[0001] REFERENCE TO SEQUENCE LISTING
[0002] This application includes a Sequence Listing filed electronically as an XML file named “Sequence listing PCT. xml, ” created on March 25, 2024, with a size of 9,998 bytes. The Sequence Listing is incorporated herein by reference.TECHNICAL FIELD
[0003] This application pertains to the field of biology, with a specific focus on a novel composition that exhibits anti-inflammatory and skin regenerative properties, as well as its preparation method and applications. The composition comprises platelet-rich plasma, β-nicotinamide mononucleotide, and sialic acid, which synergistically contribute to its therapeutic effects. The composition can be applied to various skin conditions, including skin scalds, burns, atopic dermatitis, diabetic feet, and skin aging.BACKGROUND
[0004] Blood, β-nicotinamide mononucleotide (NMN) , and telomeres are interconnected entities, with cells in the blood containing telomeres that can be affected by NMN, potentially influencing their length and stability. Research indicates that the length and stability of telomeres in blood cells are linked to the risk of various diseases. Plasma proteins, such as FGF, IGF-1, and TNF-α, have also been found to be associated with telomere length and stability. Studies have demonstrated the ability of NMN to increase NAD+ content in skin cells of mice, improving cellular energy metabolism and antioxidant capacity, thereby promoting wound healing. NMN can also facilitate wound healing by repairing DNA damage, stimulating cell proliferation, regulating immune responses, and employing other mechanisms. However, clinical applications of NMN in this regard remain limited.
[0005] Platelet-rich plasma (PRP) is a concentrated platelet solution obtained from autologous blood through centrifugation, which has demonstrated the capacity to stimulate bone and soft tissue repair. PRP has been utilized in orthopedics, oral and maxillofacial surgery, cardiothoracic surgery, neurosurgery, gynecology and obstetrics, ophthalmology, otolaryngology, general surgery, plastic, and aesthetic surgery, among other fields.
[0006] Sialic acid, also known as cubilose acid, is a key component of cubilose and has demonstrated inhibitory effects on tyrosine hydroxylase and dopamine hydroxylase, preventing melanin production and fading facial blemishes. Due to its carboxyl group, sialic acid molecules carry a negative charge, facilitating cellular participation in water and salt balance, material transport, and other processes, thus playing a crucial role in maintaining cellular function.
[0007] Despite the availability of various skin repair technologies, including those targeting skin burns, atopic dermatitis, diabetic feet, and skin rejuvenation, there remains a shortage of compositions capable of promoting skin repair while reducing inflammation, thereby increasing wound healing rates and resulting in lighter scar pigmentation.SUMMARY
[0008] In response to the technical challenge of a lack of compositions capable of promoting skin repair and reducing inflammation for various skin conditions, such as skin burns, atopic dermatitis, diabetic feet, and skin rejuvenation, the present invention provides a composition capable of anti-inflammatory and promoting skin repair, along with its preparation method and applications.

[0046] Compared with traditional technical solutions, the present invention has the following advantages and effects.
[0047] The present invention builds upon traditional PRP technology by adding β-nicotinamide mononucleotide (NMN) to the composition. NMN stabilizes telomeres of cells in the blood and promotes wound healing by directly interacting with wounded skin cells, converting into NAD+ more efficiently, enhancing energy metabolism and antioxidant ability of cells, stimulating cell proliferation, regulating immune responses, and implementing other mechanisms. In addition, the sialic acid molecule in the composition has a negative charge, which facilitates cell participation in water and salt balance, material transport, and other processes. Sialic acid can effectively inhibit the activity of tyrosine hydroxylase and dopamine hydroxylase, preventing the production of melanin. As a result, sialic acid mainly has the effect of fading stains and colors of wound scars in the present patent. Furthermore, the platelet-rich plasma in the composition contains abundant cytokines and growth factors that play a critical role in wound healing.
[0048] The composition of platelet-rich plasma, β-nicotinamide mononucleotide (NMN) , and sialic acid provided by the present invention can greatly accelerate wound healing. Compared to the separate use of platelet-rich plasma (PRP) , β-nicotinamide mononucleotide (NMN) , or sialic acid, the composition promotes faster wound healing. Additionally, after wound scar repair, the resulting scar appears lighter in color.BRIEF DESCRIPTION OF THE DRAWINGS

[0065] To aid in comprehending the present invention, a detailed description is provided below. However, it should be noted that the present invention can be implemented in multiple forms and is not restricted to the embodiments or examples described herein. The embodiments or examples are provided for the purpose of imparting a more thorough and complete understanding of the disclosed contents of the present invention.
