[0002] QUALITY OF LIFEENHANCING LIQUID COMPOSITION
[0003] TECHNICAL FIELD
[0004] This present invention generally relates to liquid compositions, and more specifically, this invention relates to a liquid composition for enhancing quality of life through a delivery system of functional gas in the form of nanobubbles.
[0005] BACKGROUND OF THE INVENTION
[0006] In various metabolic processes within the body, free radicals are formed. Free radicals are molecules with unpaired electrons and exhibit high reactivity. They play a crucial role in several physiological processes, acting as mediators in intercellular communication systems to the immune system. These free radicals need to be regulated to function optimally in maintaining cell functions, protecting against infections, mediating the regulation of cellular communication quality, neural tissue functions, and overall maintaining the homeostasis of life systems. The control of free radicals in the body is carried out by natural antioxidants in the form of enzyme systems or metabolite products such as hydrogen molecules, in addition to various complex compounds that are generally complex organic compounds. This control system regulates the amount of free radicals in alignment with normal physiological needs.
[0007] In certain situations, the quantity of free radicals increases such that it becomes unbalanced with the number of antioxidants. This condition is commonly referred to as oxidative stress, which can be caused by habits such as engaging in activities that do not align with the body’s biological clock, stress, and excessive consumption of chemical products like cosmetics, and food and drinks containing synthetic ingredients. Oxidative stress can lead to adverse effects such as premature aging and the pathological development of degenerative diseases like cancer, Alzheimer's, Parkinson's, atherosclerosis, and others. Within the cell, there are several antioxidants in the form of endogenous enzymes such as superoxide dismutase (SOD), catalase, and glutathione peroxidase (GPx) that can neutralize free radical macromolecules. However, there are no antioxidants found to neutralize small free radical molecules.
[0008] Based on research outcomes from scientific publications over the past 20 years, hydrogen (H2) has been demonstrated to act as a selective antioxidant agent capable of neutralizing small free radical molecules such as hydroxyl radicals (*OH) and peroxynitrite (ONOO-). H2possesses high cellular membrane penetration capabilities, allowing it to easily neutralize free radicals present in cell organelles and even DNA. In the studies, H2was tested on mice models of ischemia / reperfusion (I / R) to examine its effects on cardiovascular therapy, and the results indicated that H2could reduce brain damage and oxidative stress.
[0009] It has also been reported that hydrogen (H2) possesses capabilities beyond reducing the accumulation of free radicals, such as enhancing the activity of antioxidant enzymes in wound tissue, thereby accelerating wound healing. In addition to acting as a neutralizing molecule for free radicals, H2 also has the ability to suppress inflammation, regulate the body's immune system, and decrease apoptosis (cell death). Hydrogen is an element abundantly found in the universe. In its normal state, hydrogen exists as a gas with a very light molecular weight. However, hydrogen has a very low solubility in water (Shafanov&Khitrin, 2013) and easily escapes from solutions. Therefore, to improve the stability and solubility of hydrogen, nano bubble technology is employed.
[0010] Hydrogen nano bubbles are smaller than 400 nm, which is below the average size of a cell. The H2 molecules within the nano bubbles can remain stable for a long period, up to eight years of storage. Due to their high stability, hydrogen nano bubbles are extensively utilized in the health sector. Research conducted by Kato et al. (2015) has shown that solutions containing hydrogen nano bubbles are more effective at neutralizing *OH free radicals than regular hydrogen solutions. Additionally, it has been reported that hydrogen nano bubbles are effective in reducing inflammation levels and suppressing adipogenesis (the formation of fat cells or tissues) (Li et al., 2020; Xiao & Miwa, 2021).
[0011] Hydrogen peroxide (H2O2) is a molecule that is naturally produced in the body as a byproduct of oxygen metabolism. It is catalyzed by the enzymes peroxidase and catalase, which break down H202into water and free oxygen ions. Hydrogen peroxide is a ubiquitous molecule and is abundant in nature (Halliwell et al., 2000). In 1888, H2O2was reported to have been used in therapies for healing diseases such as dengue fever, diphtheria, nasal mucosa inflammation, cough, asthma fever, and tonsillitis (Love, 1888). Hydrogen peroxide infusions were able to address influenza pneumonia epidemics following World War I (Oliver et al., 1920). In 1989, researcher Willhelm promoted the use of H2O2therapy to treat cancer, skin diseases, polio, and bacterial- related diseases, and in 1998, it was reported that H202could alleviate arthritis pain (Green, 1998). Research by Cervantes et al. (2021) describes the use of H2O2in inhibiting the COVID-19 virus.
