[0002] The scientific name for cannabis is Cannabis sativa L., and it is an annual herb belonging to the cannabis family. It has been cultivated as a fiber crop in Korea and China, and in the West, it has been used as a medicinal plant for the use of cannabinoids, which are components of the terpenoid family.
[0003] Hemp seeds contain 90% of the unsaturated fatty acids oleic acid, linoleic acid, and linolenic acid, and 3% of gamma-linolenic acid, and contain 20 types of amino acids, including essential amino acids, vitamins, minerals, and dietary fiber, as well as other nutritional and functional ingredients, making them a useful substance that can have immune and antioxidant effects in the human body.
[0004] Per 100 g of hemp seeds, it contains 36 g of protein, 3-4 g of fatty acids, and 30 g of fiber. The nutritional information is 145 mg of calcium, 14 mg of iron, 483 mg of magnesium, 7 mg of manganese, 1,160 mg of phosphorus, 859 mg of potassium, 7 mg of zinc, 3800 IU of vitamin A, 0.4 mg of vitamin B1, 0.11 mg of vitamin B2, 2.8 mg of vitamin B3, 0.12 mg of vitamin B6, 2,277.5 IU of vitamin D, and 90 mg of vitamin E. It contains 1.7 times more vitamin B1 and folic acid than broccoli, 1.5 times more dietary fiber than bananas, and 1.2 times more calcium than eggs.
[0005] In February 2015, the Ministry of Food and Drug Safety of Korea regulated the THC allowable standards for hemp seeds and hemp seed oil to 5 mg or less per kg and 10 mg or less per kg, respectively, and hemp seeds are used as food and cosmetics after undergoing THC tests three times from sowing to harvesting and customs clearance.
[0006] Cordyceps militaris is a small mushroom belonging to the Clavicipitaceae family of the Ascomycetes order of the Ergot fungi order. Its name comes from its appearance, which is that it lives inside insects in the winter and sprouts like grass in the summer. The mechanism by which Cordyceps militaris is formed is that the Cordyceps militaris fungus forms conidia or ascospores, attaches to the joints, digestive organs, etc. of living insects, and then consumes nutrients from the insect's body, forming hyphae, which eventually kill the insect. When winter passes and summer comes and the temperature and humidity rise, mushrooms emerge from the insect's buried body and grow above the ground. In this way, Cordyceps militaris refers to the combination of a dead insect filled with hyphae and the mushroom that grows from its body.
[0007] Cordyceps militaris is divided into three parts: the head, the stipe, and the host. The head contains the spores used for reproduction, and the long part that supports the head is the stipe. The head and stipe together are called the fruiting body or mushroom, and the tip of the stipe protrudes from the pupa body. The head is convex, spherical, cylindrical with pointed ends, or spatula-shaped, and multiple perithecium are formed on its surface or beneath the epidermis.
[0008] Cordyceps militaris is generally composed of water, fat, crude protein, carbohydrates, and ash, and the protein contains about 18 kinds of amino acids essential for the human body. In addition, Cordyceps militaris contains special ingredients such as cordyceps polysaccharide and cordyceic acid, which are known to be used in the treatment of anemia, fatigue, high blood pressure, pulmonary tuberculosis, asthma, etc. Because it parasitizes insects, it is also very useful as a biopesticide that provides effective pest control while preventing environmental pollution. Cordyceps militaris has been passed down in China since ancient times as the "elixir of longevity for emperors," and it is recorded in the Compendium of Materia Medica that "when used regularly, it has a clear effect of strengthening weak constitutions and enhancing immunity." The mycelia obtained during spawn culture are known to produce mannitol, which is used for medicinal purposes, as well as other useful substances. Additionally, the fruiting body contains a component called cordycepin, which is known as an isomer of quinic acid. Cordycepin acts as a toxin to host insects and is an active substance with antibacterial, antiviral, and anticancer effects, and is known to be excellent for various diseases related to aging. Despite these excellent effects, collecting Cordyceps militaris fruiting bodies grown in a natural state is very difficult, so artificial cultivation technology is required to ensure a stable supply of Cordyceps militaris fruiting bodies.