[0066] Example 1: Preparation of platelet-rich plasma and a composition thereof
[0067] Specific steps are as follows.
[0068] (1) Fresh human blood (obtained from Hong Kong Red Cross) was subjected to centrifugal separation in two steps to obtain platelet-rich plasma (PRP) . The first step involved centrifugation at 1,200-2,000 rpm for 5 minutes, followed by centrifugation at 3,000-3,800 rpm for 20 minutes.
[0069] (2) The platelet-rich plasma was mixed with β-nicotinamide mononucleotide (NMN) and sialic acid in certain proportions (volume-mass ratio of platelet-rich plasma to sialic acid to NMN was 100: 1: 1 (V / W / W) , 1,000: 1: 1 (V / W / W) , 3,000: 1: 1 (V / W / W) , 5,000: 1: 1 (V / W / W) , or 10,000: 1: 1 (V / W / W) ) . NMN was purchased from Jinan Wo'erde Chemical Co., Ltd. and sialic acid was purchased from Cabio Biotechnology (Wuhan) Co., Ltd. The resulting mixture was subjected to ultrasonic treatment at 100 W for 10 minutes and uniform mixing to obtain a mixture, which was then stored in a refrigerator at a low temperature of -80℃ and subsequently freeze-dried to obtain a composition called PRP plus for later use.
[0070] Example 2: Cytotoxicity test
[0071] To determine whether the PRP plus composition has any cytotoxic effects on keratinocytes and whether it can stimulate skin cell proliferation, an MTT assay was conducted to assess cell viability. HaCaT cells (catalog No. T0020001, purchased from AddexBio Company) were maintained in DMEM culture medium supplemented with 10%FBS (fetal bovine serum) and 1%penicillin or streptomycin. Adherent cells were digested with a 0.25%trypsin solution, counted, and suspended at a concentration of 1×104 cells / mL. The cell suspension was then added to a 96-well plate and placed in a CO2 incubator with 5%CO2 at 37℃ for 24 hours. Different concentrations of PRP and PRP plus were added to the cells for treatment. After 24 hours of continuous culture, the culture medium was removed, and an MTT solution at a concentration of 0.5 mg / mL was added to the cells and incubated for 4 hours. After incubation, a DMSO solution was added and the mixture was shaken for 15 minutes. The absorbance was then measured at 570 nm to determine the cell viability.
[0072] The results, as shown in Figure 1, indicate that neither PRP plus nor PRP has any cytotoxic effects on HaCaT cells. However, when the added concentration of PRP plus or PRP is greater than 5% (v / v) in the culture medium, the medium becomes gel-like, which is unsuitable for conducting experiments such as scratch-based cell migration. The optimal experimental concentration for both PRP plus and PRP is 1% (v / v) . Therefore, in other examples, both PRP plus and PRP were used at a concentration of 1% (v / v) .
[0073] Example 3: Wound healing experiment
[0074] In this example, a wound healing experiment was carried out using HaCaT cells treated with 1%PRP plus composition, 1%PRP, 1 mM NMN, and 1 mM sialic acid. The 1%PRP plus composition was prepared using the method described in Example 1.
[0075] Specific experimental steps are as follows.
[0076] (1) The HaCaT cells were cultured in a DMEM culture medium containing 10% (v / v) of FBS (fetal bovine serum) and 1% (w / v) of penicillin or streptomycin. Adherent cells at the bottom of the culture dish were subjected to digestion with a 0.25% (w / v) trypsin solution, prepared into a cell suspension, counted and then diluted into a suspension with a cell concentration of 1×105 piece / mL, and the suspension was inoculated into a 12-well plate and placed in a CO2 incubator as a cell incubator including 5%of CO2 at 37 ℃ until the cells were fully spread densely.
[0077] (2) A cross was scratched in the center of the bottom of each 12-well plate by a 200 μL suction head of a pipette so as to simulate the formation of a wound, and the cells were photographed under a microscope with a magnification of 50x to obtain a cell layer coverage rate, recorded as T0.
[0078] (3) A blank control (no other components were added except for the culture medium) , a positive control (VEGF, vascular endothelial growth factor) , PRP, PRP plus and NMN were added into the 12-well plate separately. After the samples were added and the cells were cultured for 20 hours, the cells were photographed again to obtain a cell layer coverage rate, recorded as T20. Results are as shown in FIG. 2.