[0012] However, on the other hand, for years after its discovery, H2O2was considered a dangerous molecule to human body tissues. Although its use has sparked controversy in alternative medicine, H2O2is often regarded as a toxic molecule to various life systems. For instance, it can cause severe pathological conditions such as cancer, ischemia, and neurodegenerative diseases. Nevertheless, scientific evidence has reported a positive role of H2O2in biological processes, for example, in regulating the immune system to combat bacteria and viruses (Halliwell et al., 2000). The role of H2O2depends on its concentration within the cells. At low concentrations, H2O2can act as a signaling molecule to regulate cellular processes and survival.
[0013] Currently, there are patents related to the use of Hydrogen Peroxide (H2O2) for medical treatments, including United States Patent No. US 2004 / 0101571 Al entitled "Use of Hydrogen Peroxide for the Treatment of Warts." This patent describes the application of Hydrogen Peroxide (H2O2) for treating human papillomavirus (HPV), including verruca warts, plantar warts, flat warts, and genital warts. Another relevant patent is United States Patent No. US 2014 / 0227367 Al, titled "Treating Inflammation with a Combination of an Ellagitannin and Hydrogen Peroxide." In this patent, Hydrogen Peroxide (H2O2) is combined with Ellagitannin to treat inflammation, with a weight ratio ranging from 1:1000 to 10:1 of the extract to Hydrogen Peroxide (H2O2)• The Ellagitannin used is punicalagin, which is derived from the skin of pomegranate fruit. Additionally, the binding agent used is catechin, obtained from green tea extract.
[0014] However, none of the studies or patents thus far have shown real results concerning improvements to the cardiovascular system, which consists of the heart, blood vessels, and blood. The cardiovascular system is crucial for transporting nutrients and oxygen-rich blood to all body cells and returning deoxygenated blood back to the lungs. Any disruption in this system can lead to impaired transport processes, causing cells to suffer from a lack of nutrients and oxygen, which can eventually affect the performance of body organs. This impairment can decrease an individual's quality of life.
[0015] One factor that can disrupt the cardiovascular system is the presence of high cholesterol levels in the blood, typically resulting fromunhealthy dietary habits, such as consuming foods high in saturated fats and low in fiber. Additionally, lifestyle factors like insufficient physical activity also contribute to high cholesterol levels. Elevated blood cholesterol can lead to the formation of plaque along the walls of blood vessels.
[0016] The formation process of plaque involves a complex series of events in which cholesterol wax adheres to the walls of blood vessels, causing thickening, hardening, and narrowing of these vessels. In this context, Low-Density Lipoproteins (LDL) cholesterol plays a primary role in plaque formation. As plaque accumulates in the arteries, the body responds by dispatching white blood cells that attempt to digest the LDL cholesterol, turning into toxins. The increased presence of white blood cells at these sites can cause inflammation of the arterial walls. This process results in an increase in the number of muscle cells in the arterial walls, forming a covering layer over the soft plaque. If this soft plaque ruptures, it forms a clot that impedes blood flow to all parts of the body.
[0017] In light of these issues, the inventor conducted research to determine the effectiveness of complementary therapy using nano H2(GNH) bubble solution combined with hydrogen peroxide (H2O2). This research is evidence-based, consisting of animal trials and clinical studies focused on improvements in the cardiovascular system and quality of life.
