Description

NON-INVASIVE TEST KIT, DIAGNOSTIC SYSTEM AND METHODS FOR TARGET HEALTH ASSESSMENTCross-Reference to Related Applications:

[0001] The present application claims the priorities from the U.S. provisional patent application serial number 63 / 557,602 filed February 26, 2024, and the disclosure of which is incorporated herein by reference in its entirety.Field of the Invention:

[0002] The present invention generally relates to at least the fields of animal health monitoring, psychological health assessment, and so forth. Background of the Invention:

[0003] Conventional approaches to assessing the health of animals (e.g., pets, livestock, zoo animals) predominantly involve invasive blood tests conducted in veterinary clinics or research facilities. These procedures can induce discomfort and stress in the animals, thereby exacerbating anxiety levels and potentially leading to complications. In addition, the dependency on veterinary clinics for health assessments imposes significant financial burdens on animal caregivers. The accumulated costs from frequent clinic visits and blood tests present a considerable barrier, hindering access to routine health monitoring and preventive care. Besides, these procedures can induce stress in animals, and physical restraint poses risks to handlers, especially with dangerous species like big cats (e.g., lions) .

[0004] Caregivers are increasingly seeking convenient, non-invasive solutions to monitor the well-being of their pets from the comfort of their own homes. However, there is a noticeable gap in the market, with no existing products capable of simultaneously collecting animal saliva and detecting biomarkers. This lack of innovation hampers proactive health management and limits the ability to intervene in a timely manner.

[0005] Caregivers also seek non-invasive solutions for large-scale or high-risk settings, such as detecting infectious diseases in livestock or monitoring stress in zoo animals. However, no existing products safely collect saliva from uncooperative or dangerous animals while enabling biomarker detection.

[0006] The current state of the art fails to realize non-restrained health monitoring. Notably, the prior art lacks a comprehensive solution that integrates both saliva collection and biomarker detection in a non-invasive, animal-friendly manner. For example, in US 2023 / 0393155 A11, while lateral flow immunoassay devices are discussed, there is no consideration for the design of the sample collection component, nor does it feature the test strip cassette design. Furthermore, Damián, Juan Pablo, et al. 2 describes a sample collection method using a cotton roll only.

[0007] Prior art fails to address species-specific challenges. For examples, frequent testing for livestock in zoonotic diseases (e.g., avian influenza) requires restraining large animals like cattle, which increases injury risks to handlers and stress-induced biomarker inaccuracies.Summary of the Invention:

[0008] Existing methodologies tend to prioritize physical health assessment, thus failing to adequately address potential behavioral or emotional concerns. A burgeoning demand exists for a holistic evaluation of animals' health conditions, encompassing both physiological and psychological well-being.

[0009] In an aspect, the present invention provides a non-invasive test kit for animal biomarker detection. The kit includes a collection device for saliva collection and a plug-in test strip cassette for biomarker detection. The collection device is configured as a toy, pacifier, or feeding apparatus, etc. The collection device includes a shell with one or more openings and a collection core embedded in the shell for saliva absorption and filtration. The plug-in test strip cassette includes a housing designed to securely hold one or more test strips.

[0024] In one embodiment, the one or more test strips are designed in a multi-channel format and share a common sample pad.

[0025] In one embodiment, a multi-channel test strip configuration enables the simultaneous detection of at least three distinct biomarkers.

[0026] In one embodiment, the conjugation pad contains a specific antibody labeled with nanoparticles (e.g., gold nanoparticles or fluorescent-labelled nanoparticles) .

[0027] In one embodiment, the collection device is designed to retain a saliva volume of at least 0.5 mL within the collection chamber for accurate biomarker detection.

[0028] In one embodiment, the shell is reinforced with bite-resistant materials (e.g., thermoplastic polyurethane) for use with large or dangerous animals, such as tigers, and lions.

[0029] In another aspect, the present invention provides a diagnostic system, which includes a non-invasive test kit, and an integrated detection module. The non-invasive test kit is designed to collect saliva from a target and detect one or more biomarker present within the saliva. The integrated detection module includes colorimetric, fluorescent, or electrochemical sensors. The integrated detection module is operably connected to a digital analysis platform for quantification and data processing.

[0030] In one embodiment, the one or more test strips include multiple test lines arranged with antibody gradients, configured to generate signal intensities corresponding to analyte concentration levels.

[0032] In one embodiment, the target includes an animal.

[0033] In one embodiment, the electronic device may have a processor, a display screen, and a storage unit, and being configured to generate reports, provide real-time feedback, perform data analysis based on the test results, and store the results in cloud-based or local storage for subsequent review and monitoring.

