Description

A SUPRAMOLECULAR PANTHENOL BISABOLOL AND A PREPARATION METHOD AND APPLICATION THEREOFTechnical Field

[0001] The invention relates to the technical field of biological medicines, in particular to a supramolecular panthenol bisabolol and a preparation method and application thereof. Background

[0002] Pantothenic acid, also known as vitamin B5 and pantothenic acid, is odorless, slightly bitter in taste, and is so named because it is acidic in nature and widely exists in a variety of foods. Pantothenic acid can participate in metabolic processes in a form of acetyl-CoA; help in cell formation, maintain normal development and development of the central nervous system; manufacture antibodies, help to fight infectious diseases, mitigate the side efficacies and toxicity of various antibiotics, and help to alleviate allergic symptoms; maintain hair, skin and blood health. Pantothenic acid is stable to heat, oxidation, and reduction in neutral solution but can be decomposed under acid, alkali and dry heat conditions. The panthenol is a stable hydroxyl homolog of pantothenic acid. The panthenol is a water-soluble, stable, and small molecule that easily penetrates through the stratum corneum and has the efficacies of moisturizing, relieving, repairing, and the like. The panthenol can be easily absorbed and quickly converted into pantothenic acid through the action of an enzyme to continuously exert the efficacy of the pantothenic acid when being locally applied. Therefore, panthenol is used mainly in the biomedical and daily chemical industries.

[0003] Bisabolol is a nontoxic sesquiterpene alcohol and can be extracted from the Chrysanthemum plant. This component has good stability and skin compatibility and can improve the anti-irritation capability for skin, inhibit bacteria growth, repair skin with inflammation injury, and increase SPF value when added into sunscreen products. Bisabolol also has high permeability in the skin cortex, which is dozens of times more effective than commonly used penetrants. It can be used as a main agent or auxiliary agent to prevent and treat skin diseases such as acne, and it also has a conditioning effect on the skin.

[0004] Panthenol has the efficacies of moisturizing, relieving, repairing, and the like. Bisabolol has the efficacies of resisting inflammation, inhibiting bacteria, promoting permeation, improving skin irritation resistance, and the like. Theoretically, the combination of panthenol and bisabololcan exerts a synergistic efficacy, promotes the penetration of active ingredients, improves bioavailability, and significantly reduces their irritation, avoiding the risk of damage to the skin and achieving a gentle and efficient drug delivery.

[0005] However, panthenol and bisabolol are not mutually soluble. The combination of panthenol and bisabolol has certain difficulties, and its application is yet to be developed.

[0006] Deep Eutectic Solvents (Abbreviated as DES) refer to two-component or three-component deep eutectic mixtures composed of a certain stoichiometric ratio of hydrogen bond acceptors (such as quaternary ammonium salts) and hydrogen bond donors (compounds such as amides, carboxylic acids, and polyols) . The freezing point of the deep eutectic mixtures is significantly lower than the melting point of each component's pure substance. The deep eutectic solvent was reported by Abbott et al. in 2003 for the first time and has physical and chemical properties very similar to those of ionic liquids, so the deep eutectic solvent is also classified as a novel ionic liquid or ionic liquid analog and is a novel supramolecular solvent. However, compared with the traditional organic solvent and ionic liquid, the deep eutectic solvent has the characteristics of low melting point, low cost, low toxicity, easy preparation, reproducibility, biodegradability, and the like, has the characteristics of designability and adjustability of the structure, which common solvents do not have, and can promote the transdermal absorption of active molecules by improving the solubility of the active molecules, improving the oil-water distribution coefficient of the active molecules, and reducing the osmotic resistance and the like.

[0007] However, there is no report on the preparation of panthenol and bisabolol as deep eutectic solvents.

[0008] Disclosure of the Invention

[0009] To overcome the defects of the prior art and aim to solve the problems that panthenol and bisabolol are not mutually soluble and are not easy to prepare into a formula product, one object of the invention is to provide a supramolecular panthenol bisabolol which has good solubility, good transdermal absorption efficacy, high biocompatibility and no irritation. It can be applied to the formula product and has good repairing, relieving, and moisturizing efficacies, especially in efficacy type skin care products.