[0018] SUMMARY OF THE INVENTION
[0019] This invention pertains to a liquid composition designed to enhance quality of life through a system for delivering functional gases in the form of nano bubbles. This liquid composition is capable of improving the cardiovascular system in the human body, enabling it to function efficiently in delivering nutrient-rich and oxygenated blood to all body cells, while also returning deoxygenated blood back to the lungs. The volume percentages of the liquid composition in accordance with this invention consist of 0.59%v-2.75%v nano bubble hydrogen (H2), 0.29%v-l.37%v nano bubble oxygen (O2), 0.39%v-0.55%v hydrogen peroxide (H2O2), 0.00%v-l.83%v potassium chloride, and 91.66%v-98.33%v NaCl infusion fluid. This invention employs a nano bubble delivery system containing functional gases such as H2, O2, CO, and H2S, with bubble sizes smaller than 400nm. Functional gases are gases that have physiological roles beneficial to health. The primary goal of this invention is to improve the quality of life. Smooth blood circulation throughout the body ensures adequate nutrition and oxygen supply to all cells, which in turn keeps body cells healthy and body organs functioning properly. Consequently, this slows down the aging process and extends human lifespan expectancy. A notable benefit is that individuals over the age of 60 remain productive and independent .
[0020] Another aim of this invention is to enhance the quality of the cardiovascular system to restore its optimal function. The liquid composition is directly injected into major blood vessels. From a hematological perspective, significant improvements include an increase in lymphocytes and monocytes and a decrease in granulocytes. This condition indicates that the injected liquid composition helps regulate the body's inflammatory state. Additional benefits of the composition include the reduction of triglycerides, cholesterol, LDL, and blood glucose levels.
[0021] A further objective is to reduce blood pressure. Hydrogen molecules in the composition can decrease oxidative stress, thus providing an anti-hypertensive effect. Additionally, hydrogen molecules help correct neural imbalances by suppressing overactive sympathetic nervous system activities and enhancing the activity of the parasympathetic nervous system, which contributes to the blood pressure-lowering effect. Lowering blood pressure reduces the risk of stroke or heart disease.
[0022] Another goal of this invention is to enhance the flow through capillary vessels, the smallest blood vessels approximately 5 micrometers in diameter that connect arterial and venous blood vessels. Due to their small size, capillaries are more susceptible to blockages caused by plaque. In this invention, dissolved nano hydrogen bubbles in the infusion solution can enter capillary vessels, effectively clearing plaque and ensuring smooth blood flow. The subsequent aim is to prevent or ameliorate neurodegeneration of nerve cells by inhibiting the aggregation of a-synuclein.
[0023] BRIEF DESCRIPTION OF THE DRAWING
[0024] The embodiment of this invention will be further described in detail, referring the accompanying drawing, in wich :
[0025] FIG 1 is a block diagram illustrating changes after 10 infusions in a Phase 2 clinical trial in accordance with this invention.
[0026] DETAIL DESCRIPTION OF THE INVENTION
[0027] The composition of the liquid in volume percentage according to this invention consists of:
[0028] - H2in the form of nanobubbles 0.59%v-2.75%v;
[0029] - O2 in the form of nanobubbles 0.29%v-l.37%v;
[0030] - H2O20.39%v-0.55%v;
[0031] - MgSO40.39%v-l.83%v;
[0032] - Potassiumchloride 0.00%v-l.83%v;
[0033] - Normal saline solution 91.66%v-98.33%v.
[0034] The liquid composition according to this invention is administered into the body using a nano-bubble delivery system containing functional gas, consisting of:
[0035] - H2in the form of nano-sized bubbles smaller than 400nm,
[0036] - O2 in the form of nano-sized bubbles smaller than 400nm,
[0037] - CO in the form of nano-sized bubbles smaller than 400nm,
[0038] - H2S in the form of nano-sized bubbles smaller than 400nm.
[0039] Functional gas is a gas that has physiological functions, making it beneficial for health. In this context, functional gas is encapsulated within nano-bubbles and mixed with a NaCl infusion solution. As a result, nano-bubbles serve as a carrier medium for the functional gas into the body's cells.
[0040] Nano-bubbles are used in this invention due to their advantages over conventional bubbles and microbubbles. Their benefits include being less prone to breakage, enduring, stable, and remaining in water for an extended period (Yang & Chen, 2022). Unlike regular bubbles, which can expand, rise quickly to the surface and burst, and are unstable and disappear within seconds, microbubbles are more stable than conventional bubbles but can shrink and vanish or burst after a few minutes in water. Due to their very small size, nano-bubbles can easily enter blood vessels and surrounding cells.
