AI Technical Summary

Technical Problem

Streptavidin tetramers in affinity chromatography are susceptible to dissociation under harsh conditions, leading to streptavidin leachate that interferes with protein-protein interactions and sample quality.

Method used

Development of crosslinked streptavidin-conjugated chromatographic materials with biotinylated affinity agents and free biotin to bind to accessible binding sites, reducing streptavidin leachate through methods such as crosslinking streptavidin molecules to a polymer core and binding sites with biotinylated agents and free biotin.

Benefits of technology

The solution achieves a significant reduction in streptavidin leachate, with columns showing at least an 85% reduction in leachate absorbance and often reaching no detectable leachate, thereby improving sample quality and reducing interference.

Abstract

Disclosed herein are chromatographic materials comprising crosslinked streptavidin particles, wherein the crosslinked streptavidin molecule is conjugated to the surface material of a particle, said particle comprising a polymer core. The crosslinked streptavidin particles are bound to a biotinylated affinity agent and free biotin. The combined impact of streptavidin crosslinking and binding free biotin to accessible binding sites in streptavidin drastically reduce leaching of streptavidin from the chromatographic materials. The chromatographic materials disclosed herein may be used to prepare affinity chromatographic columns..