[0034] In another aspect, the present invention provides a non-invasive method for target health assessment. The method includes preparing a collection device; preparing a plug-in test strip cassette for biomarker detection, wherein the plug-in test strip cassette comprises a housing designed to securely hold one or more test strips; collecting saliva by using the collection device; analyzing the collected saliva for multiple biomarkers simultaneously using a plug-in lateral-flow test strip cassette; displaying biomarker results on an integrated detection module comprising colorimetric, fluorescent, or electrochemical sensors. The integrated detection module is operably connected to a digital analysis platform for quantification and data processing. The non-invasive method simultaneous collects animal saliva and detects one or more biomarkers.

[0035] In one embodiment, the step of analyzing the collected saliva for multiple biomarkers simultaneously using a plug-in lateral-flow test strip cassette includes sliding the housing along a sliding groove toward the collection chamber, thereby immersing a sample pad of a lateral-flow test strip into the saliva collected in the collection chamber; and conducting a lateral-flow testing.

[0037] In one embodiment, the one or more test strips are mounted on the test strip holder at an initial position, and the sample pad is covered by the cassette cap and prevented from pre-contact with saliva sample.

[0038] In one embodiment, the expected saliva volume for one test is approximately 0.5-1 mL.

[0039] In one embodiment, the target includes an animal selected from the group consisting of companion animals, working animals, livestock, or zoo animals.

[0040] The present invention solves the mentioned problems through the following solutions: (1) Non-invasive and convenient approach for saliva collection: The non-invasive test kit provides a non-invasive alternative to traditional  blood tests. Specifically, animal saliva, abundant in biomarkers, is used as the testing medium instead of invasive procedures. The collection process is made engaging and comfortable for animals through the design of the animal interactive products. (2) Cost-effective methods: By enabling domestic testing, the invention reduces the need for frequent  visits to veterinary clinics, thereby lowering expenses for pet owners, animal caregivers, farmers, and zookeepers. The cost-effective nature of the test kit makes regular health monitoring more accessible. (3) Comprehensive assessment to multiple biomarkers: Animal saliva contains a wide range of biomarkers that indicate both  physical diseases (e.g., influenza virus, parvovirus, rabies virus) and physiological status (e.g., stress level, nutrition assessment) . The test kit allows for the detection of these biomarkers, providing a more comprehensive assessment of animals’ health condition. (4) Domestic testing options: The non-invasive test kit enables animal caregivers to perform health  evaluations conveniently at home. The collection process is designed to be user-friendly, encouraging the caregivers to take a proactive role in their animals’ health management. The plug-in lateral-flow test strip cassette simplifies the testing process, making it accessible to a wide range of users.Brief Description of the Drawings:

[0049] In the following description, an extended design of animal-based products incorporating a non-invasive test kit is set forth as preferred examples. It will be apparent to those skilled in the art that modifications, including additions and / or substitutions may be made without departing from the scope and spirit of the invention. Specific details may be omitted so as not to obscure the invention; however, the disclosure is written to enable one skilled in the art to practice the teachings herein without undue experimentation.

[0050] There are abundant biomarkers in animal saliva, indicating certain diseases (e.g. influenza virus, parvovirus, rabies virus, etc. ) and physiological status (e.g. stress level, nutrition assessment, etc. ) . The present invention provides a test kit for animal saliva collection and biomarker detection. FIG. 1 illustrates the overall structure of the test kit, which includes several components, including a shell for animal biting and chewing; a sample collection component comprising a collection core for saliva absorption and filtration, and a collection chamber positioned within the collection core for saliva collection; and a test strip cassette for biomarker detection, and at least one test strip is / are embedded into the test strip cassette.

[0051] The shell is designed for animal interaction. it is made from materials such as natural rubber or thermoplastic rubber, ensuring flexibility, durability, and safety. The shell includes multiple strategically placed openings to facilitate saliva entry during the animal’s biting or chewing actions (FIGs. 2A-2C) . The collection core and collection chamber are positioned inside the shell. The collection core absorbs, filters, and directs saliva into the collection chamber. The chamber is designed to ensure a sufficient amount of saliva is retained for testing purposes.

[0052] When being bitten or chewed, animals’ saliva will flow through the openings on shell and be absorbed and filtered by the collection core, and eventually collected in the collection chamber. Subsequently, the liquid collected chamber can be integrated into the test strip cassette. The test strip cassette is an external plug-in component containing the lateral-flow test strip, which performs biomarker detection based on a detection principle (colorimetric, fluorescent or electrochemical) .

[0053] For instance, the collection chamber is located within the collection device. The porous material absorbs and filters the saliva during biting or chewing, facilitating efficient collection of the saliva sample. As the animal bites or chews the rubber frame, the saliva is soaked into the porous material and transported towards the collection chamber. Next, the biomarker detection is achieved using the plug-in test strip cassette (FIG. 3) .