[0010] The second object of the present invention is to provide a method for preparing the supramolecular panthenol bisabolol.

[0011] The third object of the present invention is to provide an application of the above supramolecular panthenol bisabolol in the preparation of skin care products with moisturizing and repairing efficacies.

[0012] One object of the invention is realized by adopting the following embodiment:

[0013] A supramolecular panthenol bisabolol is composed of panthenol, bisabolol, and ethoxy diglycol and has a chemical structural general:

[0014] Furthermore, in the above supramolecular panthenol bisabolol, the molar ratio of panthenol to bisabolol to ethoxy diglycol is 1-2: 1-2: 1.

[0015] The second object of the invention is realized by adopting the following embodiment:

[0016] A preparation method of supramolecular panthenol bisabolol comprises the following preparation steps:

[0017] S1) under the protection of inert gas, mix panthenol, bisabolol and ethoxy diglycol, heat and stir them until there is no stratification when standing;

[0018] S2) keep inert gas protection, and carry out ultrasonic treatment on the mixed solution obtained in S1 to obtain the supramolecular panthenol bisabolol.

[0019] Further, in the preparation method, in step S1, the inert gas is nitrogen, the nitrogen pressure is 0.1-20MPa, the heating and stirring temperature is 30-80 ℃, the heating and stirring time is 1-24 hours, and the heating and stirring rotating speed is 100-600 rpm.

[0020] Further, in the preparation method, the inert gas is nitrogen, the nitrogen pressure is 10-15 MPa, the heating and stirring temperature is 50-70 ℃, the heating and stirring time is 1-5 hours, and the heating and stirring rotating speed is 300-600 rpm.

[0021] Further, in the preparation method, in step S2, the ultrasonic power of the ultrasonic treatment is 20-3000W, the ultrasonic frequency is 5-100 kHz, and the ultrasonic treatment time is 1-4 hours.

[0022] Furthermore, in the preparation method, the ultrasonic power of the ultrasonic treatment is 100-1000W, and the ultrasonic frequency is 10-50 kHz.

[0023] Further, in the preparation method, the ultrasonic treatment method includes setting the ultrasonic power to 100‐600 W, the ultrasonic frequency to 10‐50 kHz, pausing for 2 s after each ultrasonic treatment for 2 s, and alternating and continuing the treatment for 1‐4 h.

[0024] The third object of the invention is realized by the following embodiment:

[0025] A formulated skin care product with a moisturizing and repairing efficacy containing the supramolecular panthenol bisabolol.

[0026] Furthermore, the mass proportion of the supramolecular panthenol bisabolol in the formulated skin care product is 0.5%to 2.5%. The formulated skin care product may be anhydrous essence, emulsion, cream, essence, mask, or other feasible formulated skin care products.

[0027] The invention has the following beneficial efficacies. In the present invention, by adding ethoxy diglycol as the third component, the ethoxy diglycol forms a deep eutectic solvent with panthenol and bisabolol under weak interactions such as hydrogen bonds and van der Waals interactions, thereby solving the problem that panthenol and bisabolol are immiscible, reducing the damage of bisabolol to cells, and further improving the biocompatibility of bisabolol. At the same time, since the supramolecular panthenol bisabolol has the structural designability and adjustability of a deep eutectic solvent, it promotes transdermal absorption by means such as enhancing the solubility of panthenol and bisabolol, improving the oil-water distribution coefficient, and reducing its penetration resistance, thereby showing a better repairing efficacy than panthenol monomer and a physical mixture of panthenol and bisabolol, and also laying a foundation for the combined application of panthenol and bisabolol in pharmaceutical preparations. The preparation method provided by the present invention has simple and safe steps and is conducive to industrial production.BRIEF DESCRIPTION OF THE DRAWINGS

[0074] Performance tests:

[0075] (I) Biosafety testing of supramolecular panthenol bisabolol

[0076] The biological safety of the samples is tested by performing cytotoxicity tests of supramolecular panthenol bisabolol, a physical mixture of panthenol and bisabolol, and bisabolol monomers on human fibroblasts. The survival rate of cells is measured by taking metabolic activity as an indicator. MTT (thiazole blue) is metabolically reduced in living cells to generate blue-purple insoluble formazan, and the number of the living cells is related to the chroma measured by a photometer after the formazan is dissolved in alcohol. The specific experimental steps are as follows:

[0077] I: The human skin fibroblast suspension is inoculated into a 96-well plate, 100 μL per well, and incubated for 24 h.

