[0002] The process of growth, proliferation of eukaryotic cells involves the mother cell producing two identical daughter cells through mitosis of the cell′s chromosomes by accurately replicating its genome which contains genetic information. This process of cell proliferation and division is called a cell cycle, which includes the entire process from the completion of one division to the completion of the next division. The cell cycle comprises four growth stages: the G1 phase, where proteins, RNAs and the like are synthesized in large quantities after mitosis, the S phase, where DNA is synthesized and replicated, the G2 phase, the preparatory stage before mitosis, and the M phase, where cells undergo mitosis. Depending on the condition and needs of the cell, the cell decides to divide and proliferate, or stop, through the cell cycle. Cell proliferation and division must maintain its genetic information intact and correct. Whether or not to enter the next phase of the cell cycle until the completion of the entire cell cycle is ensured and completed by multiple checkpoints during the cell cycle process.

[0293] Figure 6 shows the TGA and DSC thermograms of the crystal form D. The thermal analysis showed a 4.0%weight loss when heated to 150 ℃, 2 endothermic peaks (peak temperature) at 155.1℃ and 277.3℃, and 1 exothermic peak (peak temperature) at 192.7 ℃. In order to study the heat signal of the crystal form D before melting, a heating test was performed, which showed that the crystal form D remained unchanged after heating to 130℃ and cooling to room temperature, and then changed to the crystal form B after heating to 200℃ and cooling to room temperature. The variable temperature XRPD test of the crystal form D showed that the crystal form D did not change after 20 minutes of nitrogen purging, but changed to the crystal form M after heating to 130℃. Combined with the results of TGA, nuclear magnetic resonance (NMR) and heating / variable temperature XRPD tests, it was speculated that the crystal form D was a hydrate, which was dehydrated and turned into anhydrous crystal form M after heating under nitrogen purging, and then the crystal form M reabsorbed water and returned to the crystal form D after cooling and contact with air.

[0295] The crystal form E is obtained by the anti-solvent addition in CHCl3 / MTBE system using the compound of formula I as the starting material. XRPD was used to characterize the crystal form E, and the XRPD diffraction peak data were shown in Table 4 below, and the XRPD spectrum was shown in Figure 7.

[0297] Figure 8 shows the TGA and DSC thermograms of the crystal form E. The thermal analysis showed a weight loss of 2.2%when the sample of the crystal form E was heated to 120℃, a weight loss of 9.3%when further heated to 200℃, 2 endothermic peaks at 171.7 and 277.6℃ (peak temperature) , and 1 exothermic peak at 189.2 ℃ (peak temperature) . The NMR results showed that the molar ratio of the residual solvent CHCl3 to the compound of formula I was 0.1 (corresponding to 2.0%weight loss) , and the molar ratio of the residual solvent MTBE to the compound of formula I was 0.22 (corresponding to the weight loss of 3.2%) . In order to study the DSC thermal signal and the great TGA weight loss of the crystal form E, it was heated to 120℃ and 200℃ respectively, then cooled to room temperature, and XRPD characterization was performed, and the results showed that after the crystal form E was heated to 120℃ and cooled to room temperature, the crystal form did not change, when it was heated to 200℃ and cooled to room temperature, the crystal form E changed to the crystal form B. The NMR results showed that the residual solvent remained unchanged after the crystal form E was heated to 120℃, and it was speculated that the stepped weight loss from 120℃ to 200℃ on the TGA mainly corresponded to the removal of the organic solvent. Since when the crystal form E sample was heated to 120℃, it remained unchanged, but when heated to 200℃, the crystal form changed, it was speculated that the removal of organic solvent caused the crystal transformation, and considering that the crystal form E is obtained in the MTBE-containing system, it was speculated to be an MTBE solvate.

[0319] Figure 18 shows the TGA / DSC thermograms of the crystal form J. The thermal analysis showed a weight loss of 2.4%when the crystal form J was heated to 120℃, a weight loss of 6.8%when further heated to 200℃, and three endothermic peaks at 134.9, 161.8 and 277.9℃ (peak temperature) . The NMR results showed that the molar ratio of the residual solvent toluene to the compound of formula I was 0.58 (corresponding to a weight loss of 8.6%) . In order to study the DSC thermal signal and the great TGA weight loss of the crystal form J, the heating test was carried out. XRPD results showed that when the crystal form J was heated to 120℃ and cooled to room temperature, it was converted to the crystal form N. When the crystal form J was heated to 145℃and cooled to room temperature, it was converted to a mixture of the crystal form N and B, when the crystal form J was heated to 180℃ and cooled to room temperature, it was converted to the crystal form B. According to the characterization and heating test results of the crystal form N, it was speculated to be a toluene solvate. Since the crystal form N is obtained by heating the crystal form J, and the crystal form J had a higher toluene residue, it was speculated that the crystal form J is also a toluene solvate.