[0041] At room temperature or 25°C, nano-bubbles sized 100-200 nm contain H2gas ranging from 3.487xlCT16mL to 2.789xlCT15mL and O2gas ranging from 1.743xlCT16mL to 1.395xlCT15mL. On average, it can be stated that in 100 mL of water, there will be approximately 3 mL of H2gas and 1.5 mL of O2gas. Thus, for 500 mL of water, nano-bubbles can encapsulate up to 15 mL of H2gas and 7.5 mL of O2gas. In a volume of 4.184xlCT15mL per nanobubble, each nano-bubble can only contain a maximum of 4.184xlCT15mL of H2and O2gas. Meanwhile, the maximum solubility of H2and O2gas in nano-bubbles in water is 4.5 mL per 100 mL of water, or 4.5%. To accommodate all 4.5mL of H2and O2gas, approximately 1.075xl015nano-bubbles are required per lOOmL of water. From this data, it can be concluded that for 500mL of water, up to 5.375xl015nano-bubbles containing H2and O2gas can be accommodated.
[0042] The composition of the liquid in volume percentage according to this invention consists of:
[0043] - H2in the form of nanobubbles = 3-15mL
[0044] - O2in the form of nanobubbles = 1.5-7.5mL
[0045] - H2O2= 2-3mL
[0046] - MgSO4= 2-10mL
[0047] - Potassium chloride = 0-10mL
[0048] - Normal saline solution = 500mL.
[0049] In the liquid composition according to this invention, the nano hydrogen bubbles used are characterized by being the smallest and lightest molecules in nature, which readily penetrate cells. The utilization of these nano hydrogen bubbles provides an effect that: inhibiting aging or as an anti-aging agent,
[0050] - Anti-free radicals,
[0051] Inhibiting cell death,
[0052] - Anti-inflammatory.
[0053] Whereas hydrogen peroxide (H2O2) in small quantities offers numerous benefits for the body. The use of hydrogen peroxide (H2O2) will provide effects which include: inhibiting viral and bacterial infections, supplying oxygen to tissues, regulating processes and cell survival.
[0054] Meanwhile, nano oxygen bubbles are considered the most vital molecules for life due to their role in every cellular function. The use of these nano oxygen bubbles also provides antibacterial effects and enhances energy.
[0055] The next component in this liquid composition is MgSCq, which is known to improve the elasticity of blood vessels and maintain the health of nerve cells, heart rhythm, immune system, bones, and muscle function. Another key ingredient in this composition is potassium, which allows nerves to respond to stimuli and muscles to contract or tighten, including those in the heart. Additionally, potassium helps counteract the effects of sodium in raising blood pressure and facilitates the movement of nutrients into cells, as well as the expulsion of waste products from them. However, potassium is not added to the composition of this liquid according to this invention if the individual receiving therapy has kidney issues. Conversely, for individuals without kidney problems, potassium is incorporated into the liquid composition for their treatment.
[0056] The inventor conducted a series of trials with this liquid composition as per the invention, starting with phase 1 animal trials using male Wistar rats as subjects. In this phase 1 trial, a safety study was conducted by comparing three groups of normal rats as follows:
[0057] - ratsinjected with this fluid include,
[0058] - controlratswithout treatment (control),
[0059] - ratsinjected with physiological saline. In this phase 1 trial, research was also conducted to compare three groups of hypercholesterolemic rat models, or rats that were fed a high-fat diet (HFD), as follows:
[0060] - hypercholesterolemic rats injected with this solution,
[0061] - hypercholesterolemic rats injected with simvastatin,
[0062] - hypercholesterolemic rats without treatment (control).
[0063] This phase 1 trial has received approval from the Ethics Commission of Universitas Brawijaya with the approval number 068-KEP-UB-2022, and was conducted in the Animal Physiology
[0064] Laboratory, Department of Biology, Universitas Brawijaya.
[0065] The phase 1 trial was carried out by administering injections to rats as the experimental animal subjects.
[0066] Subsequent procedures included daily temperature measurements of the rats. The next step involved surgical procedures on the rats, during which the liver organs were examined using a microscope with a 400x magnification.