[0078] II: After the cells are cultured for 24 hours in an adherent way, the culture medium is discarded, 100 μL of the sample solution to be tested is added into each well, and a blank group and a negative group are arranged.

[0079] III: After the cells are incubated for 24 h, 10 μL MTT solution is added to each well, and the cells are cultured in an incubator for 4 h.

[0080] IV: The culture solution is discarded, 150 μL DMSO is added to each well, it is shaken for 10 min, and the absorbance at 490 nm (OD490 nm) is measured with an enzyme reader. It is compared with the control sample that is not treated with the tested substance.

[0081] The result is calculated.

[0082] In the formula:

[0083] Relative survival--relative OD490 nm, %;

[0084] Test OD490 nm--average OD490 nm of the tested substance;

[0085] Neg OD490 nm--average OD490 nm of negative controls.

[0086] Wherein, the samples to be tested are supramolecular panthenol bisabolol prepared in example 1, the physical mixture of panthenol and bisabolol prepared in comparative example 4, and commercially available bisabolol.

[0087] The cytotoxicities of supramolecular panthenol bisabolol, the physical mixture of panthenol and bisabolol, and bisabolol monomers on human fibroblasts are shown in Fig. 4, wherein the IC50 of supramolecular panthenol bisabolol is about 7.766 mg / mL (95%CI: 7.357 mg / mL-8.192 mg / mL) , its IC90 is about 1.476 mg / mL (95%CI: 0.000013 mg / mL-3.821 mg / mL) ; the IC50 of the physical mixture of panthenol and bisabolol is about 5.886 mg / mL and its IC90 is about 4.669 mg / mL (95%CI: 3.8905 mg / mL-5.0003 mg / mL) ; the IC50 of the bisabolol is about 6.124 mg / mL and its IC90 about 4.949 mg / mL (95%CI: 4.7535 mg / mL-5.1641 mg / mL) . In a lower concentration range (0.039 mg / mL-5 mg / mL) , the cytotoxicity difference of these three components is small, and when the concentration is further increased to 10 mg / mL, the cell survival rate of the supramolecular panthenol bisabolol group is significantly higher than those of the other two groups, which indicates that the supramolecular panthenol bisabolol reduces the damage of bisabolol to cells to a certain extent and further improves the biocompatibility of bisabolol.

[0088] (II) In vitro repairing efficacy test of the supramolecular panthenol bisabolol

[0089] The repairing efficacy of the sample to be detected is tested by a cell wound healing test. The cell wound healing test is a method for simply measuring cell migration movement and repair capacity. It is similar to an in vitro wound healing model. It can judge the repair capacity of each group of cell migration on a monolayer adherent cell cultured on an in vitro culture dish or a plate according to the migration capacity of cells in different groups on a wound area. The samples to be tested are supramolecular panthenol bisabolol prepared in example 1 and commercially available panthenol monomer. A medium containing 3%serum is used for Negative Control (NC) and experimental group, and a medium containing 15%serum is used for Positive Control (PC) .

[0090] It is analyzed by adopting a t-test method, wherein the test level α= 0.05; P≥0.05, which indicates that there is no statistical difference, and there is no obvious change in the relative migration rate of the cells; P <0.05, there is significant difference and the relative cell migration increased, which indicates that samples have repairing efficacy. The specific experimental steps are as follows:

[0091] I: The suspension of human keratinocytes or fibroblasts is inoculated into a 24-well cell culture plate; each well is inoculated with 500 μL of the suspension, and the suspension returns to an incubator to be cultured for 18-24 h until the cells are fused.

[0092] II: The culture medium is discarded, and the culture medium containing the corresponding sample is replaced. Simultaneously, the image is collected under the mirror and recorded as 0 h. The collected position is marked and continues to incubate.

[0093] III: At 24 h of incubation, an image is taken at the same marked positions.