[0323] Figure 20 showed the TGA / DSC thermograms of the crystal form K. The thermal analysis showed a weight loss of 2.6%when heated to 100℃ and a weight loss of 8.6%when further heated to 180℃, and DSC showed that the sample had 2 endothermic peaks at 131.2 and 275.9℃ (peak temperature) . The NMR results showed that the molar ratio of the residual solvent IPAc to the compound of formula I was 0.41 (corresponding to a weight loss of 6.9%) , and no residual solvent THF was detected. In order to study the DSC thermal signal and the great TGA weight loss of the crystal form K, it was heated to 100℃ and 180℃ respectively and then cooled to room temperature, and XRPD characterization was performed, and the results showed that after the crystal form K was heated to 100℃ and cooled to room temperature, the crystal form did not change, and after the crystal form K was heated to 180℃ and cooled to room temperature, it was converted to the crystal form B. The NMR results showed that the residual solvent remained unchanged after the crystal form K was heated to 100℃, indicating that the stepped weight loss from 100℃ to 180℃ on TGA mainly corresponded to the removal of IPAc solvent. Since when the crystal form K sample was heated to 100℃, it remained unchanged, but when heated to 180℃, the crystal form changed, it was speculated that the removal of IPAc solvent caused the crystal transformation. Thus, it was speculated to be an IPAc solvent.

[0343] Figure 29 shows the TGA / DSC thermograms of the crystal form O. The thermal analysis showed a weight loss of 29.7%when the sample was heated to 200℃, with 2 endothermic peaks at 157.8 and 267.3℃ (peak temperature) and 1 exothermic peak at 150.5℃ (peak temperature) . The NMR results showed no solvent residues and some additional signals were observed. The HPLC test results showed that the HPLC area purity of the sample was only 90.8%, indicating that the sample had been degraded, and the additional signal observed on the NMR may correspond to the degradation products.

[0345] In order to study the transformation relationship between each anhydrous crystal form and its hydrate, three anhydrous crystal forms A, B, M and one hydrate crystal form D discovered in the experiment were tested in suspension competition (the crystal form M was not used in suspension competition study because it was only obtained in the variable temperature XRPD test and was transferred after contact with air) .

[0346] (1) Study on the transformation relationship between anhydrous crystal forms

[0347] Suspension competition tests were carried out on anhydrous crystal forms A and B. A certain amount of the crystal form A was weighed into an HPLC vial and 1.0 mL of different solvents were added. After suspension stirring for 1 hour at room temperature, samples were filtered using a 0.45 μm pore size PTFE membrane, and the filtrate was transferred to HPLC vials containing the crystal form A and B solids (~5 mg each) , where they were tested for solid XRPD after suspension stirring at room temperature / 50 ℃, respectively. The results were summarized in Table 16 below. The results showed that the crystal form A was converted to the crystal form B at room temperature and at 50℃ in different solvents, indicating that the crystal form B was more thermodynamically stable than the crystal form A at room temperature to 50℃.

[0349] (2) Study on the transformation relationship between anhydrous crystal form and hydrate

[0350] Anhydrous crystal form B and hydrate crystal form D, which were more stable at room temperature, were selected for suspension competition test under different water activities. A certain amount of the crystal form A was weighed into an HPLC vial and 1.0 mL of solvent with different water activities were added. After suspension stirring for 1 hour at room temperature, the samples were filtered using a 0.45 μm pore size PTFE filter, and the filtrate was transferred to HPLC vials each containing ~5 mg of the crystal form B and D solids, and the solid XRPD was tested after suspension stirring at room temperature. The results were summarized in Table 17 below. The results showed that anhydrous crystal form B was obtained at water activity of 0.8 and less, and hydrate crystal form D was obtained in pure water, indicating that the key water activity point for the mutual conversion of anhydrous crystal form B and hydrate crystal form D at room temperature was in the range of 0.8~1.