[0067] The results of the safety testing of the injected fluid showed that the survival rate of the rats injected with this fluid was better compared to the control group (which received no treatment). Specifically, in the control group, some rats died, whereas in the group injected with the fluid, no deaths occurred. Further tests on the injected fluid in a group of hypercholesterolemic rats revealed that the fluid could reduce triglyceride and total cholesterol levels compared to the control. Additionally, it was found to increase HDL levels, but it did not significantly affect LDL levels. Meanwhile, the hypercholesterol group treated with simvastatin (a cholesterol- lowering drug) showed a decrease in cholesterol levels; however, the mortality rate in this group was high at 60%.
[0068] The percentage of rats that died in phase 1 of the trial can be viewed in Table 1 below:
[0069] Table 1
[0070] Meanwhile, the lipid profile data from this Phase 1 trial can be viewed in Table 2, which displays comparative data on total cholesterol, triglycerides, HDL, and LDL. This table presents five stages of observation, as follows:
[0071] Stage 1: Before the administration of the high fat diet (HFD),
[0072] Stage 2: 0-3 weeks after the administration of HFD,
[0073] Stage 3: 4-5 weeks after the administration of HFD,
[0074] Stage 4: 6-7 weeks after the administration of 3x hydrogen nanobubbles in the treatment group,
[0075] Stage 5: 7-8 weeks after the administration of 5x hydrogen nanobubbles in the treatment group.
[0076] Table 2 Previous studies conducted on mice have demonstrated that drinking water containing H2can increase HDL, reduce triglycerides, and even lower blood glucose levels (Kamimura et al., 2011). Liu et al. (2021) tested the administration of H2 gas on 10 mice subjected to a high-fat diet (HFD). Their research indicated that H2gas can activate antioxidant enzymes that bind with HDL, thus neutralizing free radicals and reducing inflammation. Additionally, H2was reported to significantly enhance the activity of HDL in reducing cholesterol in foam cells (Song et al., 2012). Foam cells are responsible for plaque formation in blood vessels. The results from this phase 1 trial concluded that injections containing nano hydrogen bubble components are safe and can reduce liver fat.
[0077] Beyond the phase 1 trial, the inventor also conducted a phase 2 clinical trial on humans, which was a randomized clinical trial approved by the ethics committee of the Politeknik Kesehatan Malang, under no. 41O / KEPK-POLKESMA / 2022, and carried out at Dr.Benggol Hospital. The trial involved 52 participants, including 8 randomly selected placebos, with age criteria of adults (<37 years). Participants received injections according to this composition, involving 10 infusion sessions followed by laboratory tests on blood samples and physical examinations. Additionally, participants completed the SF36 questionnaire, which covers general health, mental health, emotional limitations, vitality, body pain, and physical function.
[0078] The results of the phase 2 clinical trial are presented in Figure 1, which shows a bar chart of changes after 10 infusion sessions in non-placebo participants. This chart displays the significance values for each test parameter. Laboratory tests on participants' blood samples were conducted to gather data indicative of degenerative diseases. For blood glucose levels before and after treatment, the significance value was found to be 0.000. As this value is less than 0.05, it can be concluded that there is a significant difference between the levels before and after treatment. For LDL levels before and after treatment, the significance value was also 0.000, indicating a significant difference. However, for HDL levels before and after treatment, the significance value was 0.379, indicating no significant difference between the levels before and after treatment.
[0079] Following the results from the electrocardiography (ECG) examination, there were 2 cases of persistent sinus rhythm with inferior old myocardial infarction (OMI Inferior) before and after treatment. Additionally, there were 6 cases of persistent inferior anteroseptal ischemia before and after treatment. There was also 1 case maintaining sinus rhythm with incomplete right bundle branch block (RBBB) before and after treatment. Moreover, 34 cases showed normal ECG results both before and after treatment. From the ECG examinations for treatments involving fluid infusion, which were conducted 10 times, improvements in cardiac function have not yet been reported. Subsequently, echocardiogram results indicated an improvement in Left Ventricle Ejection Fraction (LVEF), from 53-73% before treatment to 61-78% after treatment, suggesting improved cardiac function.