[0009] Korean Patent Publication No. 2022-0088077 discloses a composition for cultivating Cordyceps militaris containing barley and red ginseng powder as active ingredients and a method for cultivating the same, and Korean Patent Registration No. 2552650 discloses a method for cultivating Cordyceps militaris using abalone. However, there is no technology in the prior art to increase the functional components of Cordyceps militaris using hemp seeds as in the present invention.
[0010] The present invention was derived from the above-mentioned needs, and in the process of conducting research to increase the content of functional ingredients contained in cultured Cordyceps militaris, the inventors prepared a medium using various materials and verified the activity thereof, and confirmed that Cordyceps militaris cultured on a hemp medium prepared using hemp seeds pretreated by the method of the present invention had increased contents of cordycepin and adenosine, which are the key effective ingredients of Cordyceps militaris, and that Cordyceps militaris with excellent palatability could be produced, thereby completing the present invention.
[0011] In order to solve the above problem, the present invention provides a method for culturing Cordyceps militaris, characterized by comprising the steps of (1) preparing a plant extract by adding water to a mixture of purslane and lotus seed and then extracting it; (2) soaking hemp seeds by adding the plant extract prepared in step (1); (3) preparing a hemp medium by adding raw sugar and water to the soaked hemp seeds in step (2) and then sterilizing them; and (4) inoculating Cordyceps militaris spores into the hemp medium prepared in step (3) and culturing them.
[0012] In addition, the present invention provides Cordyceps militaris cultured by the above method.
[0013] In addition, the present invention provides a Cordyceps militaris extract prepared by adding a solvent to the Cordyceps militaris and then extracting it, wherein the extract is characterized in that it contains 4 to 7 mg / g of cordycepin and 34 to 38 mg / g of adenosine, respectively.
[0014] Cordyceps militaris cultured using the method of the present invention has high cordycepin and adenosine contents, and thus it is believed that the Cordyceps militaris culture method using the hemp medium of the present invention can be usefully used for cultivating high-quality Cordyceps militaris.
[0015] In addition, Cordyceps militaris cultured by the method of the present invention is safe as THC (Tetrahydrocannabinol), a hallucinogenic component of cannabis, is not detected at all, and has the effect of providing Cordyceps militaris with a more desirable taste, texture, and flavor.
[0016] Figure 1 is a photo of Cordyceps militaris cultured using hemp seed and brown rice in a 25:25 ratio.
[0017] Figure 2 shows the results of culturing Cordyceps militaris for 6 weeks using hemp medium with different amounts of water added to hemp seeds soaked for 48 hours.
[0018] Figure 3 shows the results of culturing Cordyceps militaris for 6 weeks using hemp medium with different amounts of water added to hemp seeds soaked for 24 hours.
[0019] In order to achieve the above purpose, the present invention
[0020] (1) A step of preparing a plant extract by adding water to a mixture of bottle gourd and lotus seed and then extracting it;
[0021] (2) A step of adding the plant extract prepared in step (1) to hemp seeds;
[0022] (3) A step of preparing a hemp medium by adding raw sugar and water to the hemp seeds soaked in step (2) and then sterilizing them; and
[0023] (4) A method for culturing Cordyceps militaris is provided, characterized by including a step of inoculating Cordyceps militaris spores into the hemp medium manufactured in step (3) and culturing them.
[0024] In the method for culturing Cordyceps militaris of the present invention, the plant extract of step (1) can be prepared by adding 15 to 25 times (v / w) of water to a mixture of Centella asiatica and Nelumbo nucifera in a weight ratio of 0.8 to 1.2:0.8 to 1.2, and then extracting at 90 to 110°C for 10 to 30 minutes, and more preferably, adding 20 times (v / w) of water to a mixture of Centella asiatica and Nelumbo nucifera in a weight ratio of 1:1, and then extracting at 100°C for 20 minutes. Soaking hemp seeds with the plant extract prepared under the above conditions allowed Cordyceps militaris to be cultured in a manner suitable for the growth of Cordyceps militaris while increasing the content of effective ingredients.
[0025] In addition, in the method for cultivating Cordyceps militaris of the present invention, the hemp seed variety used in step (2) is the IH3 variety, which has a lower THC (Tetrahydrocannabinol) content and a higher CBD (Cannabidiol) content than the conventional variety. In addition, when the peeled hemp seed (Cannabis sativa seed) is used, Cordyceps militaris hardly grows, so it is preferable to use the harvested hemp seed as is.
