[0006] Background of the Invention
[0007] Chemical substances such as aromatic hydrocarbons such as dioxin and aromatic hydrocarbons (PAHs) are present in the air and atmosphere. Exposure such as by inhaling such chemical substances in the air can harm the human body in various aspects. For example, pollutants in the atmosphere may cause inflammation in the cells of the body, resulting in irritation, overproduction of certain substances like melanin (i.e., the natural skin pigment present in human skin) produced by cells, and even cancer.
[0008] The present inventors have surprisingly found cosmetic compositions comprising ubiquinone, and at least one skin care benefit agent are effective for combating the harmful effects of atmospheric pollution, particularly that relating to hyperpigmentation in human skin. “Hyperpigmentation,” as used herein, refers to an area of skin wherein greater pigmentation than that of an adjacent area of skin is present, such as in the case of a sun / age spot or post- inflammatory hyperpigmentation (PIH). [0076] One of ordinary skill in the art would be adept to identify any need of neutralization, such as for adequate swelling of acrylate- based polymers to achieve a desirable and / or maximum viscosity. Any pH modifier known in the field acceptable for use in personal care and / or pharmaceutical products can be employed to result in a cosmetic composition possessing a skin-acceptable pH, typically within the range of from 4 to 8. In a preferred embodiment, the pH of the cosmetic composition is from 4.25 to 7.5, and most preferably, from 5 to 7. Traditional buffers or pH modifiers include common additives such as sodium hydroxide, potassium hydroxide, hydrochloric acid, citric acid, triethanolamine, and aminomethyl propanol. Viscosity of the cosmetic composition disclosed herein is preferably 1 ,000 to 120,000 mPas, and, more preferably, 5,000 to 80,000 mPas, taken under conditions of ambient temperature and a shear rate of 1s_1with a strain controlled parallel plate rheometer made commercially available from suppliers like T.A. [0078] When making the present cosmetic composition, the desired ingredients are mixed via conventional methods. If applicable, the oil phase and water phases are separately mixed using standard mixers and mixing blades and heated to a temperature usually from 20 to 85°C under atmospheric pressure. Upon reaching the desired temperature, the oil phase is added to the water phase to create an emulsion. Other ingredients in the cosmetic composition are subsequently added and mixed in no particular order.
[0079] The cosmetic composition disclosed herein is a composition suitable for topical application to human skin, including leave-on and wash-off products. Preferably, the term encompasses a fluid liquid, and particularly a moisturizer rather than a make-up product. Most preferred are leave-on compositions. The term "leave-on" as used with reference to compositions herein means a composition that is applied to or rubbed on the skin and left thereon.
[0080] Many types of packaging can be used to store and deliver the present cosmetic composition. The selection of packaging is dependent upon the cosmetic end-use and the viscosity of the composition itself. As an example, leave-on lotions and creams for skin typically employ plastic containers with an opening at a dispense end covered by an appropriate closure. Conventional closures include flip-top hinged lids, screw-caps, and non-aerosol pumps. As another example, appropriate packaging to be used for antiperspirants, deodorants and depilatories include a container with a roller-ball applicator on a dispensing end if the composition is fluid and of a thinner viscosity. If the composition is in a stick format, a container with a propel-repel mechanism wherein the stick is fixed on a platform towards a dispensing orifice is appropriate. If the composition is in an aerosol format, then metallic cans pressurized by a propellant and having a spray nozzle is appropriate. In general, patches, bottles, tubes, roller-ball applicators, squeeze containers, or lidded jars are preferred.

[0085] The cosmetic compositions will now be described below in the context of specific non-limiting examples to facilitate a greater understanding of the present cosmetic compositions. One of ordinary skill in the art will recognize that variations of the inventive compositions that differ from the examples given may be practiced without deviating from the teachings of the present compositions.
[0086] Examples
[0087] The depigmenting effects of a combination comprising ubiquinone and a skin care benefit agent were evaluated through an in vitro pollutant-induced model. The pollutant selected was chrysene, as made commercially available by Sigma-Aldrich. The skin care benefit agent selected was N-acetyl-DL-methionine as made commercially available by Evonik. The ubiquinone selected was made commercially available by Sigma-Aldrich.
[0088] Darkly-pigmenting (DP) neonatal primary human melanocytes were sourced from Thermo Fisher Scientific. The melanocytes were maintained in Medium 254 supplemented with human Melanocyte Growth Supplement 2 (MGM, Cascade Biologicals) as well as with 10 U / rnL penicillin G and 0.1 mg / mL streptomycin sulphate. The maintenance and sub-culturing of cells were carried out in the manner set forth in the manufacturer’s instructions. Cells between passages 3-6 were used for experimentation. Cells were maintained at a temperature of about 37°C and incubated in a humidified incubator with 5% CO2 atmosphere.
[0089] For the experiments, cells were seeded at a density of 5 x 104cells per well of a 24-well plate and maintained in a 5% CO2 incubator at about 37°C and 95% humidity for twenty-four (24) hours. Cultures were dosed with different concentrations of ubiquinone or acetyl methionine alone or in combination, and were challenged by the presence of 5 pg / ml chrysene. Seventy-two (72) hours later, culture supernatants were collected to determine cellular cytotoxicity by a Lactate Dehydrogenase (LDH) assay kit made commercially available by Invitrogen, while cultured cells were harvested and processed for estimation of melanin content using conventional techniques known to one skilled in the art. P0001048 CPL
[0090] 18
[0091] Cellular cytotoxicity by LDH Assay
[0092] 30 pL of culture supernatant was collected from each treatment well and transferred to a 96-well plate. To determine maximum LDH, the lysis buffer provided in the LDH assay kit (10X) was added to the well containing cells in a 1 :10 dilution to enable release of maximum LDH from the cells. To the 30 pL of culture supernatant collected, 30 pL of a mixture of the LDH assay buffer and substrate solution was made based on the number of samples and added. The plate was incubated for thirty (30) minutes. Then, 30 pL of stop solution was added to stop the reaction. Once the reaction was stopped, the reading was recorded at a wavelength of 490 nm in a TECAN plate reader. After ascertaining non-cytotoxic doses, treatments were progressed for melanin estimation.
[0093] Melanin content estimation
[0094] After culture supernatants were collected and processed for cellular cytotoxicity by the LDH assay, adherent cells were lysed with a solution containing 1 N NaOH and 10% DMSO prepared in water and placed for one (1) hour at 60°C in a shaker incubator. The lysed cell suspension was cooled to room temperature (i.e. , about 20 to 25°C), mixed thoroughly, and transferred to a 384- well plate. Melanin content was measured at 405 nm in a TECAN plate reader. Melanin content was reported as % control ± SE.
[0095] Example 1 :
[0096] Acetyl methionine and ubiquinone were combined in different concentrations to evaluate impact on pollutant-induced melanin content in cultured primary human melanocytes. It was unexpectedly found that acetyl methionine and ubiquinone synergistically reduced chrysene- induced melanin content only at specific dose combinations and weight ratios as captured in Tables 1 and 2 of Example 1.