[0079] FIG. 2 is an image showing experimental results of the cell layer coverage rate of the HaCaT cells, where the contents correspond to the blank control, the vascular endothelial growth factor (VEGF) , the PRP plus, the PRP, the sialic acid and the NMN from left to right in sequence, and the contents are T0 and T20 from top to bottom in sequence. After the cells are cultured for 20 hours, it is found through morphological observation that the wound healing rate of the cells treated with the PRP plus composition is obviously higher than that treated with the PRP, the NMN and the sialic acid. The results show that the PRP plus composition can increase the wound healing rate.
[0080] Example 4: Study on inhibition of the production of melanin
[0081] Melan-Acells are melanocytes derived from normal mice that express normal levels of melanin production. These cells are preferred over B16 cells for studying the inhibition of melanin production. Tyrosinase is a rate-limiting enzyme in the melanin production process. To measure the content of melanin produced in Melan-Acells, colorimetry is used to measure the absorbance at 405 nm. This allows for the indirect detection of the activity of tyrosinase inhibition and reduction of melanin production for different samples. A lower melanin content indicates a stronger whitening effect of the sample.
[0082] Specific experimental steps are as follows.
[0083] A Melan-Acell line (donated by the University of Hong Kong) was placed in a CO2 incubator including 5%of CO2 for culture at 37 ℃ and then cultured in a DMEM culture medium containing 10%of FBS (fetal bovine serum) and 1%of penicillin or streptomycin for 24 hours. Then Melan-Acells were prepared into a suspension with a cell concentration of 5×105 piece / mL and inoculated into a 6-well plate. The cells were placed in a CO2 incubator including 5%of CO2 for culture at 37 ℃ for 24 hours. Then different samples (including different proportions of a PRP plus composition with a concentration of 1%, 1%PRP, 1 mM NMN and 1 mM sialic acid) were added separately. The cells were continuously cultured for 24 hours, scraped and transferred into a 1.5 ml centrifuge tube, followed by centrifugation using a centrifuge at a rotation speed of 1,000 rpm. The culture medium was sucked out, 1 mol / L of NaOH was added for dissolution, and the absorbance was measured at 405 nm. 10 μM of H-89 was used as a positive control, a blank group without drug treatment was used as a negative control, and the relative content of the melanin was 100%. The H-89, purchased from Shanghai Yuanye Biotechnology Co., Ltd., is a highly effective and selective PKA inhibitor that can inhibit the formation of the melanin.
[0084] Experimental results are as shown in FIG. 3. Inhibition effects of different proportions of the PRP plus composition on the formation of the melanin are compared. Compared with a control group, the PRP plus composition of the PRP, the sialic acid (NANA) and the NMN at a ratio of 3,000: 1: 1 (V / W / W) has the best effect of obviously inhibiting the melanin (P is less than 0.01) , which is the optimal ratio of the composition PRR plus. The PRP plus composition in different proportions ( (100-10,000) : 1: 1) has the effect of inhibiting the production of the melanin. The sialic acid (NANA) also has the effect of obviously inhibiting the production of the melanin, indicating that the sialic acid added into the PRP plus composition obviously improves the effect of inhibiting the production of the melanin and is conducive to fading of wounds.
[0085] Example 5: Promotion of the expression of mRNA in collagen COLIA1
[0086] In this study, mouse dermal fibroblast cells (mouse dermal fibroblast cells obtained by primary culture, MDF) are selected, real-time PCR is adopted, and a fluorescence dye is used for realizing luminescence detection, thereby quantifying the expression quantity of mRNA in a target gene COLIA1 in samples. Effects of a 1%PRP plus composition sample, a 1%PRP sample, a 1 mM NMN sample and a 1 mM sialic acid sample on promoting the expression of a collagen gene are compared. TGF-β1 (transforming growth factor β1) is used as a positive control.
[0087] Specific experimental steps are as follows.
