A 60% dispersion of sodium hydride in mineral oil (240 mg, 6 mmol, 1.2 equiv) was added to a solution of (lR)-l-(3,5-dichloro-4-pyridyl)ethanol (960 mg, 5 mmol, 1 equiv) in tetrahydrofuran (60 mL) at room temperature. After 30 minutes, 5-chloro-lH- pyrazolo[4,3-d]pyrimidine (927 mg, 6 mmol, 1.2 equiv) was added and the mixture was heated at 60 °C overnight. Additional 60% dispersion of sodium hydride in mineral oil (680 mg, 17 mmol, 3.4 equiv) was added and the heating continued at 73 °C for 24 hours. The reaction was cooled to 0 °C and carefully quenched with ice-cold water (50 mL). The mixture was extracted with ethyl acetate (2 x 100 mL). The combined organic layers were dried over sodium sulfate, filtered, and concentrated under reduced pressure. The residue was purified on an InterChim automated chromatography system (120 g column), eluting with a gradient of 0 to 100% ethyl acetate in heptanes to give a yellow solid (100 mg, 6% yield). [0391] Step 3. 5-[(lR)-l-(3,5-Dichloro-4-pyridyl)ethoxy]-3-[6-(2-methylsulfonyl- 2,6-diazaspiro[3.3]heptan-6-yl)-3-pyridyl]-lH-pyrazolo[4,3-d]pyrimidine. 2- (Methylsulfonyl)-6-(5-(4,4,5,5-tetramethyl-l,3,2-dioxaborolan-2-yl)pyridin-2-yl)-2,6- diazaspiro[3.3]heptane (117 mg, 0.31 mmol, 1.5 equiv), [1,1'- bis(diphenylphosphino)ferrocene]dichloropalladium(II) (15 mg, 0.02 mmol, 0.1 equiv) and potassium carbonate (58 mg, 0.42 mmol, 2.0 equiv) were added sequentially to a solution of 5-[(lR)-l-(3,5-dichloro-4-pyridyl)ethoxy]-3-iodo-lH-pyrazolo[4,3-d]pyrimidine (90 mg, 0.21 mmol, 1.0 equiv) in a 10 to 1 mixture of 1,4-dioxane and water (11 mL) in a sealed tube. After sparging with nitrogen for 10 minutes, the reaction was heated at 95 °C for 16 hours. After cooling to room temperature, the reaction mixture was filtered through Celite (2 g), which was washed with tetrahydrofuran (50 mL). The filtrate was concentrated under reduced pressure. The residue was redissolved in dimethyl sulfoxide (8 mL) and purified on an InterChim automated chromatography system (RediSep Rf GOLD 100 g HP C18 column), eluting with a gradient of 0 to 100% acetonitrile in water. The mixed products were further purified on a Gilson automated chromatography system (Waters Atlantis T3 Prep OBD column, 5 pm, 19 x 250 mm), eluting with a gradient of 0 to 100% acetonitrile in water with 0.1% formic acid. [0392] Kinase-tagged T7 phage strains were prepared in an E. coli host derived from the BL21 strain. E. coli were grown to log -phase and infected with T7 phage and incubated with shaking at 32°C until lysis. The lysates were centrifuged and filtered to remove cell debris. Streptavidin-coated magnetic beads were treated with biotinylated small molecule ligands for 30 minutes at room temperature to generate affinity resins for kinase assays. The liganded beads were blocked with excess biotin and washed with blocking buffer SeaBlock (Pierce), 1% BSA, 0.05% Tween 20, 1 mM DTT to remove unbound ligand and to reduce non-specific binding. Binding reactions were assembled by combining kinases, liganded affinity beads, and test compounds in lx binding buffer (20% SeaBlock, 0.17x PBS, 0.05% Tween 20, 6 mM DTT).

All compounds for Kd measurements are distributed by acoustic transfer (non-contact dispensing) in 100% DMSO.
