COMPOSITIONS OF BIOACTIVE AGENTSFIELD OF THE INVENTION

[0001] The present invention relates to the field of pharmaceutical composition. In particular, the present invention relates to a pharmaceutical composition ( “RM02” ) comprising Compound A, Compound B, Compound C, and Compound D, as defined herein, and their use in treating a neurodegenerative disease (e.g., Alzheimer’s disease) and improving cognition or memory.BACKGROUND

[0002] In the United States there are currently 6.2 million individuals with Alzheimer’s disease (AD) . In addition to these with AD in the dementia stages, there are about 10 million individuals with mild cognitive impairment (MCI) (Cummings J et al, Translational Research &Clinical Interventions, 2022) . In Alzheimer’s disease, the main presenting symptoms are memory loss, difficulty completing familiar tasks, confusion with time or place and challenges in planning or solving problems et al. (Alzheimer’s Association) . There is no cure for Alzheimer’s disease nowadays. A total of 6 drugs were approved by FDA so far, including Aducanumab, Lecanemab, and Donanemab. All of the treatments only slow down the Alzheimer’s progress but failed to clearly stop progression. The need for therapy to prevent and improve the symptoms of Alzheimer’s disease is compelled by the rising number of affected individuals and the growing public health crisis posed by the cost of caring for all the world.

[0004] In some embodiments, there is provided a pharmaceutical composition comprising at least three components selected from the group consisting of: (i) Compound A, being a compound of Formula (A) , or a pharmaceutically acceptable salt thereof, wherein RA1 and RA2 are each independently H; C1-C6 alkyl optionally substituted with one or  more -OH; or -RAa, wherein RAa is a monosaccharide, disaccharide, or polysaccharide moiety; RA3 are each independently H or -ORAc, wherein RAc is a monosaccharide, disaccharide,  polysaccharide moiety, or a or C1-C6 alkyl optionally substituted with one or more -OH; RA4, RA5, RA6, RA7, and RA8 are each independently H or -ORAb, wherein RAb, at each  occurrence, is independently H or C1-C6 alkyl optionally substituted with one or more -OH; and is a single or double bond; (ii) Compound B, being a compound of Formula (B) , or a pharmaceutically acceptable salt thereof, wherein RB1 is H or C1-C6 alkyl; RB2 is -OH, C (O) RBa or C (O) ORBa, wherein RBa is H or C1-C6 alkyl; and RB3, RB4, RB5, and RB6 are each independently H, -OH, or -O- (C1-C6 alkyl) ; (iii) Compound C, being a compound of Formula (C) , or a pharmaceutically acceptable salt thereof, wherein: RC1 is H, C1-C6 alkyl optionally substituted with one or more halo or -OH; C (O) RCa;  C (O) ORCb; -P (O) (ORCc) (ORCd) ; or phenyl optionally substituted with one or more halo or NO2; wherein RCa, RCb, RCc, and RCd are each independently H; C1-C6 alkyl; or phenyl optionally substituted with one or more halo or NO2; and (iv) Compound D, being a compound of Formula (D) , or a pharmaceutically acceptable salt thereof, wherein RD1, RD2, RD4, and RD4 are each independently H or C1-C6 alkyl; and L is a branched or unbranched C1-C6 alkylene optionally substituted with one or more  halogen or -NH2.

[0106] Murine model APP / PS1 is double transgenic mouse expressing a chimeric mouse / human amyloid precursor protein (Mo / HuAPP695swe) and a mutant human presenilin 1 (PS1-dE9) , both directed to CNS neurons. Both mutations are associated with early-onset Alzheimer's disease. In these transgenic mice, expression of the human APP transgene is approximately 3-fold higher than endogenous murine APP. Amyloid plaque deposition starts at approximately six weeks of age in the neocortex and approximately three to four months in the hippocampus. Seven APP / PS1 transgenic mice testing groups were established, with five transgenic APP / PS1 mice in each group. Oral administration of these compositions was carried out over a period of two months. The study included five RM02 testing groups treated with five distinct RM02 compositions as prepared in the previous step, one control group treated with RM01 composition, and one control group with saline. Additionally, one control group of C57 wildtype (WT) mouse lacking the APP / PS1 mutations and given no treatments was also included in this study. The study was approved by appropriate administrative authorities. All mice were of similar age, weight, and health status, and were housed in a controlled environment with consistent temperature, humidity, and a 12-hour light / dark cycle, and were provide ad libitum access to food and water. The mice were monitored daily for signs of adverse reactions, changes in behavior, body weight, and food and water consumption.