[0355] To a mixture of DMSO (235.0 kg, 4.0 V) , ethyl 4-chloro-2-methylthiopyrimidine-5-carboxylate (the compound of SM1, 53.0 kg, 227.77 mol, 1.0eq. ) and Na2CO3 (29.4 kg, 277.4 mol) is stirred at room temperature is added the solution ofN-tert-butoxycarbonyl-1, 2-ethylenediamine (the compound of formula SM2, 37.2 kg, 232.20 mol) in DMSO (58.3 kg, 1.0V) at 25 ℃ and stirred for 2 hours. After the addition of H2O (795.0 kg, 15.0 V) and EtOAc (715.5 kg, 15.0 V) to the mixture and phases separation, the organic phase is collected, washed twice with water (53.0 kg, 1.0 V) and concentrated to 4 V under reduced pressure at no more than 40 ℃. 4 N HC1 solution in isopropyl acetate (448.0 kg, 9.5V) is added slowly to the concentrated organic solution at no more than 20 ℃, and stirred for at least 16 hours. The mixture was filtered and the filter cake is washed with ethyl acetate. The filter cake is dried under reduced pressure at 45 ℃ for 16 hours to obtain ethyl 4- ( (2-aminoethyl) amino) -2- (methylthio) pyrimidine-5-carboxylate dihydrochloride (the compound of Formula 2, 70.65 kg, white solid, purity 99.13%, yield 89.4%) .

[0379] ADP-Glo Kinase reagent (Promega, #V9103) was used with reaction buffers of 50 mM HEPES, 1 mM EGTA, 10 mM MgCl2, 0.01%Brij 35, 2 mM DTT (pH 7.5) . The mother liquor of the compound of formula I was firstly diluted in series to 10 concentrations in DMSO at a 1: 3 ratio, then 0.05 μL of the diluted compound was added to a 384-well plate (Greiner, #784075) using an Echo 665 instrument, and 2.5 μL of Weel enzyme (Carna, #05-177) solution was added and centrifuged at 1000 rpm for 1 minute. After incubation at 25℃ for 10 minutes, 2.5 μL of ATP / substrate mixture was added to initiate the reaction and reacted at 25℃ for 60 minutes. Subsequently, 4 μL of an ADP-Glo reagent was added and incubated at 25℃ for 40 minutes, followed by the addition of 8 μL ofa kinase detection reagent and incubation at 25℃ for 40 minutes. Finally, the chemiluminescence value was measured on the PHERAstar FSX instrument from BMG.

[0383] The newly recovered LoVo cells were cultured to the third generation, and the growth state was good and the confiuency was about 90%, and they were used for experiments. The LoVo cells was digested with trypsin, centrifugated at 800 rpm for 5 min, the supernatant was discarded, the residue was resuspended with fresh medium, counted, seeded at a density of 6, 000 cells per well onto a 96-well cell culture plate, and placed overnight in a 37℃ 5%CO2 incubator. The mother liquor of the compound of formula I was diluted in series to 8 concentrations in DMSO at a ratio of 1: 3 and 1: 10 (the last concentration was a DMSO negative control) : 10 μM, 3.3 μM, 1 μM, 0.33 μM, 0.1 μM, 0.033 μM, 0.01 μM, 0 μM (the DMSO final concentration was 1‰) . 5 μL of each concentration was added to 120 μL of medium (25-fold dilution) and mixed with shaking. The cells cultured overnight were taken and the medium was removed, 195 μL of fresh medium was added to each well, and 5 μL of diluted medium containing the corresponding concentration of tested compound was added, and then the plate was incubated in a 37℃ 5%CO2 incubator for 3 days. After removing the stock solution, 90 μL of fresh serum-free 1640 medium was added to each well, and then 10 μL of CCK-8 detection reagent was added to each well, and after 2 hours of incubation, it was placed in a multi-function reader to read the absorbance value (OD value) at the wavelength of 450 / 650nm. The data were analyzed with the software Graph Pad Prism 5.0, and the inhibitory activity of the compound on cell proliferation was plotted as the coordinates of cell viability and the concentration of the compound. The IC50 values were fitted with an S-shaped dose-response curve equation as: Y= 100 /  (1+10^ (LogC-LogIC50) ) , where C is the concentration of the compound.