[0054] Referring to FIG. 4, the test strip cassette has a cassette substrate contains a sliding groove to facilitate the precise movement of the test strip, a test strip holder securely positions the lateral-flow strip during testing, and a transparent cassette cap protects the sample area and result zone, ensuring no contamination or premature exposure. Each component is constructed from durable, non-toxic materials. Safety mechanisms, such as sealed edges and snap-fit designs, prevent sample leakage. The cassette can be configured for single-use or multi-use applications, depending on user preference.

[0055] Moreover, the test strips are mounted in the test strip cassette with a sliding groove to perform time-sensitive detection according to certain biomarkers. Each of the one or more test strips has a sample pad, an absorbent pad, a conjugation pad, a test line region, a control line region, and a backing card, as shown in FIG. 5. The sample pad serves as the initial contact point for the sample, where the sample is applied. The absorbent pad functions to absorb excess sample and helps control the fluid flow through the test strip, ensuring the sample moves efficiently across the test areas. The conjugation pad contains labeled reagents or antibodies that bind to the target analyte in the sample, facilitating the detection process by forming a visible signal when the analyte is present. The test line region shows the area where the specific analyte-antibody complex is captured, forming a visible line (often due to a color change) when the target analyte is detected. The control line region is the area where a separate antibody or reagent is fixed, providing a visual control to ensure the test strip has worked properly and the sample has flowed through the strip correctly. The backing card provides structural support for the test strip and maintains the integrity of the strip during use, ensuring it remains in a flat, stable position for optimal performance.

[0056] For quantitative analysis, the test strip employs: · Multi-line gradients: Anti-analyte antibodies at 10 / 25 / 50 ng / mL thresholds. · Digital integration: A digital format that converts test line intensity (e.g.,  RGB analysis) to biomarker concentrations, referencing preloaded calibration curves

[0057] There is a fluidic channel from the surface of the shell to the collection chamber to plug in the test strip to perform biomarker detection.

[0058] Before performing biomarker detection, the test strip is mounted on the test strip holder at the initial position, where the sample pad of test strip is covered by the cassette cap and prevented from pre-contact with saliva sample. Then, sliding the test strip holder along the groove immerses the sample pad into the collected saliva in the chamber, triggering the testing process. This design ensures precise timing and controlled sample interaction. Users are instructed to wait 10 -15 minutes for the results to develop before analysing the data.

[0059] In certain embodiment, the shell is designed with a round shape and features circular openings with an average diameter ranging from 2 mm to 8 mm.

[0060] In another embodiment, the openings on the shell can be designed as various shapes and sizes.

[0061] Additionally, the shell can be customized in various colors, textures, and shapes to enhance animal engagement. Maintenance features, such as easy-to-clean surfaces and replaceable components, are also incorporated for user convenience.

[0062] In one embodiment, the collection core is made of highly absorbent material, such as sponge or any non-toxic and food-grade certified material suitable for absorption.

[0063] In one embodiment, the expected saliva volume for one test is approximately 0.5-1 mL.

[0064] In one embodiment, the test strip can be designed in a multi-channel format. Different channels are functionalized with specific antibodies and share a common sample pad, allowing for multiplexed testing (FIG. 5) .

[0065] In this invention, the biomarker detection is achieved by a conventional lateral-flow test strip, based on the principle of gold-colloid based sandwich immunoassay. When the sample pad contacts with saliva sample, the saliva sample will be soaked and flow to the conjugation pad containing specific antibody labelled gold nanoparticles. The biomarker conjugated nanoparticles will continue across to the test and control lines successively. The existence and concentration of certain biomarker in saliva sample can be detected and measured by comparing the color of the test line and the control line.

[0066] In certain application of multiple biomarker detection, the test strip can be designed for multiplexed testing in multi-channel format, in which different channels are functionalized with modification and conjugation of certain antibodies respectively and share with a common sample pad.

[0067] In one embodiment, the tested animal fall into broad animal categories, such as dogs, cats, rabbits, guinea pigs, cattle, birds, hamsters, ferrets, reptiles, fish, lions, tigers etc.

[0068] In another aspect, the present invention provides a method for simultaneous collecting animal saliva and detecting one or more biomarker. The method includes preparing said kit; sliding a test strip holder along the sliding groove toward a collection chamber, thereby immersing a sample pad of a lateral-flow test strip into saliva collected in the collection chamber; conducting a lateral-flow testing; and reading out results through direct observation or image capturing and processing.