[0107] Efficacy Assessment using Modified Morris Water Maze

[0108] The Morris water maze is a behavioral assay to measure spatial learning and memory. A modified version of the Morris water maze test, as previously described by Vorhees and Williams (Vorhees CV &Williams MT, 2006) , was used to evaluate the efficacy of RM02 compositions in the APP / PS1 murine model. A custom-written ANY-maze Behavioral Tracking software Escape was used to assess the time spent in the platform quadrant. Briefly, the time of each mouse spent in the correct quadrant of the maze was used as an indicator for cognitive function or memory retention, critical in evaluating Alzheimer's disease interventions. Experimentally, the longer the mouse spent in the correct quadrant, the better the compositions were at treating the APP / PS1 transgenic mice for the cognitive function or memory retention.

[0109] As illustrated in FIG. 1, the WT C57 mouse group, lacking the APP / PS1 mutations, averaged 22 seconds in the correct quadrant. In comparison, the saline control group, bearing the double APP / PS1 mutations, averaged about 12 seconds, an approximately 50%drop compared to the WT mouse group. The data indicated a severe cognitive impairment in the APP / PS1 murine model. With the RM01 or the RM02 treatments, all the APP / PS1 mice in these groups showed an improvement on an average compared to those untreated transgenic mice in the saline control group. The double APP / PS1 transgenic mice in the RM01 treatment group showed an improvement, averaged 15 seconds. The double APP / PS1 transgenic mice in the five RM02 treatment groups also showed improvements, averaged 13-19 seconds. Notably, the RM02_BCD, RM02_ABC, and RM02_ABD combinations scored as the top three efficacious treatments, exceeding those in the RM01 treatment group. All the RM02_BCD, RM02_ACD, RM02_ABC, RM02_ABD, and RM02_ABCD combinations were effective at treating the APP / PS1 transgenic mice for the cognitive function or memory retention. Example 2 Immunohistochemistry Study of Mice Brain Tissues

[0112] Three groups of mice were included in this study. Group 1 (Saline Control) included four APP / PS1 transgenic mice and were administered with a saline solution. Group 2 (RM02 Test) included four APP / PS1 mice and were administered with a RM02 pharmaceutical composition. The RM02 composition was prepared with an exemplary Compound A (Hesperidin; designated as A) , an exemplary Compound B (4-hydroxybenzoic acid; designated as B) , an exemplary Compound C (5-hydroxymethyfufural; designated as C) , and an exemplary Compound D (Betaine; designated as D) . Group 3 (RM01 Control) included four C57 mice and were administered with a RM01 pharmaceutical composition as prepared in Example 1. For a period of two months, the mice in these three groups were given a daily oral dose of 0.8ml of a saline solution, an RM02, and an RM01, respectively. Following the two-month treatments, the mice were euthanized and were subject to immnunohistochemistry analysis.

