The method of any of claims 2, 3, 5, 6, 8, 9, 11, 12, 14, 15, 17, or 18 wherein the anti- connexin Cx43 hemichannel activating antibody or fragment thereof comprises a light chain immunoglobulin variable region comprising: a) a complementarity determining region 1 (CDR1) comprising a sequence having at least 60% identity to a sequence set forth in SEQ ID NO: 49; b) a complementarity determining region 2 (CDR2) comprising a sequence having at least 60% identity to a sequence set forth in SEQ ID NO: 50; and / or c) a complementarity determining region 3 (CDR3) comprising a sequence having at least 60% identity to a sequence set forth in SEQ ID NO: 51.

27. The method of any of the preceding claims, comprising administering to the subject an effective amount an expression vector encoding the anti-connexin Cx43 hemichannel activating antibody.

28. The method of any of the preceding claims, wherein the anti-connexin Cx43 hemichannel activating antibody is administered in a pharmaceutically acceptable composition.

29. The method of any of the preceding claims, wherein the anti-connexin Cx43 hemichannel activating antibody is administered systemically.

30. The method of claim 29, wherein the anti-connexin Cx43 hemichannel activating antibody is administered intravenously, intradermally, intratumorally, intramuscularly, intraperitoneally, subcutaneously, or locally.

31. The method of claim 28, wherein the anti-connexin Cx43 hemichannel activating antibody is a humanized antibody.

32. The method of any of claims 1-26, further comprising administering at least a second therapeutic agent to the subject.

33. The method of claim 32, wherein the second therapeutic agent is a myostatin inhibitor, a MAS receptor agonist, or an Apelin receptor agonist.

34. The method of any of the preceding claims, wherein the anti-connexin Cx43 hemichannel activating antibody or fragment thereof binds to a Cx43 hemichannel.

35. The method of any the preceding claims, wherein the anti-connexin Cx43 hemichannel activating antibody or fragment thereof further comprises a tag sequence.

36. The method of any of the preceding claims, wherein the anti-connexin Cx43 hemichannel activating antibody or fragment thereof stimulates the opening of a Cx43 hemichannel.

37. The method of any of the preceding claims, wherein the fragment thereof is a Fab fragment, an Fab’ fragment or an F(ab’)2 fragment.

38. The method of claim 28, wherein the pharmaceutical acceptable composition is lyophilized.

39. The method of claim 3, 6, 9, 12, 15, or 18, wherein the anti-connexin Cx43 hemichannel activating antibody or fragment thereof comprises: a) a heavy chain immunoglobulin variable region comprising: i) a first complementarity determining region 1 comprising a sequence having at least 60% identity to SEQ ID NO: 19; ii) a second complementarity determining region 2 comprising a sequence having at least 60% identity to SEQ ID NO: 20; and iii) a third complementarity determining region 3 comprising a sequence having at least 60% identity to SEQ ID NO: 21; and b) a light chain immunoglobulin variable region comprising: i) a first complementarity determining region 1 comprising a sequence having at least 60% identity to SEQ ID NO: 49; ii) a second complementarity determining region 1 comprising a sequence having at least 60% identity to SEQ ID NO: 50; and iii) a third complementarity determining region 1 comprising a sequence having at least 60% identity to SEQ ID NO: 51.

40. The method of claim 39, wherein the anti-connexin Cx43 hemichannel activating antibody is a humanized antibody.

41. The method of any of claims 1, 3, 4, 6, 7, 9, 10, 12, 13, 15, 16, or 18, wherein the anti- connexin Cx43 hemichannel activating antibody or fragment thereof comprises a heavy chain immunoglobulin variable region comprising:a) a complementarity determining region 1 (CDR1) comprising a sequence having a single amino acid change compared to a sequence set forth in SEQ ID NO: 19; b) a complementarity determining region 2 (CDR2) comprising a sequence having a single amino acid change compared to a sequence set forth in SEQ ID NO: 20; and / or c) a complementarity determining region 3 (CDR3) comprising a sequence having a single amino acid change compared to a sequence set forth in SEQ ID NO: 21.

42. The method of any of claims 2, 3, 5, 6, 8, 9, 11, 12, 14, 15, 17, or 18, wherein the anti- connexin Cx43 hemichannel activating antibody or fragment thereof comprises a light chain immunoglobulin variable region comprising: a) a complementarity determining region 1 (CDR1) comprising a sequence having a single amino acid change compared to a sequence set forth in SEQ ID NO: 49; b) a complementarity determining region 2 (CDR2) comprising a sequence having a single amino acid change compared to a sequence set forth in SEQ ID NO: 50; and / or c) a complementarity determining region 3 (CDR3) comprising a sequence having a single amino acid change compared to a sequence set forth in SEQ ID NO: 51.

43. The method of claim 3, 6, 9, 12, 15, or 18, wherein the anti-connexin Cx43 hemichannel activating antibody or fragment thereof comprises: a) a heavy chain immunoglobulin variable region comprising: i) a first complementarity region 1 comprising a sequence having a single amino acid change compared to SEQ ID NO: 19; ii) a second complementarity region 2 comprising a sequence having a single amino acid change compared to SEQ ID NO: 20; and iii) a third complementarity region 3 comprising a sequence having a single amino acid change compared to SEQ ID NO: 21; andb) a light chain immunoglobulin variable region comprising: i) a first complementarity region 1 comprising a sequence having a single amino acid change compared to SEQ ID NO: 49; ii) a second complementarity region 1 comprising a sequence having a single amino acid change compared to SEQ ID NO: 50; and iii) a third complementarity region 1 comprising a sequence having a single amino acid change compared to SEQ ID NO: 51.

56. The method of any of the preceding claims, wherein the subject is not on a high fat diet.

57. The method of any of the preceding claims, wherein the subject is not obese or does not have type II diabetes.

58. The method of any of claims 10-11 or 19-57, wherein the muscle disease or musclewasting disorder is Duchenne muscular dystrophy, Becker’s muscular dystrophy, distal, Emery - Dreifuss, facio-scapulo-humeral, limb-girdle, myotonic, oculopharyngeal, sacropenia, fraility, polymyositis, muscle stem cell senescence, myotonia, nemaline myopathy, multi-minicore myopathy, centronuclear myopathy, metabolic myopathy, cachexia, spinal muscular atrophy, amyotrophic lateral sclerosis, critical illness myopathy, myasthenia gravis, dermatomyositis, inclusion body myositis, pompe disease (glycogen storage disease type II), McArdle disease (glycogen storage disease type V), carnitine palmitoyltransferase II deficiency, or inflammatory myopathy.
