihalo C1-C6 alkyl, hydroxy C1-C6 alkyl, alkoxy C1-C6 alkyl, C3-C8cycloalkyl, hetero C2-C6alkyl, amino C1-C6alkyl, hydroxy C1-C6alkyl, alkoyx C1- C6 alkyl, cyano C1-C6 alkyl, C1-C6 dialkylamino C1-C6 alkyl, morpholinyl C1-C6 alkyl, piperazinyl C1-C6alkyl, C1-C6dialkylaminocarbonyl, C1-C6dialkylaminosulfonyl, carboxylic acid, and ester; wherein alkyl is optionally substituted with C1-C6 alkyl or hetero C1-C6 alkyl; and a bond between any two X is a single or double bond; and 12 70378-02 each E is independently N or CRc, wherein Rc is H or halogen; (ii) P, a 5 / 6 bi-cyclic aryl moiety represented by a structure: wherein each A iseach G is independently CH or N; W is N or CRb, wherein Rb is selected from H, cyano, halo, C1-C6 alkyl, trifluoro C1-C6 alkyl, halo C1-C6alkyl, dihalo C1-C6alkyl, hydroxy C1-C6alkyl, alkoxy C1-C6alkyl, C3-C8cycloalkyl, hetero C2-C6 alkyl, amino C1-C6 alkyl, cyano C1-C6 alkyl, C1-C6 dialkylamino C1-C6 alkyl, morpholinyl C1-C6alkyl, piperazinyl C1-C6alkyl, C1-C6dialkylaminocarbonyl, C1-C6dialkylaminosulfonyl, hydroxyaryl, alkoyxaryl, carboxylic acid, and ester; wherein alkyl is optionally substituted with C1-C6alkyl or hetero C2-C6alkyl; and B is as defined above; (iii) Y is H or a 4- to 12-membered monocyclic ring or bi-cyclic ring, wherein the bi-cyclic ring is a fused bi-cyclic ring, a bridged bi-cyclic ring, or a spiro bi-cyclic ring, substituted with one or more groups selected from R1-R8 based on the ring size and represented by a structure: wherein T is sp3- carbon, each R1, R2, R3, R4, R5, R6, R7,and R8is independently selected from H, halo, nitro, cyano, C1-C6 alkyl, amino C1-C6 alkyl, hydroxy C1-C6 alkyl, alkoxy C1-C6 alkyl, C1-C6 alkylcarbonyl, C1-C6alkylsulfonyl, C3-C8cycloalkyl, hetero C2-C6alkyl, aryl, and heteroaryl; or any two R groups from R1-R8, together with the carbon atom to which they are attached, form a 3- to 8-membered cyclic ring; and Q is O, S, S=O, SO2, NRa or CRbRb, wherein Ra is as defined above and each Rb is independently as defined above or the two Rb, together with the carbon atom to which they are 13 70378-02 attached, form a 3- to 6- membered cyclic ring optionally containing one or more heteroatoms, wherein each heteroatom is independently N, S or O.

[0048] In some embodiments, provided is a compound of formula (IA), or a pharmaceutically acceptable salt, hydrate, tautomer, or optical isomer thereof: HN N 5 Y wherein each R9 and R10 isfrom hydrogen, deuterium, C1-C6 alkyl, and hetero C2-C6alkyl; each E is independently N or CRc, wherein Rcis H or halogen; and Y is H or a 4- to 12-membered monocyclic ring or bi-cyclic ring, wherein the bi-cyclic ring is a fused bi-cyclic ring, a bridged bi-cyclic ring, or a spiro bi-cyclic ring, substituted with one or more groups selected from R1-R8based on the ring size and represented by a structure: wherein T is sp3- carbon, each R1, R2, R3, R4, R5, R6, R7, and R8is independently selected from H, halo, nitro, cyano, C1-C6 alkyl, amino C1-C6 alkyl, hydroxy C1-C6 alkyl, alkoxy C1-C6 alkyl, C3-C8 cycloalkyl, hetero C2-C6alkyl, aryl, and heteroaryl; or any two R groups from R1-R8, together, with the carbon atom to which they are attached, form a 3- to 8-membered cyclic ring; and 14 70378-02 Q is O, S, S=O, SO2, NRa or CRbRb, wherein Ra is as defined above and each Rb is independently as defined above or the two Rbtogether, with the carbon atom to which they are attached, form a 3- to 6- membered cyclic ring optionally containing one or more heteroatoms, wherein each heteroatom is independently N, S or O.
[0049] In some embodiments, the Y is: N N N N N N N .
[0051] In some embodiments, the Y is: 15 70378-02 .
[0052] In some embodiments, the Y is: 70378-02 .