[0113] Immunohistochemistry Procedure and Analysis

[0114] Following the two-month treatments, the mice were euthanized by transcardial perfusion with phosphate buffered saline (PBS) while under anesthesia (2: 1 of ketamine / xylazine) . Following the cold saline perfusion, the brain was rapidly extracted. Half of the brain was then fixed in a solution of 4%paraformaldehyde in PBS (0.1 M, pH 7.4) and was kept at 4-℃ overnight. To ensure cryoprotection, the part of brain tissue was submerged in a solution containing 25%sucrose in PBS (0.1M, pH7.4) , subsequently the brain tissue was embedded in OTC, and were sectioned into 15 μm thick slices using a Leica CM1950 cryostat. The brain slice sections were permeabilized and blocked in PBS with 0.3%Triton X-100 and 10%donkey serum at room temperature for 2 hours. Next, the sections were incubated overnight at 4℃ with one or more primary antibodies in PBS with 0.3%Triton X-100 and 10%donkey serum. The primary antibody was selected from one of the following: anti-b -amyloid (Abcam; ab126649) , anti-Iba1 (Wako Chemicals; 019-19741) , anti-voltage-dependent anion channel 1 (VDAC1) (Proteintech; 55259-1-AP) , anti-glial fibrillary acidic protein (Proteintech; 16825-1-AP) . After washing away the unbound primary antibody, each brain tissue section was subsequently incubated with fluorescently conjugated secondary antibodies (Invitrogen) for 2 hours at room temperature. Nuclei in these brain tissue sections were stained using Hoechst 33342 (Invitrogen) . Fluorescent images were acquired using Leica microscope (DM5000; Leica) with a 20x objective. Identical settings were maintained for all brain tissue sections to ensure consistent results.

[0118] The brain tissue sections were also immunohistochemically analyzed by other biomarkers, including amyloid beta (Aβ; white circles in FIG. 2B) , ionized calcium-binding adaptor molecule 1 (Iba1; grey dots in image b and e in FIG. 2B) , glial fibrillary acidic protein (GFAP; grey dots in image c and f in FIG. 2B) and voltage-dependent anion-selective channel 1 (VDAC1; grey dots in image a and d in FIG. 2B) . Brain tissue sections of the untreated APP / PS1 transgenic mice consistently showed a higher number of any of these biomarkers, including the amyloid beta, comparing to the brain tissue sections from the RM02 treated APP / PS1 transgenic mice. In the image a in FIG. 2B, few VDAC1 grey dots were observed and the VDAC1 biomarker did not overlap with the amyloid beta biomarker. In the image d in FIG. 2B, no VDAC1 grey dots were observed. The data indicated that the expression of the VADC1 biomarker was reduced in the RM02 treated group and the VADC1 and the amyloid beta biomarkers did not appear to be co-expressed in the same locations. In the image b in FIG. 2B, many Iba1 grey dots were observed and approximately 50%of the Iba1 biomarker overlapped with the amyloid beta biomarker. In the image e in FIG. 2B, much less of the Iba1 grey dots were observed, compared to image b in FIG. 2B, and the Iba1 biomarker overlapped with the amyloid beta biomarker. The data indicated that the expression of the Iba1 biomarker was reduced in the RM02 treated group. The data indicated that the expression of the Iba1 biomarker was reduced in the RM02 treated group and the Iba1 and the amyloid beta biomarkers did appear to be co-expressed in the same locations. In the image c in FIG. 2B, many GFAP grey dots were observed and the GFAP biomarker did not overlap with the amyloid beta biomarker. In the image f in FIG. 2B, reduced number of GFAP grey dots were observed and the GFAP biomarker did not overlap with the amyloid beta biomarker. The data indicated that the expression of the GFAP biomarker was reduced in the RM02 treated group and the GFAP and the amyloid beta biomarkers did not appear to be co-expressed in the same locations.