[0088] (1) Primary culture of mouse dermal fibroblast cells (MDF) :
[0089] Newly born healthy C57BL / 6 mice were selected and disinfected with alcohol, and the back skin was clipped with scissors while the subcutaneous fat and connective tissues were not clipped as far as possible. Removed skin tissues were soaked in a dispase II (purchased from Shanghai Yuanye Biotechnology Co., Ltd. ) and digested overnight at 4 ℃. The skin tissues were taken out on the next day and clipped until the skin tissues were not observed by naked eyes to obtain a mixture. The mixture was transferred into a 0.25%trypsin solution and subjected to digestion culture in a water bath pot at 37 ℃, followed by filtration and centrifugation. The cells were re-suspended in a DMEM culture medium containing 20%of FBS, and a suspension was inoculated into a 100 mm culture dish and then placed in a CO2 incubator including 5%of CO2 for culture at 37 ℃ for 48 hours. The MDF cells were prepared into a suspension with a cell concentration of 2×104 piece / mL by using a DMEM culture medium containing 10%of FBS and 1%of penicillin or streptomycin. The suspension was inoculated into a 6-well plate, placed in a CO2 incubator including 5%of CO2 for culture at 37 ℃ for 24 hours and then placed in a CO2 incubator including 5%of CO2 for culture at 37 ℃ for 24 hours. Then a PRP plus sample, a PRP sample, a sialic acid sample and an NMN sample were separately added. A group added with nothing was used as a negative control, TGF-β1 was added to serve as a positive control, and mRNA was extracted from the MDF cells after the cells were continuously cultured for 24 hours.
[0090] (2) Extraction of total RNA from MDF cells:
[0091] After the MDF cells were treated with each sample for 24 hours, the culture medium was sucked out, RNA was extracted by an RT RNA separation reagent, and the concentration of the RNA was measured by Nanodrop. The quality and quantity of the RNA were determined by measuring the absorbance ratio at A260 / A280 and A260 / A230 (the ratio was greater than 1.8) .
[0092] (3) Design of primer sequences was as follows:
[0093] a COLIA1 F forward primer (SEQ ID NO: 1) : 5'-TCT GCG ACA ACG GCA AGG TG-3', and
[0094] a COL1A1 R reverse primer (SEQ ID NO: 2) : 5'-GAC GCC GGT GGT TTC TTG GT-3'.
[0095] (4) Real-time PCR:
[0096] An extracted total RNA sample was normalized to 2 μg, and any contaminated genomic DNA was removed by DNase I. According to an experimental instruction manual, 500 ng of the RNA sample was subjected to reverse transcription to obtain cDNA. Relative gene quantification was performed by 480. Amplification was performed for 45 cycles, and each cycle was performed by denaturation at 95 ℃ for 30 seconds, annealing at 55 ℃ for 30 seconds and extension at 72 ℃ for 20 seconds. Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) was used as an internal reference, and the mRNA level was determined by calculating a 2-ΔΔCt value. Results are as shown in FIG. 4.
[0097] When the skin is wounded, most of hypodermal cells are stimulated to secrete collagen so as to regenerate the skin structure. From FIG. 4, it can be seen that the PRP plus composition has an obvious effect of promoting the expression of mRNA in collagen COLIA1 (compared with a control group, *P is less than 0.05) , which is conducive to wound healing of the skin. According to the positive control TGF-β1, **P is less than 0.01. The PRP, the NMN and the sialic acid have no obvious differences.
[0098] Example 6: Inhibition effect on inflammatory factors
[0099] When the skin is wounded, skin cells release a variety of different cytokines, leading to skin inflammation. In this experiment, human skin keratinocytes (HaCaT cells) having inflammation induced by lipopolysaccharide (LPS) is used as a cell model to investigate and compare inhibition effects of PRP, a PRP plus composition, NMN and sialic acid on inflammation. HaCaT cells were used and pretreated with added samples for 3 hours first, followed by real-time PCR. During detection, the HaCaT cells were treated with the sample prepared in Example 1, LPS (100 ng / ml) was added into the cells to induce inflammation, and mRNA expression levels of inflammatory factors IL-6, TNF-α and COX-2 secreted by the HaCaT cells were detected by real-time PCR and compared.
[0100] Specific experimental steps are as follows.
[0101] (1) The HaCaT cells were prepared into a suspension with a cell concentration of 3×105 piece / mL and then inoculated into a 12-well plate. The cells were placed in a CO2 incubator including 5%of CO2 for culture at 37 ℃ for 24 hours, then a 1%PRP plus composition, a 1%PRP sample, a 1 mM sialic acid sample and a 1 mM NMN sample were separately added for pretreatment of the cells for 3 hours, and lipopolysaccharide (LPS, 100 ng / mL) was added to induce inflammation responses. A culture medium was used as a blank control, and dexamethasone (10 μM) was used as a positive control (Dex) . After the cells were cultured for 24 hours, mRNA was extracted from the HaCaT cells. The cells treated with each sample were cultured in 3 multiple wells, and the experiment was repeated for 3 times.