[0026] In addition, in the method for culturing Cordyceps militaris of the present invention, the step (2) may preferably be soaked at 20 to 25°C for 20 to 28 hours by adding a plant extract to 30 to 70 parts by weight of hemp seeds, and more preferably, may be soaked at 20 to 25°C for 24 hours by adding a plant extract to 50 parts by weight of hemp seeds. If the hemp seeds are used without soaking, the Cordyceps militaris growth rate decreases, so it is preferable to soak the hemp seeds under the conditions described above. If the hemp is soaked in more than the above range, there is a problem that the hemp seeds sprout or are rather unsuitable for the growth of Cordyceps militaris.
[0027] In addition, in the method for culturing Cordyceps militaris of the present invention, the hemp medium of step (3) can be prepared by adding 1 to 10 parts by weight of raw sugar and 20 to 40 parts by weight of water to soaked hemp seeds, and then sterilizing at 110 to 130°C for 20 to 40 minutes, and more preferably, by adding 5 parts by weight of raw sugar and 30 parts by weight of water to soaked hemp seeds, and then sterilizing at 121°C for 30 minutes. Preparation of the hemp medium under the above conditions is suitable for the growth of Cordyceps militaris, and can also increase the content of effective ingredients such as cordycepin and adenosine.
[0028] In addition, in the method for culturing Cordyceps militaris of the present invention, the step (4) may preferably be performed by inoculating 15 to 25 parts by weight of Cordyceps militaris spores into a hemp medium and culturing them at 19 to 21°C for 5 to 10 weeks, and more preferably by inoculating 20 parts by weight of Cordyceps militaris spores into a hemp medium and culturing them at 19 to 21°C for 6 to 8 weeks. Culturing under the above conditions maximizes the content of effective ingredients of Cordyceps militaris, and allows sufficient growth in a state suitable for consumption in terms of flavor, taste, and texture.
[0029] More specifically, the method for cultivating Cordyceps militaris of the present invention is
[0030] (1) A step of preparing a plant extract by adding 15 to 25 times (v / w) of water to a mixture of bottle gourd and lotus seed in a weight ratio of 0.8 to 1.2:0.8 to 1.2 and then extracting at 90 to 110°C for 10 to 30 minutes;
[0031] (2) A step of adding the plant extract prepared in step (1) to 30 to 70 parts by weight of hemp seeds and soaking them at 20 to 25°C for 20 to 28 hours;
[0032] (3) A step of preparing a hemp medium by adding 1 to 10 parts by weight of raw sugar and 20 to 40 parts by weight of water to the hemp seeds soaked in step (2) above and sterilizing at 110 to 130°C for 20 to 40 minutes; and
[0033] (4) It may include a step of inoculating Cordyceps militaris spores into the hemp medium manufactured in step (3) above and culturing them at 19 to 21°C for 5 to 10 weeks.
[0034] More specifically
[0035] (1) A step of preparing a plant extract by adding 15 to 25 times (v / w) of water to a mixture of bottle gourd and lotus seed in a weight ratio of 0.8 to 1.2:0.8 to 1.2 and then extracting at 90 to 110°C for 10 to 30 minutes;
[0036] (2) Step of adding the plant extract prepared in step (1) to 30 to 70 g of hemp seeds and soaking them at 20 to 25°C for 20 to 28 hours;
[0037] (3) A step of preparing a hemp medium by adding 1 to 10 g of raw sugar and 20 to 40 mL of water to the hemp seeds soaked in step (2) above and sterilizing at 110 to 130°C for 20 to 40 minutes; and
[0038] (4) It may include a step of inoculating Cordyceps militaris spores into the hemp medium manufactured in step (3) above and culturing them at 19 to 21°C for 5 to 10 weeks.
[0039] Most specifically
[0040] (1) A step of preparing a plant extract by adding 20 times (v / w) of water to a mixture of bottle gourd and lotus seed in a 1:1 weight ratio and then extracting at 100°C for 20 minutes;
[0041] (2) Step of adding the plant extract prepared in step (1) to 50 g of hemp seeds and soaking them at 20 to 25°C for 24 hours;
[0042] (3) A step of preparing a hemp medium by adding 5 g of raw sugar and 30 mL of water to the hemp seeds soaked in step (2) and sterilizing at 121°C for 30 minutes; and
[0043] (4) It may include a step of inoculating Cordyceps militaris spores into the hemp medium manufactured in step (3) and culturing them at 19 to 21°C for 6 to 8 weeks.