[0069] The present invention revolutionizes the conventional approach of disease-related biomarker screenings, which typically rely on invasive blood tests conducted in veterinary clinics. This innovative method enables pet owners, animal caregivers, farmers, and zookeepers to perform domestic testing on animal common diseases, physiological indicators, etc., using a non-invasive and friendly manner.

[0070] During detection, by sliding the test strip holder along the sliding groove on the cassette substrate toward collection chamber, the sample pad of the test strip is immersed into the saliva sample present in the collection chamber, initiating the lateral-flow testing, as shown in FIG. 6.

[0071] The result read-out is achieved through either direct observation or image capturing and processing. In case of qualitative virus detection, the colors of the test line and control line on the test strip are compared to determine a positive or negative result.

[0072] For quantitative detection (e.g. hormone level, cortisol, oxytocin, vasopressin, etc. ) , images are captured with a camera and processed using grey processing and grey value extraction. The grey value of the test line is normalized by that of the control line and fitted with a built-in standard curve to calculate the quantitative level of the biomarker (FIG. 7) .

[0073] Compared to traditional diagnostic methods, which often rely on invasive procedures or animal testing to assess target health, the non-invasive method proposed in this invention represents a significant advancement in both scientific and ethical terms. Current diagnostic systems and health assessments frequently require the use of animals to simulate human conditions, subjecting them to potential harm and suffering. In contrast, the non-invasive approach outlined herein offers a viable alternative that reduces demand for animal testing, aligning with the growing global movement towards more humane and ethical practices in medical research and diagnostics.

[0074] By simplifying the testing process and reducing stress on animals, this invention promotes proactive health management and strengthens the bond between owners and their animals. The process to collect animal saliva and the design of the collection device are considered as novelties.

[0075] This invention not only provides more accurate, human-relevant data but also reduces the environmental and ethical concerns associated with animal testing. By minimizing the reliance on animal models, this invention contributes to the broader scientific and regulatory trend of adopting alternatives to animal experimentation, thus meeting both regulatory requirements and public expectations for responsible, compassionate research methodologies.

[0076] EXAMPLE

[0077] Example 1

[0078] Fabrication of a collection device

[0079] In this example, a saliva collection device is fabricated using a natural rubber shell molded into a spherical shape with circular openings (2-8 mm in diameter) to facilitate saliva entry. A food-grade certified sponge serves as the collection core, designed to absorb and filter saliva efficiently. The sponge is embedded within the shell, aligning its porous surface with the openings, and featured a hollow collection chamber capable of holding at least 1 mL of saliva. A fluidic channel connects the sponge surface to the chamber, enabling integration with a lateral-flow test strip cassette. The device is assembled using non-toxic, water-resistant adhesive, ensuring durability and functionality during repeated biting and chewing actions. Stress and absorption tests confirms the device's resilience and effectiveness, while usability tests demonstrate its friendly design. Customization options, including variations in shape, texture, and color, further enhanced engagement and adaptability to different preferences. For dangerous species (e.g., lions) , the shell is fortified with reinforcement elastic materials and designed for remote deployment (e.g., attached to feeding apparatus) . Saliva collection occurs during natural chewing behavior, eliminating direct handler contact.

[0080] Example 2

[0081] Diagnostic system for biomarker detection A diagnostic system is provided for monitoring the health status of an animal by  detecting biomarkers present in saliva. The system includes the non-invasive test kit described in the Example 1 for collecting and detecting biomarkers from the dog’s saliva. The system further includes an integrated detection module that contains colorimetric, fluorescent, or electrochemical sensors for sensitive and specific biomarker analysis. The integrated detection module is operably connected to a digital analysis platform, which facilitates real-time quantification, data processing, and result interpretation. The digital platform may be equipped with machine learning algorithms for enhanced biomarker trend analysis, remote health monitoring, and early disease detection. Additionally, the system may support wired or wireless connectivity to external electronic devices, allowing for seamless data transmission, storage, and sharing with veterinarians or pet owners for timely intervention and health management.

[0082] Example 3

[0083] Non-invasive method for target health assessment

[0084] Saliva collection reduces discomfort compared to invasive blood sampling. According to Vincent I.C. et al. 5, salivary cortisol levels in dogs correlate strongly with plasma cortisol concentrations. Salivary cortisol can reflect emotional stress even when behavioral signs are subtle or have subsided, making it a valuable tool for monitoring mental well-being. Overall, using saliva for cortisol measurement offers significant advantages in convenience, comfort, and sensitivity to emotional changes in dogs.

[0085] The foregoing description of the present invention has been provided for the purposes of illustration and description. It is not intended to be exhaustive or to limit the invention to the precise forms disclosed. Many modifications and variations will be apparent to the practitioner skilled in the art.