[0102] (2) Extraction of total RNA: The total RNA was extracted by RT RNA, and doped DNA was removed by DNase I enzyme. The quality and quantity of the RNA were determined by measuring the absorbance ratio using NanoDrop at A260 / A280 and A260 / A230 (the ratio was greater than 1.8) . Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) was used as a reference, and the content of the total RNA sample in an extract was standardized.
[0103] (3) Design of qRT-PCR primer sequences of different factors was as follows:
[0104] an IL-6 F-forward primer (SEQ ID NO: 3) : 5'-GAGAGTAGTGAGGAACAAGCCAGAGC-3, and
[0105] an IL-6 R-reverse primer (SEQ ID NO: 4) : 5'-CTA CAT TTG CCG AAG AGC CCT CAG G-3', where results are as shown in FIG. 5;
[0106] a TNF-α F-forward primer (SEQ ID NO: 5) : 5'-ATG AGC ACT GAA AGC ATG ATC CGG-3', and
[0107] a TNF-α R-reverse primer (SEQ ID NO: 6) : 5'-GCAATGATCCCAAAGTAGACCTGCCC-3', where results are as shown in FIG. 6;

[0112] (4) qRT-PCR (real-time PCR) :
[0113] According to an experimental instruction manual, 500 ng of the total RNA sample was subjected to reverse transcription to obtain cDNA. Relative gene quantification was performed by 480. PCR amplification was performed for 45 cycles, and each cycle was performed by denaturation at 95 ℃ for 30 seconds, annealing at 55 ℃ for 30 seconds and extension at 72 ℃ for 20 seconds. Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) was used as an internal reference, and the mRNA level was determined by calculating a ΔΔCt value.
[0114] Results are as shown in FIG. 5, it can be seen that the inhibition effect of the PRP plus on the inflammatory factor IL-6 is obviously better than that of the PRP (P is less than 0.05) and the NMN, and the inhibition effect of the sialic acid on the inflammatory factor IL-6 is equivalent to that of the positive control (dexamethasone) . In the PRP plus composition, the sialic acid has an obvious anti-inflammation effect.
[0115] Results are as shown in FIG. 6, it can be seen that the inhibition effect of the PRP plus composition on the inflammatory factor TNF-α is obviously better than that of the PRP and the NMN, and the sialic acid and the positive control have an obvious anti-inflammation effect (P is less than 0.001) .
[0116] Results are as shown in FIG. 7, it can be seen that the inhibition effect of the PRP plus (P is less than 0.01) on the inflammatory factor COX-2 is obviously better than that of the PRP and the NMN, and the sialic acid and the positive control have an obvious anti-inflammation effect (P is less than 0.001) .
[0117] By comparing the anti-inflammation effects of the PRP plus, the PRP, the sialic acid and the NMN on the inflammatory factors IL-6, TNF-α and COX-2, it is found that the anti-inflammation effect of the PRP plus composition added with the NMN and the sialic acid is obviously better than that of the PRP.
[0118] Example 7: Determination of the content of nitric oxide (NO)
[0119] iNOS (inducible nitric oxide synthase) is an enzyme that catalyzes the production of reactive nitrogen species free radicals in the body, including nitric oxide (NO) . NO is an indicator of inflammation in the body and is highly unstable, making iNOS levels easier to detect. Therefore, iNOS levels can be used as an indicator of the degree of inflammation in the body or cells. The iNOS levels can be induced by lipopolysaccharide (LPS) , and a positive control, such as dexamethasone (Dex) , can inhibit iNOS levels by up to 40%.
[0120] In Example 6, HaCaT cells were treated as described, and the supernatant of the cells was collected. The iNOS levels were detected using an NO test kit (batch No. GR3311102-1, purchased from Abcam Company) , and the amount of NO release was obtained. The results are presented in Figure 8.
[0121] Based on the results presented in FIG. 8, it is evident that the PRP plus composition has a significant effect in inhibiting iNOS levels and reducing the amount of NO released compared to the other components in the composition. This anti-inflammatory effect of the PRP plus composition is useful in preventing infections and accelerating wound healing by inhibiting the release of NO. The statistical analysis shows that the difference between the PRP plus composition and the control group is highly significant (P<0.001) , which is better than the effects observed with PRP, NMN, and sialic acid. The effect of the PRP plus composition is comparable to the positive control, dexamethasone (Dex) .
[0122] Example 8: Antioxidant effect of PRP plus
[0123] Skin wounds often become infected by bacteria, which further hinders wound healing due to the production of reactive oxygen species (ROS) . Patients with diabetes mellitus are especially affected by impaired wound surface healing.