[0044] The present invention also provides Cordyceps militaris cultured by the above method.
[0045] The methanol extract of Cordyceps militaris of the present invention is characterized by containing 4 to 7 mg / g and 34 to 38 mg / g of cordycepin and adenosine, respectively.
[0046] The present invention also provides a Cordyceps militaris extract prepared by adding a solvent to the Cordyceps militaris and then extracting it, wherein the extract contains 4 to 7 mg / g of cordycepin and 34 to 38 mg / g of adenosine, respectively.
[0047] The method of extracting cordycepin and adenosine from Cordyceps militaris of the present invention may specifically include a step of adding 2 to 3 times (v / w) a 40 to 60% (v / v) methanol aqueous solution to Cordyceps militaris powder obtained by drying Cordyceps militaris and then pulverizing Cordyceps militaris and performing ultrasonic extraction at 20 to 30°C for 50 to 70 minutes, and more specifically, a step of adding 2.5 times (v / w) a 50% (v / v) methanol aqueous solution to Cordyceps militaris powder obtained by drying Cordyceps militaris and then pulverizing Cordyceps militaris and performing ultrasonic extraction at 25°C for 60 minutes.

[0084]
[0085] 1. Materials
[0086] The applicant of the present invention, 'Jang Sun-beom', holds a cannabis cultivation license, and the cannabis (Cannabis sativa L.) used was grown in a licensed facility.
[0087] The hemp variety used in the present invention is 'IH3', which is an industrial hemp with a hallucinogenic THC (Δ9-Tetrahydrocannabinol) content of 0.11%, which is significantly lower than the 1.74% of conventional varieties, and a hallucinogenic inhibitory CBD (Cannabidiol) content of 2.14%, which is higher than the 0.58% of conventional varieties. Therefore, although the IH3 variety is an industrial hemp with a THC content of less than 0.11%, no THC component was detected during the processing (sterilization, cultivation) process, making it safe. The applicant requested the Gyeongsangbuk-do Institute of Health and Environment to analyze the THC content of hemp seeds, and the result was determined to be suitable for use.
[0088] In addition, the Cordyceps militaris used in the present invention was obtained from a mushroom cultivation bank.
[0089]
[0090] Example 1. Evaluation of Cordyceps militaris growth according to the amount of hemp seed soaking and water addition.
[0091] In a preliminary experiment, the inventor of the present invention found that when hemp seeds were used as a medium without being soaked, the Cordyceps militaris fruiting bodies were not properly cultured, so hemp seeds were soaked in water and used as a medium.
[0092] Hemp seeds with the outer hulls removed can be used as a medium without soaking them in water, but when sterilized, they do not take the form of cooked rice, but rather a rice cake-like state with a film formed, so there are no air gaps (pores) for the Cordyceps militaris to penetrate, and the Cordyceps militaris did not grow well. In addition, when hemp seeds and brown rice were used as a medium in a 25:25 ratio, the Cordyceps militaris had difficulty penetrating and various bacteria proliferated easily (Fig. 1), so the hemp seeds were used without removing the outer hulls.
[0093] First, 50 g of hemp seeds were soaked in water at 20-25℃ for 48 hours, then taken out and washed in running water. 0 mL, 10 mL, 20 mL, and 30 mL of water were added to the seeds, respectively, and sterilized at 121℃ for 30 minutes to prepare a hemp medium. 20 mL of Cordyceps militaris liquid spawn was inoculated into the prepared hemp medium, and cultured for 6 weeks under conditions of 19-21℃ and 80-90% relative humidity. As a result, when 10 mL of water was added, the most Cordyceps militaris fruiting bodies were generated, but when 30 mL of water was added, a film was formed on the top and fruiting bodies were not formed. As a result, it was confirmed that soaking hemp seeds for 48 hours was not suitable for culturing Cordyceps militaris liquid spawn (Fig. 2).