1.A pharmaceutical composition comprising at least three components selected from the group consisting of:(i) Compound A, being a compound of Formula (A) ,or a pharmaceutically acceptable salt thereof, whereinRA1 and RA2 are each independently H; C1-C6 alkyl optionally substituted with one or more -OH; or -RAa, wherein RAa is a monosaccharide, disaccharide, or polysaccharide moiety;RA3 are each independently H or -ORAc, wherein RAc is a monosaccharide, disaccharide, polysaccharide moiety, or a or C1-C6 alkyl optionally substituted with one or more -OH;RA4, RA5, RA6, RA7, and RA8 are each independently H or -ORAb, wherein RAb, at each occurrence, is independently H or C1-C6 alkyl optionally substituted with one or more -OH; andis a single or double bond;(ii) Compound B, being a compound of Formula (B) ,or a pharmaceutically acceptable salt thereof, whereinRB1 is H or C1-C6 alkyl;RB2 is -OH, C (O) RBa or C (O) ORBa, wherein RBa is H or C1-C6 alkyl; andRB3, RB4, RB5, and RB6 are each independently H, -OH, or -O- (C1-C6 alkyl) ;(iii) Compound C, being a compound of Formula (C) ,or a pharmaceutically acceptable salt thereof, wherein:RC1 is H, C1-C6 alkyl optionally substituted with one or more halo or -OH; C (O) RCa; C (O) ORCb; -P (O) (ORCc) (ORCd) ; or phenyl optionally substituted with one or more halo or NO2; wherein RCa, RCb, RCc, and RCd are each independently H; C1-C6 alkyl; or phenyl optionally substituted with one or more halo or NO2;and(iv) Compound D, being a compound of Formula (D) ,or a pharmaceutically acceptable salt thereof, whereinRD1, RD2, RD4, and RD4 are each independently H or C1-C6 alkyl; andL is a branched or unbranched C1-C6 alkylene optionally substituted with one or more halogen or -NH2.2.The pharmaceutical composition of claim 1, comprising the Compound A, or a pharmaceutically acceptable salt thereof, and at least two components selected from the group consisting of the Compound B, or a pharmaceutically acceptable salt thereof, Compound C, or a pharmaceutically acceptable salt thereof, and the Compound D or a pharmaceutically acceptable salt thereof.3.The pharmaceutical composition of claims 1 or 2, comprising the Compound A, or a pharmaceutically acceptable salt thereof, the Compound B, or a pharmaceutically acceptable salt thereof, and the Compound C, or a pharmaceutically acceptable salt thereof.4.The pharmaceutical composition of claims 1 or 2, comprising the Compound A, or a pharmaceutically acceptable salt thereof, the Compound B, or a pharmaceutically acceptable salt thereof, and the Compound D, or a pharmaceutically acceptable salt thereof.5.The pharmaceutical composition of claims 1 or 2, comprising the Compound A, or a pharmaceutically acceptable salt thereof, the Compound C, or a pharmaceutically acceptable salt thereof, and the Compound D, or a pharmaceutically acceptable salt thereof.6.The pharmaceutical composition of any one of claims 1-5, comprising the Compound A, or a pharmaceutically acceptable salt thereof, the Compound B, or a pharmaceutically acceptable salt thereof, the Compound C, or a pharmaceutically acceptable salt thereof, and the Compound D, or a pharmaceutically acceptable salt thereof.7.The pharmaceutical composition of any one of claims 1-6, consisting essentially of the Compound A, or a pharmaceutically acceptable salt thereof, the Compound B, or a pharmaceutically acceptable salt thereof, the Compound C, or a pharmaceutically acceptable salt thereof, and the Compound D, or a pharmaceutically acceptable salt thereof.8.The pharmaceutical composition of claim 1, comprising the Compound B, or a pharmaceutically acceptable salt thereof, the Compound C, or a pharmaceutically acceptable salt thereof, and the Compound D, or a pharmaceutically acceptable salt thereof.9.The pharmaceutical composition of any one of claims 3, 4, 6, or 7, wherein the Compound A or a pharmaceutically acceptable salt thereof, and Compound B or a pharmaceutically acceptable salt thereof, are present in the pharmaceutical composition in a weight ratio of about 1: 100 to about 100: 1.10.The pharmaceutical composition of any one of claims 3, 4, 6, 7, or 9, wherein the Compound A or a pharmaceutically acceptable salt thereof, and Compound B or a pharmaceutically acceptable salt thereof, are present in the pharmaceutical composition in a weight ratio of about 1: 1.11.The pharmaceutical composition of any one of claims 3 or 5-7, wherein the Compound A or a pharmaceutically acceptable salt thereof, and Compound C or a pharmaceutically acceptable salt thereof, are present in the pharmaceutical composition in a weight ratio of about 1: 100 to about 100: 1.12.The pharmaceutical composition of any one of claims 3, 5-7, or 11, wherein the Compound A or a pharmaceutically acceptable salt thereof, and Compound C or a pharmaceutically acceptable salt thereof, are present in the pharmaceutical composition in a weight ratio of about 1: 1.13.The pharmaceutical composition of any one of claims 4-7, whe