[0056] In some embodiments, each X in the structures 7a-7u is independently CH2, O, S, S=O, SO2, NH, N-alkyl, N-heteroalkyl, amide, sulfonamide, urea, or carbamate.
[0057] In some embodiments, the compound of formula (I) or (IA) is:70378-02 HNN.
[0058] In some embodiments, the compounds of formula (I) and (IA) are selective FLT3 inhibitors. The compounds can inhibit FLT3 at nanomolar concentrations. The compounds do not inhibit other commonly targeted kinases and exhibit very low activity against immune or cardiovascular anti-targets, such as c-KIT, KDR, FGFR1 / 2, PDGFR ^ / ^, and RET. Thus, compounds avoid co-inhibition of FLT3 with other targeted kinases, including c-KIT and KDR, which are particularly problematic as this leads to synthetic lethality.
[0059] In some embodiments, the compound of formula (I) and (IA) inhibit FLT3 with IC50 value between about 0.01 nM to about 30 nM, such as about 0.01 nM to 30 nM or 0.01 nM to about 30 nM or 0.01 nM to 30 nM.
[0060] In some embodiments, the compound of formula (I) or (IA) has low activity against anti- targets KDR, c-KIT, FGFR1 / 2, PDGFR ^ / ^, and / or RET. The compound of formula (I) or (IA) inhibits said anti-targets with an IC50 value greater than 300 nM, which is at least ten times higher than the IC50value for FLT3.
[0061] In some embodiments, the compound of formula (I) and (IA), for example, compound 17 inhibits FLT3-ITD mutant with IC50 of about 1 nM (Fig.4A). It shows low inhibition for KIT and RET with IC50 values about 912 ± 67 nM and 18400 ± 9100 nM (Figs. 4B-4C) and very low activity against cardiovascular kinases FGFR1 / 2 and PDGFR ^ / ^, with IC50 values >2690 nM 20 70378-02 (Figs.4E-4H). Compound 17 can be FLT3 type I inhibitor. It shows IC50 of about 0.41±0.12 nM and about 1 nM against mutants FLT3-D835 and FLT3-D835Y, which are similar to known FLT3 type I inhibitors e.g., gliteritinib (0.19±0.06 nM and 1 NM).
[0062] Thus, the compounds of the disclosure evade the inhibition of these critical anti-targets while targeting FLT3, which can allow for inhibition of the oncogenic pathway associated with FLT3 mutations without interfering with the normal physiological functions of essential receptors, such as c-KIT, KDR, and RET.
[0063] Provided is a pharmaceutical composition comprising a compound of formula (I) or (IA) or a pharmaceutically acceptable salt, hydrate, tautomer, or optical isomer of either of the foregoing, and a pharmaceutically acceptable carrier, excipient, or diluent. The carrier, excipient, or diluent can vary based on the particular route of administration (see, e.g., Remington’s The Science and Practice of Pharmacy, 23rded. (2020))
[0064] Provided is a method of inhibiting a FLT3 in a patient, which method comprises administering to the patient (i) a therapeutically effective amount of compound of formula (I) or (IA), (ii) a pharmaceutically acceptable salt, hydrate, tautomer, or optical isomer of (i), or (iii) a pharmaceutical composition comprising (i) or (ii) and a pharmaceutically acceptable carrier, excipient, or diluent, whereupon FLT3 in the patient is inhibited.

[0106] A new kinase inhibitor library, which consisted of compounds composed of known hinge binders (see compounds 1-10 below) connected to a pyrazole, was designed. Additionally, some compounds within the library contained various cyclic heterocycles, such as morpholine, piperazine, and tetrahydrofuran (for example, compounds 4-10). These heterocycles are also considered privileged moieties, as they could impact a compound’s aqueous solubility, as well as engage in hydrogen-bonding interactions with different protein residues, including the backbone amide. 29 70378-02 HN N HN NHNNHNNQ
[0107] Compoundsreactions (Scheme 1). Final compounds with substitutions at the C6 position, denoted “Q”, were formed via a two- step synthesis (see Scheme 1, steps A and B). Intermediates were synthesized by reacting various amine / alcohol substituents with 3-bromo-6-chloroimidazo[1,2-b]pyridazine. Subsequent Suzuki cross-coupling between the pyrazolo boronic acid and the bromide afforded the final compounds. 70378-02 Scheme 1: Synthesis of analogs via two-step synthesis. (A) synthesis of the intermediate by addition of amine / alcohol to aryl chloride to afford “Q” substituted pyrazolopyrimidine derivatives. (B) formation of final compounds via Suzuki coupling.