[0094] Next, hemp seeds were soaked in water for 24 hours, taken out, and water was added in amounts of 10 mL, 20 mL, and 30 mL, respectively, and sterilized at 121°C for 30 minutes to prepare hemp medium. Liquid spawn was inoculated into the prepared hemp medium under the same conditions and cultured. As a result, the occurrence of fruiting bodies increased as the amount of water added increased, but it was determined that only hemp seeds were insufficient in nutrients for the growth of Cordyceps militaris (Fig. 3).
[0095] Therefore, in the present invention, as a result of experiments conducted by pretreating hemp seeds and forming a medium using various materials, it was confirmed that when hemp seeds were soaked in an organic plant extract as in cultivation example 1 and an appropriate amount of organic raw sugar was added, Cordyceps militaris grew well and the functional components of the produced Cordyceps militaris were also enhanced.
[0096]
[0097] Example 2. Active ingredient content of Cordyceps militaris
[0098] The Cordyceps militaris extract of Manufacturing Example 1 and the Cordyceps militaris extract prepared by the method of Manufacturing Example 1 but using Cordyceps militaris of Comparative Example 1 instead of Cultivation Example 1 were subjected to experimental analysis to compare the contents of cordycepin and adenosine. The detection method for cordycepin and adenosine was performed using high performance liquid chromatography (HPLC) equipped with a UV DAD (Diode Array Detector) to measure the contents of the main components.
[0099] In the step of measuring the above sample, the retention time of the peak, which is the measurement result, was measured using the elution time and absorbance of the peak component using the analysis equipment, high-performance liquid chromatography (HPLC). The absorbance was performed at 260 nm, and a comparative evaluation was performed based on whether the retention times of the main components in the standard solution and the sample solution were consistent. Acetonitrile and an aqueous solution containing 0.5% acetonitrile were used as the mobile phase. The mobile phase mixing ratio composition was 10% acetonitrile and 90% aqueous solution in the initial stage, and the separation of cordycepin and adenosine was confirmed by adjusting the mixing ratio of acetonitrile by time unit. The separation of each component was performed using a reversed-phase C18 column, and the column temperature was set to 35℃ and the injection amount was set to 15 ㎕.
[0100] Active ingredient content of Cordyceps militaris extract (mg / g) Cordyceps militaris type Cordyceps militaris Cultivation example 15.5236.19 Comparative example 1 Not detected 8.04
[0101] As a result, cordycepin was not detected in the extract using Cordyceps militaris of Comparative Example 1, but cordycepin was detected in the extract of Cordyceps militaris of Cultivation Example 1, and it was confirmed that the adenosine content was increased by more than 4 times compared to Cordyceps militaris of Comparative Example 1. In addition, THC (Tetrahydrocannabinol), a hallucinogenic component of marijuana, was not detected in the cultivated Cordyceps militaris. These results suggest that the present invention can be a novel cultivation method capable of increasing the content of effective ingredients during Cordyceps militaris cultivation.
[0102]
[0103] Example 3. Sensory evaluation of Cordyceps militaris
[0104] A sensory evaluation was conducted using Cordyceps militaris of Cultivation Example 1 and Comparative Examples. After each Cordyceps militaris was blanched in boiling water, 30 sensory evaluation agents were asked to classify the aroma, taste, texture, and overall preference, and the 7-point preference scale was used to indicate 1 point: very bad, 4 points: bad, 3 points: average, 4 points: good, 5 points: very good. The average was calculated and compared in Table 2 below.
[0105] Sensory evaluation of Cordyceps militaris according to substrate type Type Aroma Taste Texture Overall preference Cultivation example 16.46.26.06.2 Comparative example 14.84.24.64.4 Comparative example 25.05.05.25.0 Comparative example 33.63.03.03.2 Comparative example 45.45.65.25.4 Comparative example 55.45.65.45.6
[0106] As a result, Cordyceps militaris of Comparative Example 3, which was cultured on a medium using only hemp seeds, showed low scores in all items, and in a preliminary experiment, Cordyceps militaris did not grow well on a medium using only hemp seeds (Comparative Example 3), so it was not suitable for growth. On the other hand, Cordyceps militaris cultured by the method of Cultivation Example 1 showed the highest scores in all items including aroma, taste, and texture, indicating that the Cordyceps militaris culture conditions of Cultivation Example 1 were most suitable for consumer preferences.