[0080] Further Doppler ultrasound examinations for vascular assessments related to Intima Media Thickness (IMT) revealed 11 cases where the carotid artery bulb thickness remained unchanged before and after treatment, while the remaining 32 cases remained normal, showing no overall change. However, Doppler ultrasound examinations related to Flow-Mediated Dilation (FMD) indicated an increase in all cases to above 7.0 after treatment, signifying a decrease in arterial resistance and improved elasticity. These findings are supported by the observed decreases in blood pressure and heart rate before and after treatment.
[0081] The illustrations demonstrate that this fluid infusion can balance body homeostasis and enhance quality of life based on variables assessed by the SF36 questionnaire (general health, mental health, emotional limitations, vitality, body pain, and physical function). The data indicate notable improvements in general health, increased vitality, and reduced body pain after the fluid treatment. These findings are consistent with research conducted by Kang et al. (2011) on liver tumor patients undergoing radiotherapy, where consuming hydrogen-rich water for six weeks significantly improved life quality scores compared to placebo patients.
[0082] From a hematological perspective, notable improvements include an increase in lymphocytes and monocytes, along with a decrease in granulocytes. These changes suggest that this fluid infusion regulates inflammatory conditions in the body. This is also supported by the fact that H2O2can enhance lymphocytes and monocytes (Tang et al., 2007; Yoo&Huttenlocher, 2009). On the other hand, although not prominently, this fluid infusion can reduce triglycerides, cholesterol, LDL, and blood glucose levels. These findings are consistent with previous clinical studies, which showed that solutions rich in hydrogen can reduce lipid and glucose levels compared to a placebo (Song et al., 2013; LeBaron et al., 2020). Based on prior studies, it has also been reported that H2O2at low concentrations has insulin-like properties and plays a role in glucose transport activity in muscles (Henriksen, 2013). With its ability to lower cholesterol, triglycerides, and LDL, it can reduce the potential for degenerative diseases.
[0083] A variable that clearly shows improvement after administering this fluid infusion is a decrease in blood pressure. This is attributed to the reduction of oxidative stress by hydrogen molecules (Kawakami et al., 2022). Testing with mice has shown that hydrogen molecules provide an antihypertensive effect not only during daytime rest but also during nighttime activity. Moreover, hydrogen molecules can correct neural imbalances by suppressing an overly active sympathetic nervous system and enhancing the activity of the parasympathetic nervous system, which occurs concurrently with the blood pressure-lowering effect (Sugai et al., 2020). Based on the findings above, it can be concluded that this fluid infusion is safe for application, can provide positive effects for the cardiovascular system, and improve quality of life. Besides infusion injections, the composition can also be made into a drink, applied as a spray, or administered via injection.
[0084] The liquid composition for enhancing quality of life in accordance with this invention may also consist o:
[0085] - H2in the form of nanobubbles 0.59%v-2.72%v,
[0086] - O2in the form of nanobubbles 0.29%v-l.36%v,
[0087] - CO in the form of nanobubbles 0.00%v-0.54%v,
[0088] - H2S in the form of nanobubbles 0.00%v-0.54%v,
[0089] - MgSO40.39%v-0.81%v,
[0090] - Potassium chloride 0.00%v-l.81%v,
[0091] - Normal saline solution 90.66%v-98.33%v.
[0092] In the composition of the fluid, there is a noticeable reduction of H2O2, but an addition of CO in the form of nano bubbles and H2S in the form of nano bubbles. The function of the CO and H2S gases is to act as neurotransmitters for nerve communication. The composition includes:
[0093] - H2in nanobubble form = 3-15mL
[0094] - O2in nanobubble form = 1.5-7.5mL
[0095] - H2O2= 2-3mL
[0096] - MgSO4= 2-10mL
[0097] - Potassium = 0-1OmL
[0098] - CO = 0-3mL
[0099] - H2S = 0-3mL
[0100] - Normal saline solution = 500mL.
[0101] The explanations and details provided in the description are intended for illustrative purposes only, and are not limitations on the invention, as modifications can be made without departing from the patent protection claimed in this invention.