[0124] In this study, a total antioxidant capacity (TAC) colorimetric assay kit, K274-100, purchased from Biovision, was selected for detection. A standard curve was established based on the copper ion oxidation resistance ability of Trolox (tocopherol acetate: a derivative of vitamin E) , and the total antioxidant ability (expressed as the equivalent concentration of Trolox) of different proportions of a 1%PRP plus composition, 1%PRP, 1 mM sialic acid and 1 mM NMN was calculated based on the standard curve. In this test, a higher Trolox equivalent indicates that a sample has a better antioxidant ability.
[0125] Results are as shown in FIG. 9. Different proportions of the PRP plus have an antioxidant effect, where the antioxidant effect of the PRP, the sialic acid (NANA) and the NMN at a ratio of 3,000: 1: 1 (V / W / W) is obviously better than other groups such as the PRP, the sialic acid (P is less than 0.001) and the NMN (P is less than 0.05) . Therefore, the PRP plus composition can obviously improve the antioxidant effect of the PRP and reduce the ROS level, thereby promoting the wound surface healing. Example 9: Wound healing experiment using blood taken from different animal sources
[0126] In this experiment, fresh blood was collected from cattle, horse, sheep, pig, deer, chicken, and rabbit. PRP plus samples were prepared from each of these animals using the method described in Example 1. A wound healing experiment was then carried out as per Example 3. The results of the experiment are presented in FIG. 10. The results indicate that PRP plus samples prepared from the blood of different animals have a certain effect in promoting wound healing. However, PRP plus samples prepared from human and deer blood exhibited better wound healing effects.
[0127] Example 10: Experiment of promoting wound healing of mice
[0128] In this experiment, 12 healthy 6-week-old C57BL / 6 mice were used. These mice had a body weight of 20-25 g and were free from specific pathogens. The mice were housed in a laboratory under controlled environmental conditions, including 12 hours of light and 12 hours of darkness, a temperature of (25±1) ℃ and a relative humidity of 50-70%. The mice were allowed free access to food and water, and the experiment was conducted 1 week after the animals had adapted to the environment. The use and handling of experimental animals were in accordance with the requirements of the Hong Kong University of Science and Technology for animal ethics.
[0129] The experimental steps involved anesthetizing the mice with chloral hydrate, shaving the back hair, and creating 0.5 cm diameter holes on the left and right sides of the back using a hole puncher. Normal saline was added into the holes on the left side as a control, while 1%PRP plus or 1%PRP (v / v, with normal saline as a solvent) was added into the holes on the right side. Drug administration was carried out once a day.
[0130] The results of the experiment are presented in FIG. 11. After treating the mice with PRP plus and PRP for 6 days, it was observed that the PRP plus had a significant effect in promoting wound healing compared to normal saline, and was better than PRP.
[0131] Example 11: Promotion of healing of scalded wounds
[0132] In this example, a 2-year-old child was accidentally scalded during the Spring Festival in 2021. The child received medical treatment for 7 days, but exudation and inflammation of the wound still occurred. The child's parents decided to try using PRP plus (with a concentration of 1% (v / v) and with normal saline as a solvent) . The results of the treatment are presented in FIG. 12. After applying PRP plus for three days, there was a reduction in inflammation and better formation of scabs. After discontinuing the use of PRP plus for four days, the wounds were allowed to heal.
[0133] Example 12: Reduction of scars and relief of itching
[0134] In this example, a police officer was burned by a strong water shower during patrol and received medical treatment in different hospitals. Although wound healing was achieved, the police officer experienced unbearable pain and itching. Therefore, the police officer decided to try using PRP plus (with a concentration of 1% (v / v) and with normal saline as a solvent) . The results of the treatment are presented in FIG. 13. It was observed that the thickness of the wound scars was reduced, and the itching was relieved after using PRP plus.
[0135] Example 13: Accelerated wound healing of a diabetic foot
[0136] In Case 1, a patient suffering from diabetes mellitus agreed to use PRP plus (with a concentration of 1% (v / v) and with normal saline as a solvent) after consulting with a doctor in 2023. After using PRP plus for 7 days, wound scabs were formed, and the results of the treatment are presented in FIG. 14. In Case 2, a patient suffering from diabetes mellitus agreed to use PRP plus (with a concentration of 1% (v / v) and with normal saline as a solvent) after consulting with a doctor in 2023. After using PRP plus for 16 days, wound scabs were formed, and the results of the treatment are presented in FIG. 15.