[0094] IV: Data analysis: The IPP image analysis software is used to analyze the wound area of each image, and the relative migration is calculated by taking 0 h as 100%.

[0095] The supramolecular panthenol bisabolol is used for carrying out a cell wound test on human keratinocytes. The detection results of the relative content of different groups are shown in Table 1-1, and the relative migration rate of different groups is shown in Figure 5-1. The inter-group statistical results are shown in Table 1-2, and the image of cells of different groups are shown in Figure 5-2.

[0098] As shown in Table 1-2, compared to the group NC, the relative migration rate of the group PC was increased at 24 h (P < 0.05) , indicating that the experiment was effective. As shown in Table 1-1, Fig. 5-1, and Fig. 5-2, compared to the group NC, the relative migration rate of supramolecular panthenol bisabolol was significantly increased (P < 0.05) at the test concentrations of 0.07 mg / mL, 0.035 mg / mL, and 0.01mg / mL, and the repairing rates were 37.75%, 27.39%, and 13.81%, respectively, indicating that supramolecular panthenol bisabolol promoted cell migration and achieved the repairing efficacy at the concentrations of 0.07 mg / mL, 0.035 mg / mL, and 0.01 mg / mL.

[0099] The cell wound healing test of panthenol was performed on human keratinocytes. The test results of the relative contents of different groups are shown in Table 2. The relative migration rates of different groups are shown in Fig. 6-1, and the images of cells of different groups are shown in Fig. 6-2.

[0100] Table 2. Relative migration rates of panthenol on humam keratinocyte

[0101] As shown in Table 2, compared to the group NC, the relative migration rate of the group PC was increased at 24 h (P < 0.05) , indicating that the experiment was effective. As shown in Fig. 6-1 and 6-2, compared to the Group NC, the relative migration rates of panthenol at the test concentrations of 0.07 mg / mL, 0.035 mg / mL, and 0.01 mg / mL did not increase significantly, indicating that panthenol didn't promote cell migration at the concentrations of 0.07 mg / mL, 0.035 mg / mL, and 0.01 mg / mL.

[0102] The cell wound healing test of supramolecular panthenol bisabolol was performed on human keratinocytes. The test results of the relative content of different groups are shown in Table 3. The relative migration rate of different groups is shown in Fig. 7-1, and the cell image of different groups is shown in Fig. 7-2.

[0103] Table 3. Relative migration rates of supramolecular panthenol bisabolol on human keratinocytes

[0104] As shown in Table 3, compared to the group NC, the relative migration rate of the group PC was increased at 24 h (P < 0.05) , indicating that the experiment was effective. As shown in Fig. 7-1 and 7-2, compared to the group NC, the relative migration rate of supramolecular panthenol bisabolol was significantly increased (P < 0.05) at test concentrations of 2.5 mg / mL, 1.25 mg / mL, and 0.625 mg / mL, and the repairing rates were 36.02%, 23.96%, and 18.22%, respectively, indicating that supramolecular panthenol bisabolol promoted cell migration and achieved repairing efficacy at concentrations of 2.5 mg / mL, 1.25 mg / mL, and 0.625 mg / mL.

[0105] The cell wound healing tests of panthenol were performed on human fibroblasts. The testing results for the relative content of different groups are shown in Table 4. The comparison of relative migration rates of different groups is shown in FIG. 8-1, and the cell image of different groups is shown in FIG. 8-2.

[0106] Table 4. Relative migration rates of panthenol on human keratinocytes

[0107] As shown in Table 4, compared to the group NC, the relative migration rate of the group PC was increased at 24 h (P < 0.05) , indicating that the experiment was effective. As shown in Fig. 8-1 and 8-2, compared to the group NC, the relative migration rate of panthenol at the test concentrations of 1.25 mg / mL and 0.625 m / mL was not significantly increased (P > 0.05) , indicating that panthenol didn't promote cell migration at the concentrations of 1.25 mg / mL and 0.625 m / mL, and at the test concentration of 2.5 mg / mL, the relative migration rate was obviously increased (P<0.05) , and the repairing rate was 10.45%, indicating that the panthenol promoted the cell migration and achieved the repairing efficacy at the concentration of 2.5 mg / ml.