[0108] After synthesis of the initial pyrazole-containing compounds (1-10 above), the compounds were screened at concentration of 100 nM against FLT3 via an ADP Glo Kinase Assay (Fig. 2). Among the various hinge binders, the imidazo[1,2-b]pyridazine seemed to perform the best, when substituted at the 6-position (denoted Q) with 6-membered and not 5-membered heterocycles. For example, compound 3, the unsubstituted imidazo[1,2-b]pyridazine showed 0% inhibition of FLT3, and analogs 1 and 2, which have alternate hinge binders, only inhibited FLT3 at 11.8 ± 7.4% and 8.3 ± 2.2%, respectively. In terms of Q, FLT3 inhibition was lower with 5-membered heterocycles, as compounds 4 and 5 showed inhibitory activities of 9.6 ± 0.8% and 8.7 ± 2.5%, respectively. On the contrary, 6-membered heterocycles substituted at the 6-position (Q) appeared to have significant inhibitory activity against FLT3, as seen with compounds 6-10. The morpholine substituent (6) showed 30.2 ± 6.5% inhibition of FLT3, but this was improved with a trans-dimethyl substitution, as seen in compound 7 (46.2 ± 4.9%), and inhibition increased even further with the cis-dimethyl substitution in compound 8 (61.6 ± 0.7%). Additionally, two substituted piperazine analogs (9 and 10) were evaluated but showed less inhibitory activity against FLT3 (33.4 ± 4.2% and 29.8 ± 6.7%, respectively) compared to the dimethylmorpholines.
[0109] After the preliminary FLT3 screening, analog 8, containing the trans-2,6- dimethylmorpholine, emerged as the most potent inhibitor (Fig.2). The optimization of analog 8 were carried out to probe selectivity for FLT3 over the aforementioned KDR and c-KIT anti- targets. In addition to being the most potent FLT3 inhibitor, analog 8 had a higher fraction of sp3- hybridized carbon atoms and thus was deemed more drug-like than the other preliminary compounds, such as 1, 2, and 6. A structure-activity relationship (SAR) study was carried out to explore how modifications affected FLT3 and off-target inhibition. The new pyrazolo-based compounds (11-20) that are potent, yet selective, FLT3 inhibitors were identified. 31 70378-02 NC HNNNNNO The above compounds (11-20) were screened against FLT3 and primary anti-targets c-KIT and KDR (Fig. 3). Different substitutions in the pyrazole ring were explored. Compound 11, which contains the 1H-pyrazol-3-yl as opposed to the 1H-pyrazol-4-yl in lead compound 8, showed a significant decrease in FLT3 inhibition (28.3 ± 0.8%). The -NEt substitution (13) showed similar inhibitory activity against FLT3 (59.1 ± 2.9%) compared to compound 8, but less selectivity over c-KIT and KDR. Other modifications to the pyrazole, including the addition of a nitrile moiety (12) and a morpholine ring (14), also resulted in loss of FLT3 inhibition. When substituting with a phenyl ring, as in 15, increased inhibitory activity was found against FLT3 but not the selectivity over the c-KIT and KDR anti-targets. Placing a phenyl linker between the imidazo[1,2- b]pyridazine and the pyrazole (17) increased FLT3 inhibition (95.6 ± 0.6%), while significantly decreasing inhibitory activity against c-KIT (27.2 ± 2.9%) and KDR (9.9 ± 1.8%). Under similar conditions, compound 17 showed better selectivity against these off-target enzymes than ponatinib, which has been approved for CML and trialed for AML, and gilteritinib (Fig. 3). 32 70378-02 Substituting the pyrazole moiety in compound 17 with a methyl group (18) resulted in reduced FLT3 inhibition (43.4 ± 2.7%).
[0111] IC50 values for FLT3 and the anti-targets c-KIT, KDR, (see Fig. 5), RET, FGFR1 / 2, and PDGFR ^ / ^ were determined. While compound 17 and gilteritinib inhibited FLT3-ITD with similar IC50values (~1 nM), compound 17 had very little activity against the anti-targets (see Figs. 4A-4H), whereas gilteritinib potently inhibited c-KIT and RET with IC50 values of 59.7 ± 4.4 nM and 16.7 ± 1.2 nM, respectively (compared to 912 ± 67 nM and 18400 ± 9100 nM, respectively, for compound 17). Additionally, compound 17 showed very limited activity against cardiovascular kinases FGFR1 / 2 and PDGFR ^ / ^, with IC50 values >2690 nM. Avoiding the inhibition of these critical anti-targets while targeting FLT3 could allow for inhibition of the oncogenic pathway associated with FLT3 mutations without interfering with the normal physiological functions of essential receptors, such as c-KIT, KDR, and RET.
[0112] Kinase inhibitor type and cellular activity The reported FLT3 inhibitors are either type I (for example gilteritinib or midostaurin) or type II (for example quizartinib or ponatinib). The FLT3-D835 mutation generally leads to induced “DFG-in” conformation, which favors type I inhibitor binding. On the contrary, most FLT3-D835 mutants are resistant to type II inhibitors. The IC50values for gilteritinib (a type I inhibitor) and compound 17 against mutated FLT3-D835Y are very similar (0.19 ± 0.06 nM and 0.41 ± 0.12 nM, respectively). Under similar experimental conditions, both compounds inhibited FLT3- D835Y with IC50values of approximately 1 nM, so it appears that the D to Y mutation, which favors the DFG-in conformation, did not affect the activity of compound 17; therefore, compound 17 could be a type I inhibitor. Docking studies of 17 (Fig.6) to the active conformer of FLT3 (the “DFG-in” homology model) were performed. In the docked model, the imidazo[1,2-b]pyridazine moiety in compound 17 (a known hinge-binding moiety) interacts with the FLT3 Cys-694 residue. Similarly, the hinge binding moiety pyrazine-2-carboxamide found in gilteritinib interacts with the hinge Cys-694.
[0113] The antiproliferative activities of compound 17 against FLT3-driven AML cell line Molm- 14 as well as the Molm-14 FLT3-D835Y and FLT3-F691L mutant cell lines were also examined. As expected for a potential type I inhibitor, compound 17 inhibited mutated Molm-14 FLT3-ITD- D835Y better (GI50= 9.58 ± 1.33 nM) than the mutated Molm-14-FLT3-ITD-F691L (GI50= 23.1 ± 6.8 nM), (Fig. 8). Immunoblotting confirmed that compound 17 could reduce the levels of phosphorylated FLT3 in Molm-14 at similar levels to gilteritinib (Figs.7A-7B). 33 70378-02
[0114] Kinase Assay The ADP-Glo Kinase Assay Kit (Promega®) was utilized for determination of FLT3, c-KIT, and KDR inhibition. Kinase Buffer (25 mM MOPS, pH 7.2, 12.5 mM β -glycerol-phosphate, 25 mM MgCl2, 2 mM EDTA) was supplemented with DTT (0.2 mM) and BSA (40 µg / mL) immediately prior to use. Reaction volumes of 5 µL containing enzyme, inhibitor, 10 µM ATP, and the appropriate peptide substrate (20 µM) were incubated for 3 hours, and then quenched with 5 µL of GLO reagent. After 40 min, 10 µL DET reagent was added and incubated for 1 hour before visualization on a Cytation™ 5 Cell Imaging Multi-Mode Reader (BioTek).
[0115] Docking Gilteritinib and compound 17 structures were converted to mol2 files, uploaded to Maestro, and prepared using LigPrep protocol. PDB: 6JQR was used for the FLT3 inactive conformation. FLT3 homology model was used for the FLT3 active conformation. Protein structures were prepared for docking using the Protein Preparation Wizard. Compounds were docked to each proteins structure using the Induced Fit Docking protocol. Docking conformations were examined Visual representations were generated using The PyMol Molecular Graphics System, Version 2.0. Ligands were extracted from the docking poses and crystal structure in Maestro. The function use_neutral_scaffold was utilized to ignore multiple bonds patterns and ionization states.
[0116] Cell Cultures Molm-14, Molm-14 (D835Y), and Molm-14 (F691L) cell lines. Cells were cultured to confluence in RPMI supplemented with 10% fetal bovine serum and 1X Penicillin-Streptomycin (Gibco™). Cells were plated at 5000 cells / well in 96 well plates. After 24-hour incubation at 37 °C in a 5% CO2 incubator, compounds diluted in the same media were added to the indicated final concentration and allowed to incubate at 37 °C in a 5% CO2incubator for 72 hours. Viability was determined using CellTiter-Blue® Cell Viability Assay (Promega) with absorbance readings at 570 nm performed on a Cytation™ 5 Cell Imaging Multi-Mode Reader (BioTek) 3 hours after addition.
[0117] Immunoblotting Molm-14 cells were treated with 17 or control for 2 hours. Cells were collected and lysed, and protein concentrations were quantified relative to BSA standards. After protein denaturation, proteins were separated via SDS-PAGE electrophoresis and transferred to NC membranes. After blocking, overnight incubation with specific primary antibodies, and room temperature incubation for 2 hours with peroxidase-conjugated secondary antibodies, peroxidase activity was detected with SuperSignal West Pico reagents using a CCD camera LAS-4000. Bands were normalized 34 70378-02 relative to loading control. Values reported represent the means of triplicates and error bars represent SD. All specific antibodies were purchased from Cell Signaling.