[0108] In conclusion, at the concentrations of 0.07 mg / mL, 0.035 mg / mL, and 0.01 mg / mL, the supramolecular panthenol bisabolol promoted the migration of human keratinocytes and achieved the repairing efficacy. The repairing rates were 37.75%, 27.39%, and 13.81%, respectively. At the same concentration, panthenol didn't exhibit the repairing efficacy. At the concentrations of 2.5 mg / mL, 1.25 mg / mL, and 0.625 mg / Ml, the supramolecular panthenol bisabolol promoted the migration of human fibroblasts and achieved the repairing efficacy. The repairing rates were 36.02%, 23.96%, and 18.22%, respectively. Only at the concentration of 2.5 mg / ml, the panthenol promoted the migration of human fibroblasts and achieved the repairing efficacy. The repairing rate was only 10.45%. Compared to panthenol, supramolecular panthenol bisabolol showed better repairing efficacy. On one hand, the bisabolol has a certain repairing efficacy and acts together with panthenol, so that the supramolecular panthenol bisabolol has a better repairing efficacy. On the other hand, the supramolecular panthenol bisabolol promotes transdermal absorption through enhancing the solubility of panthenol and bisabolol, improving the oil-water distribution coefficient, reducing the self-permeation resistance and the like by virtue of the characteristics of structural designability and adjustability of the deep eutectic solvent, thereby showing better repairing efficacy.

[0109] (III) Relieving efficacy tests of supramolecular panthenol bisabolol

[0110] The relieving efficacy of the samples to be tested is evaluated by LPS-induced mouse macrophage RAW 264.7 cell model and human keratinocyte TRPV1 protein expression test. The samples to be tested are the supramolecular panthenol bisabolol prepared in Example 1, the physical mixture of panthenol and bisabolol prepared in Comparative Example 4, and the commercially available panthenol monomer.

[0111] LPS-induced mouse macrophage RAW 264.7 cell model

[0115] I (cell plating) : Inoculating mouse macrophage RAW 264.7 suspension into 24-well plates and incubated for 24 h.

[0116] II (liquid preparation) : Preparing the tested substance working solution according to the test protocols (Table 5) , respectively.

[0117] Table 5. Inflammatory factor testing program

[0118] III (dosing) : According to the testing protocol of Table 5, groups are dosed. Each well is added with 1 mL, each group has 3 duplicate wells, and the reaction is maintained for 2 h. Group NC adds culture medium, other groups add drug-containing culture medium containing LPS.

[0139] (2) Human keratinocyte TRPV1 protein expression test

[0140] Capsaicin receptors are a class of widely distributed ion channels, a key protein that causes burning and stinging of the skin. When exogenous stimuli such as capsaicin and the like activates TRPV1, it causes Ca2+ inflow, causing stinging and burning sensations in the skin. The sample may exert a relieving efficacy by inhibiting TRPV1 expression.

[0141] A result is judged as follow: Analyzing with a t-test method, wherein the test level α= 0.05; P≥0.05, indicating that there is no statistical difference, and that the sample has no inhibitory effect on the change of TRPV1 protein content; P<0.05, indicating that there is a significant difference and the content of TRPV1 protein is significantly reduced, and that the sample has the efficacy of relieving. The specific experimental steps are as follows:

[0181] When the product is used for 14 days and 28 days, the redness a*of the experimental group is significantly reduced by 23.30% (p<0.001) and 23.47% (P<0.001) , respectively. The difference of the SPSS data analysis result of the 28th day is extremely remarkable, indicating that 2.5%supramolecular panthenol bisabolol essence emulsion (the experimental group) has the efficacy of reducing the redness of the skin. The redness a*of the control group is significantly reduced by 28.17% (P < 0.001) and 27.42% (P < 0.001) , respectively. The difference of the SPSS data analysis results on the 28th day is extremely remarkable, indicating that the 3%physical mixture of panthenol and bisabolol essence emulsion (control group) has the efficacy of reducing the redness of the skin. There is no significant difference (P≥0.05) in the inter-group difference data at the 14th day and 28th day, indicating that 2.5%supramolecular panthenol bisabolol essence emulsion and 3%physical mixture of panthenol and bisabolol essence emulsion have the same relieving efficacy.
